Preparation of PEI/La2O3/Polypropylene fiber filter for household water purifiers and investigation of its phosphate reduction and bacterial control

Linlin Hao,Gewei Feng,Zengli Zhai,Mingxuan Li

It has been accepted that limiting phosphate is a prerequisite for bacterial inactivation in tap water. La 2 O 3 nanoparticles were loaded onto polypropylene (PP) fibers through the method of melt spinning. Hydrophilic modification of the adsorbent surface was performed using polyethyleneimine (PEI) to prepare PEI/La 2 O 3 /PP. The results indicated that the optimal loading amount of La 2 O 3 was 1 %, and the optimal concentration of PEI was 1 %. The successful loading of La 2 O 3 resulted in a roughened fiber surface and increased fiber diameters . After PEI modification the water contact angle was reduced from 98° to 50°. The pH zpc of PEI/La 2 O 3 /PP was determined to be 8.3. The co-existing ions have little effect on phosphate adsorption. Phosphate adsorption can be well described by Redlich-Peterson model with a maximum K R-P of 3.98 × 10 −5  L/mg. Adsorption kinetics fit well with the pseudo-second-order reaction equation. The activation energy (E a) is determined to be 57.9 kJ/mol, providing the evidence of chemical nature of the reaction involving phosphate ions on the surface of PEI/La 2 O 3 /PP. The PEI/La 2 O 3 /PP adsorbent can reduce the phosphate concentration in tap water to below 0.01 mg/L, leading to a reduction in Escherichia coli concentration from approximately 10,000 CFU/mL to around 10 CFU/mL. The survival rates of Escherichia coli in a multi-nutrient environment are higher than in a single phosphate source environment. Our research provides a new feasible approach for ensuring microbial water quality safety in household water purifiers.

Construction of novel benzoxazine-linked covalent organic framework with antimicrobial activity via postsynthetic cyclization

Q. Ma,X. Liu,H. Wang,Q. Zhuang,J. Qian

Organic framework materials have shown increasingly promising applications in biomedicine, such as drug delivery and release. In this work, we first synthesized a new hydroxyl-containing imine-linked two-dimensional covalent organic framework (COF) through solvothermal synthesis . Then, the imine group was converted into a benzoxazine group using a cyclization reaction. The results show that the postsynthetic modification did not change the basic framework of the original COF and did not affect the basic properties of the original COF. At the same time, the new benzoxazine group obtained by cyclization gave the COF good antibacterial activity against Escherichia coli and Staphylococcus aureus . The COF efficiency after cyclization was improved, and its antibacterial activity against both bacteria was over 90% compared with the imine-linked COF. Moreover, the benzoxazine-linked COF crystal structure and pore structures were retained, leaving the drug delivery and release functions unaffected. A benzoxazine-linked COF has never been reported because it cannot be synthesized by a direct reaction method. The work in this paper shows that the COFs that cannot be directly synthesized can be obtained through specific postsynthetic modification reactions. This means that more functional COFs can be obtained based on existing COFs, and the diversity of COF types and their potential applications can be further enriched and expanded.

Citrus pectin derived silver nanoparticles and their antibacterial activity

Wenlin Zhang,Xi Juan Zhao,Yanyan Jiang,Zhiqin Zhou

Herein, a green, simple, and cost-effective synthesis method for silver nanoparticles (Ag NPs) has been established by using citrus pectin as reducing and capping agent. Importantly, no toxic chemicals were added into the synthesis process. The Ag NPs were characterized by UV-vis, TEM, XRD, and FTIR. The results indicated the Ag NPs were spherical shape with small, uniform size of 3 nm and excellent dispersity. Additionally, the Ag NPs have good antibacterial activity against Escherichia coli and Staphylococcus aureus. Using citrus pectin for preparation of Ag NPs was potentially a new path for the efficient utilization of citrus resources.

