Augmentation of chloramphenicol degradation by Geobacter-based biocatalysis and electric field

Leilei Xiao,Jiajia Li,Eric Lichtfouse,Zhenkai Li,Quan Wang,Fanghua Liu

Electroactive microorganisms and electrochemical technologies have been separately used for environmental remediation such as antibiotics removal, yet the efficiency of coupling these two methods for chlorinated antibiotics removal is poorly known. Here we tested the synergy of Geobacter sulfurreducens PCA, an electroactive bacteria, and an electrical field, on chloramphenicol removal. Removal is increased two-fold by increasing the temperature from 30°C to 37°C. The cyclic voltammograms and chronoamperometry tests demonstrated that G. sulfurreducens PCA catalyzed chloramphenicol chemical reduction with electrode as excusive electron donor. A critical voltage, −0.6 to −0.5 V vs. Ag/AgCl, was discovered for chloramphenicol degradation with an increase of removal rate about 2.62-folds, from 31.06% to 81.41%. Combined removal with both G. sulfurreducens PCA and an electrical field increased the apparent rate constant and reached 82.77% removal at −0.5 V. Specially, the combined removal at −0.5 V even presented more robust removal efficiency compared to −0.6 V (78.64%) without G. sulfurreducens  PCA. Mass spectrometry of degradation products indicates the reduction of nitro into amine groups, and dechlorination into less toxic compounds. Overall, combined biocatalysis and an electrical field is a promising method to remove antibiotics from polluted environments.

Revealed mechanism of 3D-open-microarray boosting exoelectrogens Geobacter enrichment and extracellular electron transfer for high power generation in microbial fuel cells

Da Liu,Chao-qun Xu,Wen-kai Fang,Cheng-yu Li

The   anode enables raised microbial fuel cells (MFCs) performance via in-situ growth electroactive material. However, the role of fabricated microstructures in electroactive bacteria loading and extracellular electron transfer (EET) has been paid less attention. Here, MoS2 nanosheets are custom grown on carbon cloth to construct anode models with diverse surface microstructures. Surprisingly, the 3D-MoS2/NS-CC anode only 0.85 d enables the MFC to be started and achieves a maximum power density of 3.85 W/m 2 , which is significantly faster and higher than that of 2D-MoS2/NS-CC (3.6 d, 2.75 W/m 2 ) and CC (4.46 d, 1.98 W/m 2 ). As for the mechanism of 3D-MoS2/NSCC   boosting MFC performance, this is attributed to the 3D-open-microarray preventing electroactive bacteria from shedding and facilitating to the establishment of excellent EET channels through the formed hybrid cell-electrode systems and Geobacter enrichment of up to 86.1 %. This research provides promising guidance for integrating nanomaterials and architecture to construct high-performance anodes in MFCs.

Flavin mononucleotide-stimulated microbial fuel cell for efficient gaseous toluene abatement

Yue Lang,Yanan Yu,Hongtao Zou,Jiexu Ye,Shihan Zhang,Jianmeng Chen

Chemical park is regarded as a major contributor of VOCs emissions in China. Currently, a green and safe technology, microbial fuel cells (MFCs), is being developed for the VOCs abatement. Noting that effective electron transfer is critical to the MFC performance. In this work, flavin mononucleotide (FMN) was dosed as an electron shuttle to improve the removal of the typical toxic VOCs, toluene. The experimental results revealed that the performance of toluene removal and power generation were accelerated with the dosage of 0.2–2 μM FMN. With the addition of 1 μM FMN, the removal efficiency, the maximum output voltage and the coulombic efficiency of MFC were increased by 18.4%, 64.4% and 56.3%, respectively. However, a further increase in FMN concentration to 2 μM caused a reduction in the removal efficiency and coulombic efficiency. The images of scanning electron microscopy and confocal laser scanning microscopy showed that the presence of FMN greatly promoted the microbial growth and its activity. Furthermore, microbial community analysis also implied that the moderate dosage of FMN (0.2–1 μM) was beneficial for the growth of the typical exoelectrogens, Geobacter sp., and thus the coulombic efficiency was increased. In addition, an electron transfer pathway involving in cytochrome b , OMCs, cytochrome c , and MtrA was proposed based on the cyclic voltammetry analysis. This work will provide a fundamental theoretical support for its application of toxic VOCs abatement from the chemical park.

