Triadimefon is ubiquitous in various environmental media. Although toxicity of triadimefon to individual of aquatic organisms has been confirmed, its effect on organisms at population level remain poorly understood. In this study the long-term effect of triadimefon on individual and population of Daphnia magna were studied using multi-generational experiments and matrix model. Development and reproduction of three generations of F1 and F2 were significantly inhibited with the triadimefon concentration of 0.1 mg/L ( p < 0.01). Toxicity of triadimefon to the offspring was stronger than to the parent ( p < 0.05). When triadimefon concentration was higher than 0.1 mg/L, both population number and intrinsic rate of increase showed a decreasing trend with the increasing exposure concentration. Age structure of the population also tended to decline. Toxicity threshold derived on population-level was between mortality-based LC 50 and reproduction-based NOEC of Daphnia magna , and also between acute toxicity and chronic toxicity derived from species sensitivity distribution (SSD). The risk of population level derived from risk quotient was low for most areas, and the results derived from probability risk showed that the expected loss of intrinsic rate of increase of population was 0.0039 without considering other factors. Compared to the individual-level, the ecological risks at the population level were closer to the actual situation of the ecosystem response to the chemical pollution.

Nonylphenol (NP) is a toxic xenobiotic compound, which is persistent in the aquatic environment and is extremely toxic to aquatic organisms. Although the exact molecular mechanisms of its toxic effect are well understood, the multigenerational reproduction and multigenerational - gene expression changes caused by NP still remain unclear. The following work investigated the effect of NP on four consecutive generations of zebrafish by examining their growth and several reproductive parameters, the degree of gonad damage, and the expression of related reproduction related genes. The results showed that high concentrations (20 and 200 μg·L −1 ) of NP could decrease growth and induce gonad damage in zebrafish. In addition, gnrh2 and gnrh3 genes were up-regulated, and fshβ and lhβ genes were downregulated in the hypothalamus in male zebrafish; while in female fish, the fshβ and lhβ were upregulated in P and F1 generations, and then down-regulated in the F2 generation. Meanwhile, the cyp19a1a gene was downregulated in the gonad of male fish, while the genes of fshr , lhr and esr showed a downward trend in females. Compared to P generation, F2 generation was more tolerant to higher NP concentrations (20 and 200 μg·L −1 ), as was also more sensitive to lower concentrations of NP (2 μg·L −1 ). Consequently, stress and damage caused by environmentally-relevant concentrations of aquatic pollutants in a vertebrate model were measured and predicted. Prevention and control measures can be actively and effectively proposed, which might be transversal to other exposed organisms, including humans. After several generations, typical transgenerational genetic phenomena might occur, which should be addressed by further studies.

Nonylphenol (NP) is a persistent environmental chemical that can disrupt the organism’s endocrine system, and is detected in the surface water and sea. In this study, we investigated whether NP can alter transcriptional expression of sexual differentiation-related genes. Three generations of zebrafish were exposed to 0, 2, 20 and 200 μg·L −1 of NP, and transcriptional expression of sexual differentiation genes were assessed in 10, 20 and 40 dpf in the F1 and F2 generations. Growth of zebrafish exposed to 200 μg·L −1 of NP was inhibited at 125 dpf in the F1 generation. 20 μg·L −1 of NP resulted in 80% females in the F1 generation, but had no effect on the F2 generation. In terms of the sexual differentiation genes, the transcriptional expression of cyp19a1a and esr1 genes were upregulated in 20 μg·L −1 of NP in the F1 generation. But expression of the sexual differentiation genes were not affected in the F2 generation. Overall, NP could affect sexual differentiation and gene transcriptional expression in the F1 generation. The tolerance of contaminant in the offsprings was improved at low concentration.

The novel brominated flame retardant 1, 2-bis (2,4,6 tribromophenoxy) ethane (BTBPE) has been widely used since the mid-1970s and detected in the aquatic environment and organisms; however, little information is available regarding its biotoxicity. In this study, the life-history parameters, transcriptomic regulation, multi-generational offspring size, and population dynamics of rotifers were determined in response to BTBPE (0, 0.01, 1, and 100 μg/L). The results showed that BTBPE (>1 μg/L) significantly increased the pre-reproductive time, shortened both reproduction periods and the lifespan, and decreased the total number of rotifer offspring. Transcriptomic analysis revealed that the expression of key genes involved in the pathways of citrate cycle, 2-oxocarboxylic acid metabolism, carbon metabolism, and propanoate metabolism was affected. Moreover, BTBPE has transgenerational effects on rotifers, the size of the F1 to F5 generations decreased significantly in response to BTBPE. Finally, the population density in the 100 μg/L BTBPE treatment group was significantly reduced by 30 % compared to the control group, suggesting that the population growth of rotifers was inhibited by BTBPE exposure. The findings of this study indicate that BTBPE has effects on the individual reproduction, population growth, offspring quality, and energy metabolism of rotifers. These findings highlighted the potential impacts of BTBPE on dominant zooplankton species, providing useful baseline information for evaluating the potential ecological risk of novel brominated flame retardant.