Polyphenolic Nanoparticle-Modified Probiotics for Microenvironment Remodeling and Targeted Therapy of Inflammatory Bowel Disease

Qinglian Hu,Jingyu Li,Tong Wang,Xiangchi Xu,Yuxuan Duan,Yuanxiang Jin

Inflammatory bowel diseases (IBDs) refer to multifaceted disorders in the intestinal microenvironment and microbiota homeostasis. In view of the broad bioactivity and high compatibility of polyphenols, there is considerable interest in developing a polyphenol-based collaborative platform to remodel the IBD microenvironment and regulate microbiota. Here, we demonstrated the coordination assembly of nanostructured polyphenols to modify probiotics and simultaneously deliver drugs for IBD treatment. Inspired by the distinctive structure of tannic acid (TA), we fabricated nanostructured pBDT-TA by using a self-polymerizable aromatic dithiol (BDT) and TA, which exhibited excellent antioxidant and anti-inflammatory capability in vitro. We thus coated pBDT-TA and sodium alginate (SA) to the surface of Escherichia coli Nissle 1917 layer by layer to construct the collaborative platform EcN@SA-pBDT-TA. The modified probiotics showed improved resistance to oxidative and inflammatory stress, which resulted in superior colon accumulation and retention in IBD model mice. Further, EcN@SA-pBDT-TA could alleviate dextran sulfate sodium (DSS)-induced colitis by controlling the inflammatory response, repairing intestinal barriers, and modulating gut microbiota. Importantly, EcN@SA-pBDT-TA-mediated IBD drug delivery could achieve an improved therapeutic effect in DSS model mice. Given the availability and functionality of polyphenol and prebiotics, we expected that nanostructured polyphenol-modified probiotics provided a solution to develop a collaborative platform for IBD treatment.

Structural characterization and prebiotic activity evaluation of novel fucosylated disaccharides prepared from bacterial exopolysaccharides

Shihao Nan,Xinmiao Ren,Mengshi Xiao,Lin Zhu,Haijin Mou,Ningyang Li,Rong Li,Shaoping Nie,Xiaodan Fu

The deoxyhexose sugar l -fucose and fucosylated oligosaccharides (FCO) are essential for various biological processes in human body and associated gut microbiota. Bacterial exopolysaccharides offer a promising new source for preparing functional oligosaccharides. Here, FCO were obtained through acidolysis of fucose-containing exopolysaccharides with a molecular weight of 5.9 × 10 3  kDa, derived from Clavibacter michiganensis M2. Structural analysis revealed that the main disaccharide components of FCO exhibited a molecular weight of 326 Da and a monosaccharide composition of fucose, galactose, and glucose in a 3:1:2 ratio. Nuclear magnetic resonance indicated that the disaccharide fragments consisted of two backbones of α- Galp -(1 → 2)-α- Fucp and β- Glcp -(1 → 4)-β- Fucp , respectively. In vitro fermentation of FCO with human fecal cultures resulted in an increased abundance of Bifidobacterium and Bacteroides , and a reduction in Escherichia-Shigella and Fusobacterium . This research outlines a novel structural fucosylated oligosaccharide and its preparation method, suggesting its potential application as a promising new prebiotic.

Curdlan-polyphenol complexes prepared by pH-driven effectively enhanced their physicochemical stability, antioxidant and prebiotic activities

Huan Li,Saiqing Xu,Ying Xie,Qun Zhang,Shenghua Ding,Rongrong Wang,Fuhua Fu,Xiaobei Zhan

In this study, the curdlan-polyphenol complexes were constructed by a pH-driven method. The interaction between curdlan and various hydrophobic polyphenols (curcumin, quercetin, and chlorogenic acid) was investigated. Curdlan could self-assemble into particles for loading polyphenols through hydrogen bonding and hydrophobic interactions. The three polyphenols were embedded in curdlan in an amorphous state. The curdlan-curcumin complex showed the lowest viscoelasticity but exhibited the highest curcumin loading ability (34.04 ± 1.73 mg/g). However, the curdlan-chlorogenic acid complex emerged the opposite trend, indicating that the loading capacity was associated with the hydrophobicity of polyphenols. The antioxidant activity of curdlan significantly increased after combining with polyphenols, which could be maintained during in vitro simulated gastrointestinal digestion. In particular, the curdlan-quercetin complex exhibited the highest antioxidant activity and short-chain fatty acid concentration, which could influence gut microbiota composition by promoting the proliferation of Prevotella and inhibiting the growth of Escherichia_Shigella . In conclusion, the curdlan-polyphenol complexes prepared by an alcohol-free pH-driven method could effectively enhance the gastrointestinal stability of polyphenols as well as increase the antioxidant and prebiotic activities of curdlan, which could be applied as a functional ingredient to improve gut health.

