An Urchin-Shaped Copper-Based Metalloporphyrin Nanosystem as a Sonosensitizer for Sonodynamic Therapy

Aiqing Ma,Hui Ran,Jiaxing Wang,Rui Ding,Chengyu Lu,Lanlan Liu,Yingmei Luo,Huaqing Chen,Ting Yin

Sonodynamic therapy (SDT), as a novel cancer therapy strategy, might be a promising approach due to the depth-penetration property in tissue. Sonosensitizers are the key element for efficient SDT. However, the development of sonosensitizers with strong sonosensitization efficacy is still a significant challenge. Herein, an urchin-shaped copper-based metalloporphyrin liposome nanosystem (FA–L–CuPP) is constructed and identified as an excellent sonosensitizer. Under ultrasound (US) irradiation, FA–L–CuPP can be highly excited to generate several reactive oxygen species (ROS), such as singlet oxygen (1O2) and free radicals (⋅OH). The molecular orbital distribution calculations reveal that a strong intramolecular charge transfer might occur in the CuPP complex under US irradiation, which could afford enough energy to the surrounding O2and H2O to concert1O2, O2−and ⋅OH. Working as “ammunitions”, the largely produced ROS can kill 4T1 tumor cells, effectively inhibiting tumor growth. This work provides an urchin-shaped nanosonosensitizer based on a copper complex, which might provide an idea to design a novel sonosensitizer for noninvasive and precise SDT antitumor applications. Keywords:sonodynamic therapy;copper-based metalloporphyrin;sonosensitizer;urchin-shaped;noninvasive therapy

Renal-Clearable Metalloporphyrin Complex-Based Nanosonosensitizers Using Photoacoustic Imaging Guiding to Enhance Sonodynamic Cancer Therapy

Haiying Wang,Lifeng Hang,Hong Qu,Peiru Wu,Kelei Hua,Rongjian Jiang,Yanzhao Diao,LaiPing Fang,Sulian Su,Guihua Jiang

Sonodynamic therapy (SDT) is a noninvasive approach to tumor treatment, with ongoing efforts being focused on developing highly effective sonosensitizers with low toxicity. Herein, a liquid-phase stripping technique was introduced as a simple reflux method for synthesizing ultrasmall Mn-PCN-224 nanodots (MM NDs). Compared with PCN-224 nanodots, the synthesized MM NDs, which function as renal-clearable nanoagents, produced 2.42 times more reactive oxygen species (ROS) under identical ultrasound (US) irradiation conditions. In vivo and in vitro experiments revealed that A549 lung cancer cells treated with MM NDs under US irradiation and H2O2 exhibited a relative cell viability of ∼9% and a tumor inhibition rate of ∼91%. This result demonstrates that MM NDs can efficiently increase the effectiveness of SDT by leveraging their catalase-like activity and ultrasmall size (4 nm) to prevent ROS quenching. Furthermore, these nanoagents could be effectively utilized for photoacoustic (PA) imaging to track their accumulation in tumors and monitor the alleviation of the hypoxic tumor microenvironment. Notably, MM ND-mediated SDT demonstrated superior penetration depth compared to PDT, making it more effective in inhibiting contralateral tumors while facilitating deep-tissue treatment. Thus, this study introduces renal-clearable nanoagents with promising potential for PA-guided SDT, thereby paving the way for more effective tumor treatment strategies.

Piezo-Augmented Sonosensitizer with Strong Ultrasound-Propelling Ability for Efficient Treatment of Osteomyelitis

Xiaobo Feng,Liang Ma,Jie Lei,Qunle Ouyang,Yuxuan Zeng,Yue Luo,Xiaoguang Zhang,Yu Song,Gaocai Li,Lei Tan,Xiangmei Liu,Cao Yang

