Discovery of Arbutin as Novel Potential Antiviral Agent Against Tomato Yellow Leaf Curl Virus

Ying Xu,Tingting Zhang,Shimei Mu,Yanqun Peng,Duanpu Wu,Li Yang,Qing Li,Zengxue Wu,Jian Zhang

Tomato yellow leaf curl virus (TYLCV), a major plant virus, infects multiple plant species, severely threatening global food security. Arbutin, a natural product used in cosmetics to reduce pigmentation, also exhibits antibacterial and anti-inflammatory properties. However, its potential in plant protection remains undocumented. This study reveals arbutin’s ability to inhibit TYLCV infection. In Nicotiana benthamiana, 100 μg/mL arbutin inhibited viral gene accumulation by up to 76.8%, surpassing ningnanmycin (65.8%) and ribavirin (39.5%). Besides, microscale thermophoresis indicated that arbutin bound strongly to the TYLCV coat protein (CP). Molecular docking indicated that arbutin interacted with ARG58, VAL65, and CYS69. RT-qPCR and Western blot experiments confirmed the crucial roles of these amino acids, especially VAL65, in viral infection. Transcriptome analysis revealed that mutating VAL65 affected plant–pathogen interaction pathways and MAPK signaling in host defense mechanisms. This study unveils arbutin’s novel antiviral function, providing crucial insights for developing new biopesticides against plant viruses.

PABPC4 Inhibits SADS-CoV Replication by Degrading the Nucleocapsid Protein Through Selective Autophagy

Chenchen Zhao,Yan Qin,Haixin Huang,Wei Chen,Yanqing Hu,Xinyu Zhang,Yuying Li,Tian Lan,Wenchao Sun

Simple SummarySADS-CoV is a newly emerging coronavirus affecting the swine industry worldwide. However, our understanding of its pathogenic pathways remains limited. In this study, the researchers aimed to determine the role of the host factor, poly(A)-binding protein 4 (PABPC4), in SADS-CoV infection. This study found that PABPC4 inhibits SADS-CoV replication by degrading the nucleocapsid protein through selective autophagy. This discovery could potentially contribute to developing novel antiviral strategies to prevent and control SADS-CoV infections, protecting the pig industry from significant economic losses.Swine acute diarrhea syndrome coronavirus (SADS-CoV) is a novel enteric coronavirus that causes severe clinical diarrhea and intestinal pathological injury in pigs. Selective autophagy is an important mechanism of host defense against virus invasion. However, the mechanism through which SADS-CoV-mediated selective autophagy mediates the innate immune response remains unknown. Here, we report that the host protein PABPC4 can inhibit SADS-CoV replication through targeting and degrading its N protein. Furthermore, we demonstrate that PABPC4 recruits MARCHF8 (an E3 ubiquitin ligase), which ubiquitinates the N protein and is degraded via NDP52/CALCOCO2 (a selective autophagy cargo receptor). Taken together, these findings reveal a new mechanism by which PABPC4 inhibits virus replication, and reveal a new target for antiviral drug development.

Aptamer-Based Activatable Tyramide Signal Amplification for Low-Background Detection of SARS-CoV-2 Nucleocapsid Protein

Zhiyong Huang,Ziyan Du,Juan Li,Da Han,Jiaxuan He,Yunben Yang,Dan Wang,Yu Liang,Yunshan Yang,Ruizi Peng,Weihong Tan

As severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection posed a significant threat to public health and the global economy, in vitro diagnosis of the SARS-CoV-2 nucleocapsid protein proved to be an effective way for SARS-CoV-2 infection control in the past years. Tyramide signal amplification (TSA) has been extensively utilized in tissue imaging and pathological diagnosis owing to the powerful signal enhancement. However, the elevated “ALWAYS ON” fluorescence background limited the accuracy and sensitivity of the conventional TSA assay. To achieve an activated “TURN ON” signal, herein, a small molecule, termed dichlorodihydrofluorescein tyramide (T-DCFH), was synthesized for activatable TSA. Under the catalysis of horseradish peroxidase (HRP) with hydrogen peroxide (H2O2), this T-DCFH facilitates the “TURN ON” fluorescence signal. Additionally, as a recognition tool, DNA aptamer has been used for developing in vitro diagnostic approaches. Hence, based on HRP-labeled aptamers binding with SARS-CoV-2 nucleocapsid protein, we achieved aptamers-based activatable TSA detection with a higher signal-to-noise ratio than that of fluorescent dye (FITC)-labeled aptamers, while showing lower background than traditional fluorescein tyramide with “ALWAYS ON”. The results demonstrated that the activated T-DCFH significantly enhances the fluorescence signal while diminishing the background noise. By employing multiple aptamers targeting, we offered a timely and accurate in vitro diagnostic approach for future emergent infectious diseases.

