Carboxylesterase 1 regulates peroxisome proliferator-activated receptor gamma to inhibit the growth and metastasis of breast cancer cells

Wen Jingli,Geng Lei,Wang Ruohan,Zhang Xiaolei,Sui Yanmin,Liu Xiaofang,Han Xin

Breast cancer is a common malignancy in women, and it has an absence of effective therapies. Carboxylesterase 1 (CES1), a member of the carboxylesterase family, has anti-tumor properties in several types of cancer. However, the function of CES1 in breast cancer remains unclear. Peroxisome proliferator-activated receptor gamma (PPARG) is a downstream regulator of CES1 and exhibits anti-breast cancer properties. Both CES1 and PPARG were downregulated in breast cancer tissues. Low CES1 and PPARG expression were linked to poorer breast cancer survival. We constructed CES1 knockdown and overexpression models of breast cancer cells by CES1 overexpressing plasmids and plasmids containing short hairpin RNA. High expression of CES1 inhibited breast cancer cell proliferation, evidenced by diminished cell viability, decreased DNA replication, and G1 phase arrest. CES1 overexpression decreased the protein levels of CDK2, CDK6 and cyclin B1 in breast cancer cells. CES1 inhibited the Bcl-2/Bax axis and increased Cleaved caspase-3 levels. Transwell assays showed that CES1 inhibited cell migration and invasion. CES1 increased E-cadherin protein expression and decreased Vimentin protein expression. CES1 knockdown facilitated the proliferation, migration, and invasion of breast cancer cells. CES1 was found to regulate PPARG expression in breast cancer cells positively. We transfected PPARG-interfering plasmids into breast cancer cells with CES1 overexpression. Inhibition of PPARG abrogated the anti-growth and anti-metastasis functions of CES1 in breast cancer cells. This study elucidates that CES1 inhibits the malignant progression of breast cancer by up-regulating the expression of PPARG. Graphical

The critical role of transcription factor GATA1 in erythrocyte proliferation and morphology in blood clam Anadara granosa

Jiacheng Wang,Yuhang Zhang,Yingbin Xu,Hongyu Jin,Can Yang,Hongxing Liu,Yongbo Bao

The blood clam ( Anadara granosa ) exhibits innovative red blood compared to other molluscs, with erythrocytes constituting 79 % to 89 % of the total hemocyte count. Previously, we identified that GATA-binding protein 1 (designated as Ag GATA1) was significantly up-regulated in the transcriptomes of blood clam populations with high hemocyte counts. However, the function of Ag GATA1 in erythropoiesis remains unknown. In this study, we conducted GATA1 knockdown experiments in A. granosa , which resulted in a significant reduction of both erythrocytes and other cells during the S phase, as well as significantly decreased proliferative activity in the gill tissue. The cell proliferation assay demonstrated that overexpression of Ag GATA1 increased the DNA replication activity of K562 cells. Additionally, o-dianisidine staining revealed defects in erythropoiesis within the gill tissue following Ag GATA1 knockdown, while protein analyses confirmed a significant reduction in Ag GATA1 expression. Furthermore, decreased cytoskeletal fluorescence and morphological changes in erythrocytes suggest that Ag GATA1 influenced cytoskeletal protein expression, leading to changes in erythrocyte morphology. This study provides new insights into the role of Ag GATA1 in hemocyte formation and highlights its significance in understanding erythropoiesis within invertebrate developmental tissues.

Propofol inhibited gastric cancer proliferation via the hsa-miR-328-3p/STAT3 pathway

Bai Z. M.,Li X. F.,Yang Y.,Yang Y. F.,Lv D. R.,Tang L. L.