Amelioration effects of chlorogenic acid on mice colitis: Anti-inflammatory and regulation of gut flora

Peng Xu,Shuai Chen,Qi Fu,Siyi Zhu,Zheng Wang,Jie Li

This research endeavors to investigate the potential preventive and protective effects of chlorogenic acid (CGA), a bioactive component found in various foods, in the context of dextran sodium sulfate (DSS)-induced colitis in mice. The study reveals that CGA administration, at dosages of 150 and 300 mg/kg, significantly retards the progression of colitis. This is substantiated by observable enhancements in colon length, reduction in the disease activity index, and mitigation of colon tissue damage. Meanwhile, CGA acts as an antioxidant by reducing the oxidative stress associated with colitis inflammation and increasing the activity of antioxidant enzymes. Furthermore, CGA demonstrates anti-inflammatory properties by inhibiting the inflammatory response associated with colitis and regulating the excessive expression of cytokines. Notably, CGA contributes to the restoration of intestinal barrier function by augmenting the levels of claudin-1 and ZO-1. Moreover, the investigation uncovers CGA's capacity to modulate the gut microbiota composition in colitic mice, fostering the proliferation of beneficial bacteria, notably Lactobacillus , while concurrently impeding the growth of detrimental bacteria like Bacteroides , Enterobacteriaceae, and Escherichia_Shigella . To conclude, these findings substantiate the viability of CGA for diverse applications in the realms of food and medicine, thereby bolstering its potential as a therapeutic strategy within dietary interventions.

Modified montmorillonite armed probiotics with enhanced on-site mucus-depleted intestinal colonization and H2S scavenging for colitis treatment

Jiali Yang,Shengchan Ge,Shaochong Tan,Hua Liu,Mingzhu Yang,Wei Liu,Kaixiang Zhang,Zhenzhong Zhang,Junjie Liu,Jinjin Shi,Zhi-Hao Wang,Jitian Li

Inflammatory bowel diseases (IBD) are often associated with dysregulated gut microbiota and excessive inflammatory microenvironment. Probiotic therapy combined with inflammation management is a promising approach to alleviate IBD, but the efficacy is hindered by the inferior colonization of probiotics in mucus-depleted inflammatory bowel segments. Here, we present modified montmorillonite armed probiotic Escherichia coli Nissle 1917 (MMT-Fe@EcN) with enhanced intestinal colonization and hydrogen sulfide (H 2 S) scavenging for synergistic alleviation of IBD. The montmorillonite layer that can protect EcN against environmental assaults in oral delivery and improve on-site colonization of EcN in the mucus-depleted intestinal segment due to its strong adhesive capability and electronegativity, with a 22.6-fold increase in colonization efficiency compared to EcN. Meanwhile, MMT-Fe@EcN can manage inflammation by scavenging H 2 S, which allows for enhancing probiotic viability and colonization for restoring the gut microbiota. As a result, MMT-Fe@EcN exhibits extraordinary therapeutic effects in the dextran sulfate sodium-induced mouse colitis models, including alleviating intestinal inflammation and restoring disrupted intestinal barrier function, and gut microbiota. These findings provide a promising strategy for clinical IBD treatment and potentially other mucus-depletion-related diseases.

Simulated digestion and fecal fermentation behaviors of exopolysaccharides from Paecilomyces cicadae TJJ1213 and its effects on human gut microbiota

Juanjuan Tian,Xiaomeng Wang,Xueliang Zhang,Xiaohong Chen,Xin Rui,Qiuqin Zhang,Mingsheng Dong,Wei Li

This study aimed to explore the in vitro simulated digestion and fecal fermentation behaviors of two purified exopolysaccharide fractions (EPS1 and EPS2) from Paecilomyces cicadae TJJ1213 and its effects on human gut microbiota composition. Results showed that EPS1 and EPS2 could not be digested by saliva-gastrointestinal. After fecal fermentation, however, the molecular weight of EPS1 and EPS2 significantly decreased, and the molar ratios of the monosaccharide composition remarkably changed, indicating that EPS1 and EPS2 could be degraded and consumed by human gut microbiota. Notably, EPS1 and EPS2 could significantly modulate the composition, via increasing the relative abundances of Bacteroides and Phascolarctobacterium and decreasing the pathogenic bacteria Escherichia-Shigella, Klebsiella and Fusobacterium . In addition, EPS1 and EPS2 also could promote the production of short-chain fatty acids during fermentation for 24 h. These results suggested that EPS from Paecilomyces cicadae TJJ1213 can be used as a functional food to improve health and prevent diseases by promoting gut health.