In order to understand how bisphenol A (BPA) exposure acts on the evolutionary dynamics of populations and changes of stress response across generations, the model animal Caenorhabditis elegans was used to conduct the multigenerational testing. Multiple endpoints at the physiological (growth, reproduction, and locomotion behaviors) and molecular (stress-related gene expressions) levels were examined by multigenerational exposure to low-concentration BPA (0.001–10 μM) across four generations. The results showed that changes of physiological-level effects across four generations varied in magnitude and direction, depending on the exposure concentrations. C. elegans individuals in the first generation grew smaller, moved slower, and produced less offsprings than the controls by BPA exposure. As for each trait tested, the first generation response could be commonly mirrored in the subsequent generations at the highest concentration of 10 μM. However, at lower concentrations, response of parental generation was a relatively poor predictor of the effects on progeny, as acclimation or cumulative damage could occur in the subsequent generations. The integrated gene expression profiles visually illustrated that the tested gene expressions at low concentrations (0.001–0.01 μM) were more obviously changed in both G 1 and G 4 generations, and the G 1 generation showed a much greater degree of increase in stress-related gene expressions than the G 4 generation. The multigenerational toxicity data emphasize the need of considering biological effects over multiple generations to conduct accurate assessment of environmental risks of toxicants on population dynamics.

Although ketamine (KET) has been widely detected in aquatic environments, the ecotoxicity data in aquatic invertebrates and associated risk remained unclear. This study aimed to investigate the adverse effects on benthos ( Caenorhabditis elegans ( C.elegans )) posed by KET from chronic (10 days) and multigenerational (four generations) exposure. Such exposure induced dose-dependent alterations on apoptosis, reactive oxygen species (ROS) induction, locomotion activity, feeding rate, chemotaxis, and brood size of nematodes, showing a cumulative damage through generations. KET posed vulva deformations and worm bags of C. elegans with a dosed-dependent increase. As a consequence, the fecundity and viability of worms would be impaired, which could eventually impact aquatic ecosystem equilibrium. Meanwhile, the bioactivation/detoxification process of xenobiotics and longevity regulating pathway induced by KET might be responsible for the physiological function disorders. Accordingly, the risk quotients (RQ) of KET in surface water in China were calculated using the 90% indicator protection concentration (C 0.1 ) derived from multiple toxicity indicators cumulative analyses. The results would be more objective considering numerous biomarkers changes of one species in comparison with traditional method using no observed effect concentrations (NOEC) of teratogenesis . The risk in surface water in southern China was up to high level (RQ > 1), suggesting long-term monitoring was imperative.

Tris(1,3-dichloro-2-propyl) phosphate (TDCIPP) is considered a re-emerging environmental pollutant, and exposure to environmentally relevant concentrations has been shown to cause individual developmental toxicity in zebrafish and the water flea ( Daphnia magna ). However, multigenerational effects during exposure to TDCIPP and after subsequent recovery were unknown. In the present study, individuals of a model aquatic organism, the ciliated protozoan, T. thermophila were exposed to environmentally-relevant concentrations of TDCIPP (0, 300 or 3000 ng/L) for 60 days (e.g., theoretically 372 generations) followed by a 60-day period of recovery, during which T. thermophila were not exposed to TDCIPP. During exposure and after exposure, effects at the molecular, histological, individual and population levels were examined. Multigenerational exposure to 300 or 3000 ng TDCIPP/L for 60 days significantly decreased numbers of individuals, sizes of individuals, expressed as length and width of bodies, number of cilia, and depth and diameter of basal bodies of cilia, and up-regulated expressions of genes related to assembly and maintenance of cilia. Complete or partial recoveries of theoretical sizes of populations as well as sizes of individuals and expressions of genes were observed during the 60-day recovery period. Effects on number of cilia and depth and diameter of basal body of cilia were not reversible and could still be observed long after cease of TDCIPP exposure. Collectedly, and shown for the first time, multigenerational effects to T. thermophila were caused by exposure to environmentally relevant concentrations of TDCIPP. Also, there were multi-generational effects at the population level that were not caused by carry-over exposure to TDCIPP. The “permanent” alterations and their potential significance are discussed.