Improving power output in microbial fuel cells with free-standing CoCx/Co@CC composite anodes

Guilian Li,Minqiao Lu,Fan Wang,Yueteng Hou

Microbial fuel cells (MFCs) are being recognized as a promising solution for sustainable energy generation and wastewater treatment . Despite their potential, MFCs face certain challenges that must be overcome, such as the problem of low power output caused by inefficient extracellular electron transfer (EET). In this study, we develop a nanoporous CoC x /Co@CC composite anode using microwave-assisted and high-temperature calcination methods to enhance EET efficiency and bacterial adhesion in MFCs. Compared with pure carbon cloth (CC) anodes, MFCs with CoC x /Co@CC anodes exhibit a 1.24-fold increase in power density, with a maximum output of 2693 mW m −2 . The improved performance can be attributed to the lower charge transfer resistance (R ct ) and higher redox activity of the CoC x /Co@CC anode. Cobalt's multiple valence states enhance its ability to transfer electrons between microorganisms and electrodes, ultimately increasing the overall redox activity. The CoC x /Co@CC anode also promotes the growth and enrichment of exoelectrogens, with Geobacter being enriched up to 75 % at the anode, compared with 46 % at the CC electrode. These findings demonstrate the potential of CoC x /Co@CC anodes for improving the EET efficiency of MFCs, which can have significant implications for sustainable energy and wastewater treatment.

Evaluation of the respective contribution of anode and cathode for triclosan degradation in a bioelectrochemical system

Min-Hua Cui,Qian Zhang,John Justo Ambuchi,Lan-Ying Liu,Lei Chen,Shi-Ming Niu,Chao Zhang,Hong-Bo Liu,Chao Tie,Xue-Juan Bi,He Liu,Ai-Jie Wang

In this work, the bioelectrochemical system (BES) is a feasible alternative for successfully degrading typical refractory emerging contaminant triclosan (TCS). A single-chamber BES reactor with an initial TCS concentration of 1 mg/L, an applied voltage of 0.8 V, and a solution buffered with 50 mM PBS degraded 81.4 ± 0.2% of TCS, exhibiting TCS degradation efficiency improvement to 90.6 ± 0.2% with a biocathode formed from a reversed bioanode. Both bioanode and biocathode were able to degrade TCS with comparable efficiencies of 80.8 ± 4.9% and 87.3 ± 0.4%, respectively. Dechlorination and hydrolysis were proposed as the TCS degradation pathway in the cathode chamber, and another hydroxylation pathway was exclusive in the anode chamber. Microbial community structure analysis indicated Propionibacteriaceae was the predominant member in all electrode biofilms, and the exoelectrogen Geobacter was enriched in anode biofilms. This study comprehensively revealed the feasibility of operating BES technology for TCS degradation.

Humic and fulvic acid-mediated dissimilatory iron reduction coupled with phosphate solubilizing bacteria inducing secondary mineral formation for efficient cadmium immobilization

Gongting Wang,Wenqing Luo,Zedong Teng,Xin Zhao,Yuxin Cheng,Yanzhong Wang,Bin Liu,Mengnan Lu,Min Li

Cadmium (Cd) pollution in sediments has become an urgent environmental problem. Humus mediated dissimilatory iron reduction and phosphorus solubilizing bacteria (PSB) were widely used for Cd immobilization. Here, we utilized humic acid (HA) and fulvic acid (FA) to promote the efficiency of iron reduction by Geobacter metallireducens GS-15 to immobilize Cd with the involvement of PSB. The results showed that both HA and FA can facilitate the reduction of Fe(III) and the release of P in ferric phosphate minerals, and then the immobilization of Cd. The immobilization of dissolved Cd 2 + in HA, FA and BCK groups were 76.25 %, 65.25 % and 63.58 %, respectively. HA and FA promoted the growth of PSB and the secretion of acetic acid, high concentration of acetic acid provided sufficient nutrients for the growth of iron-reducing bacteria. Thereby, high concentration of Fe(II) facilitated the recrystallization, which effectively immobilized Cd 2+ . Compared to the CK group, the HA and the FA groups increased the proportion of exchanged Cd converted to combined Cd in secondary minerals by 25.32 % and 19.16 %, respectively. The findings reveal that enhanced effect of HA on microbial iron reduction coupled with PSB can facilitate the immobilization of Cd, providing a promising strategy for the remediation of Cd-contaminated sediments.

