Characterisation of zinc-chelating peptides derived from scallop (Patinopecten yessoensis) mantle and their intestinal transport in everted rat sacs

Xiaoyang Liu,Jiatong Sun,Yue Li,Xuening Yu,Fawen Yin,Deyang Li,Pengfei Jiang,Yoshimasa Nakamura,Dayong Zhou

Summary Recently, food-derived peptides were frequently studied and used as metal ion chelator against negative effects of trace element absorption induced by various human gastrointestinal environments. In this study, the scallop mantle hydrolysates (SMHs) were produced and characterised to study their intestinal promotion of zinc absorption. During production of hydrolysates, the results of degree of hydrolysis (DH) and proteins solubility suggested that the alcalase was suitable for the production of peptides from scallop mantle proteins where the chelating conditions were settled (30 °C, 10 min and 3 mg mL −1 ). The molecular distribution of SMH was mainly 1–5 kDa and chelation of zinc with SMH (involved in carboxyl and amine group of SMH) triggered no significant alteration on the secondary structure of SMH. Zinc morphologically altered the surface structures of SMH. Moreover, SMH could successfully carry and transport zinc while the fluorescence staining indicated free zinc accumulated inside the tissue of everted rat sacs. Finally, several representative transported peptides (intact or cleaved) were identified, such as FTGEPGPSGPT, AINDPFIDL and ALPHAILRI. This study provided a theoretical basis for the investigation of absorption of marine animal-derived peptides as zinc carriers.

Lipid profiles in different parts of two species of scallops (Chlamys farreri and Patinopecten yessoensis)

Xiao-Pei Hu,Qing-Da An,Da-Yong Zhou,Ting Lu,Fa-Wen Yin,Liang Song,Qi Zhao,Jiang-Hua Zhang,Lei Qin,Bei-Wei Zhu,Fereidoon Shahidi

Different classes of lipids and phospholipids (PL) as well as fatty acid (FA) compositions, and glycerophospholipid (GP) molecular species in edible parts (adductor muscles) and by-products (gonads and mantles) of scallops Patinopecten yessoensis and Chlamys farreri were characterized. The lipids contained high levels of polyunsaturated FA (PUFA) (20.44–58.81% of total FAs) and PL (54.99–78.26% of total lipids). Among PL, phosphatidylcholine (41.39–51.86 mol%) was dominant. More than 230 GP species belonging to glycerophosphocholine, glycerophosphoethanolamine, glycerophosphoserine, glycerophosphoinositol, lysoglycerophosphocholine, lysoglycerophosphoethanolamine and lysoglycerophosphoinositol were characterized. Individual GP molecular species in different body parts of scallops were also partially quantified. Most of the dominant GP species contained PUFA, especially eicosapentaenoic acid (EPA, 20:5n-3) and docosahexaenoic acid (DHA, 22:6n-3). Considering the high level of PUFA enriched GP, edible parts and by-products (gonads and mantles) of scallops Patinopecten yessoensis and Chlamys farreri provide great potential as health-promoting food for human consumption.

Transcriptional responses to DHA-induced lipid droplet accumulation in primary adherent mantle cells of Sinonovacula constricta (Lamarck, 1818)

Bowen Yan,Zhaoshou Ran,Qiang Zhou,Jiaxin Zhu,Fei Kong,Jilin Xu

Docosahexaenoic acid (DHA), an essential omega-3 long-chain polyunsaturated fatty acid, is crucial for the development of marine mollusks. Despite its importance, the molecular mechanisms of DHA action in these organisms are poorly understood, primarily due to the lack of reliable in vitro models. This study utilizes the razor clam Sinonovacula constricta as a model organism, establishing a primary adherent cell culture from various tissues, with mantle tissue identified as the optimal source for its rapid migration and low contamination risk. Cells incubated with varying concentrations of DHA (0–400 μM) for 6 h exhibited a concentration-dependent accumulation of lipid droplets (LDs), highlighting the model's suitability for investigating lipid metabolism. Transcriptomic analysis of cells treated with 50 μM DHA identified 848 differentially expressed genes (DEGs), with 747 upregulated and 101 downregulated, indicating the complex effects of DHA. The top 20 significantly upregulated and downregulated DEGs revealed significant involvement in cell signaling transduction, extracellular matrix organization, and substance transport and metabolism. Particularly, lipid metabolism genes, including perilipin 2 and abhydrolase domain containing 2, as well as pathways such as the PPAR signaling pathway and sphingolipid metabolism, were significantly altered, suggesting a reprograming of fatty acid metabolism contributing to LD accumulation. Additionally, pathways related to immune response were notably affected, potentially enhancing cellular health. Collectively, this study provides the first comprehensive insights into the molecular mechanisms underlying DHA-induced LD accumulation in a marine mollusk model at the cellular level, laying a foundation for developing precise nutritional strategies involving DHA in S. constricta aquaculture.

