Cerium-Organic Framework and Resveratrol Composite Hydrogel Scaffold with Dual Antioxidant Activity for Enhanced Bone Regeneration

Yao Lu,Lan Zhang,Bin Yang,Hansheng Hu,Hao Sun,Dan Guo

The repair of large bone defects remains a significant challenge in the field of orthopedics, as traditional bone substitutes face issues such as limited supply and structural incompatibility. Therefore, the development of novel biomaterials to promote bone repair is of great significance. This study proposes a composite material based on a gelatin/alginate dual-network hydrogel scaffold (Gel/AlgMA), which regulates oxidative stress in the local microenvironment of bone defects by loading cerium metal–organic frameworks (Ce-UiO-66) and resveratrol (Res). The cerium ions in Ce-UiO-66 exhibit excellent antioxidant properties in their multivalent states, capable of scavenging excess reactive oxygen species (ROS), improving mitochondrial function, and enhancing stability through a polydopamine (PDA) coating. The Gel/Alg@Ce-Res/PDA composite scaffold was prepared via photopolymerization and its in vitro biocompatibility, antioxidant properties, and osteogenic potential were evaluated. The results indicated that the composite scaffold effectively scavenged ROS, alleviated oxidative stress, and promoted the proliferation and differentiation of osteoblasts. Moreover, in vivo experiments further confirmed the favorable effects of the Gel/Alg@Ce-Res/PDA scaffold in bone defect repair. This study provides an innovative biomaterial design for bone defect treatment, with promising clinical application prospects.

Preparation and Properties of Partial-Degradable ZrO2–Chitosan Particles–GelMA Composite Scaffolds

Yang Ji,Mengdie Hou,Jin Zhang,Meiqi Jin,Tianlin Wang,Huazhe Yang,Xiaodong Zhang

In the field of bone repair, the inorganic–organic composite scaffold is a promising strategy for mimicking the compositions of the natural bone. In addition, as implants for repairing load-bearing sites, an inert permanent bone substitute composites with bioactive degradable ingredients may make full use of the composite scaffold. Herein, the porous zirconia (ZrO2) matrix was prepared via the template replication method, and the partial degradable ZrO2–chitosan particles–GelMA composite scaffolds with different chitosan/GelMA volume ratios were prepared through the vacuum infiltration method. Dynamic light scattering (DLS) and the scanning electron microscope (SEM) were adopted to observe the size of the chitosan particles and the morphologies of the composites scaffold. The mechanical properties, swelling properties, and degradation properties of the composite scaffolds were also characterized by the mechanical properties testing machine and immersion tests. The CCK-8 assay was adopted to test the biocompatibility of the composite scaffold preliminarily. The results show that chitosan particles as small as 60 nm were obtained. In addition, the ratio of chitosan/GelMA can influence the mechanical properties and the swelling and degradation behaviors of the composites scaffold. Furthermore, improved cell proliferation performance was obtained for the composite scaffolds. Keywords:porous zirconia matrix;chitosan particles;GelMA;composite scaffold

Preparation and in vitro evaluation of polyurethane composite scaffolds based on glycerol esterified castor oil and hydroxyapatite

