Effect of substrate stiffness on interfacial Schallamach wave of flexible film/substrate bilayer structure: Cohesive contact insight

Shouyao Liu,Jian Wu,Shixue He,Xuebo Yuan,Stanisław Stupkiewicz,Youshan Wang

As the critical feature of the stick-slip for soft materials, the interfacial Schallamach waves of flexible composite structures are essential for smart tactile sensors to realize sliding perception. Herein, the Schallamach waves of polydimethylsiloxane film/substrate bilayer structures with three substrate stiffnesses regulated by porosities are investigated by setting up in-situ sliding tests and establishing finite element models with mixed-mode cohesive contact. Inhomogeneity in microcontact stiffness disrupts the continuity and synchronization of the Schallamach waves, resulting in non-periodic fluctuations in the contact force. The buckling phenomenon of the film structure marks the transition from stick to slip. This buckling induces a shift at the crack front from normal compressive stress to tensile stress, leading to mixed-mode damage.

The rate of fluid shear stress is a potent regulator for the differentiation of mesenchymal stem cells

Danyang Yue,Mengxue Zhang,Juan Lu,Jin Zhou,Yuying Bai,Jun Pan

We have previously demonstrated that the rate of fluid shear stress (ΔSS) can manipulate the fate of mesenchymal stem cells (MSCs) to osteogenic or chondrogenic cells. However, whether ΔSS is comparable to other two means of induction medium and substrate stiffness that have been proven to be potent in differentiation control is unknown. In this study, we subjected MSCs to 1–7 days of osteogenic or chondrogenic chemical induction, or 1–4 days of 37 or 86 kPa of substrate stiffness induction, followed by 20 min of Fast ΔSS (0–0′) or Slow ΔSS (0–2′), which is a laminar FSS that linearly increased from 0 to 10 dyn/cm 2 in 0 (Fast) or 2 min (Slow) and maintained at 10 dyn/cm 2 for a total of 20 min. We found that 20 min of ΔSS could compete with 5 days' chemical and 2 days' substrate stiffness inductions. Our study confirmed that ΔSS is a powerful tool to control the differentiation of MSCs, which stressed the possible application in MSCs linage specification.

Improved Endothelial Function of Endothelial Cell Monolayer on the Soft Polyelectrolyte Multilayer Film with Matrix-Bound Vascular Endothelial Growth Factor

Hao Chang,Mi Hu,He Zhang,Ke-feng Ren,Bo-chao Li,Huan Li,Li-mei Wang,Wen-xi Lei,Jian Ji

Endothelialization on the vascular implants is of great importance for prevention of undesired postimplantation symptoms. However, endothelial dysfunction of regenerated endothelial cell (EC) monolayer has been frequently observed, leading to severe complications, such as neointimal hyperplasia, late thrombosis, and neoatherosclerosis. It has significantly impeded long-term success of the therapy. So far, very little attention has been paid on endothelial function of EC monolayer. Bioinspired by the microenvironment of the endothelium in a blood vessel, this study described a soft polyelectrolyte multilayer film (PEM) through layer-by-layer assembly of poly(l-lysine) (PLL) and hyaluronan (HA). The (PLL/HA) PEM was chemically cross-linked and further incorporated with vascular endothelial growth factor. It demonstrated that this approach could promote EC adhesion and proliferation, further inducing formation of EC monolayer. Further, improved endothelial function of the EC monolayer was achieved as shown with the tighter integrity, higher production of nitric oxide, and expression level of endothelial function related genes, compared to EC monolayers on traditional substrates with high stiffness (e.g., glass, tissue culture polystyrene, and stainless steel). Our findings highlighted the influence of substrate stiffness on endothelial function of EC monolayer, giving a new strategy in the surface design of vascular implants.

Role of Emerin in regulating fibroblast differentiation and migration at the substrate of stiffness coupled topology: Emerin regulates fibroblast differentiation and migration

Tiantian Yang,Li Wang,Haiyang Ma,Kailun Li,Yajing Wang,Wenjie Tang,Zichen Wang,Meiwen An,Xiang Gao,Ludan Xu,Yunyun Guo,Jiqiang Guo,Yong Liu,Hugen Wang,Yang Liu,Quanyou Zhang

In hypertrophic scars, the differentiation and migration of fibroblasts are influenced by the extracellular matrix microenvironment, which includes factors such as stiffness, restraint, and tensile force. These mechanical stresses incite alterations in cell behavior, accompanied by cytoskeletal protein reorganization. However, the role of nucleo-skeletal proteins in this context remains underexplored. In this study, we use a polyacrylamide hydrogel (PAA) to simulate the mechanical stress experienced by cells in scar tissue and investigate the impact of Emerin on cell behavior. We utilize atomic force microscopy (AFM) and RNA interference technology to analyze cell differentiation, migration, and stiffness. Our findings reveal that rigid substrates and cellular restriction elevate Emerin expression and diminish differentiation. Conversely, reducing Emerin expression leads to attenuated cell differentiation, where stiffness and constraining factors exert no notable influence. Furthermore, a softening of cells and an enhanced migration rate are also markedly observed. These observations indicate that variations in nuclear skeletal proteins, prompted by diverse matrix microenvironments, play a pivotal role in the pathogenesis of hypertrophic scars (HSs). This research offers novel insights and a reference point for understanding scar fibrosis formation mechanisms and preventing fibrosis.