Dual-Mode Lateral Flow Immunoassay Based on “Pompon Mum”-Like Fe3O4@MoS2@Pt Nanotags for Sensitive Detection of Viral Pathogens

Meimei Xu,Shuai Zhao,Chenglong Lin,Yanyan Li,Weida Zhang,Yusi Peng,Rui Xiao,Zhengren Huang,Yong Yang

Lateral flow immunoassay (LFIA) has been widely used for the early diagnosis of diseases. However, conventional colorimetric LFIA possesses limited sensitivity, and the single-mode readout signal is easily affected by the external environment, leading to insufficient accuracy. Herein, multifunctional Fe3O4@MoS2@Pt nanotags with a unique “pompon mum”-like structure were triumphantly prepared, exhibiting excellent peroxidase (POD)-like activity, photothermal properties, and magnetic separation capability. Furthermore, the Fe3O4@MoS2@Pt nanotags were used to establish dual-mode LFIA (dLFIA) for the first time, enabling the catalytic colorimetric and photothermal dual-mode detection of severe acute respiratory syndrome coronavirus 2 nucleocapsid protein (SARS-CoV-2 NP) and influenza A (H1N1). The calculated limits of detection (cLODs) of SARS-CoV-2 NP and H1N1 were 80 and 20 ng/mL in catalytic colorimetric mode and 10 and 8 ng/mL in photothermal mode, respectively, demonstrating about 100 times more sensitive than the commercial colloidal Au-LFIA strips (1 ng/mL for SARS-CoV-2 NP; 1 μg/mL for H1N1). The recovery rates of dLFIA in simulated nose swab samples were 95.2–103.8% with a coefficient of variance of 2.3–10.1%. These results indicated that the proposed dLFIA platform showed great potential for the rapid diagnosis of respiratory viruses.

Enhanced targeting: Production of filamentous bacteriophage monoclonal antibodies using the major coat protein pVIII as anchor

Qing Li,Jianyu Zhu,Ting Yuan,Leina Dou,Xuezhi Yu,Jianzhong Shen,Huijuan Yang,Zhanhui Wang

Filamentous bacteriophage display technology has been employed in antibody discovery, drug screening, and protein–protein interaction study across various fields, including food safety, agricultural pollution, and environmental monitoring. Antifilamentous bacteriophage antibodies for identifying filamentous bacteriophage are playing a pivotal role in this technology. However, the existing antifilamentous bacteriophage antibodies lack sensitivity and specificity, and the antibodies preparation methods are cumbersome and hyposensitive. The major coat protein pVIII of filamentous bacteriophage has an advantage in quantification, which is benefit for detecting signal amplification but its full potential remains underutilized. In this study, the partial polypeptide CT21 of the major coat protein pVIII of filamentous bacteriophage was intercepted as the targeted immunogen or coating antigen to prepare antifilamentous bacteriophage antibodies. Six filamentous bacteriophage-specific monoclonal antibodies (mAbs) M5G8, M9A2, P6B5, P6D2, P8E4, and P10D4 were obtained. The limit of detections of the prepared six mAbs for detecting filamentous bacteriophage was 1.0 × 10 7 pfu mL −1 . These mAbs stayed stable under different pH, temperature, and exhibited high specificity in real application. This study not only provides a new idea for simplifying the preparation of antifilamentous bacteriophage antibodies which could apply in filamentous bacteriophage display, but it also presents a novel strategy for preparing antibodies against protein-specific epitopes with high sensitivity.

