Summary Objective We assessed the expression of glucose transporter (GLUT)-1 and hexokinase (HK)-Ⅱ in vocal cord leukoplakia (VCL) and investigated its clinical significance. Methods We collected 127 archival pathological specimens from patients with VCL, 29 from vocal cord polyps, and 31 with laryngeal carcinoma at our hospital between December 2018 and December 2022. GLUT-1 and HK-II expression was detected using immunohistochemistry. Results Significant differences were observed between the VCL and vocal cord polyp groups according to sex ( P < 0.0001), smoking status ( P = 0.001), and alcohol consumption ( P = 0.007). GLUT-1 expression was observed in 36.2%, 20.7%, and 90.3% of patients with VCL, vocal cord polyps, and laryngeal carcinoma, respectively. GLUT-1 was highly expressed in severe dysplasia or carcinoma in situ (CIS) compared with vocal cord polyps ( P < 0.001). GLUT-1 expression was lower in the VCL group than in the laryngeal carcinoma group ( P < 0.001). HK-II expression was observed in 42.5%, 34.5%, and 80.6% of patients with VCL, vocal cord polyps, and laryngeal carcinoma, respectively. HK-II expression was significantly higher in severe dysplasia or CIS compared with vocal cord polyps ( P = 0.004). HK-II expression was lower in the VCL group than in the laryngeal carcinoma group ( P < 0.001). Both GLUT-1 expression ( P < 0.001) and HK-II expression ( P = 0.03) increased significantly with the degree of dysplasia. A significant correlation was observed between GLUT-1 and HK-II expression (r = 0.313, P < 0.001). Conclusion GLUT-1 and HK-II are significantly overexpressed in VCL patients. In VCL patients, GLUT-1 and HK-II expression levels increase proportionally with the degree of dysplasia.

Vocal fold (VF) scarring results from injury to the unique layered structure and is one of the main reasons for long-lasting dysphonia. A minimally invasive procedure with injectable hydrogels is a promising method for therapy. However, current surgical techniques or standard injectable fillers do not yield satisfactory outcomes. In this work, an injectable hybrid hydrogel consisting of oxide hyaluronic acid and hydrazide-modified waterborne polyurethane emulsion was injected precisely into the injury site and cross-linked in situ by a dynamic hydrazone bond. The prepared hydrogel displays excellent injectability and self-healing ability, showing favorable biocompatibility and biodegradability to facilitate endogenous newborn cell migration and growth for tissue regeneration. With the aim of evaluating the antifibrosis and regeneration capacity of the hybrid hydrogel in the VF scarring model, the morphology and vibration characteristics of VFs, inflammatory response, and healing status were collected. The hybrid hydrogel can decrease the inflammation and increase the ratio of collagen III/collagen I to heal damaged scar-free tissue. Fascinatingly, the mucosal wave oscillations of healing VF by injecting the hybrid hydrogel were vibrated like the normal VF, achieving functional restoration. This work highlights the utility of hybrid hydrogels consisting of synthetic biodegradable waterborne polyurethane emulsions and natural hyaluronic acid as promising biomaterials for scarless healing of damaged VFs.

Summary Biological tissues, such as heart valves and vocal cords, function through complex shapes and high fatigue resistance. Achieving both attributes with synthetic materials is hitherto an unmet challenge. Here we meet this challenge with hydrogels of heterogeneous structures. We fabricate a three-dimensional hydrogel skeleton by stereolithography and a hydrogel matrix by cast. Both the skeleton and matrix are elastic and stretchable, but the skeleton is much stiffer than the matrix, and their polymer networks entangle topologically. When such a hydrogel is stretched, the compliance of the matrix deconcentrates stress in the skeleton and amplifies fatigue resistance. We fabricate a homogeneous hydrogel and a heterogeneous hydrogel, each in the shape of a human heart valve. Subject to cyclic pressure, the former fractures in ∼560 cycles but the latter is intact after 50,000 cycles. Soft materials of complex shapes and high fatigue resistance open broad opportunities for applications.

In recent years, flexible high-performance piezoresistive pressure sensors have attracted considerable attention for the important application potential in the emerging fields of smart robots, wearable electronics and electronic skin. Herein, inspired by human skin, a new strategy was proposed for the fabrication of a double-layer piezoresistive pressure sensor with wide sensing range and high sensitivity. It was based on the utilization of sandpaper as template and MXene for the constructions of micro-protrusion rough surface on polydimethylsiloxane film and electrically conductive pathways, respectively. The prepared sensor demonstrated high sensitivity of 2.6 kPa −1 in wide linear range of 0–30 kPa, fast response/recovery time of 40/40 ms and excellent repeatability. Importantly, the sensor was successfully applied for the real-time detection of radial artery heart rate, limb movement, handwriting and vocal cord vocalization. Moreover, the integrated device by the sensors had the capability of identifying and visualizing spatial pressure distribution. The findings conceivably stand out a new methodology to prepare flexible high-performance piezoresistive pressure sensors for wearable electronics, human-computer interaction, intelligent robots and health monitoring.
