Cloning and Characterization of a New Chitosanase From a Deep-Sea Bacterium Serratia sp. QD07

Zheng Qiuling,Meng Xiangjun,Cheng Mingyang,Li Yanfeng,Liu Yuanpeng,Chen Xuehong

Chitosanase is a significant chitosan-degrading enzyme involved in industrial applications, which forms chitooligosaccharides (COS) as reaction products that are known to have various biological activities. In this study, the gene csnS was cloned from a deep-sea bacterium Serratia sp. QD07, as well as over-expressed in Escherichia coli, which is a new chitosanase encoding gene. The recombinant strain was cultured in a 5 L fermenter, which yielded 324 U/mL chitosanases. After purification, CsnS is a cold-adapted enzyme with the highest activity at 60°C, showing 37.5% of the maximal activity at 0°C and 42.6% of the maximal activity at 10°C. It exhibited optimum activity at pH 5.8 and was stable at a pH range of 3.4–8.8. Additionally, CsnS exhibited an endo-type cleavage pattern and hydrolyzed chitosan polymers to yield disaccharides and trisaccharides as the primary reaction products. These results make CsnS a potential candidate for the industrial manufacture of COS.

Evaluation of Chito-Oligosaccharide (COS) in Vitro and in Vivo: Permeability Characterization in Caco-2 Cells Monolayer and Pharmacokinetics Properties in Rats

Zhang Pengpeng,Zhang Miaomiao,Dong Kaiyu,Zhang Yicong,Yang Shuang,Wang Yuanhong,Jiang Tingfu,Yu Mingming,Lv Zhihua

Chito-oligosaccharide (COS) had shown a variety of biological activities and potential biomedical implications. The present study investigated the pharmacokinetics, bioavailability, and in vitro absorption of COS with degrees of polymerization (DPs) 2–7 and explored the influence of DPs on them. From Caco-2 cell permeation studies, COS were low permeability compounds with no directional effects, suggesting a low in vivo absorption mediated by facilitation diffusion and paracellular absorption. After an intragastrical administration to rats, COS2 showed the highest systemic exposure in six oligosaccharides. The bioavailability of COS2–7 was 7.33%, 6.11%, 4.67%, 4.13%, 4.02%, 0.99%, respectively. Differences in bioavailability for each COS correlated to structural variations, with high DPs contributing to a decrease in bioavailability. In conclusion, COS could be absorbed by the intestinal tract both in vitro and in vivo . The very low oral bioavailability of COS could be due to low permeability. DPs can affect absorption and bioavailability of COS2–7. This study provided evidence for the absorption characteristics of COS2–7 to help us better understanding the pharmacological actions.

Encapsulation of fucoxanthin in pickering emulsion with improved stability and bioaccessibility for inflammatory bowel disease prevention

Bowen Jiao,Nanting Zhu,Decheng Bi,Lijun Yao,Yan Wu,Beiwei Zhu,Xu Xu

Fucoxanthin (FUC), a lipid-soluble carotenoid with various bioactivities, demonstrates promise as a preventive agent against inflammatory bowel disease (IBD). Nevertheless, the inherent instability and low aqueous solubility of FUC limit its application in functional food formulations and pharmaceutical applications. In this study, a FUC-loaded pickering emulsion (G-SPI-COS/FUC emulsion) with high encapsulation efficiency and small particle size was developed by using genipin-crosslinked soy protein isolate (SPI)-chitooligosaccharide (COS) nanoparticles as stabilizers. G-SPI-COS/FUC emulsion exhibited excellent stability under varying pH values, ionic strengths, and storage conditions. In vitro digestion experiments showed that G-SPI-COS/FUC emulsion controlled FUC release and enhanced its intestinal absorption and bioavailability. Furthermore, G-SPI-COS/FUC emulsion was more effective in preventing dextran sulfate sodium (DSS)-induced colitis than free FUC and FUC-loaded pickering emulsion stabilized by native SPI. These findings indicated that G-SPI-COS/FUC emulsion could serve as a promising delivery system for FUC, providing a novel approach to the prevention of IBD.