J. J. Du,Y. Zuo,Q. Zou,B. Sun,M. B. Zhou,L. M. Li,Y. Man,Y. B. Li

Sponge-like polyurethane (PU) and composite have recently attracted attention in biomedical field as scaffolds for bone repair or tissue engineering research. In this study, castor oil was transesterified with glycerol to increase its functional hydroxyl groups and improve the mechanical properties of the resulting composite scaffolds. The glyceride of castor oil (GCO) was used to copolymerise with isophorone diisocyanate to generate PU, meanwhile blending with hydroxyapatite (HA) powder to prepare porous HA/PU composite scaffolds. The HA/glyceride based PU (GCPU) scaffolds were compared with pristine castor oil (PCO) based PU scaffolds (PCPU) by investigating the physicochemical properties and cytocompatibility using Fourier transform infrared, X-ray diffraction, SEM, mechanical testing and methylthiazolyldiphenyl-tetrazolium bromide assay. The results showed that the number of hydroxyl groups of GCO was nearly twice as high as that of PCO, which enhanced efficiently the crosslinking density of PU matrix. The compressive strength of HA/GCPU scaffold with 40 wt-%HA was ∼4·6 MPa, much higher than that (0·058 MPa) of HA/PCPU scaffolds. The in vitro biological results demonstrated that both GCPU and HA/GCPU scaffolds exhibited desirable cytocompatibility. The novel synthetic route is highly effective for the preparation of HA/GCPU porous scaffolds, which have good prospect in the repair of bone defects or for bone tissue engineering.

Preparation and Properties of 3D-Printed Hydrogel-Embedded PLA Composite Scaffolds

Zhiliang Huang,Jinming Qian,Qingyuan Li,Lijuan Zhu,Shanshan Nie,Qianqian Ju,Tingguo Liu

This study presents the development of a biodegradable composite scaffold designed to address the limitations associated with conventional bone repair materials. 3D-printing filaments were developed using fish scale powder (FSP) or fish scale-derived hydroxyapatite (HAP), polylactic acid (PLA), and polycaprolactone (PCL), with a focus on the optimal effect of 5 wt% HAP on the mechanical properties of PLA. Composite scaffolds were further prepared by integrating 3D-printed grid scaffolds with chitosan/gelatin composite hydrogels through vacuum freeze-drying. The properties of the composite scaffolds were evaluated through mechanical property tests, thermal analysis (including DSC and TGA), absorption rate assessments (conducted via immersion experiments in various solutions), and surface morphology analyses (utilizing SEM and EDS). The results show that the addition of HAP significantly improves the mechanical properties of 3D-printed filaments. The high absorption rate exhibited by the HAP/PLA composite scaffold suggests improved biocompatibility and its potential for in vivo tissue fluid absorption. The HAP/PLA composite scaffold showed visible surface mineralization and degradation after 10 days of immersion in simulated body fluid (SBF), highlighting its potential for bone repair and regeneration.

Stepwise 3D-spatio-temporal magnesium cationic niche: Nanocomposite scaffold mediated microenvironment for modulating intramembranous ossification

Jie Shen,Bo Chen,Xinyun Zhai,Wei Qiao,Shuilin Wu,Xuanyong Liu,Ying Zhao,Changshun Ruan,Haobo Pan,Paul K. Chu,Kenneth M.C. Cheung,Kelvin W.K. Yeung

The fate of cells and subsequent bone regeneration is highly correlated with temporospatial coordination of chemical, biological, or physical cues within a local tissue microenvironment. Deeper understanding of how mammalian cells react to local tissue microenvironment is paramount important when designing next generation of biomaterials for tissue engineering. This study aims to investigate that the regulation of magnesium cationic (Mg 2+ ) tissue microenvironment is able to convince early-stage bone regeneration and its mechanism undergoes intramembranous ossification. It was discovered that moderate Mg 2+ content niche (~4.11 mM) led to superior bone regeneration, while Mg 2+ -free and strong Mg 2+ content (~16.44 mM) discouraged cell adhesion, proliferation and osteogenic differentiation, thereby bone formation was rarely found. When magnesium ions diffused into free Mg zone from concentrated zone in late time point, new bone formation on free Mg zone became significant through intramembranous ossification. This study successfully demonstrates that magnesium cationic microenvironment serves as an effective biochemical cue and is able to modulate the process of bony tissue regeneration. The knowledge of how a Mg 2+ cationic microenvironment intertwines with cells and subsequent bone formation gained from this study may provide a new insight to develop the next generation of tissue-repairing biomaterials.