Enhanced electroactive bacteria enrichment and facilitated extracellular electron transfer in microbial fuel cells via polydopamine coated graphene aerogel anode

Wei Guo,Yingying Chen,Jiayi Wang,Liang Cui,Yunhui Yan

The structure and surface physicochemical properties of anode play a crucial role in microbial fuel cells (MFCs). To enhance the enrichment of exoelectrogen and facilitate extracellular electron transfer (EET), a three-dimensional macroporous graphene aerogel with polydopamine coating was successfully introduced to modify carbon brush (PGA/CB). The three-dimensional graphene aerogel (GA) with micrometer pores improved the space utilization efficiency of microorganisms. Polydopamine (PDA) coating enhanced the physicochemical properties of the electrode surface by introducing abundant functional groups and nitrogen-containing active sites. MFCs equipped with PGA/CB anodes (PGA/CB-MFCs) demonstrated superior power generation compared to GA/CB-MFCs and CB-MFCs (MFCs with GA/CB and CB anodes respectively), including a 23.0 % and 30.1 % reduction in start-up time, and an increase in maximum power density by 2.43 and 1.24 times respectively. The higher bioelectrochemical activity exhibited by the biofilm of PGA/CB anode and the promoted riboflavin secretion by PGA modification imply the enhanced EET efficiency. 16S rRNA high-throughput sequence analysis of the biofilms revealed successful enrichment of Geobacter on PGA/CB anodes. These findings not only validate the positive impact of the synergistic effects between GA and PDA in promoting EET and improving MFC performance but also provide valuable insights for electrode design in other bioelectrochemical systems.

Weak electrostimulation enhanced the microbial transformation of ibuprofen and naproxen

Zhenfei Wang,Bin Liang,Yanan Hou,Si Li,Li Xie,Liang Peng,Peng Zhang,Aijie Wang,Hui Yun,Xiangkai Li

Ibuprofen (IBU) and naproxen (NPX) are commonly used non-steroidal anti-inflammatory drugs (NSAIDs) with high-risk quotients and are frequently detected in various aquatic environments. A weak electrostimulated biofilm not only had improved removal efficiencies to IBU and NPX, but also transformed different enantiomers with comparable efficiency and without configuration inversion. IBU was transformed mainly by oxidation (hydroxyl-IBU, carboxy-IBU), while NPX was mainly detoxified. The microbial analysis of IBU and NPX biofilm showed that the shared core consortia (> 1%) contained typical electro-active bacteria ( Geobacter , Desulfovibrio ), fermenters ( Petrimonas , Acetobacterium ) and potential degraders ( Pandoraea , Nocardiaceae ), which exhibited synergistic interactions by exchanging the additional electrons, H + , coenzyme NAD(H) or NAD(P) (H) and energy. The fungal community has a significant correlation to those core bacteria and they may also play transformation roles with their diverse enzymes. Plenty of nonspecific oxidoreductase, decarboxylase, hydrolase, cytochrome P450, and other enzymes relating to xenobiotic degradation were high-abundance encoded by the core consortia and could potentially participate in IBU and NPX biotransformation. This study offers new insights into the functional microbes and enzymes working on complex NSAIDs biotransformation and provided a feasible strategy for the enhanced removal of NSAIDs (especially IBU and NPX).

Photocathode optimization and microbial community in the solar-illuminated bio-photoelectrochemical system for nitrofurazone degradation

Yanping Hou,Guiyun Yuan,Shanming Qin,Lingli Tu,Yimin Yan,Zebin Yu,Hongfei Lin,Yongli Chen,Hongxiang Zhu,Hainong Song,Shuangfei Wang

To further enhance the bio-photoelectrochemical system (BPES) performance for nitrofurazone (NFZ) degradation and current output, the g-C 3 N 4 /CdS photocathode was optimized, and microbial community shift from inoculation to the BPES was analyzed. Results showed that photocathode with g-C 3 N 4 /CdS (mass ratio of 1:9) loading of 7.5 mg/cm 2 exhibited the best performance, with NFZ removal of 83.14% (within 4 h) and current of ~9 mA in the BPES. Proteobacteria accounted for the largest proportion: 66.53% (inoculation), 71.89% (microbial electrolysis cell (MEC) anode), 74.67% (BPES anode) and 57.31% (BPES cathode), respectively. In addition, Geobacter was the most dominant genus in MEC and BPES anode and cathode, which occupied 31.64%, 67.73% and 41.34%, respectively. The microbial compositions of BPES anode and cathode were similar, but different from that of MEC anode. Notably, Rhodopseudomonas , a photosynthetic species, was detected in the BPES. Cognition of microbial community in the BPES is important for advancing its development.

