Successful cryopreservation and revival rely on effective cooling and rewarming strategies. Rewarming processes are more vital, because insufficient rewarming normally leads to devitrification or recrystallization, resulting in irreversible damage to biomaterials. Conventional rewarming methods often fail to achieve the required super-high rates. Laser warming has emerged as a promising approach for achieving ultra rapid and internalized heating. Most studies on laser rewarming rely heavily on simulations and lack experimental validation. Moreover, conventional procedures require the manual transfer of vitrified samples from liquid nitrogen to laser exposure spot, which introduces operational risks. To address these limitations, an integrated platform combining vitrification-based cryopreservation with automated laser rewarming was developed. A fast-response temperature measurement system was incorporated to monitor the real-time thermal changes in samples during laser irradiation. The laser rewarming rate of a 1 μL droplet was measured to be 3.5 × 10 6 K/min, which is far beyond those achieved by conventional water bath methods and significantly higher than the required critical warming rate. Three distinct temperature evolution modes were observed: insufficient, appropriate, and excessive heating. We directly confirmed a temperature overshoot followed by rapid thermal decay immediately after laser exposure, which has never been reported before. By varying the laser energy and beam size, the extent of temperature overshoot and subsequent thermal relaxation were controlled, which enabled the optimization of laser rewarming strategy. The findings can be used for the refinement of numerical models and will contribute to the development of safer and more effective rewarming protocols in the cryopreservation field.

Robust platforms and advanced biocompatible materials having diverse performances are in tremendous demand for cryopreservation of biocells, which are greatly limited by the crystallization, formation, and growth of ice crystals. The fickle structure and the arduous extraction process of modern attainable antifreezing proteins cause fatal cryoinjury of the cells making it challenging to develop anti-icing materials. Thus, designing Au colloids is an effective way to combat cell-damaging concerns during the ice freezing–thawing process. Herein, we propose an emerging biomimetic hybrid nanomaterial (AuNR@SiO2-CDs) prepared by combining the photoheating and rewarming controlling characteristics of carbon dots (CDs) and gold nanorods (AuNRs), respectively, via a SiO2 scaffold that has an optimal aspect ratio of ∼4.4. The performance of the material is applied in the freezing and resuscitation of Hela cells. The typical linkage between the AuNR and CDs not only retains the stable structure but also possesses the symmetric functional characteristics of affirmative cryoprotectant materials and sustained low cytotoxicity of cell viability >90%. The cell recovery rate of the Hela cell is significantly improved to ∼60%, which is propped up to >4% higher by the laser irradiation dose. The above hybrid material is paving a path toward novel bionic antifreezing proteins and is envisioned for ice recrystallization inhibition and rapid rewarming.

The Hydrogel wound dressing with good rewarming property, anti-freezing property, and adhesion property has important practical significance in medical care for the first and second grade frostbite. The ears, hands, mouth, and nose of field workers on duty and training high cold region are prone to frostbite due to long-term exposure to in severe cold. Therefore, rapid rewarming of wound surface is the key to the treatment of frostbite. In this paper, antifreeze and rewarming are achieved by photothermal conversion materials and quaternary ammonium salts. Herein, the PAM/CS/MXene30/Betaine5/AgNPs200(PCM30B5A200) hydrogel based on chitosan (CS) and acrylamide (AM) is designed and fabricated by thermally initiated free radical polymerization. The dressing has excellent rewarming properties, anti-freezing properties, tensile properties, repeatable self-adhesion and antibacterial properties. In this system, hydrogels are mainly cross-linked by hydrogen bonds between CS and AM. Betaine is added to the material to make the hydrogel have good freeze-resistance and flexibility at −24 °C, and can be combined with AgNPs and MXene, to achieve a triple antibacterial. The PCM30B5A200 hydrogel reached the rewarming temperature of 40.5 °C at 9 min. PCM30B5A200 multifunctional hydrogels have good application value in the first and second grade frostbite wounds.

Cryopreservation is essential to store living cells and tissues for future use while maintaining the proper levels of cell functions. The use of cryoprotective agents (CPAs) to inhibit intracellular ice formation during cryopreservation is vital for cell survival, but the addition and removal of CPAs and ice recrystallization during rewarming will cause fatal injury to cells. The conventional CPA loading and unloading methods generate osmotic shocks and cause mechanical injury to biological samples, and the conventional method of rewarming using a water bath also leads to ice recrystallization and devitrification. A new CPA-loaded microparticle-based method for loading and photothermal rewarming under near-infrared (NIR) laser irradiation was proposed to overcome these difficulties. We have successfully achieved the controlled release of CPAs (2 M EG, 2 M PG, and 0.5 M trehalose) with a graphene oxide (GO, 0.04% w/v) core from a 1.5% (w/v) sodium alginate shell to the human umbilical vein endothelial cells (HUVECs) within 60 s using NIR laser irradiation (808 nm Lasever at 5000 mW/cm2) and successfully recovered the CPA-loaded cells with 0.04% (w/v) GO in 8–10 s using the same NIR irradiation. The results show that this method achieved 25% higher viability of HUVECs compared to the conventional method. In short, this study proposes a new approach for achieving controlled CPA loading to cells with a photothermal-induced strategy for cell cryopreservation.

Strong hydration of cryoprotective agents reduces the glass transition temperature of water and suppresses ice formation. However, lethal cooling and warming remain a critical obstacle to cross-scale cryopreservation of clinical biospecimens. Herein, a snowman-like Janus nanohybrid composed of magnetic iron tetraoxide and photothermal polypyrrole is reported. Its cranial–corporal asymmetry enables heterogeneous hydration for a record high efficiency of ice confinement, reducing mean ice crystal area by 98.4%. Molecular dynamics simulations reveal that Janus architecture simultaneously enhances interactions with ice and strengthens the local hydration anisotropy, accounting for the effective inhibition of ice growth. Superflash warming over 920 °C min ‒1 by magnetically rotating its anisotropic structure for uniform heat dissipation narrows the hostile temperature window in micro-/macroscopic scenarios, as further confirmed by Monte Carlo modeling. This design enables cost-effective post-thaw magnetic retrieval, eliminating the need for heavy centrifuges, well-suited for scalable and on-site applications. As a result, cryopreserved samples from single cells, bacteria, to porcine trachea retain near-complete viability and functionality. Therefore, this study offers a promising technique to bridge the gap between microscale cell storage and whole-organ preservation.