A smart scaffold composed of three-dimensional printing and electrospinning techniques and its application in rat abdominal wall defects

Yang Zhi,Song Zhicheng,Nie Xin,Guo Kaijin,Gu Yan

Background Biological composite scaffolds are increasingly being used in abdominal wall reconstruction but still have certain shortcomings. The present study describes here a novel three-dimensional (3D) scaffold fabricated by combining 3D printing (3DP) and electrospinning (ESP). Methods Biological composite scaffolds are composed of integrated 3DP interconnected macrofiber and random ESP microfiber networks. The 3DP scaffold retains intact 3D architecture and mechanical properties, while the ESP network serves as a cell entrapment system at the extracellular matrix (ECM) scale. Biological composite scaffolds are implanted in a defective rat abdominal wall to detect if it could induce early vascularization and reconstruction of the tissue defect. Results SEM analysis reveals a pore diameter of 424.47 ± 58.49 μm and a porosity of 70.46 ± 2.48% for biological composite scaffolds. In the in vitro test of cell proliferation, biological composite scaffolds do not affect rat dermal fibroblast proliferation in a time- and dose-dependent manner. The animal experiments show tissue remodeling and early angiogenesis as compared to 3DP scaffolds. Conclusions Our experiment prepares a biological scaffold with both a macro- and microscale structure by ESP and 3DP technology. Thus, the integration of 3DP and ESP techniques provides a new set of smart scaffolds for abdominal wall defect and hernia repair.

Preparation and Synergistic Effect of Biomimetic Poly(lactic acid)/Graphene Oxide Composite Scaffolds Loaded with Dual Drugs

Shuqiong Liu,Wanzhu Li,Zhenyi Xu,Jiapeng Hu,Fangfang Wu,Yuying Zheng

To promote the bone repair ability of drug-loaded scaffolds, poly(lactic acid) (PLA)/graphene oxide (GO)/Salvianolic acid B (Sal-B)/aspirin (ASA) dual drug-loaded biomimetic composite scaffolds were prepared. The results showed that the addition of these two drugs delayed the gel formation of the composite system, but a biomimetic nanofiber structure could still be obtained by extending the gel time. The addition of Sal-B increased the hydrophilicity of the scaffold, while an increase in ASA reduced the porosity. Dual drug-loaded scaffolds had good haemocompatibility and synergically promoted the proliferation of MC3T3-E1 cells and enhanced alkaline phosphatase activity. Sustained-release experiments of the two drugs showed that the presence of ASA slowed the cumulative release of Sal-B, while Sal-B promoted the release of ASA. Kinetic modeling showed that the release of both drugs conforms to the Korsmeyer–Peppas model, but Sal-B conforms to the Fick diffusion mechanism and ASA follows Fick diffusion and carrier swelling/dissolution.

Influence of pore architectures of silk fibroin/collagen composite scaffolds on the regeneration of osteochondral defects in vivo

Xue Feng,Peifang Xu,Tao Shen,Yihan Zhang,Juan Ye,Changyou Gao

The regeneration of osteochondral defects faces great challenges because of the limited self-regenerative capabilities of cartilage tissues. In situ inductive regeneration can be realized using bioactive scaffolds combined with endogenous reparative cells. Cell migration could be significantly facilitated by scaffolds with oriented channels. For this purpose, silk fibroin (SF) was composited with collagen (Col) to fabricate extracellular matrix (ECM)-mimetic SF/Col composite scaffolds with random pores, radially aligned pores or axially aligned pores by ice-templated assembly and temperature gradient-guided thermally-induced phase separation. Scanning electron microscopy (SEM) observation confirmed the random and aligned architectures in the respective scaffolds. The three kinds of SF/Col composite scaffolds exhibited a porous structure with a porosity of ∼85%, an appropriate elastic modulus with mechanical anisotropy in the aligned scaffolds, and good biocompatibility. The oriented channels could improve in vivo cell migration and infiltration. During the tissue remodeling processes, the regeneration of osteochondral tissues particularly cartilage was obviously faster in the radially aligned scaffold group than in the other two groups. Nevertheless, satisfactory regeneration was achieved in the two aligned scaffold groups with hyaline cartilage formation at 18 weeks post-surgery, while a hybrid of hyaline cartilage and fibrocartilage was formed in the random scaffold group.