Enhancing pearl luster by targeted delivery of astaxanthin using submicron hollow ordered mesoporous silica to the mantle of pearl oysters

Yuan-shuai Fu,Xi-ke Li,Yue Lin,Yun-ao Li,Yong Zhao,Wei-hao Wu,He Wang,Xue-feng Lv,Yuan Zhang,Yong-heng Zhu,Zhi-yi Bai

Pearl luster is a key determinant of pearl quality, and enhancing pearl luster remains a major challenge in current pearl production. Despite astaxanthin holding great potential in regulating pearl luster, its poor bioavailability and insufficient accumulation at target sites significantly limit its practical application. Herein, we developed a submicron hollow ordered mesoporous silica (HOMSS) carrier highly loaded with ASX to achieve enhanced and controlled delivery for pearl luster improvement. The HOMSS system demonstrated high ASX loading capacity and enabled sustained release of ASX. In vivo fluorescence imaging confirmed targeted accumulation of HOMSS in the mantle and visceral mass of Hyriopsis cumingii . Notably, pearl oysters treated with HOMSS-ASX demonstrated significantly enhanced antioxidant capacity and a 35.75 % increase in purple luster intensity. Integrated transcriptomic and molecular docking analyses revealed that ASX specifically interacts with the aryl hydrocarbon receptor (AHR), triggering upregulation of cytochrome P450 family genes expression and promoting the biosynthesis of hydroxyserotonin. This metabolite deposits in the mantle, intensifying the purple luster of pearls. In summary, our innovative nanodelivery strategy based on the HOMSS system not only promoted physiological status and metabolic balance of pearl oysters but also significantly enhanced pearl luster. These findings provide valuable insights for the development of novel nanodelivery applications in shellfish aquaculture and the pearl industry.

Examination of the roles of CgTH in tyrosine metabolism and pigmentation of Pacific oyster (Crassostrea gigas)

Kunyin Jiang,Hong Yu,Lingfeng Kong,Shikai Liu,Shaojun Du,Qi Li

The visually appealing shell colours and intricate patterns of shellfish not only attract consumers but also enhance their economic value. Understanding the inheritance pattern of shell colour and the molecular mechanism of pigmentation is highly important for enhancing breeding efficiency. This study involved the identification of a tyrosine hydroxylase gene ( CgTH ) and the investigation of its function in the metabolism of tyrosine and pigmentation in Pacific oysters. CgTH contains a conserved Biopterin_H domain that consists of a binding site for iron atoms and a binding site for tetrahydrobiopterin (BH4). Quantitative PCR (qPCR) and immunofluorescence analysis revealed that the expression of CgTH was higher in the mantle of black shell oysters in comparison to the white strain. The in vitro and in vivo experiments confirmed that Cg TH is involved in tyrosine metabolism through the catalysis of the conversion of L-tyrosine to L-DOPA. In order to examine its functions in melanin synthesis in oysters, 3-Iodo-L-tyrosine (3-IT) treatment, RNA interference (RNAi), and L-DOPA treatment assays were conducted. The results of our study showed that 3-IT effectively inhibited Cg TH activity, leading to a suppression of tyrosine metabolism. Knockdown of Cg TH hindered the cAMP-CREM-MITF axis and decreased pigmentation. On the other hand, L-DOPA stimulated melanin production through cAMP signaling and counteracted the inhibitory effects of 3-IT on the metabolism of tyrosine. These data indicated that there may be a feedback loop involving Cg TH and the cAMP-CREM-MITF axis that regulates tyrosine metabolism and melanogenesis in Pacific oysters. This study enhances our understanding of the molecular mechanism that governs oyster pigmentation and offers a possible molecular indicator for the targeted breeding of particular strains of Pacific oysters ( Crassostrea gigas ).