NanoFe3O4 accelerates methanogenic straw degradation in paddy soil enrichments

Cunpu Qiu,Youzhi Feng,Meng Wu,Jianwei Zhang,Xiaofen Chen,Zhongpei Li

Methanogenesis is crucial in the carbon cycle , and conductive iron oxide minerals play an important role in methanogenesis in anoxic environments. The influence of iron oxides is ubiquitous, but the strength and nature of iron oxides effects are believed to vary in response to environmental conditions, especially the soil iron concentration. We evaluated the influence of conductive nanoFe 3 O 4 on straw degradation in high-iron (HR) and low-iron (LR) paddy soil enrichments, as well as the corresponding responses of related microorganisms . Under conductive nanoFe 3 O 4 amendment, methane production and straw degradation increased by 184%–285% and 7.8%–12.7%, respectively, in the two soil enrichments. The increase apparently coincided with increased interspecies interactions within the microorganismal community, especially enhanced syntrophic cooperation among fermenting bacteria, syntrophic bacteria and methanogenic archaea , and facilitated interspecies electron transfer , which improved carbon conversion and alleviated end-product inhibition. The methanogenic archaeal community also exhibited changes with nanoFe 3 O 4 amendment, and the relative abundance of Methanosarcina increased. Pairwise comparisons revealed that although methanogenic straw degradation was higher in HR soil than in LR soil, the enhancement induced by the nanoFe 3 O 4 amendment showed to a larger degree and the microbial responses were more distinct in LR soil than in HR soil. The abundances of Geobacter and Methanosarcina greatly increased, and the microbial community shifted towards a composition similar to that in HR soil. This work indicates that iron oxides facilitate the methanogenic degradation of rice straw and that the indigenous iron content significantly influences the microbial community composition. This study provides a technique for agricultural residue degradation and has clear implications for our understanding of soil C-Fe biogeochemical coupling.

Lignocellulose reconstituted shape-controllable self-supporting carbonaceous capacitance-anodes with high electron transfer rates for high-performance microbial electrochemical system

Pinpin Yang,Yaqian Gao,Lefei Xue,Yu Han,Jingkun An,Weihua He,Yujie Feng

Natural biomass is a promising candidate for manufacturing an efficient anode in the microbial electrochemical system (MES) for its abundance and low cost. However, the structure and performance of the electrode highly depend on the biomass species. A simple and sustainable method for creating a self-supporting electrode is proposed by freeze-drying and carbonizing a blend of cellulose, lignin, and hemicellulose. This strategy leads to a cork-like structure and improved mechanical strength of the lignocellulose carbon. A power density of 4780 ± 260 mW m −2 (CLX-800) was achieved, which was the highest record for unmodified lignocellulose-based anodes in the microbial fuel cells. The morphological as lamellar multilayer and rich in hydrophilic functional groups could facilitate the formation of thick electroactive biofilms and enrich Geobacter with the highest abundance of 92.3%. The CLX material is expected to be the ideal electrode for high performance and functionally controllability.

Molecular insight into electron transfer properties of extracellular polymeric substances of electroactive bacteria by surface-enhanced Raman spectroscopy

Tan Bin,Zhou ShaoFeng,Wang Yi,Zhang BeiPing,Zhou LiHua,Yuan Yong

Extracellular polymeric substances (EPS) extracted from electroactive bacteria show promising redox activity, but the electron transfer (ET) mechanism of the EPS has been rarely elucidated because of their structural complexity and lack of efficient methodologies. In this study, the charge transfer theory of surface-enhanced Raman spectroscopy (SERS) was applied to evaluate the redox properties of EPS adsorbed on Ni and Ag nanoparticles (NPs). These metal NPs were used to simultaneously magnify Raman signals and reduce/oxidize the redox moieties in EPS. As a result, the ET between EPS and metal NPs was evaluated through the changes in Raman signals. In this regard, we compared the redox activity of EPS extracted from two typical electroactive bacteria ( Shewanella oneidensis and Geobacter sulfurreducens ) and another two nonelectroactive strains ( Escherichia coli and Bacillus subtilis ). Electrochemical measurements show that the electroactive strains have higher redox capabilities than nonelectroactive strains. The SERS analysis shows that the porphyrin present in cytochrome c is the dominating redox moiety in the EPS of electroactive bacteria. The results of this study show that SERS with active metal substrates is a sensitive tool to probe the redox response of EPS, offering an opportunity to better understand the redox nature of biomolecules.