Smartphone-Based Electrochemical Immunoassay for Point-of-Care Detection of SARS-CoV-2 Nucleocapsid Protein

Ruijin Zeng,Minghao Qiu,Qing Wan,Zhisheng Huang,Xiaolong Liu,Dianping Tang,Dietmar Knopp

Large-scale, rapid, and inexpensive serological diagnoses of severe acute respiratory syndrome-coronavirus-2 (SARS-CoV-2) are of great interest in reducing virus transmission at the population level; however, their development is greatly plagued by the lack of available point-of-care methods, leading to low detection efficiency. Herein, an ultrasensitive smartphone-based electrochemical immunoassay is reported for rapid (less than 5 min), low-cost, easy-to-implement detection of the SARS-CoV-2 nucleocapsid protein (SARS-CoV-2 N protein). Specifically, the electrochemical immunoassay was fabricated on a screen-printed carbon electrode coated with electrodeposited gold nanoparticles, followed by incubation of anti-N antibody (Ab) and bovine serum albumin as the working electrode. Accompanied by the antigen–antibody reaction between the SARS-CoV-2 N protein and the Ab, the electron transfer between the electroactive species [Fe(CN)6]3–/4– and the electrode surface is disturbed, resulting in reduced square-wave voltammetry currents at 0.075 V versus the Ag/AgCl reference electrode. The proposed immunoassay provided a good linear range with SARS-CoV-2 N protein concentrations within the scope of 0.01–1000 ng/mL (R2 = 0.9992) and the limit of detection down to 2.6 pg/mL. Moreover, the detection data are wirelessly transmitted to the interface of the smartphone, and the corresponding SARS-CoV-2 N protein concentration value is calculated and displayed. Therefore, the proposed portable detection mode offers great potential for self-differential diagnosis of residents, which will greatly facilitate the effective control and large-scale screening of virus transmission in resource-limited areas.

Synthesis of novel silica-encapsulated core-satellite nanotags for enhanced SERS detection

Haoquan Wang,Qi Yang,Yan Zhang,Bo Peng,Feng Wu,Zewei Quan

Developing a SERS nanotag with high signal intensity, excellent signal stability, and biocompatibility is of significant importance in the fields of food safety, drug testing, and virus detection. In this work, silica-encapsulated, indocyanine green (ICG)-modified Au octahedral core-small Au spherical nanoparticles (NPs) satellite nanostructures (Au/Au@ICG@SiO 2 ) are uniformly synthesized as surface-enhanced Raman scattering (SERS) tags, which provide abundant “hot spots” and also increase the stability of Raman reporter molecule ICG to greatly facilitate subsequent functionalization and detection. Both experimental and numerical results demonstrate that this Au/Au@ICG nanostructure achieve a detection limit of 10⁻⁷ M for ICG in aqueous solution ascribed to the increased number of hot spots compared with Au octahedral NPs. As-prepared Au/Au@ICG@SiO 2 SERS nanotags are further integrated with a lateral flow immunoassay (LFIA) for the detection of the SARS-CoV-2 nucleocapsid protein. Compared to the traditional bio-recognition method on LFIA strips with the detection limit of 8 pg/mL, the detection capability for the SARS-CoV-2 nucleocapsid protein is improved approximately 16 times (500 fg/mL) based on this novel SERS tag and custom-built adaptive Raman mapping spectrometer. This Au/Au@ICG@SiO 2 SERS tag is expected to play a significant role in early detection, prevention, and control of similar infectious viruses in the future.

Sialic acid serves as a functional receptor for grass carp reovirus

Qian Wang,Zichao Peng,Bin Gui,Yongming Li,Lanjie Liao,Zuoyan Zhu,Yaping Wang,Libo He

