Characterization of a postbiotic exopolysaccharide produced by Lacticaseibacillus paracasei ET-22 with antioxidant and anti-inflammatory efficacy

Jingwei Wu,Zhihui Li,Zhuoting Zhang,Jian Zhang,Hangyu Hu,Hanglian Lan,Weilian Hong,Zhennai Yang

Exopolysaccharides (EPSs) produced by lactic acid bacteria have been extensively studied, but the EPSs as postbiotics are much less investigated. In this study, the EPS, designated as ET22-EPS, was extracted and purified from the culture broth of Lacticaseibacillus paracasei ET-22 previously characterized with strong postbiotic activities. ET22-EPS was determined with a molecular weight of 5.12 × 10 5  Da, and a monosaccharide composition of mannose, galactose, glucose, and N -acetylglucosamine in a molar ratio of 0.662:0.103:0.095:0.026. ET22-EPS exhibited a sheet-like and fibrous stacking morphology with a networked structure. ET22-EPS also showed high thermal stability with a degradation temperature of 295.33 °C. ET22-EPS was identified to consist of an octasaccharide repeating unit with a backbone structure of four consecutively linked →2)-α-D-Man p (1 → together with →2,6)-α-D-Man p (1 → 2)-α-D-Glc p (1→, and a branch chain of α-D-Gal p (1 → 6)-α-D-Man p (1→. The in vitro bioassay indicated that ET22-EPS effectively scavenged the free radicals of diammonium 2,2′-azino-bis(3-ethylbenzothiazoline-6-sulfonate), 1,1-diphenyl-2-picrylhydrazyl, hydroxyls, and superoxide anion. ET22-EPS showed strong anti-inflammatory effects by reducing cell phagocytosis, nitric oxide production, pro-inflammatory cytokine secretion, and the TNF-α/IL-10 ratio in a RAW264.7 cell model. Therefore, ET-22-EPS shows good prospects as potential postbiotics in the development of health foods.

Structural Characterization of Exopolysaccharide Produced by Leuconostoccitreum B-2 Cultured in Molasses Medium and Its Application in Set Yogurt

Lu Liang,Min Xu,Lei Pan,Zhijiang Zhou,Ye Han

Sugarcane molasses is an agricultural by-product containing sucrose. In this study, the exopolysaccharide (M-EPS) produced byLeuconostoc citreumB-2 in molasses-based medium was characterized, optimized, and its application in set yogurt was investigated. The structure analysis, including gel permeation chromatography, Fourier transform infrared spectroscopy, and nuclear magnetic resonance, revealed that the M-EPS was a linear dextran composed of D-glucose units, which were linked by α-(1→6) glycosidic bonds with 19.3% α-(1→3) branches. The M-EPS showed a lower molecular weight than that produced from sucrose. The M-EPS was added into the set yogurt, and then the water holding capacity, pH, and microstructure of set yogurt were evaluated. Compared with the controls, the addition of M-EPS improved the water holding capacity and reduced the pH of set yogurt. Meanwhile, the structure of the three-dimensional network was also observed in the set yogurt containing M-EPS, indicating that M-EPS had a positive effect on the stability of set yogurt. The results provide a theoretical basis for the cost-effective utilization of sugarcane molasses. Keywords:exopolysaccharide;molasses;structure;optimization;set yogurt

Weissella confusa XG-3 EPS: production intensification by co-culturing with Candida shehatae and partial characterization

Shuhan Yang,Shouqi Zhao,Yingying Ning,Dan Zhao

Previous studies have shown that co-culture methods can enhance exopolysaccharide production, no studies have reported this enhancement in co-cultures between Weissella confusa and Candida shehatae. This study developed an efficient co-culture system combining W. confusa XG-3 and C. shehatae (1:1 ratio), and employed response surface methodology to optimize both exopolysaccharide yield and properties. The initial co-culture produced 56.82 ± 0.12 g/L exopolysaccharide, a 1.63-fold increase over single culture. Optimization (sucrose: 105.97 g/L; yeast extract: 5.81 g/L; peptone: 6.24 g/L) further increased the yield to 115.66 g/L, a 2.04-fold improvement. The co-cultured exopolysaccharide had high water solubility (60.45 ± 0.70%) and water holding capacity (614.74 ± 9.26%). The co-cultured EPS showed significant antioxidant activity. At 5 mg/mL, the scavenging rates of ABTS, DPPH, hydroxyl, superoxide, H2O2 and total reducing power reached the highest 52.22 ± 0.47%, 50.79 ± 2.86%, 55.80 ± 2.56%, 36.05 ± 2.07%, 66.85 ± 0.30%, and 0.39 ± 0.01. It also had a superior effect on the proliferation of several probiotics compared with commercial prebiotics and exhibited high thermostability with a degradation threshold at 292.5 °C. These results demonstrate a novel, scalable strategy for producing multifunctional EPS with broad applications in the food industry.

