Magnetic microswarms capable of performing navigation to targeted lesions show great potential for in vivo medical applications. However, using the swarms for lesion cavity filling encounters challenges from precise delivery and sealing. Herein, this work develops a magneto-thermal hydrogel swarm consisting of magnetic hydrogel particles, which can perform phase transition induced by temperature change. The particles are prepared using a temperature-responsive hydrogel matrix, tissue adhesive monomers, and magnetic microparticles. The swarms can be remolded to various shapes, and it can be used to seal perforation in phantom and gastric tissue. The swarms can also serve as drug carriers, and their drug release profiles induced by temperature changes are characterized. Finally, the targeted delivery, adaptive filling, and sealing of a gastric ulcer using the swarms are achieved in ex vivo and in vivo environments.

Near-infrared (NIR) in vivo fluorescence imaging has exhibited the distinct advantage of high optical resolution at deeper penetration into biological tissues. Ag2Te quantum dots (QDs), with a relatively narrow band gap, show great promise for fluorescence emission at long wavelengths in the second near-infrared (NIR-II) window for bioimaging. However, existing Ag2Te QDs have severely hindered the application of in vivo bioimaging due to their poor fluorescence brightness and stability, so it is important to prepare Ag2Te QDs with high quantum yield and stability as well as high biocompatibility in the NIR-II window. Herein, we designed an integrated method for the preparation of water-soluble Ag2Te QDs by mutual adaptation of QD synthesis and surface modification. We first synthesized high-quality Ag2Te QDs with different NIR-II emission wavelengths and the photoluminescence quantum yields (PLQYs) up to 6.51% by rapidly injecting the TBP-Te precursor into a hot solvent to form a highly fluorescent Ag2Te core. Then water-dispersible Ag2Te QDs were obtained by direct exchange of the hydrophobic Ag2Te QD surface ligands with thiol ligands. The PLQY of the water-soluble Ag2Te QDs obtained by this method can still be maintained at 4.94%. With these highly bright and stable Ag2Te QDs, the abdominal vessels, hindlimb arterial vessels, venous vessels, sacral lymph nodes, and tumor vessels were visualized non-invasively in vivo in the NIR-II window in mice. The results demonstrate that the integrated strategy of QD synthesis and modification provides valuable technical support for further in-depth applications of Ag2Te QDs.

Electrochemical sensors based on molecularly imprinted polymer (MIP) provide an exceptional platform for sensitive and selective analysis of analytes in complex matrix through incubation while in vivo analysis is critical for achieving quantitative information for the biological study in real time. However, the deployment of MIP electrochemical sensors for in vivo analysis highly depends on their forms and fabrication. Here blade-style MIP electrochemical sensors were designed and fabricated in batches based on the technique of lamination, focusing on their applications for in vivo analysis of trace indole-3-acetic acid (IAA) in plants. The blade-style electrochemical sensors were fabricated using stainless steel sheets as supporting and conductive substrates for the working electrodes. The MIP sensors were prepared in batches using cyclic voltammetry (CV) based electropolymerization of pyrrole (the functional monomer) and IAA (the template) followed by elution under a constant potential at a multichannel electrochemical station with improved throughput. After optimization, the sensors could stably and reproducibly quantify IAA with the detection limit of 0.1 μmol·L −1 . The blade-style MIP sensors could be either plugged in tomato fruits for in vivo analysis of IAA or coupled with a piece of hydrogel for non-invasive analysis of IAA in tomato leaves. The blade-style MIP electrochemical sensors may provide a platform for in vivo analysis by not only obviating the time-consuming sample treatment but also simplifying the analytical procedure. This study may pave the way for extensive applications of MIP electrochemical sensors for real-time, spatially resolved analysis of endogenous biomolecules in living organisms in the near future.