The successful treatment of osteomyelitis remains a great challenge in the field of orthopedics. The clinical method for treating refractory bone infection requires a combination therapy of long-term systemic antibiotics administration and surgical debridement. It is highly desirable to develop an antibiotic-free, noninvasive, rapid strategy to eradicate osteomyelitis. Herein, we fabricate a piezoelectric-enhanced sonosensitizer that consists of a porphyrin-based hollow metal–organic framework (HNTM), MoS2 nanosheets, and a red cell (RBC) membrane. We find that the ultrasound (US)-induced piezoelectric polarization of MoS2 can improve the charge transfer of HNTM at the heterointerface of HNTM-MoS2, increasing the production of reactive oxygen species (ROS). Besides, MoS2 increases the asymmetric shape of HNTM, leading to the strong US-propelling ability of HNTM–MoS2. The produced ROS and strong mechanical force can kill methicillin-resistant Staphylococcus aureus (MRSA) with an antibacterial efficiency of 98.5% under 15 min of US treatment, resulting in intracellular DNA damage and increased oxidative stress and disturbance of purine metabolism, tryptophan metabolism, and pantothenate and CoA biosynthesis of MRSA. Together with the toxin neutralization ability, the RBC-HNTM-MoS2 successfully eliminates the bone infection and suppresses inflammation and bone loss. This work provides another strategy for developing an efficient sonosensitizer through piezoelectric-assisted sonocatalysis and enhancing US-propelling ability.

Engineering Molybdenum-Assisted Tellurium Nanosonosensitizers for Enhanced Sonodynamic Tumor Nanotherapy

Lile Dong,Wenjuan Li,Yu Luo,Chaoran Liu,Kai Li,Yu Chen,Guobin Hong

Materdicine-augmented sonodynamic therapy has emerged as an efficient noninvasive strategy for cancer treatment by generating reactive oxygen species associated with the cavitation effect and sonosensitizers. However, semiconductor-based inorganic nanosonosensitizers have undesirable nondegradability, which can cause unsatisfactory therapeutic efficacy and potential toxicity. With the aim of overcoming this obstacle, in this study, Mo-assisted Te (MT) nanosonosensitizers with high oxidative degradation abilities are engineered for photoacoustic imaging (PA)-guided sonodynamic tumor nanotherapy. Intriguingly, the engineered MT nanosonosensitizers can generate abundant singlet oxygen under ultrasound irradiation to eradicate tumor cells, thereby demonstrating its ability to achieve high tumor growth inhibition rates. In particular, MT nanosonosensitizers exhibit oxidative degradation properties that guarantee desirable biocompatibility and potentiate clinical translation. Therefore, this study validates a paradigm for theory-oriented design of sonodynamic tumor nanotherapy.

Bacteria-driven nanosonosensitizer delivery system for enhanced breast cancer treatment through sonodynamic therapy-induced immunogenic cell death

Du Meng,Wang Ting,Peng Wangrui,Feng Renjie,Goh MeeiChyn,Chen Zhiyi

Background Sonodynamic therapy (SDT) has shown promise as a non-invasive cancer treatment due to its local effects and excellent tissue penetration. However, the limited accumulation of sonosensitizers at the tumor site hinders its therapeutic efficacy. Although nanosonosensitizers have improved local tumor accumulation through passive targeting via the enhanced permeability and retention effect (EPR), achieving sufficient accumulation and penetration into tumors remains challenging due to tumor heterogeneity and inaccurate targeting. Bacteria have become a promising biological carrier due to their unique characteristic of active targeting and deeper penetration into the tumor. Methods In this study, we developed nanosonosensitizers consisting of sonosensitizer, hematoporphyrin monomethyl ether (HMME), and perfluoro-n-pentane (PFP) loaded poly (lactic-co-glycolic) acid (PLGA) nanodroplets (HPNDs). These HPNDs were covalently conjugated onto the surface of Escherichia coli Nissle 1917 (EcN) using carbodiimine chemistry. EcN acted as an active targeting micromotor for efficient transportation of the nanosonosensitizers to the tumor site in triple-negative breast cancer (TNBC) treatment. Under ultrasound cavitation, the HPNDs were disrupted, releasing HMME and facilitating its uptakes by cancer cells. This process induced reactive oxygen species (ROS)-mediated cell apoptosis and immunogenic cell death (ICD) in vitro and in vivo. Results Our bacteria-driven nanosonosensitizer delivery system (HPNDs@EcN) achieved superior tumor localization of HMME in vivo compared to the group treated with only nanosonosensitizers. This enhanced local accumulation further improved the therapeutic effect of SDT induced-ICD therapeutic effect and inhibited tumor metastasis under ultrasound stimulation. Conclusion sOur research demonstrates the potential of this ultrasound-responsive bacteria-driven nanosonosensitizer delivery system for SDT in TNBC. The combination of targeted delivery using bacteria and nanosonosensitizer-based therapy holds promise for achieving improved treatment outcomes by enhancing local tumor accumulation and stimulating ICD.Graphical Abstract

