Background Acetaminophen, a widely used analgesic and antipyretic drug, has become a significant aquatic micro-pollutant due to its extensive global production and increased consumption, particularly during the COVID-19 pandemic. Its high-water solubility leads to its pervasive presence in wastewater treatment plants (WWTPs), posing substantial risks to the environment and human health. Biological treatment is one of the promising approaches to remove such pollutants. Although previous studies have isolated acetaminophen-degrading pure cultures and proposed catabolic pathways, the interactions between microbiotas and acetaminophen, the distribution feature of acetaminophen degradation genes, and the gene-driven fate of acetaminophen in the real-world environment remain largely unexplored. Results Among the water samples from 20 WWTPs across China, acetaminophen was detected from 19 samples at concentrations ranging from 0.06 to 29.20 nM. However, p -aminophenol, a more toxic metabolite, was detected in all samples at significantly higher concentrations (23.93 to 108.68 nM), indicating the presence of a catabolic bottleneck in WWTPs. Metagenomic analysis from both the above 20 samples and global datasets revealed a consistently higher abundance of initial acetaminophen amidases compared to downstream enzymes, potentially having explained the reason for the bottleneck. Meanwhile, a close correlation between initial amidases and Actinomycetota revealed by genome-based taxonomy suggests a species-dependent degradation pattern. Additionally, a distinct amidase ApaA was characterized by newly isolated Rhodococcus sp. NyZ502 (Actinomycetota), represents a predominant category of amidase in WWTPs. Significant phylogenetic and structural diversity observed among putative amidases suggest versatile acetaminophen hydrolysis potential in WWTPs. Conclusions This study enhances our understanding of acetaminophen’s environmental fate and highlights the possible occurrence of ecological risks driven by imbalanced genes in the process of acetaminophen degradation in global WWTPs. Microbial technologies hold great promise for in situ remediation of polycyclic aromatic hydrocarbons (PAHs)-contaminated soils. However, the selection of enhancement measures and corresponding remediation strategies remains insufficiently understood. In this study, a series of enhancement treatments, including bacterial consortium inoculation (comprising Achromobacter denitrificans BP1, Rhodococcus aetherivorans BW2, and Lysinibacillus sp. BS3), nutrient addition, and bio-ventilation, were implemented to develop effective in situ remediation strategies for PAHs-contaminated soil. Over a 60-day incubation, the enhancement treatments achieved phenanthrene (PHE) degradation efficiencies of 68.0–94.7 % and benzo[a]pyrene (BaP) degradation efficiencies of 12.9–82.4 %. Degradation rates across soil layers followed the pattern: upper layer > lower layer > middle layer. Enhancement treatments significantly boosted soil dehydrogenase (DH) and fluorescein diacetate (FDAH) activities. Among these, the sequential consortium inoculation with nutrient addition treatment (T6) demonstrated the highest degradation efficacy. In the treatment T6, the relative abundance of consortium genera was significantly increased, playing critical roles in PAHs degradation. The connectivity and stability of the soil bacterial network were enhanced, providing greater resilience to pollutants. Quantitative PCR analysis showed that the enhancement strategy increased RHDα-GN gene abundance by 1.98-fold at the initial and maintained a positive correlation with PAHs residues throughout the process (p < 0.05), and the phe gene exhibited a continuous upward trend during remediation, ultimately reaching 1.61–1.96 times its initial abundance. Overall, this study provides a strong candidate of integrated enhancement strategies to advance in situ bioremediation of PAH-contaminated sites.

In this study, a microbial fuel cell powered electro-Fenton system (MFCⓅEFs) was established in order to overcome the shortcomings of low electron flux and unexpected methane production, while simultaneously treating excess sludge (ES, substrate) and refractory syringic acid (SA). A strategy of co-adding lysozyme (LZ, as ES degradation catalyst) and 2-bromoethanesulfonate (BES, as methane inhibitor) into ES was optimized in MFCⓅEFs to maximize electron flux, microbial community diversity and functional gene abundance. The removal of sludge total chemical oxygen demand (TCOD) achieved 81.69% in 25 d under an optimal co-addition strategy (40.41 mg/gSS of LZ, 27.03 mmol/L of BES, adding on 22.8 h of the7th day), with a simultaneous high degradation of SA (99.30% in 25 h). Correspondingly, a maximum power density of 3.35 W/m 3 was achieved (only 0.62 W/m 3 from the control), which effectively realizes in-situ micro-electricity generation and utilization for bioelectric Fenton processes. Moreover, 42.25% of the total charges were employed for bio-electricity generation. The electricigens of Pseudomonas , Acinetobacter and Chlorobium showed effective enrichment, while the abundance of methanogenesis archaea was extremely decreased. Functional genes associated with methanogenesis including mtaA, hdra, and mcrA were effectively inhibited. The life cycle assessment along with an optimized co-addition strategy illustrated a beneficial environmental effect , particularly in terms of ecosystem quality and climate change. Above all, an enhanced synchronous degradation of excess sludge and refractory pollutants had been realized in a green and environmentally friendly way.