Totally-green cellulosic fiber with prominent sustained antibacterial and antiviral properties for potential use in spunlaced non-woven fabric production

Jinxin Lan,Yao Wu,Changmei Lin,Jiazhen Chen,Ruiqi Zhu,Xiaojuan Ma,Shilin Cao

The worldwide spread of COVID-19 has put a higher requirement for personal medical protective clothing, developing protective clothing with sustained antibacterial and antiviral performance is the priority for safe and sustaining application. For this purpose, we develop a novel cellulose based material with sustained antibacterial and antiviral properties. In the proposed method, the chitosan oligosaccharide (COS) was subjected to a guanylation reaction with dicyandiamide in the presence of Scandium (III) triflate; because of the relatively lower molecular weight and water solubility of the COS, GCOS (guanylated chitosan oligosaccharide) with high substitution degree (DS) could be successfully synthetized without acid application. In this instance, the minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) of the GCOS were only 1/8 and 1/4 of that of COS. The introduction of GCOS onto the fiber endowed the fiber with extremely high antibacterial and antiviral performance, showing 100% bacteriostatic rate against Staphylococcus aureus and Escherichia coli and 99.48% virus load reduction of bacteriophage MS2. More importantly, the GCOS modified cellulosic fibers (GCOS-CFs) exhibit excellent sustained antibacterial and antiviral properties; namely, 30 washing cycles had negligible effect on the bacteriostatic rate (100%) and inhibition rate of bacteriophage MS2 (99.0%). Moreover, the paper prepared from the GCOS-CFs still exhibited prominent antibacterial and antiviral activity; inferring that the sheeting forming, press, and drying process have almost no effect on the antibacterial and antiviral performances. The insensitive of antibacterial and antiviral activity to water washing (spunlace) and heat (drying) make the GCOS-CFs a potential material applicable in the spunlaced non-woven fabric production.

Fabrication of 3D printed trabecular bone-templated scaffolds modified with rare earth europium (III)-based complex for enhancing mitochondrial function in bone regeneration

Xiuhong Huang,Liqin Zheng,Pengfei Li,Ziling Lin,Shaohong Huang,Changren Zhou

Bone defects have been a tough clinical challenge. The natural trabecular tissue reflects a balance between the minimal metabolic cost of maintenance and maximal network stability, with mitochondria being the energy suppliers for osteogenesis. Herein, a 3D printed trabecular bone-templated scaffold composed of polylactide (PLA) modified with europium (III)-organic ligands (polydopamine and chitooligosaccharide, PDA/COS@Eu) was fabricated (denoted as Tra-PLA/PDA/COS@Eu) to augment bone repair by facilitating mitochondrial function. First, the trabecular bone templated-scaffold provides superior mechanical performance and permeability in contrast to a computer aided designed (CAD) scaffold under a same volume fraction. Tra-PLA/PDA/COS@Eu scaffold up regulated PI3K/AKT/S6K/HIF-1α pathway and significantly promoted BMSCs proliferation and osteogenic differentiation. In addition, Tra-PLA/PDA/COS@Eu scaffold provided anti-inflammatory environment by promoting macrophages M2 polarization through COS, and significantly stimulated BMSCs osteogenesis with enhanced mitochondrial function and biosynthesis. Remarkably, Eu ions are the key ingredient in enhancing mitochondrial function and biosynthesis in BMSCs. Further studies showed that the above-mentioned facilitations in a rabbit bone defect model were found to be more significant with trabecular bone-templated scaffolds, as opposed to CAD scaffolds. In summary, this novel trabecular bone-templated scaffold targeting mitochondrial function has the potential to become an effective method in the treatment of large bone defects.

An Injectable Hydrogel Co-Delivering Propranolol and Imiquimod to Synergistically Enhance Immunogenic Cell Death after Melanoma Resection

Yunfeng Tang,Xiangtian Deng,Min Yi,Dong Wang,Cheng Zheng,Shichang Zhao,Renliang Zhao

Melanoma remains one of the most aggressive skin cancers with high recurrence rates following surgical resection. To overcome the challenges of residual tumor cells and postoperative immune resistance, an injectable hydrogel is developed that co-delivers propranolol (Pro) and imiquimod (IMQ). The rationale is to achieve a synergistic therapeutic effect by enhancing immunogenic cell death (ICD) and strengthening antitumor immunity. The hydrogel is synthesized via boronate ester crosslinking between HA-EGCG and CMCS-PBA and demonstrates excellent injectability, self-healing, biodegradability, and tissue adhesion. In vitro, Pro potentiated IMQ-induced reactive oxygen species (ROS) production, enhancing ICD markers such as calreticulin exposure (CRT), high mobility group box 1 (HMGB1), and adenosine triphosphate (ATP) release, thereby promoting dendritic cell maturation and T cell activation. In vivo, Pro/IMQ@ Hydrogel (PIH) effectively inhibited tumor recurrence in an incomplete melanoma resection model, significantly reducing tumor volume and inducing apoptosis without systemic toxicity. Immunohistochemical analyses revealed increased CD8⁺ and CD4⁺ T cell infiltration, elevated inflammatory cytokine levels, and enhanced memory T cell responses. This work highlights the potential of PIH as a potent localized chemo-immunotherapy platform for melanoma by converting “cold” tumors into “hot” immunogenic lesions.