Neutrophil membrane-encapsulated nanosonosensitizer with ultrasound-reinforced ferroptosis in Pseudomonas aeruginosa pneumonia

Du Chunhong,Wang Shuai,Cheng Yijie,Li Jie,Zhang Yufei,Li Zhuohao,Zhu Baolin,Wu Zhongming,Zhang Xinge,Zhou Lingyi

Pneumonia caused by Pseudomonas aeruginosa ( P. aeruginosa ) infection remains a formidable clinical challenge due to persistent biofilm formation and intrinsic antibiotic resistance, exacerbated by bacterial iron homeostasis that stabilizes biofilm architecture and neutralizes oxidative stress. Herein, we present Fe/TNT@NM, a biomimetic nanosonosensitizer activated by ultrasound (US) to dismantle biofilms through dual extracellular-intracellular mechanisms. The nanosonosensitizer features an iron-doped titanate nanotube (Fe/TNT) core encapsulated within a neutrophil membrane (NM). Under US irradiation, Fe/TNT@NM generates sonodynamic reactive oxygen species (ROS) extracellularly and enhances Fe 3+ release. These ions catalyze the Fenton reaction extracellularly to amplify chemodynamic effects and disrupt intracellular iron homeostasis, triggering bacterial ferroptosis. The NM coating enables immune evasion and biofilm-targeted delivery. This ultrasound-reinforced ferroptosis strategy synchronizes extracellular ROS storms with intracellular iron dyshomeostasis, achieving dual-action biofilm dismantling and eradication of drug-resistant P. aeruginosa . In a murine pneumonia model, Fe/TNT@NM suppresses biofilms and mitigates pulmonary injury. By converging biomimetic targeting, sonodynamic-chemodynamic cascades, and ultrasound-augmented ferroptosis, this nanosonosensitizer presents a paradigm-shifting approach to combat refractory biofilm infections and antibiotic resistance. Ultrasound Microbubbles Mediated Sonosensitizer and Antibody Co-delivery for Highly Efficient Synergistic Therapy on HER2-Positive Gastric Cancer

Lihong Sun,Jinxia Zhang,Menghong Xu,Lulu Zhang,Qingshuang Tang,Jing Chen,Ming Gong,Suhui Sun,Huiyu Ge,Shumin Wang,Xiaolong Liang,Ligang Cui

Trastuzumab combined with chemotherapy is the first-line treatment for advanced HER2-positive gastric cancer, but it still suffers from limited therapeutic efficiency and serious side effects, which are usually due to the poor delivery efficiency and the drug resistance of tumor cells to the chemotherapeutic drugs. Herein, a type of ultrasound microbubble for simultaneous delivery of sonosensitizers and therapeutic antibodies to achieve targeting combination of sonodynamic therapy and antibody therapy of HER2-positive gastric cancer was constructed from pyropheophorbide-lipid followed by trastuzumab conjugation (TP MBs). In vitro and in vivo studies showed that TP MBs had good biological safety, and their in vivo delivery can be monitored by ultrasound/fluorescence bimodal imaging. With ultrasound (US) located at the tumor area, TP MBs can be converted into nanoparticles (TP NPs) in situ by US-targeted microbubble destruction; plus the enhanced permeability and retention effects and the targeting effects of trastuzumab, the enrichment of sonosensitizers and antibodies in the tumor tissue can be greatly enhanced (∼2.1 times). When combined with ultrasound, TP MBs can not only increase the uptake of sonosensitizers in HER2-positive gastric cancer NCI-N87 cells but also efficiently generate singlet oxygen to greatly increase the killing effect on cells, obviously inhibiting the tumor growth in HER2-positive gastric cancer NCI-N87 cell models with a tumor inhibition rate up to 79.3%. Overall, TP MBs combined with US provided an efficient way for co-delivery of sonosensitizers and antibodies, greatly enhancing the synergistic therapeutic effect on HER2-positive gastric cancer while effectively reducing the side effects.