Interface engineering of a 3D carbon nanofiber/iron oxide scaffold for room-temperature ethanol sensing

Chengcheng Xu,Xiaosong Du,Wenjun Huang,Yin Long,Yang Wang

Ethanol is one of the most widely consumed chemicals and has high volatility. Long-term exposure to ethanol can cause irritation and damage to human organs and even lead to cancer. Therefore, reliable, low-cost and low-power ethanol gas sensors are urgently needed. However, due to the high barrier associated with the reaction between adsorbed oxygen and ethanol gas, current commonly used chemiresistive sensors are difficult to detect low concentration of ethanol gas at room temperature. Herein, we developed a 3D composite scaffold sensor, i.e. iron oxide nanoparticles decorated carbon nanofiber scaffolds (CNFs) from both the inside and outside (named Fe 2 O 3 @FCNFs) through interface engineering. The Fe 2 O 3 @FCNFs composite scaffold exhibited a 6-fold increase in specific surface area as compared to that of the FCNFs, and its carrier concentration of was one order higher than that of its counterpart. Therefore, the typical Fe 2 O 3 @FCNFs sensor exhibited an unanticipated good response to ethanol gas at room temperature, including high sensitivity (-6.268 % toward 100 ppm) and low limit of detection (10 ppm). This study provides an effective solution for preparing room-temperature chemiresistive ethanol gas sensors, which can be extended to other gas detection by tuning the semiconductor sensitive materials.

3D-Printed PCL/SrHA@DFO Bone Tissue Engineering Scaffold with Bone Regeneration and Vascularization Function

Kai Chen,Liu Luo,Ruolan Tao,Muzi Li,Shuqi Qu,Xiaofang Wu,Xinyue Zhang,Haiyan Feng,Ziqiang Zhu,Dekun Zhang

The application of a three-dimensional (3D)-printed biological functional scaffold in the repair of bone defects is a promising strategy. In this study, strontium-containing hydroxyapatite (SrHA) powder was synthesized by the hydrothermal method, and then poly(ε-caprolactone) (PCL)/HA and PCL/SrHA composite scaffolds were prepared by the high-temperature melt extrusion 3D printing technology. The basic physical and chemical properties, in vitro biological properties, osteogenesis, and angiogenesis abilities of the scaffold were studied. The results showed that HA and SrHA were uniformly embedded in the composite scaffold, and the scaffold exhibited a 3D interconnected porous structure and rough microsurface. The in vitro release curve showed that Sr2+ and Ca2+ were continuously released from the PCL/SrHA scaffold. In order to verify the performance of the composite scaffold in bone regeneration, the proliferation and osteogenic differentiation of mouse embryonic osteoblasts (MC3T3E1) grown on the scaffold were evaluated. The experimental results showed that the incorporation of SrHA significantly promoted cell proliferation. Compared with the PCL/HA scaffold, the PCL/SrHA scaffold could better promote cell osteogenic differentiation. Deferoxamine (DFO) was loaded on the surface of the PCL/SrHA scaffold. By studying the proliferation, angiogenesis, and expression of osteogenesis and angiogenesis-related genes of human umbilical vein endothelial cells (HUVECs) on PCL/SrHA@DFO scaffold, it was verified that DFO had the ability to promote angiogenesis. It could induce angiogenesis in vitro in combination with Sr2+. Therefore, we believe that the composite scaffold has potential application prospects in the field of bone tissue engineering.