Purpose The aim of the present study was to elucidate the functional role of hsa-miR-328-3p/STAT3 pathway in the effects of propofol on gastric cancer proliferation. Methods Bioinformatics was used to analyze the molecular expression differences of hsa-miR-328-3p/STAT3 axis in stomach adenocarcinoma ( n  = 435) and normal samples ( n  = 41) from TCGA database. The expression of the above molecules in gastric cancer cells SGC-7901 and normal gastric mucosal cells GES-1 was verified via qPCR. The dual-luciferase assay was carried out to confirm the interaction between hsa-miR-328-3p and STAT3. Subsequently, the cell proliferation and the expression of the above molecules in SGC-7901 and GES-1 cells were evaluated after 10 μM propofol treatment. Finally, we analyzed whether propofol still inhibited the proliferation of gastric cancer by suppressing STAT3 pathway after hsa-miR-328-3p down-regulation. Results Compared with normal samples, the expression of hsa-miR-328-3p was significantly down-regulated in stomach adenocarcinoma samples, while the expression of STAT3 and downstream target genes (MMP2, CCND1 and COX2) was up-regulated. The results were consistent with those in GES-1 and SGC-7901 cell lines. Meanwhile, we found that hsa-miR-328-3p can bind to the 3′-UTR of the potential target gene STAT3. Furthermore, propofol significantly inhibited the proliferation of gastric cancer cell line SGC-7901, where hsa-miR-328-3p was up-regulated and the expression of STAT3 and downstream proliferation-related target genes were down-regulated. However, the growth inhibition of propofol on SGC-7901 cell was significantly reversed after the inhibition of hsa-miR-328-3p. Conclusions To sum up, propofol suppressed the STAT3 pathway via up-regulating hsa-miR-328-3p to inhibit gastric cancer proliferation.

Overexpressed LINC00467 promotes the viability and proliferation yet inhibits apoptosis of gastric cancer cells via raising ITGB3 level

Limao Xu,Chengmin Liu,Zhiyao Ye,Chengfeng Wu,Yuhang Ding,Juan Huang

Long non-coding RNA (lncRNA) LINC00467 plays a proto-oncogenic role in non-small cell lung cancer. However, its effect and modulatory mechanism in gastric cancer (GC) are unknown. Thereby, we elucidated the mechanism of LINC00467 in GC. LINC00467 level in GC tissues was assessed by bioinformatic analysis, and clinicopathological parameters from GC patients were collected. The levels of LINC00467, integrin subunit beta 3 (ITGB3), proliferating cell nuclear antigen (PCNA), cleaved caspase-3 and cleaved poly (ADP-ribose) polymerase 1 (PARP1) in tissue samples or treated GC cells were assessed by quantitative real-time polymerase chain reaction (qRT-PCR), fluorescence in situ hybridization (FISH), or Western blot. The viability, proliferation and apoptosis of GC cells were detected by methyl thiazolyl tetrazolium assay, colony formation assay, and flow cytometry. Levels of LINC00467 and ITGB3 were up-regulated in GC, and highly expressed LINC00467 was positively associated with tumor size, differentiation, N stage, and T stage in GC patients. LINC00467 was enriched in cytoplasm of GC cells, and overexpressed LINC00467 promoted the viability and proliferation as well as levels of ITGB3 and PCNA, while suppressing the apoptosis and levels of cleaved caspase-3 and cleaved PARP1 in GC cells. Besides, the effects of shLINC00467 on inhibiting cell viability, proliferation of GC cells and PCNA level and promoting apoptosis as well as levels of cleaved caspase-3 and cleaved PARP1 were all partially reversed by overexpressed ITGB3. Overexpressed LINC00467 enhanced the viability and proliferation but inhibited apoptosis of GC cells via increasing ITGB3 level.

Cellular responses to electrospun membranes made from blends of PLLGA with PEG and PLLGA-b-PEG

Chi Ma,Di Pang,Zuochun Xiong,Wei Bai,Chengdong Xiong

Control of cellular responses is crucial for the use of electrospun membranes in biomedical applications, including tissue engineering or biomedical devices. However, it is still unclear whether adhesion and proliferation of fibroblasts is stimulated or inhibited on polyethylene glycol (PEG)-modified electrospun membranes. In this study, poly( L -lactide-co-glycolide) (PLLGA)–PEG copolymer and pure PEG were blended with PLLGA, and then electrospun onto nonwoven membranes. The effects of blending of PLLGA–PEG or pure PEG on the adsorption of proteins, and further on the adhesion and proliferation of L929 fibroblasts on the electrospun membranes were investigated. Addition of PLLGA–PEG or PEG significantly improved the hydrophilicity of the electrospun membranes. Pure PEG had no obvious effects on the growth of L929 fibroblasts; in contrast, PLLGA–PEG significantly inhibited the adsorption of proteins and the proliferations of the cells on the electrospun membranes. In response to diminished protein adsorption, mRNA expression of genes related to cell adhesion and migration was up-regulated. The limited effects of pure PEG were probably caused by its preferential dissolution, whereas membrane-confined PLLGA–PEG displayed excellent performance on the inhibition of protein adsorption and cell proliferation. © 2012 Wiley Periodicals, Inc. Didymin Inhibits Proliferation and Induces Apoptosis in Gastric Cancer Cells by Modulating the PI3K/Akt Pathway