Bioorthogonal conjugation and responsive nanocoating of probiotics for inflammatory bowel disease

Pandi Peng,Tao Feng,Xue Yang,Rui Ding,Junru Wang,Pengyu Chen,Yuxin Guo,Peng Li

Inflammatory bowel disease (IBD) is closely associated with dysregulated immune response, gut mucosal barrier, and microbiota. Conventional treatments suffer from inferior bioavailability and inadequate efficiency. Herein, we present a synergistic therapeutic strategy based on multifunctionalized probiotics to mitigate IBD through single oral administration. The probiotic ( Escherichia coli Nissle 1917) is bioorthogonally conjugated with immunomodulators and subsequently encapsulated by an enteric coating. The viability and bioactivity of probiotics are not affected by the modifications. And the armored probiotics are able to resist the harsh environment of the stomach and shed their enteric coating in the intestinal tract, exposing immunomodulators to polarize pro-inflammatory M1-type macrophages into anti-inflammatory M2-type. In a mouse colitis model, orally administered multifunctionalized probiotics cooperatively alleviate IBD with increased body weight to 1.13 folds and decreased disease activity index to 0.43 folds, through downregulating the pro-inflammatory cytokines expression, upregulating the epithelial tight junction-associated proteins levels to restore the intestinal barrier, and increasing the microbiota richness and abundance. This work exhibits a feasible approach to construct functionalized orally administered probiotics for enhanced synergistic therapy of IBD.

Microfluidic-based multifunctional microspheres for enhanced oral co-delivery of probiotics and postbiotics

Xinyuan Yang,Weimin Nie,Chong Wang,Zhonglin Fang,Luoran Shang

Probiotic-based therapies have shown great potential in the prevention and treatment of many diseases by positively regulating intestinal flora homeostasis. However, the efficacy of oral probiotics is severely limited due to the loss of bioactivity, short intestinal retention time, and insufficient therapeutic effect. Here, based on droplet microfluidics, we developed a hydrogel microsphere with colonic targeting and mucoadhesive capabilities as a multifunctional delivery platform, which can be used for co-delivery of probiotics ( Escherichia coli Nissle 1917, EcN) and auxiliary molecules (indole-3-propionic acid, IPA), achieving synergistic therapeutic effects. In vivo studies shown that the integrated multifunctional microspheres can significantly reduce intestinal inflammation, repair intestinal barrier function, enhance probiotic colonization in the intestine, and modulate disordered intestinal flora, demonstrating enhanced therapeutic effects in a mouse model of colitis. This work reveals that microfluidic-based smart droplet microspheres can provide a versatile platform for advanced microbial therapies.

Characterization and in vitro digestion of alkali-extracted polysaccharides from Grifola frondosa and its impacts on human gut microbiota

Baoming Tian,Xue Zhou,Yan Geng,Jiangning Hu,Bangwei Ye,Peilong Sun,Kai Yang

Grifola frondosa is a highly valued, edible and biologically active fungus in which the polysaccharide component plays a major role. To investigate the fundamental structure, in vitro digestion and fermentation characteristics of Grifola frondosa polysaccharides (GFP), GFP from the waste residue of Grifola frondosa after water extraction was re-extracted by alkaline solution. The carbohydrate and β-glucan contents of GFP were 91.61% and 60.57%, respectively, and were mainly composed of glucose. GFP can form triple supercoils and combine with Congo red, and the surface of GFP was smooth and dense, and the particles were in the shape of long column, sheet and ellipse. Analysis of carbohydrate, reducing sugars, and further chromatogram data also showed that GFP was not degraded by the simulated digestion process of the mouth, stomach and small intestine. Fecal fermentation of GFP significantly increased the abundance of Bacteroidota, and decreased Proteobacterias in both normal weight and obese subjects in vitro . The short-chain fatty acid (SCFA)-producing bacteria, Faecalibacterium , Erysipelotrichaceae_UCG_003 , Lactococcus were increased, and Dorea and Escherichia-Shigella were reduced after GFP treatment. Thus, the molecular of GFP was utilized and degraded by the gut microbiota and metabolized into SCFAs. This decreased pH, H 2 S, and NH 3, as well as increased CO 2 in the fermentation broth. In conclusion, GFP can modulate the composition of the gut microbiota and is a potentially functional product.