Ethnopharmacological relevance Acute pancreatitis (AP) is an inflammatory disease caused by the premature activation of digestive enzymes, leading to pancreatic self-digestion. In about 20 % of cases, AP progresses to severe acute pancreatitis (SAP), a life-threatening condition with high mortality. In traditional Chinese medicine (TCM), SAP corresponds to the “Yangming Fu-organ Excess Syndrome,” attributed to internal damp-heat and toxin accumulation, Fu-organ qi obstruction, and blood stasis. Dahuang Mudan Decoction (DHMDD), a classic formula from Zhang Zhongjing's Eastern Han Dynasty text Synopsis of the Golden Chamber , can clear heat, detoxify, promote digestion, activate blood circulation, and resolve stasis. Despite its proven clinical efficacy, the molecular mechanisms of DHMDD remain unclear and require further investigation. Aim of the study This research sought to systematically elucidate the molecular mechanisms of DHMDD against SAP using an integrated approach that combines network pharmacology, transcriptome sequencing, and in vivo animal validation, and to identify its core bioactive components. Materials and methods To investigate the effect of DHMDD on severe acute pancreatitis (SAP), we first established a mild-to-moderate acute pancreatitis (MAP) mouse model via caerulein injection to determine the optimal therapeutic dose of DHMDD. Subsequently, an SAP model was induced with L-arginine for mechanistic studies. We used liquid chromatography-mass spectrometry (LC-MS) to identify the blood-absorbed components of DHMDD in SAP mice. The potential mechanisms of DHMDD against SAP were screened by integrating network pharmacology (based on the absorbed components) with transcriptomic sequencing; intersection analysis was performed to identify core targets and pathways. Core bioactive components were pinpointed through molecular docking. Finally, in vivo validation was conducted: SAP mice were treated with DHMDD and its key component (rhein) at different doses to verify the therapeutic effects and regulation of the core pathways, thereby confirming DHMDD's anti-SAP efficacy and material basis. Results Initial experiments showed that all doses of DHMDD alleviated pancreatic injury and lowered serum lipase and amylase levels in MAP mice, with high-dose showing the greatest benefit. In SAP mice, high-dose DHMDD also markedly reduced pathological injury in pancreatic and lung tissues, decreased acinar cell apoptosis, and lowered inflammatory cell infiltration and cytokine levels. LC-MS identified the blood-absorbed components of DHMDD, and subsequent network pharmacology analysis, based on these components, pinpointed 366 potential therapeutic targets for SAP. Intersection analysis of these targets with transcriptomic differentially expressed genes (DEGs) from pancreatic tissues confirmed the core regulatory targets. KEGG enrichment analysis highlighted the involvement of inflammation-related MAPK signaling and oxidative stress-related pathways, a finding corroborated by transcriptomic data. Notably, animal experiments validated that the core bioactive component, rhein, at two tested doses, alleviated SAP-induced pancreatic and lung injury, reduced apoptosis and inflammation, mirroring the effects of DHMDD. Both DHMDD and rhein significantly suppressed MAPK pathway activation in macrophages while enhancing the antioxidant response in acinar cells via activation of the Nrf2/HO-1 signaling cascade. Conclusion We have elucidated a dual mechanism for DHMDD in SAP: enhancing the Nrf2/HO-1 antioxidant axis in acinar cells and suppressing pro-inflammatory MAPK signaling in macrophages. The identified core component, rhein, mediates these effects, confirming its role as the key material basis. This work provides a scientific foundation for DHMDD's clinical use and positions rhein as a candidate for targeted drug development against SAP.

The construction of reliable and sensitive wearable sensors capable of monitoring specific targets in sweat remains a challenge highly relevant to a range of sensor/diagnostic applications. Herein, we develop a wearable microfluidic platform to monitor sweat lactate, in which spherical colloidal Photonic Crystal Hydrogel (PCH) is synthesized to construct the sensing part. Thanks to the structural color of PCH, lactate concentrations can be obtained via the eye-recognizable and low angle-dependent color variation. Accurate quantitative analysis is performed on a smart phone with Matlab code by integrating the linear relationship between hue value of sample photos and lactate concentrations. The developed sweat sensor presents good selectivity and long-term stability, and it is capable of detecting lactate accurately in human sweat within the physiological range. On-body measurement performed on volunteers successfully demonstrates the applicability of the developed device. This wearable analytical platform provides a promising tool for sweat lactate detection in sports science and medical diagnosis. And it is expected to contribute to developing user-friendly wearable biochemical sensor devices.