Tong Zhang,Bin Xu

Gastric cancer (GC) is a malignant tumor with high morbidity and mortality rates worldwide. This study aimed to investigate the effects and mechanisms of action of didymin, a dietary flavonoid glycoside, on GC treatment. Human GC cell lines Hs-746T and AGS were used to assess the effects of didymin on cell viability, cell proliferation, and cell cycle. The results showed that didymin decreased the proliferative capacity of GC cells and blocked cell cycle. Didymin decreased wound healing, invasion, and migration capacities of GC cells. Mitochondrial reactive oxygen species (ROS) levels and mitochondrial membrane potentials were reduced in cells treated with didymin. Network pharmacology analysis revealed that the therapeutic effects of didymin on AGS cells were related to the phosphatidylinositol 3-kinase (PI3K)/Akt pathway. In vivo mouse xenograft studies confirmed that didymin treatment decreased tumor cell proliferation, cell cycle protein levels, and Akt phosphorylation. The present study demonstrated that didymin regulates mitochondrial function and the PI3K/Akt pathway to inhibit cell proliferation and induce apoptosis in GC cells in vitro and in vivo. Therefore, didymin is a promising drug for the treatment of GC.

Semi-Synthesis of Harringtonolide Derivatives and Their Antiproliferative Activity

Xiutao Wu,Lijie Gong,Chen Chen,Ye Tao,Wuxi Zhou,Lingyi Kong,Jianguang Luo

Harringtonolide (HO), a natural product isolated fromCephalotaxus harringtonia, exhibits potent antiproliferative activity. However, little information has been reported on the systematic structure−activity relationship (SAR) of HO derivatives. Modifications on tropone, lactone, and allyl positions of HO (1) were carried out to provide 17 derivatives (2–13, 11a–11f). The in vitro antiproliferative activity against four cancer cell lines (HCT-116, A375, A549, and Huh-7) and one normal cell line (L-02) was tested. Amongst these novel derivatives, compound6exhibited comparable cell growth inhibitory activity to HO and displayed better selectivity index (SI = 56.5) between Huh-7 and L-02 cells. The SAR results revealed that the tropone and lactone moieties are essential for the cytotoxic activities, which provided useful suggestions for further structural optimization of HO.Keywords:harringtonolide;troponoids;structure-activity relationship (SAR);antiproliferative activity

Beta-sitosterol regulates PTGS1 to inhibit gastric cancer cell proliferation and angiogenesis

Jindao Wang,Minghui Zhou,Qiuli Zhou,Guangyang Sun,Yu Zhang,Feng Tao,Minfeng Ye

Background Gastric cancer (GC) is the third leading culprit of cancer-related deaths around the world. Beta-sitosterol (BS) is an important phytosterol that has been proven to have anti-proliferative effects on GC and other tumors. However, mechanisms and targets of BS in cancer are rarely explored. Methods In this investigation, the targets of BS in the treatment of GC were analyzed by network pharmacology. Molecular docking and cellular thermal shift assay were introduced to validate the binding relationship between BS and PTGS1. The impacts of BS on GC cell viability, half maximal inhibitory concentration, proliferation ability, apoptosis level, and angiogenesis ability were detected by using Cell Counting Kit-8, clone formation assay, flow cytometry, and angiogenesis experiment, respectively. In addition, the expression levels of angiogenic factors (VEGF, FGF, PAI-1) were detected by using western blot. Results In this project, through cell experiments, PTGS1 was identified as a protein that directly binds to BS. In vitro cell experiments revealed that BS promoted apoptosis and inhibited GC cell proliferation and angiogenesis. Importantly, treatment with BS attenuated the promoting influence of PTGS1 overexpression on GC cell proliferation and angiogenesis. Conclusion This investigation highlighted PTGS1 as the target of BS in GC cells. BS can regulate PTGS1 to inhibit GC cell proliferation and angiogenesis, providing new evidence for the potential use of BS as a therapeutic agent for GC.