Grass carp reovirus (GCRV) causes hemorrhagic disease and substantial economic losses in the aquaculture of grass carp ( Ctenopharyngodon idella ), a commercially important fish species in China. Although viral entry depends on interactions between viral proteins and host receptors, the specific host molecules mediating this process have not been fully elucidated. Here, we identify cell surface sialic acid (SA) as a critical functional receptor for GCRV. Enzymatic removal of SA markedly impaired viral attachment and infection. Competitive inhibition using SA-binding lectins or soluble SA confirmed that GCRV targets SA moieties on host cells. Genetic knockdown of SA biosynthesis attenuated viral binding and replication, whereas overexpression of SA pathway genes enhanced susceptibility. Surface plasmon resonance demonstrated direct binding between GCRV capsid proteins and soluble SA, and mutational analysis identified key amino acid residues involved. Notably, pretreatment of GCRV with soluble SA significantly improved fish survival and reduced virus-induced immune overactivation in vivo . To assess receptor specificity, parallel experiments using Rana grylio virus (RGV), a phylogenetically unrelated ranavirus , showed that RGV infection was unaffected by SA-targeted interventions, highlighting the specificity of SA utilization by GCRV. Together, these findings identify SA as a functional and specific receptor for GCRV, offering new insights into virus-host interactions and potential antiviral strategies in aquaculture. Grass carp hemorrhagic disease, caused by grass carp reovirus (GCRV), poses a major threat to freshwater aquaculture in China. Despite extensive studies on GCRV, the identity of host cell receptors mediating viral entry remained unknown. In this study, we demonstrate that GCRV directly binds to sialic acids-sugar molecules commonly found on cell surfaces-to attach to and infect host cells. By using a combination of enzymatic removal, lectin blocking, genetic manipulation, and biochemical binding assays, we show that sialic acid is essential for GCRV attachment and infection. Furthermore, pre-incubating GCRV with soluble sialic acids significantly reduced infection in cultured cells and improved the survival of infected grass carp. These findings reveal a key mechanism of viral entry and suggest new strategies for disease prevention and selective breeding of resistant fish. Our study not only deepens understanding of aquareovirus biology but also provides practical insights for controlling viral outbreaks in aquaculture.

Electrochemiluminescence Lateral Flow Immunoassay Using Ruthenium(II) Complex-Loaded Dendritic Mesoporous Silica Nanospheres for Highly Sensitive and Quantitative Detection of SARS-CoV-2 Nucleocapsid Protein

Wenxuan Fu,Xuexue Wang,Xudong Ying,Tao Sun,Yafeng Wang,Jing Wang,Bin Su

Lateral flow immunoassays (LFIA) are widely used for the cost-effective and rapid detection of diverse analytes. However, traditional LFIA suffers from difficulties in providing quantitative results and has low sensitivity. Herein, LFIA is combined with electrochemiluminescence (ECL), a leading transduction technique with high sensitivity and wide dynamic range, to achieve highly sensitive and quantitative detection of severe acute respiratory syndrome coronavirus nucleocapsid protein (SARS-CoV-2 N protein). Ruthenium(II)-based complexes are synthesized and loaded into dendritic mesoporous silica nanospheres (PEI-Ru/dSiO 2 ), which possessed central-radial pore channels and served as tags for ECL-LFIA. The electrodes are fabricated on a nitrocellulose (NC) membrane, which simplifies the structure of the ECL-LFIA. PEI-Ru/dSiO 2 is captured on the electrode surface via a sandwich immunoreaction, which enhances the ECL signal by decreasing the distance between PEI-Ru/dSiO 2 and the electrode surface. Using 2,2-bis(hydroxymethyl)-2,2′,2′'-nitrilotriethanol (BIS-TRIS) as coreactant, the ECL-LFIA is used for detecting SARS-CoV-2 N protein, with a linear range of 1–10 4 ng mL −1 and a limit of detection (LOD) of 0.52 ng mL −1 . ECL-LFIA can also be used to detect analytes in complex matrices. These results demonstrate that the prepared ECL-LFIA has great potential as a point-of-care testing platform for the rapid and quantitative detection of disease biomarkers.

Aptamers targeting SARS-CoV-2 nucleocapsid protein exhibit potential anti pan-coronavirus activity

Yang Minghui,Li Chunhui,Ye Guoguo,Shen Chenguang,Shi Huiping,Zhong Liping,Tian Yuxin,Zhao Mengyuan,Wu Pengfei,Hussain Abid,Zhang Tian,Yang Haiyin,Yang Jun,Weng Yuhua,Liu Xinyue,Wang Zhimin,Gan Lu,Zhang Qianyu,Liu Yingxia,Yang Ge,Huang Yuanyu,Zhao Yongxiang