An electrospun fiber reinforced scaffold promotes total meniscus regeneration in rabbit meniscectomy model

Shuang Gao,Mingxue Chen,Pei Wang,Yan Li,Zhiguo Yuan,Weimin Guo,Zengzeng Zhang,Xueliang Zhang,Xiaoguang Jing,Xu Li,Shuyun Liu,Xiang Sui,Tingfei Xi,Quanyi Guo

Low vascularization in meniscus limits its regeneration ability after injury, and tissue engineering is the most promising method to achieve meniscus regeneration. In this study, we fabricated a kind of composite scaffold by decellularized meniscus extracellular matrix/polycaprolactone (DMECM/PCL) electrospinning fibers and porous DMECM, in which DMECM/PCL fibers were used as reinforcing component. The tensile modulus of the composite scaffold in longitudinal and crosswise directions were 8.5 ± 1.9 and 2.3 ± 0.3 MPa, respectively. Besides that, the DMECM/PCL electrospinning fibers enhanced suture resistance of the composite scaffold more than 5 times than DMECM scaffold effectively. In vitro cytocompatibility showed that the porous structure provided by DMECM component facilitated meniscus cells’ proliferation. DMECM was also the main component to regulate cell behaviors, which promoted meniscus cells expressing extracellular matrix related genes such as COL I, COL II, SOX9 and AGG . Rabbits with total meniscectomy were used as animal model to evaluated the composited scaffolds performance in vivo at 3 and 6 months. Results showed that rabbits with scaffold implanting could regenerate neo-menisci in both time points. The neo-menisci had similar histology structure and biochemical content with native menisci. Although neo-menisci had inferior tensile modulus than native ones, its modulus was improved with implanting time prolonging. MRI imaging showed the signal of neo-meniscus in the body is clear, and X-ray imaging of knee joints demonstrated the implantation of scaffolds could relief joint space narrowing. Moreover, rabbits with neo-menisci had better cartilage condition in femoral condyle and tibial plateau compared than meniscectomy group. Statement of Significance We fabricated the meniscus scaffold by combining porous decellularized meniscus extracellular matrix (DMECM) and DMECM/PCL electrospinning fibers together, which used the porous structure of DMECM, and the good tensile property of electrospinning fibers. We believe single material cannot satisfy increasing needs of scaffold. Therefore, we combined not only materials but also fabrication methods together to develop scaffold to make good use of each part. DMECM in electrospinning fibers also made these two components possible to be integrated through crosslinking. Compared to existing meniscus scaffold, the composite scaffold had (1) soft structure and extrusion would not happen after implantation, (2) ability to be trimmed to suitable shape during surgery, and (3) good resistance to suture.

Biological evaluation of porous aliphatic polyurethane/hydroxyapatite composite scaffolds for bone tissue engineering

Wanxun Yang,Sanne K. Both,Yi Zuo,Zeinab Tahmasebi Birgani,Pamela Habibovic,Yubao Li,John A. Jansen,Fang Yang

Biomaterial scaffolds meant to function as supporting structures to osteogenic cells play a pivotal role in bone tissue engineering. Recently, we synthesized an aliphatic polyurethane (PU) scaffold via a foaming method using non-toxic components. Through this procedure a uniform interconnected porous structure was created. Furthermore, hydroxyapatite (HA) particles were introduced into this process to increase the bioactivity of the PU matrix. To evaluate the biological performances of these PU-based scaffolds, their influence on in vitro cellular behavior and in vivo bone forming capacity of the engineered cell-scaffold constructs was investigated in this study. A simulated body fluid test demonstrated that the incorporation of 40 wt % HA particles significantly promoted the biomineralization ability of the PU scaffolds. Enhanced in vitro proliferation and osteogenic differentiation of the seeded mesenchymal stem cells were also observed on the PU/HA composite. Next, the cell-scaffold constructs were implanted subcutaneously in a nude mice model. After 8 weeks, a considerable amount of vascularized bone tissue with initial marrow stroma development was generated in both PU and PU/HA40 scaffold. In conclusion, the PU/HA composite is a potential scaffold for bone regeneration applications. © 2014 Wiley Periodicals, Inc. J Biomed Mater Res Part A: 103: 2251–2259, 2015.