Anti-Diabetic Activity of a Novel Exopolysaccharide Produced by the Mangrove Endophytic Fungus Penicillium janthinellum N29

Zhuling Shao,Yingying Tian,Shan Liu,Xiao Chu,Wenjun Mao

Marine microorganisms often produce exopolysaccharides with novel structures and diverse biological activities due to their specific marine environment. The novel active exopolysaccharides from marine microorganisms have become an important research area in new drug discovery, and show enormous development prospects. In the present study, a homogeneous exopolysaccharide from the fermented broth of the mangrove endophytic fungusPenicillium janthinellumN29, designated as PJ1-1, was obtained. The results of chemical and spectroscopic analyses showed that PJ1-1 was a novel galactomannan with a molecular weight of about 10.24 kDa. The backbone of PJ1-1 was composed of →2)-α-d-Manp-(1→, →4)-α-d-Manp-(1→, →3)-β-d-Galf-(1→ and →2)-β-d-Galf-(1→ units with partial glycosylation at C-3 of →2)-β-d-Galf-(1→ unit. PJ1-1 had a strong hypoglycemic activity in vitro, evaluated using the assay of α-glucosidase inhibition. The anti-diabetic effect of PJ1-1 in vivo was further investigated using mice with type 2 diabetes mellitus induced by a high-fat diet and streptozotocin. The results indicated that PJ1-1 markedly reduced blood glucose level and improved glucose tolerance. Notably, PJ1-1 increased insulin sensitivity and ameliorated insulin resistance. Moreover, PJ1-1 significantly decreased the levels of serum total cholesterol, triglyceride and low-density lipoprotein cholesterol, enhanced the level of serum high-density lipoprotein cholesterol and alleviated dyslipidemia. These results revealed that PJ1-1 could be a potential source of anti-diabetic agent. Keywords:mangrove endophytic fungus;exopolysaccharide;structure;anti-diabetic activity

Exopolysaccharides produced by Antrodia cinnamomea using microparticle-enhanced cultivation: Optimization, primary structure and antibacterial property

Jia-Hui Fan,Lin-Qiang Xiong,Wei Huang,Jia-Qi Hong,Huang-Kai Guo,Ka-Hing Wong,Peter C.K. Cheung,Qiong-Qiong Yang,Bo-Bo Zhang

Microparticle-enhanced cultivation was used to enhance the production of exopolysaccharides (EPSs) from Antrodia cinnamomea . The structure and antibacterial activity of two EPSs produced by A. cinnamomea treated with Al 2 O 3 [EPS-Al (crude) and EPS-Al-p (purified)] and without Al 2 O 3 [EPS-C (crude) and EPS-C-p (purified)] were compared. It was observed that the addition of 4 g/L Al 2 O 3 at 0 h resulted in the highest EPS yield of 1.46 g/L, possible attributed to the enhanced permeability of the cell membrane. The structural analysis revealed that EPS-C-p and EPS-Al-p had different structures. EPS-C-p was hyperbranched and spherical with a Mw of 10.8 kDa, while EPS-Al-p was irregular and linear with a Mw of 12.5 kDa. The proportion of Man in EPS-Al-p decreased, while those of Gal and Glc increased when compared to EPS-C-p. The total molar ratios of 6-Glc p and 4-Glc p in EPS-Al-p are 1.45 times that of EPS-C-p. Moreover, EPSs could alter bacterial cell morphology, causing intracellular substance leakage and growth inhibition, with EPS-Al having a stronger antibacterial activity than EPS-C. In conclusion, A. cinnamomea treated with Al 2 O 3 could produce more EPSs, changing monosaccharide composition and glycosidic linkage profile, which could exert stronger antibacterial activity than that produced by untreated A. cinnamomea .

Plant allelochemicals inhibit the growth and colony formation of Microcystis

Ma Xiao,Wang Xueli,Zhou Shaoqi,Ma Jianrong,Wang Jingfu,Chen Jingan,Zeng Yan,Chen Qiao,Qin Boqiang,Li Ming

Genus Microcystis is the most common dominant species of bloom-forming cyanobacteria. Growth and colony formation are critical processes for the occurrence of Microcystis blooms. To identify allelochemicals that can inhibit the growth and colony formation of Microcystis, Microcystis aeruginosa was cultivated and treated with various concentrations of gramine, catechol, berberine, and sanguinarine in laboratory. Results indicate that the growth, colony formation, and extracellular polysaccharides (EPS) content of Microcystis were significantly inhibited when treated with gramine, catechol, and berberine in appropriate concentrations at high iron level (6.8 mg/L). Microcystis growth was inhibited and colony formation and EPS content were promoted when treated with sanguinarine at high iron level. These findings suggest that cyanobacterial blooms can be controlled by regulating the input of gramine, catechol, and berberine. Furthermore, this study revealed that plant allelochemicals could affect the growth and morphology of algae in aquatic ecological restoration, which may be potentially important for improving water quality.

