Confined interface vibration for femtoliter droplets generation and manipulation

Dege Li,Bingfang Huang,Yi Cao,Molong Han,Xinlei Wu,Qiang Sun,Chi Ma,Lilong Zhao,Peng Liu,Chao Zheng,Hang Dong,Xiaolong Wang,Yonghong Liu,Yanzhen Zhang

The precise and effective generation and manipulation of micron-sized droplets is crucial for chemical and biological research and applications. Normally, droplets generation and manipulation are achieved by two separate devices/systems with their respective operation strategies. Pipette, inkjet nozzle, and microfluidic channels equipped with programed syringes are commonly used for droplets generation whereas the subsequent manipulation is normally achieved by leveraging of forces originating either from optical, electric, magnetic or acoustic field and the corresponding complicated devices. Here we report a new paradigm that can realize simultaneous droplets generation and manipulation in an immiscible liquid environment using only the confined interface vibration generated by the ordinary inkjet nozzle. Droplets, ranging from femtoliter to picoliter, can be generated and manipulated without interruption since they are actuated by the same nozzle. Our strategy provides a platform for precise generation and manipulating the micro-sized droplets that may be valuable for applications requiring simple setups and seamless connection between droplets generation and droplets manipulation.

Micro-sonobot-based decontamination for the renewable sweat glucose sensors

Ying Wei,Xiaolong Lu,Shuting Zhang,Long Cao,Bincheng Zhao,Yuliang Chen,Yuekuan Sun,Wenjuan Liu,Nan Zhu

The increase in the diabetic population and requirements for glucose monitoring devices accelerates the promotion of tremendous glucose biosensors. Aside from blood, alternative body fluids were explored to indicate the glucose profiles in a non-invasive manner. Sweat sensing using electrochemical biosensors offers an ideal solution for glucose monitoring, but their widespread application is now greatly hindered by the performance decreasing over the operation period. Here, we present a micro-sonobot-based decontamination strategy for the sweat glucose sensor to gently clean the sensor and achieve the recovery of its sensing activity. A screen-printed electrochemical biosensor with Cu 2 O decoration was proposed to detect sweat glucose and unveil the relationship between sweat and blood glucose levels. The sweat pollutant removal and sensor regeneration were realized using an acoustic manipulation platform and the bubble-powered micro-sonobots. The specified vibration mode of the acoustic manipulation platform generates the desired acoustic field for the control of bubble-powered micro-sonobots. The self-generated and swarm-enhanced acoustic streaming as well as the motion of micro-sonobots on the fouled sensor effectively remove the sweat pollutant and realize the renewal. The proposed decontamination method is expected to be a step toward the widespread application of sweat glucose biosensors and a general solution for the cleaning and regeneration of miniaturized surface-based biosensors.

Secondary Vortex Induced by Gigahertz Acoustic Streaming and its Applications for 3D Particles Manipulations

Yangchao Zhou,Haitao Zhang,Wenlan Guo,Chen Sun,Xuexin Duan

Acoustic streaming serves as a fast-developing contactless tool for manipulation of micro/nanoparticles. However, traditional Eckart streaming has low manipulation precision and Rayleigh streaming only works in a very close distance to the solid boundaries, which limits their applications over large spatial distance in fluid. In this paper, we propose a novel boundary-independent secondary vortex induced by the high frequency (GHz) acoustic streaming and demonstrate its application for precise three-dimensional (3D) particles manipulation. The size of the Eckart streaming generated by the micro-fabricated acoustic transducer can be efficiently compressed by increasing the viscosity of the solution, which results in a more stable and controllable secondary convection streaming in the chamber. Arbitrary and precise manipulation of different types of particles in 3D space is approached by controlling the position and power of the transducer. Moreover, multilayer assembly of cell-encapsulating hydrogels are achieved to validate the high throughput and good biocompatibility of the secondary vortex. This secondary vortex-based manipulation tool exhibits capacity of versatility toward different objects, high efficiency of assembly, as well as good biocompatibility for handling viable samples, which shows potentials in bio-fabrication, material synthesis and tissue engineering etc

Mixing during Trapping Enabled a Continuous-Flow Microfluidic Smartphone Immunoassay Using Acoustic Streaming

Xian Chen,Yuan Ning,Shuting Pan,Bohua Liu,Ye Chang,Wei Pang,Xuexin Duan

Smartphone-enabled microfluidic chemiluminescence immunoassay is a promising portable system for point-of-care (POC) biosensing applications. However, due to the rather faint emitted light in such a limited sample volume, it is still difficult to reach the clinically accepted range when the smartphone serves as a standalone detector. Besides, the multiple separation and washing steps during sample preparation hinder the immunoassay’s applications for POC usage. Herein, we proposed a novel acoustic streaming tweezers-enabled microfluidic immunoassay, where the probe particles’ purification, reaction, and sensing were simply achieved on the same chip at continuous-flow conditions. The dedicatedly designed high-speed microscale vortexes not only enable dynamic trapping and washing of the probe particles on-demand but also enhance the capture efficiency of the heterogeneous particle-based immunoassay through active mixing during trapping. The enriched probe particles and enhanced biomarker capture capability increase the local chemiluminescent light intensity and enable direct capture of the immunobinding signal by a regular smartphone camera. The system was tested for prostate-specific antigen (PSA) sensing both in buffer and serum, where a limit of detection of 0.2 ng/mL and a large dynamic response range from 0.3 to 10 ng/mL using only 10 μL of sample were achieved in a total assay time of less than 15 min. With the advantages of on-chip integration of sample preparation and detection and high sensing performance, the developed POC platform could be applied for many on-site diagnosis applications.