Novel Spatially Asymmetric Copper Bismuthate-Mediated Augmentation of Energy Conversion to Realize “Three-Step” Tumor Suppression

Jiarui Wang,Haoqin Zheng,Guangyao Hu,Xujian Yang,Hongpeng You,Lile Dong,Shuyan Song

The generally undesirable bandgap and electron–hole complexation of inorganic sonosensitizers limit the efficiency of reactive oxygen species (ROS) generation, affecting the effectiveness of sonodynamic therapy (SDT). Comparatively, the novel polyvinylpyrrolidone-modified copper bismuthate (PCBO) sonosensitizers are manufactured for a “three-step” SDT promotion. In brief, first, the strong hybridization between Bi 6s and O 2p orbitals in PCBO narrows the bandgap (1.83 eV), facilitating the rapid transfer of charge carriers. Additionally, nonequivalent [CuO 4 ] 6− layers reduce crystal symmetry, confer PCBO unique piezoelectricity, and improve electron–hole separation under ultrasonic (US) excitation. This allows PCBO to convert US energy into chemical energy to produce ROS, achieving the accumulation of abundant ROS, resulting in apoptosis and tumor suppression. Concurrently, PCBO also acts as a glutathione scavenger to reduce tumor antioxidant capacity and improve efficacy. To the best of authors understanding, this study reveals PCBO as an innovative piezoelectric sonosensitizer and provides a meaningful paradigm for designing energy conversion strategies for tumor suppression.

Ultrasound Activated Copper-Based Sonosensizer for Dual-Amplified ROS-Mediated Tumor Therapy and Cuproptosis Induction

Penghui Wang,Runming Zhong,Yingzhe Wu,Lingli Jin,Yanan Zhang,Kailong Zhang,Fan Zhao

Sonodynamic therapy (SDT) is a promising noninvasive cancer treatment with deep tissue penetration, but its therapeutic efficacy is limited by insufficient reactive oxygen species (ROS) production. Herein, we developed copper-based sonosensitizer (CCG MPNs) by coordinating copper ions (Cu2+) with chlorin e6 (Ce6) and gallic acid (GA) for dual-amplified ROS-mediated tumor therapy and cuproptosis induction. CCG MPNs dissociate in the high-glutathione (GSH) tumor microenvironment, releasing Cu ions, Ce6, and GA. GA rapidly depletes GSH, enhancing tumor cell sensitivity to ROS and cuproptosis. Additionally, the released Cu+ ions generate cytotoxic hydroxyl radicals via a highly efficient Fenton reaction and ultrasound (US) activation of Ce6 produces singlet oxygen, amplifying ROS-mediated apoptosis by evoke chemodynamic therapy (CDT) and SDT. Moreover, the yielded ROS would cut off the ATP supply, resulting in a decrease in the expression of the copper transporter protein ATP7A. Large accumulations of intracellular Cu+ ions can be more readily induce cuproptosis by promoting lipoylated protein aggregation. In vitro and in vivo anti-tumor studies showed that CCG MPNs group under US treatment exhibited better anti-tumor performance. This multimodal approach overcomes the limitations of the tumor microenvironment, offering a synergistic strategy to enhance tumor therapy efficiency through combined ROS generation and cuproptosis activation.

Fluorinated Chitosan To Enhance Transmucosal Delivery of Sonosensitizer-Conjugated Catalase for Sonodynamic Bladder Cancer Treatment Post-intravesical Instillation

Guangzhi Li,Shupeng Wang,Dashi Deng,Zhisheng Xiao,Ziliang Dong,Zhiping Wang,Qifang Lei,Shan Gao,Guixiao Huang,Enpu Zhang,Guohua Zeng,Zhong Wen,Song Wu,Zhuang Liu