Alzheimer's disease (AD) is a major type of dementia in the elderly. Clinical medication at present can only relieve symptoms but has no therapeutical effect. Thus, it is urgent to develop an effective drug for the treatment of AD. In this study, 2-month-old triple transgenic AD model mice (3 × Tg-AD) was treated with 0.2 and 1.0 mM bis(ethylmaltolato) oxidovanadium IV (BEOV) in drinking water for 2 months respectively. The results of open field test, elevated plus-maze test and Y maze test showed that BEOV could significantly improve the exploratory ability, memory capacity and exercise ability and relieve the anxiety of 4-month-old 3 × Tg-AD mice. The dendritic spines of pyramidal neuron in neocortex were detected by in vivo two-photon imaging of YFP and AD-YFP mice (the offspring of 3 × Tg-AD and YFP mice). The numbers of dendritic spines increased in YFP mice but decreased in AD-YFP mice from 2.5-month-old to 3.5-month-old. Treatment with BEOV (1.0 mM) in the AD-YFP mice significantly increased the numbers of total dendritic spines, mushroom spines and thin spines, which suggested that BEOV could protect cortical neurons and reduce the loss of dendritic spines. The contents of metal ions in the brain of mice were further measured by inductively coupled plasma mass spectrometry (ICP-MS). The results showed that BEOV (1.0 mM) could significantly increase the contents of V and Se and decrease the contents of Fe, Zn, Hg, Pb, Bi and Ni in AD brains, implying that vanadium might synergize with selenium to regulate the contents of metal ions in AD mice. Summarily, BEOV could mediate the homeostasis of multiple metal ions in AD brain and protect cortical neurons by reducing the loss of dendritic spines, thus to interfere with the pathological process of AD.

The cerebral lymphatic drainage plays an important role in the occurrence and development of central nervous system diseases. Recent studies have shown that cerebral lymphatic drainage is regulated by circadian rhythm and anesthesia state; however, the regulating mechanism is still unclear. In this study, we used the second near-infrared region in vivo imaging to explore the regulation of cerebral lymphatic drainage in mice at different states. At first, by injection of a tracer at different times, we confirmed that the drainage of the meningeal lymphatic system was the fastest at zeitgeber time 2, while the internal flow of the glymphatic system was the slowest. Under anesthesia with isoflurane, administration of dexmedetomidine, an anesthetic that inhibits norepinephrine (NE) release, enabled mice to enter the stage of non-rapid eye movement sleep, at which time the influx of the glymphatic system increased, the efflux of the meningeal lymphatic system decreased, and the clearance rate of the brain parenchyma decreased. However, following the exogenous NE supplement, mice quickly changed from a non-rapid eye movement stage into an awake state with the meningeal lymphatic drainage retrieval. The results showed whether the drainage of the glymphatic system and meningeal lymphatic vessels, or parenchymal clearance, has made rapid adjustments based on sleep status that is regulated by NE. This study reveals that the NE-regulated sleep–wake cycle is a powerful regulator of cerebral lymphatic drainage and provides a potential therapeutic target for related central nervous system diseases.

As an efficient alternative for harnessing the energy from human's biofluid, the wearable energy harvesting-storage hybrid supercapacitor–biofuel cell (SC-BFC) microfluidic system is established with one multifunctional electrode. The electrode integrates the metal–organic framework (MOF) derived carbon nanoarrays with embedded Au, Co nanoparticles on the flexible substrate, and is used for symmetric supercapacitor as well as enzyme nanocarriers of biofuel cell. The electrochemical performance of the proposed electrode is evaluated, and the corresponding working mechanism is studied in depth according to the cyclic voltammetry and density functional theory calculation. The multiplexed microfluidic system is designed to pump and store natural sweat to maintain the continuous biofuel supply in hybrid SC-BFC system. The biofuel cell module harvests the electricity from lactate in sweat, and symmetric supercapacitor module to accommodate the bioelectricity for subsequent utilization. W e develop a numerical model to validate the normal operation in poor and rich sweat under variable situations for microfluidic system. One single SC-BFC unit can be self-charged to ∼0.8 V with superior mechanical durability in on-body testing, as well as the energy and power value of 7.2 mJ and 80.3 μW, respectively. It illustrates the promising scenery of energy harvesting-storage hybrid microfluidic system. This article is protected by copyright. All rights reserved