Exopolysaccharide from endophytic fungi of Cinnamomum burmannii leaves: structural characterization and hepatoprotective effects

Wenjun Zhang,Shuoyue Yuan,Ruoxin Chen,Pengzhen Li,Qinyu Huang,Zhenlong Zhou,Penghui Fan,Jianyu Su

Exopolysaccharides derived from endophytic fungi inhabiting medicinal plants are promising bioactive agents. Herein, a neutral exopolysaccharide termed DEPS was extracted and purified from an endophytic fungus isolated from Cinnamomum burmannii leaves, with a yield of 51.12 %. Structural analysis by HPGPC, UV, FTIR, NMR, and SEM revealed that DEPS is primarily composed of glucose, with minor amounts of mannose and galactose, and features →6)-α- d -Glcp-(1→ and →4)-α- d -Glcp-(1→ linkages. Functionally, DEPS exhibited a superior moisture-retention rate of 26.4 %, comparable to sodium alginate and glycerol, and showed moderate antioxidant activity. In vitro , DEPS effectively suppressed IL-6 expression in macrophages by approximately twofold, and inhibited zebrafish neutrophil migration by 50 %. In an acetaminophen (APAP)-induced liver injury model, DEPS treatment significantly alleviated hepatic damage by decreasing necrosis area from 55.2 % to 24.1 %, and reducing IL-1β expression from 273.3 % to 118.6 %. Additionally, DEPS promoted an anti-inflammatory environment through enhanced IL-10 expression and reduced TNF-α levels. These findings highlight DEPS as a structurally unique, multifunctional polysaccharide with hepatoprotective, anti-inflammatory, and antioxidant potential for food, pharmaceutical, and cosmetic applications.

Isolation, structural characterization and neuroprotective activity of exopolysaccharide from Paecilomyces cicada TJJ1213

Juanjuan Tian,Xiaomeng Wang,Xueliang Zhang,Cangping Zhang,Xiaohong Chen,Mingsheng Dong,Xin Rui,Qiuqin Zhang,Yong Fang,Wei Li

Two exopolysaccharide fractions (EPS1 and EPS2) were obtained from Paecilomyces cicadae TJJ 1213, and their structures were elucidated. The EPS1 and EPS2 were mainly composed of mannose and galactose with molar ratios of 3.2: 1.0 and 2.7: 1.0, respectively. They possessed average molecular weights of 1.69 × 10 6 and 8.06 × 10 5 Da, respectively. Structural characterization indicated that the backbone of EPS1 was consisted of →4)-α-D-Man p (1→, →3,4)-α-D-Man p (1 → and →2,6)-α-D-Man p (1→, →6)-α-D-Gal p (1→, →6)-β-D-Gal p (1→, and side chain was consisted of α-D-Man p residue. The backbone of EPS2 was composed of →6)-β-D-Gal p -(1→, →4)-α-D-Man p -(1→, →2,6)-α-D-Man p -(1 → and →6)-α-D-Gal p -(1→, and the branching point was also consisted of α-D-Man p residue. In addition, EPS1 and EPS2 had potential in protective effects of PC12 cells against hydrogen peroxide induced oxidative stress by inhibiting the production of ROS, reducing LDH leakage and alleviating mitochondrial damage. These results indicated that EPS1 and EPS2 might serve as therapeutic agents for neuronal disorders.

Interaction of the Exopolysaccharide from Lactobacillus plantarum YW11 with Casein and Bioactivities of the Polymer Complex

Min Zhang,Tiantian Lai,Mengke Yao,Man Zhang,Zhennai Yang

There has been an increased application of exopolysaccharide (EPS)-producing lactic acid bacteria (LAB) in fermented dairy products, but interactions between EPS and casein (CAS), and bioactivities of their complex are poorly studied. In this study, EPS produced byLactobacillus plantarumYW11 (EPS-YW11) was studied for interactions with CAS in a simulated fermentation system acidified by D-(+)-gluconic acid δ-lactone. The results showed that there was interaction between EPS-YW11 and CAS when EPS (up to 1%,w/v) was added to the casein solution (3%,w/v) as observed with increased viscoelasticity, water holding capacity, ζ-potential and particle size of EPS-YW11/CAS complex compared with CAS alone. Microstructural analysis showed that a higher concentration of EPS facilitated more even distribution of CAS particles that were connected through the polysaccharide chains. Infrared spectroscopy further confirmed interactions between EPS and CAS by intermolecular hydrogen bonding, electrostatic and hydrophobic contacts. Further evaluation of the bioactivities of EPS-YW11/CAS complex revealed significantly increased antibiofilm, antioxidation, and bile acids binding capacity. The present study provides further understanding on the mechanism of interactions between EPS produced by LAB and CAS, which would benefit potential applications of EPS in fermented dairy products with enhanced functionality. Keywords:Lactobacillus plantarumYW11;exopolysaccharide;casein;interaction;bioactivities