Emerging and recurrent infectious diseases caused by human coronaviruses (HCoVs) continue to pose a significant threat to global public health security. In light of this ongoing threat, the development of a broad-spectrum drug to combat HCoVs is an urgently priority. Herein, we report a series of anti-pan-coronavirus ssDNA aptamers screened using Systematic Evolution of Ligands by Exponential Enrichment (SELEX). These aptamers have nanomolar affinity with the nucleocapsid protein (NP) of Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) and also show excellent binding efficiency to the N proteins of both SARS, MERS, HCoV-OC43 and -NL63 with affinity K D values of 1.31 to 135.36 nM. Such aptamer-based therapeutics exhibited potent antiviral activity against both the authentic SARS-CoV-2 prototype strain and the Omicron variant (BA.5) with EC 50 values at 2.00 nM and 41.08 nM, respectively. The protein docking analysis also evidenced that these aptamers exhibit strong affinities for N proteins of pan-coronavirus and other HCoVs (−229E and -HKU1). In conclusion, we have identified six aptamers with a high pan-coronavirus antiviral activity, which could potentially serve as an effective strategy for preventing infections by unknown coronaviruses and addressing the ongoing global health threat.

Ningnanmycin Activates Defense Systems against Potato Virus Y in Nicotiana benthamiana

Zilin Wu,Min Huang,Junmei Jiang,Chun Zhang,Gefei Hao,Moxian Chen,Qing X. Li,Mengao Jia,Junfeng Liu,Xiangyang Li

Ningnanmycin has been proven to effectively inhibit infection by potato virus Y (PVY), although its underlying mechanisms remain unclear. In this study, we report a novel finding that ningnanmycin affects PVY replication. Two approaches were employed: studies using PVY movement-deficient mutants suggest that ningnanmycin, at a concentration of 500 μg/mL, inhibits PVY replication. Ningnanmycin interacts with the PVY-encoded coat protein (CP) with a binding constant of 1.34 μmol/L, and key amino acids Glu168 and Thr206 are involved in this interaction. Additionally, ningnanmycin induces the expression of antiviral response genes in Nicotiana benthamiana, including PRXIIB, PRXIIE, PUB4, and PER42. Furthermore, studies revealed that the overexpression of PUB4 in N. benthamiana confers resistance to PVY infection. These findings highlight the mechanisms by which ningnanmycin activates the PUB4 gene and suppresses CP assembly, thereby inhibiting PVY in N. benthamiana. This study represents an important step toward elucidating the molecular mechanism underlying the antiviral activity of ningnanmycin.

Metal–Organic Framework Sub-Nanochannels within the Confined Micropipettes: Precise Construction Makes It a Universal Aptamer-Based Sensing Platform

Haoran Tang,Shuai Zhang,Binbin Yang,Xia Qiu,Hao Wang,Yongxin Li

It is crucial to precisely construct metal–organic framework (MOF) sub-nanochannels at the tip of micro/nanopipettes for fundamental research and sensing applications. The quality of the MOF modification plays a significant role in influencing subsequent research, particularly in sensing applications. In this work, we present a precise method of constructing MOF sub-nanochannels at the tip of glass micropipettes, which serve as a universal aptamer-based sensing platform for the selective detection of proteins. In situ scanning electron microscopy (SEM), energy-dispersive spectroscopy (EDS) mapping, and fluorescence microscopy results demonstrate that the synthesized MOF (UiO-66) nanocrystals fully block the orifice of glass micropipettes (UiO-66-GMs) without forming any nanometer-scale cracks and remain confined within the geometric boundaries of the orifice. The terminal phosphate-modified aptamer readily binds to the surface of UiO-66-GMs through metal (Zr)–phosphate coordination, ultimately forming the aptamer sensor (Apt-UiO-66-GMs). The selective quantification of proteins is achieved via a decrease in current resulting from protein binding to the aptamer. Our results indicate that the precisely constructed Apt-UiO-66-GMs sensor enables highly selective and sensitive detection of SARS-CoV-2 nucleocapsid protein and holds potential for real sample detection. Furthermore, given the sharp tip of the micropipets and the external sensing interface we have constructed, our aptamer-based sensing platform also opens avenues for single-cell analysis and in vivo sensing.