Mechanisms of heat-activated soy protein isolate-chitosan oligosaccharide complexation: Synergistic interactions for enhanced gelation

Wei Wan,Rui Zhang,Lingyi Gao,Aijun Jian,Xianbing Xu,Xiaokang Na,Ming Du,Jianyu Zhu,Beiwei Zhu,Chao Wu

The strategic utilization of natural polysaccharides presents a critical approach for enhancing the gelation properties of soy protein isolate (SPI), which is a key factor in developing superior plant-based food textures. This study demonstrates a strategic technique to enhance SPI gel properties via preheating treatment and electrostatic complexation with chitosan oligosaccharide (COS). Structural analyses revealed that preheating and electrostatic complexation induced SPI unfolding, facilitating conformational transitions toward β-sheet (increased from 53.00 % to 65.35 %) and β-turn (increased from 5.10 % to 7.60 %) configurations while exposing buried tryptophan residues. Fourier-transform infrared spectroscopy, surface hydrophobicity, measurement of interaction force, and molecular docking confirmed that SPI-COS complexation was driven by synergistic electrostatic attractions, hydrogen bonds, van der Waals forces, and hydrophobic interactions. Increasing COS concentration (0–0.16 %, w / v ) progressively enlarged complex particle size (100–350 nm) but reduced colloidal stability, as evidenced by turbidity, ζ-potential, and cryo-scanning electron microscopy. Critically, COS incorporation at 0.08–0.16 % ( w / v ) significantly elevated storage modulus (increased by 275 %) and viscosity while optimizing the regulation of bound and immobilized water in a concentration-dependent manner. These findings establish that COS-mediated electrostatic complexation restructures SPI networks into mechanically robust gels, providing a green strategy for next-generation plant-based foods.

Preparation of high-strength, colored and multifunctional chitosan fibers via a novel strategy of enzyme-catalyzed phenolic-amine crosslinking within chitosan fibers in a swollen state

Zhichao Song,Shujie Xu,Bo Xu,Man Zhou,Ying Liu,Ying Zhang,Li Cui,Ping Wang,Qiang Wang,Yuanyuan Yu

This study prepared high-strength, colored and multifunctional polyphenol-chitosan fibers via a novel strategy of laccase-mediated phenolic-amine crosslinking within chitosan fibers in a swollen state. A dense covalent network within the fiber was formed through Michael addition and Schiff base reactions. Compared to unmodified chitosan fibers, the enzymatically cross-linked fibers exhibited significantly enhanced mechanical properties, with an increase of about 270% in dry tensile strength (up to 156.8 MPa) and excellent wet stability. In addition, the polyphenol-chitosan fibers were given color and different properties by the covalent cross-linking of polyphenols into the fiber. The polyphenol-chitosan fibers had stable colors and did not fade in the environment. They also had an exceptional antibacterial rate of 99.99% and excellent UV protection. This laccase-mediated phenolic-amine crosslinking strategy provides an effective, sustainable method for producing high-performance, bio-based functional fibers.

Phycocyanin–chitooligosaccharide interactions drive the formation of blue soft wheat starch gels with modified digestibility: Linking multiscale structure to functional performance

Wei Liang,Xuhui Huang,Marat Muratkhan,Chengrong Wen,Lei Qin

Designing starch-based soft gels with visual functionality and modulated digestibility is important for developing texture-modified foods. In this study, blue wheat starch composite gels were constructed by incorporating phycocyanin (Phy, 5% and 10%) and chitooligosaccharide (COS, 2.5% and 5%), and their formation, multiscale structure, physicochemical properties, and in vitro digestibility were systematically investigated. Phy imparted a blue to blue-green appearance to the gels, while COS further modulated their color characteristics. Phy and COS reduced pasting viscosity and viscosity rebuilding and promoted the transition from a dense, starch-dominated gel network toward a looser and more heterogeneous composite structure. Structural and thermal analyses revealed reduced short-range order, relative crystallinity, and gelatinization enthalpy, accompanied by weakened viscoelasticity and lower textural strength. The incorporation of Phy and COS also redistributed the digestible starch fractions, with increased RDS and decreased SDS and RS observed in several composite formulations. Molecular docking suggested that starch-derived glucan chains and COS could potentially recognize partially overlapping regions on the Phy surface, providing a possible molecular basis for local component association and subsequent structural reorganization. Overall, these findings demonstrate that Phy and COS regulate wheat starch gel properties through formulation-dependent structural reorganization, providing a basis for designing visually distinctive and soft-textured starch-based composite gels with modulated digestion profile.