Owing to their potential to transform traditional medical diagnostics and health monitoring, wearable biosensors have become an alternative evolutionary technology in the field of medical care. However, it is still necessary to overcome some key technique challenges, such as the selectivity, sensitivity, and stability of biometric identification. Herein, a novel, wearable electrochemical sensor based on a molecularly imprinted polymer (MIP) integrated with a copper benzene-1,3,5-tricarboxylate metal–organic framework (MOF) was designed for the detection of stress through the on-body monitoring of cortisol in sweat. The MOF was used as the substrate for MIP deposition to enhance the stability and sensitivity of the sensor. The sensor consisted of two layers, with a microfluidic layer as the top layer for spontaneous sweating and a modified electrode as the bottom layer for sensing. The sensor measured cortisol levels by detecting the current change that occurred when the target molecules bound to the imprinted cavities, using Prussian blue nanoparticles embedded in the MIP framework as the REDOX probe. The proposed sensor exhibited a linear detection range of 0.01–1000 nM with a detection limit of 0.0027 nM, and favorable specificity over other analogies. This facile anti-body free sensor showed excellent stability, and can be successfully applied for in situ cortisol monitoring.

Sweat pH is a critical indicator for evaluating human health. With the extensive attention on the wearable and flexible biosensing devices, the technology for the monitoring of human sweat can be realized. In this study, a sensitive, miniaturized, and flexible electrochemical sweat pH sensor was developed for the continuous and real-time monitoring of the hydrogen-ion concentration in human sweat. A flexible electrode was fabricated on the poly(ethylene terephthalate) (PET) substrate by a simple and low-cost screen-printing technology, which was based on the integration of fluoroalkyl silane-functionalized Ti3C2Tx (F–Ti3C2Tx) and the polyaniline (PANI) membrane technology instead of the traditional ion-sensitive membrane. The surface functionalization strategy for Ti3C2Tx with perfluorodecyltrichlorosilane can provide environmental stability. Functionalized Ti3C2Tx (F–Ti3C2Tx) was doped with PANI to obtain improved responsiveness, sensitivity, and reversibility. The constructed microsize, portable, and wearable F–Ti3C2Tx/PANI pH sensor aimed to real-time monitor the pH value of human sweat during exercise. On-body sweat pH monitoring for females and males, respectively, exhibited high accuracy and continuous stability compared with ex situ analyses. This study thus offers a facile and practical solution for developing a highly reliable MXene-based mini-type pH sensor to realize the online monitoring of human sweat pH.

The clinical treatment of ulcerative colitis (UC) faces great challenges due to lifetime medication. In this study, Gingerol oil was extracted and purified by the process easily scale-up and cost effective, with productivity 2.72 ± 0.38% ( w /w, versus crude drugs). The quality control of gingerol oil was fully established by HPLC fingerprint with 4 common peaks identified as 6-gingerol, 8-gingerol, 6-shogaol and 10-gingerol. The similarities of 6 batches of gingerol oil are within 0.931–0.999. The protective effects of gingerol oil are equivalent to or even stronger than that of 6-gingerol on inflammation and oxidative stress of HT-29 cells induced by lipopolysaccharide and H 2 O 2 , as well as on UC in mice caused by dextran sulfate sodium salt (DSS). Our research conclusions coincide well with the holistic view of Traditional Chinese Medicine and network pharmacology . The absorption kinetics of gingerol oil were conducted using the in situ intestinal perfusion in rats and comparable absorption were achieved in the jejunum , ileum and colon segments within 2 h. Thus, gingerol oil colon targeting pellets were prepared by extrusion-spherization technique. The cumulative dissolution behaviors and mechanisms were observed and analyzed by fitting to dissolution model. Our studies provided reliable theoretical and experimental support for the gingerol oil as reliable therapeutic choice of UC.