Sonodynamic therapy (SDT) is a noninvasive ultrasound-triggered therapeutic strategy for site-specific treatment of tumors with great depth penetration. The design of nano-sonosensitizers suitable for SDT treatment of bladder cancer (BCa) post-intravesical instillation has not yet been reported. Herein, a transmucosal oxygen-self-production SDT nanoplatform is developed to achieve highly efficient SDT against BCa. In this system, fluorinated chitosan (FCS) is synthesized as a highly effective nontoxic transmucosal delivery carrier to assemble with meso-tetra(4-carboxyphenyl)porphine-conjugated catalase (CAT-TCPP). The formed CAT-TCPP/FCS nanoparticles after intravesical instillation into the bladder cavity exhibit excellent transmucosal and intratumoral penetration capacities and could efficiently relieve hypoxia in tumor tissues by the catalase-catalyzed O2 generation from tumor endogenous H2O2 to further improve the therapeutic efficacy of SDT to ablate orthotopic bladder tumors under ultrasound. Our work presents a nano-sonosensitizer formulation with FCS to enhance transmucosal delivery and intratumoral diffusion and CAT to improve tumor oxygenation, promising for instillation-based SDT to treat bladder tumors without the concern of systemic toxicity.

Synergistic therapy for orthotopic gliomas via biomimetic nanosonosensitizer-mediated sonodynamic therapy and ferroptosis

Mingting Zhu,Pengying Wu,Yan Li,Lei Zhang,Yujin Zong,Mingxi Wan

Ferroptosis is an emerging form of programmed cell death, and its combination with sonodynamic therapy (SDT) for anti-tumor activity is gradually attracting attention. However, their application against gliomas has not been studied. Herein, multifunctional cancer homologous targeting biomimetic nanoparticles (PIOC@CM NPs) encapsulating both Fe3O4 and Ce6 were constructed as a nanosonosensitizer. Based on focused ultrasound (US) combined with circulating microbubbles (MBs) to open the blood–brain barrier (BBB) in a safe and transient manner, the development of a therapeutic strategy to integrate the biomimetic nanosonosensitizer-mediated SDT and ferroptosis could achieve synergistic therapeutic effects against gliomas. We demonstrated that the glioma C6 cell membrane (CM) on the surface of the NPs allowed the nanosonosensitizer to accumulate selectively in tumors through homologous targeting in vitro. After efficient internalization in C6 cells, the PIOC@CM NPs could significantly increase the level of reactive oxygen species (ROS) and deplete glutathione (GSH) upon ultrasonic irradiation, resulting in the loss of glutathione peroxidase-4 (GPX4) activity, which facilitated SDT and ferroptosis to kill glioma C6 cells. Furthermore, the PIOC@CM NPs were intravenously injected after noninvasively opening the BBB via US-MBs, which enhanced the accumulation of the nanosonosensitizer in tumor tissues. Crucially, an attractive phenomenon of the significant reduction in orthotopic gliomas after the second US pulse-triggered SDT and ferroptosis was observed. Taken together, this study presents a novel combinatorial glioma therapeutic strategy based on noninvasive BBB opening with a biomimetic sonotheranostic system-mediated SDT and ferroptosis.

Poly(Amino Acid) Coordination Nanoparticle as a Potent Sonosensitizer for Cancer Therapy

Lintao Bai,Mingkun Wang,Lin Zhang,Meijun Zhao,Mei Ren,Lirong Zheng,Ming Lei,Heyun Shen

Sonodynamic therapy (SDT) has gained widespread attention owning to its much deeper tissue penetration depth compared with phototherapy. However, developing sonosensitizers with high sonosensitization efficacy in a tumor hypoxic environment is still a significant challenge. Herein, we fabricated a poly(amino acid) coordination nanoparticle (PFHF) as a potent sonosensitizer with great biocompatibility and high hydroxyl radical (•OH)-generation capacity for efficient SDT. PFHF was identified as a hexa-coordination structure by X-ray absorption fine structure and density functional theory. Furthermore, PFHF could be excited by ultrasound, which directly cleaved H2O into •OH, achieving oxygen-independent SDT. In addition, doxorubicin-loaded PFHF achieved a significant synergistic chemo-sonodynamic therapeutic effect with a tumor inhibition rate of 89%. The poly(amino acid) coordination nanoparticle with good biocompatibility and oxygen-independent ability could provide an idea for the rational design of sonosensitizers.
