Structure identification and biological activity of a novel sulfated glycosaminoglycan-like polysaccharide from Sepia recurvirostra ink

Nanqi Cheng,Na Gao,Pin Wang,Yujun Ma,Pengfei Li,Xiaolu Wu,Jiaying Sun,Yulan Dai,Jinhua Zhao

A novel sulfated glycosaminoglycan-like polysaccharide (SIP), with a molecular weight of 13.78 kDa, was isolated from Sepia recurvirostra ink. SIP displayed a sulfate content of 6.82 % and consisted of GlcNAc, GalNAc, GlcA and Fuc. The distinct structural sequence of SIP was identified by 1D/2D NMR assignments of the backbone released by deaminative treatment and the oligosaccharides generated by mild acid hydrolysis. The backbone was built up of disaccharide repeating blocks structured as [−D-GlcA-β1,4-L-Fuc-α1,3-]. Notably, the non-sulfated GalNAc was attached to C-3 of Fuc, while the 4, 6-O-disulfated GlcNAc was pendant at C-4 in 20 % of GlcA residues, as revealed by the well-defined structures of trisaccharide, tetrasaccharides, heptasaccharides and decasaccharides. Except for the trisaccharide, the non-reducing ends of the other oligosaccharides were GlcNAc 6 S or GlcNAc 4S6S , suggesting selectivity of glycosidic bond cleavage and desulfation during mild acid hydrolysis. SIP showed negligible anticoagulant and heparanase inhibitory activity. Moreover, the effects of SIP on the generation of short-chain fatty acids (SCFAs) by colonic microbiota were evaluated. The addition of SIP resulted in a significant increase in levels of acetate, propionate, isobutyric acid, and isovaleric acid, offering insights into the contribution of the colonic microbiota to the health benefits of dietary squid ink.

Design of mechanically robust, recyclable, multi-stimuli-responsive shape memory elastomers based on biological phytic acid and cuttlefish ink

Jiarong Huang,Xiao Wang,Haonan Wu,Shuping Xiao,Yong Liang,Huiwen Yu,Baiping Xu,Wenhua Xu,Lingcao Tan

Facilitating the utilization of biomass in the rubber field is a facile strategy to reduce environmental pollution and the carbon emission. Herein, biological phytic acid (PA) was served as an eco-friendly curing agent, and cuttlefish ink (CI) was acted as a photothermal trigger switch to fabricate multi-stimuli-responsive shape memory biobased epoxidized natural rubber (ENR) with mechanical robustness and recyclable ability. ENR was vulcanized by PA via the ring-opening reaction between phosphate groups of PA and epoxy group of ENR, leading to the construction of an impeccable cross-linking network and an adjustable T g gradient, which endowed the elastomers with improved mechanical properties and shape memory effect. Meanwhile, the topology rearrangement occurred at elevated temperatures relied on phosphate transesterification, leading to excellent thermo-activated recyclable ability and reconfigurable shape memory performance of the elastomers. Additionally, CI acted as reinforced filler to enhance the ENR matrix, enabling elastomers to achieve a robust tensile strength of 21.4 MPa and tensile toughness of 30.4 MJ/m 3 , which was nearly 26.8 times and 11.3 times of the neat ENR, respectively. Exhilaratingly, CI also acted as photothermal trigger switch for the response to near-infrared light and sun, which realized the remotely multistage shape memory performance, demonstrating the potential application of ENR in the field of actuators without environmental pollution.

Design and construction of cuttlefish ink derived melanin complexed microneedles for microenvironment regulation to improve vitiligo treatment

Li Weimiao,Shi Yan,Zhan Ruimin,Liu Lu,Wang Jiarui,Lee Minhyeock,Zhang Bingqiang,Liang Shaoshuai,Wang Zhiguo,Kong Ming

Vitiligo is an autoimmune disorder marked by melanocyte destruction and epidermal depigmentation, primarily driven by inflammatory and oxidative stress within the affected skin lesions. Consequently, there is an urgent need for therapeutic strategies focused on protecting melanocytes and replenishing melanin for effective vitiligo management. In this study, a novel microneedle-based therapeutic platform (C/D/E@MN) was fabricated that was composed of cuttlefish ink nanoparticles (CINPs) for melanin supplementation, dipotassium glycyrrhizinate (DPG) for inflammation regulation, and skin-derived exosomes (EXO) to promote melanocyte proliferation. In addition, microneedles with varying dissolution profiles (swellable, slow-dissolving, and fast-dissolving) were designed and evaluated their performance to optimize therapeutic efficacy. In vitro results demonstrated that fast-dissolving microneedles (FDMN) significantly reduced cellular reactive oxygen species (ROS) and the secretion of vitiligo-related inflammatory cytokines and chemokines, such as IL-8, CXCL-16, and HMGB-1. Upon a vitiligo mice model, C/D/E@FDMN treatment group generated a significant increase in skin melanin content and a 15.5% reduction of whitening degree. The microneedles protected melanocytes and promoted lesion repigmentation through synergistic antioxidant, anti-inflammatory and cyto-proliferative mechanisms, offering a promising strategy for improved vitiligo therapy.

Sulfated polysaccharide of Sepiella Maindroni ink inhibits the migration, invasion and matrix metalloproteinase-2 expression through suppressing EGFR-mediated p38/MAPK and PI3K/Akt/mTOR signaling pathways in SKOV-3 cells

Wenjie Jiang,Yanna Cheng,Na Zhao,Lian Li,Yikang Shi,Aizhen Zong,Fengshan Wang

Previous studies demonstrated that SIP-SII, a sulfated derivative of SIP that is isolated from the ink of Sepiella maindroni , showed significant inhibition of tumor growth and metastasis. In this study, the effects of SIP-SII on the migration, invasion and molecular mechanism in ovarian cancer cell line, SKOV-3 cells, were investigated. The flow cytometry, confocal microscope observation, western blot and RT-PCR results indicated that SIP-SII located on cell membrane and inhibited the expression and activation of epidermal growth factor receptor (EGFR). Moreover, the binding capacity of SIP-SII with EGFR was confirmed by surface plasmon resonance (SPR) analysis and co-localization of EGFR and SIP-SII. Accordingly, SIP-SII was proved to attenuate the EGF-induced EGFR phosphorylation and migration by western blot and wound healing assay, respectively. Additionally, SIP-SII inhibited p38/MAPK and PI3K/Akt/mTOR signaling pathways in SKOV-3 cells significantly. What is more, SIP-SII showed amplified inhibitory activity on migration, invasion, and MMP-2 expression in combination with p38-specific inhibitor, PI3K-specific inhibitor or mTOR-specific inhibitor in SKOV-3 cells. Therefore, the mechanism that SIP-SII suppressed EGFR-mediated p38/MAPK and PI3K/Akt/mTOR signaling pathways to inhibit migration and invasion of SKOV-3 cells was demonstrated. These findings suggested that SIP-SII might be used as a potential inhibitor against tumor metastasis.