Optimization and Characterization of an Antioxidant Exopolysaccharide Produced by Cupriavidus pauculus 1490

Xia Mingchen,Zhang Shishi,Shen Li,Yu Runlan,Liu Yuandong,Li Jiaokun,Wu Xueling,Chen Miao,Qiu Guanzhou,Zeng Weimin

In the present study, exopolysaccharides (EPS) production by Cupriavidus pauculus 1490 was optimized by response surface methodology. The results showed that sodium gluconate (4.15 g/L), NH 4 Cl (0.52 g/L), and Na 2 HPO 4 ·12H 2 O (0.04 g/L) were the optimal medium components and concentrations. The actual EPS yield of 293.2 m g/L in the optimized medium was in close agreement with the predicted value of 283.35 m g/L. Analysis of fourier transform infrared spectroscopy indicated the EPS contained abundant functional groups, such as –OH, C=O and C–O–C, and all of them were attributed to the characteristics of polysaccharides. Mannose, glucuronic acid, glucose and xylose were detected as the main monosaccharide composition of EPS. Rheological analysis suggested that the rheogram of EPS has similar trend with Xanthan and presented the property of non-Newtonian fluid. Moreover, the addition of NaCl and KCl would partly weaken the shear stress of EPS. Three in vitro assays were conducted to evaluate the antioxidant potential of the EPS. Results demonstrated that the EPS possessed scavenging capacity on hydroxyl radical, DPPH radical and superoxide anion radical in a dose-dependent way. As indicated by above results, the EPS isolated from C. pauculus 1490 might serve as a potential antioxidant agent to be applied in nutraceutical and pharmaceutical industries.

Effects of exopolysaccharide fractions with different molecular weights and compositions on fecal microflora during in vitro fermentation

Yu-Heng Mao,Ang-Xin Song,Long-Qing Li,Ka-Chai Siu,Zhong-Ping Yao,Jian-Yong Wu

This study was to investigate the potential prebiotic function of exopolysaccharide (EPS) from a medicinal fungus and the relationship to the molecular properties by in vitro human fecal fermentation. The EPS from Cordyceps sinensis Cs-HK1 mycelial fermentation was processed into three fractions with different monosaccharide contents, a higher molecular weight (MW) and a lower MW attained by two-step ethanol precipitation , and an intermediate MW by ultrasound-degradation of EPS. All the EPS fractions were well utilized during 24–48 h of fecal fermentation, leading to significant increases in the short chain fatty acid (SCFA) production. The consumption rate and production level of SCFAs varied slightly with the different EPS fractions. The EPS also influenced the composition and diversity of the fecal microflora , increasing the relative abundance of Firmicutes but suppressing that of Proteobacteria , which may be a beneficial effect for human health. Overall the results have shown that the Cs-HK1 EPS has significant prebiotic activity which is dependent on its molecular properties.

Flagellar Motility Is Critical for Salmonella enterica Serovar Typhimurium Biofilm Development

Wang Feiying,Deng Le,Huang Fangfang,Wang Zefeng,Lu Qiujun,Xu Chenran

The food-borne pathogen Salmonella enterica serovar Typhimurium (S. Typhimurium) causes self-limiting gastroenteritis in humans and is not easily eradicated because it often attaches to suitable surfaces to form biofilms that have high resistance to disinfectants and antimicrobials. To develop an alternative strategy for the treatment of biofilms, it is necessary to further explore the effects of flagellar motility on the development process of Salmonella biofilms. Here, we constructed flagella mutants (ΔflgE and ΔfliC) to systematically study this process. By comparing them with wild-type strains, we found that these mutants lacking flagellar motility form fewer biofilms in the early stage, and the formed mature biofilms contain more cells and extracellular polymeric substances (EPS). In addition, fewer mutant cells adhered to glass plates compared with wild-type cells even after 6 h of incubation, suggesting that flagellar motility plays a significant role in preliminary cell-surface interactions. More importantly, the motility of wild-type strain was greatly decreased when they were treated with carbonyl cyanide m-chlorophenylhydrazone, which inhibited flagellar motility and reduced biofilm formation, as in the case of the ΔflgE mutant. Overall, these findings suggest that flagellar motility plays an important role in Salmonella biofilm initiation and maturation, which can help us to counteract the mechanisms involved in biofilm formation and to develop more rational control strategies.