Preparation and properties of PLCL/OM-Laponite materials with potential applications in orthopedic bandage

Yan Ren,Yonggang Lin,Yabei Ding,Ruimin Tang,Minglong Yuan,Hongli Li,Yongming Chuan

Poly( l -lactide- co -caprolactone) (PLCL) is the polymeric material with good biodegradability and biocompatible properties, but it is insufficient in strength properties that the pure PLCL is performed as orthopedic bandage. The PLCL has been blended with the laponite which is modified by cetyltrimethylammonium bromide (CTAB) to prepare composited bandage splines. The organic-modified laponite (OM-laponite) results of Fourier transform infrared (FTIR) and thermogravimetry analysis (TG) indicate that the laponite was successfully grafted with CTAB and the thermal stability was improved with increasing CTAB. The mechanical properties reveal that elongation at break strengthen with the increasing OM-laponite. When the content of OM-laponite is 2.0 %, the elongation at break and tensile strength of composited spline are superior to the medical orthopedic bandage. The results of thermal stability show that it was helpful to improve the crystallization properties of PLCL with the addition of OM-laponite. The water contact angle results demonstrate that the hydrophilicity is enhanced and is beneficial to cell adhesion. The results of cell experiment illustrate that the composite materials have a certain effect on cell proliferation when the content of OM-laponite is less than 2 %. Compared with the medical orthopedic bandage, it is satisfied with the application requirements.

Extraction and Isolation of Two Polysaccharides from Chloranthus japonicus Sieb. and Evaluation of Their Anti-Gastric Cancer Activities

Ju Liu,Wenfeng Li,Lu Jin,Yingchao Wang,Xinjun Xu,Enyao Ma,Depo Yang,Zhimin Zhao

Two unreported heteropolysaccharides, denoted as YCJP–1 and YCJP–2, were isolated from the herbs ofChloranthus japonicus. YCJP–1 was a heteropolysaccharide composed of glucose, galactose, arabinose, mannose, rhamnose, and a minor proportion of uronic acids, with the molecular weight mainly distributed in the 74,475–228,443 Da range. YCJP–2 was mainly composed of glucose, mannose, and galactose, with the molecular weights ranging from 848 to 5810 Da. To further evaluate the anti-gastric cancer effects ofC. japonicus, the inhibitory effects of the crude polysaccharide (YCJP) and the purified polysaccharides (YCJP–1 and YCJP–2) were determined using a CCK-8 assay and colon-forming assay on MGC-803 and AGS gastric cancer cell lines. Our results showed that YCJP, YCJP–1, and YCJP–2 possess prominent inhibitory effects on the proliferation of MGC-803 and AGS cells, and the AGS cell was more sensitive to YCJP, YCJP–1, and YCJP–2. Moreover, YCJP–2 demonstrated superior anti-gastric cancer effects compared to YCJP–1. This could potentially be attributed to YCJP–2’s higher glucose content and narrower molecular weight distribution.

Combinational Overexpression of Foxa3 and Hnf4a Enhance the Proliferation and Prolong the Functional Maintenance of Primary Hepatocytes

Fan J. Y.,Dama G.,Liu Y. L.,Guo W. Y.,Lin J. T.

Abstract In an in vitro culture system, primary hepatocytes usually display a low proliferation capacity, accompanied with a decrease of viability and a loss of hepatocyte-specific functions. Previous studies have demonstrated that the combination introductions of certain hepatocyte-specific transcription factors are able to convert fibroblasts into functional hepatocyte-like cells. However, such combinational usage of transcription factors in primary hepatocytes culture has not yet sufficiently studied. The forkhead box protein A3 (FoxA3) and hepatocyte nuclear factor 4α (Hnf4α) are liver-enriched transcription factors that play vital roles in the differentiation, and maintenance of hepatocytes. Thus, we simultaneously overexpressed the two genes, Foxa3 and Hnf4a , in rat hepatocytes and observed that the combinational augmentation of these two transcription factors have enhanced the proliferation and stabilized the hepatocyte-specific functions of primary hepatocytes over a long-term culture period.