Bioinspired Ion-Proton Bipathway All-Natural Hydrogel With Enhanced Conductivity for Bioelectronic Interfaces

Lijun Lu,Xingwei Chen,Yujie Zhang,Jing Liu,Chang Liu,Yun Hu,Yanchao Mao

Sustainably derived all-natural conductive hydrogels offer a new class of soft materials with intrinsic biocompatibility, biodegradability, and tunable mechanical properties, showing great promise for flexible bioelectronic interfaces. However, their practical application remains limited by inherently low electrical conductivity. Ion channels in the human nervous system, composed of proteins and oligosaccharides, can reduce the effective radius of hydrated ions by inducing partial dehydration of the hydration shell, thereby enabling highly efficient and ordered ion-proton cooperative transport. Inspired by this ability, an all-natural highly conductive ion-proton bipathway hydrogel (IPBH) composed of bovine serum albumin (BSA), chitosan oligosaccharide (COS), and Ca 2 + was developed to mimic natural ion transport environments and significantly enhance the conductivity. The resulting IPBH achieves a remarkable conductivity of 40.59 mS/cm, which is 1272% and 384% higher than that of the proton-only and ion-only conductive hydrogels, respectively. Besides, the IPBH exhibits complete biodegradability within 36 h and exceptional biocompatibility. Moreover, the IPBH exhibits excellent low-noise performance in physiological signal monitoring, enabling accurate EMG-based gesture recognition integrated with a convolutional neural network (CNN) model for real-time robotic arm control. This synergistically conductive, ion-proton coupled hydrogel provides a versatile and sustainable platform for next-generation high-performance bioelectronic interfaces.

Preparation and mechanism study of Al2O3-CeO2 mixed abrasive slurry for A-plane sapphire chemical mechanical polishing

Kui Ding,Anqi Sheng,Jing Song,Shuyan Song,Hongjie Zhang,Lang Zhao

To address the low removal rate, severe surface damage, and unclear mechanism of A-plane sapphire during chemical mechanical polishing, a novel Al 2 O 3 -CeO 2 mixed-abrasive slurry was developed by integrating the mechanical abrasion of Al 2 O 3 with the chemical reactivity of CeO 2 . The optimal formulation, containing 2 wt% Al 2 O 3 , 0.2 wt% CeO 2 , 0.15 wt% sodium hexametaphosphate, 0.7 wt% isooctanol polyoxyethylene ether, and 0.5 wt% sodium D-gluconate with aminomethyl propanol as pH regulator, yields an atomically smooth surface (Ra = 0.079 nm) and a high removal rate (MRR = 905 nm/h). XPS analysis reveals that a hydrated Al-OH layer and a CeAl x O y soft phase form on the sapphire via interfacial solid-state reaction, and are removed by mechanical shearing of the mixed abrasives.

Structural dichotomy in soy protein isolate-chitosan oligosaccharide gels: unraveling the distinct roles of 7S and 11S globulins

Wei Wan,Xiaokang Na,Ming Du,Jianyu Zhu,Beiwei Zhu,Chao Wu

7S and 11S globulins exhibit distinct binding modes with chitosan oligosaccharide. • The 7S/11S ratio determines the gelation pathway and network architecture. • 11S-rich systems form rigid, covalently cross-linked hydrogels. • Globulin composition modulates water mobility and mechanical properties. Soy protein isolate (SPI) is widely used in food applications, but the natural variation in its 7S/11S globulin ratio leads to inconsistent gel properties. In this study, we investigated how the 7S/11S ratio influences the structural and rheological properties of composite hydrogels formed with chitosan oligosaccharide (COS). Spectroscopic analyses revealed that 7S-rich systems maintain a more flexible conformation with higher α-helix content (12.7%), whereas 11S-rich systems adopt a rigid, β-sheet-rich structure with β-sheet content increasing to 49.6% (compared to 34.7% in 7S-rich systems). Rheological measurements showed that increasing the 11S fraction progressively enhances the storage modulus, which reaches approximately five times that of 7S-rich gels, transforming the gel network from physically entangled to covalently cross-linked. Microstructural and water mobility analyses confirmed that 11S-rich gels form dense, compact structures that restrict water mobility, with transverse relaxation time decreasing from 238 ms to 180 ms, whereas 7S-rich gels exhibit looser structures. These findings demonstrate that the 7S/11S ratio is a critical determinant of the macroscopic properties of SPI-COS composite gels, providing a practical strategy for tailoring plant protein gel textures.