In order to understand multi-generational effects and changes of stress response by hexabromocyclododecane (HBCD) exposure, the animal model Caenorhabditis elegans was chosen for toxicity study. Multiple endpoints, including the physiological levels (growth, reproduction, and locomotion behaviors), stress-related gene expressions, reactive oxygen species (ROS) production and degree of cell apoptosis, were evaluated on exposed nematodes and their progeny . Prolonged exposure to HBCD at concentrations of 2 nM–200 nM caused adverse physiological effects in the parental generation (F0), and these effects were also observed in the offspring under HBCD-free conditions (F1). HBCD-induced toxicities could be transferred from parent to offspring. The integrated gene expressions profiles showed that exposure to HBCD at concentrations of 20–200 nM resulted in obvious changes in stress-related gene expressions, which were more increased in F0 generation than in F1 generation. The increased expressions were pronounced in several genes related to oxidative stress and cell apoptosis, e.g., hsp-16.2 , hsp-16.48 , sod-1 , sod-3 and cep-1 genes. Exposure to 200 nM of HBCD could significantly increase ROS production and degree of cell apoptosis in the F0 and F1 generations. Therefore, it was speculated that HBCD exposure induced oxidative stress and cell apoptosis, which resulted in the adverse physiological effects. This finding is helpful for understanding the multi-generational effects and evaluating the potential risk of HBCD.

Cytochrome P450 enzymes (cyt P450s) with an active center of iron protoheme are involved in most clinical drugs metabolism process. Herein, an electrochemical platform for the investigation of drug metabolism in vitro was constructed by immobilizing cytochrome P450 2D6 (CYP2D6) with cyt P450 reductase (CPR) on graphene modified glass carbon electrode. Direct and reversible electron transfer of the immobilized CYP2D6 with the direct electron transfer constant of 0.47 s–1 and midpoint potential of -0.483 V was obtained. In the presence of substrate tramadol, the electrochemical-driven CYP2D6 mediated catalytic behavior toward the conversion of tramadol to o-demethyl-tramadol was confirmed. The Michaelis–Menten constant (Kmapp) and heterogeneous reaction rate constant during the metabolism of tramadol were calculated to be 23.85 μM and 1.96 cm s–1, respectively. The inhibition effect of quinidine on CYP2D6 catalyze-cycle was also investigated. Furthermore, this system was applied to studying the metabolism of other drugs.

Tetrabromobisphenol A (TBBPA) is one of the most prevalently used brominated flame retardants. Due to its persistence, it is predominantly found in environmental matrices and has the potential to generate multi-generational toxicity. However, knowledge of its adaptive response or long-term residual effect in multi-generations, and molecular mechanisms remain understudied. In the current study, the model animal nematode Caenorhabditis elegans ( C. elegans ) was exposed to TBBPA at environmentally realistic concentrations (0.1–1000 μg L −1 ) for four consecutive generations (G 0 to G 3 ). Degenerative age-related multiple endpoints including lifespan, locomotion behaviors, growth, reproduction, oxidative stress-related biochemical responses, cell apoptosis, and stress related gene expressions were assessed in the continuous exposure generations (G 0 and G 3 ) and the discontinuously exposed generations (T 3 and T′ 3 ). The results showed that changes in degenerative age-related response monitored four generations varied in direction and magnitude depending on the TBBPA concentrations, and the response intensify ranked as G 0 > T′ 3 /G 3 > T 3 . TBBPA at 1 μg L −1 dosage was detected as the lowest observed effect concentration in multi-biomarkers. The underlying mechanism of aging phenotypes was that reactive oxygen species accumulation led to cell apoptosis regulated by gene ape-1 , and confirmed catalase enzyme and superoxide dismutase activity played a crucial role in the detoxification process of TBBPA at the molecular level. This study provided insights into the underlying mechanism of TBBPA-interfered longevity and its environmental multi-generational potential risks.

The reproductive toxicities over generations are essential to assess the long-term impacts environmental fluoroquinolone antibiotics (FQs). In the present study, the multi-generational effects of ofloxacin (OFL) and norfloxacin (NOR) on reproduction were studied on Caenorhabditis elegans from 9 successive generations (F1-F9). Results showed that OFL showed no effects in F1, stimulation in F2 to F4, and inhibition F5 to F9. The effects of NOR also showed oscillation between stimulation and inhibition across generations. Further biochemical analysis demonstrated that the reproductive toxicities of OFL and NOR were more closely connected with total cholesterol (TCHO), progesterone (P) and testosterone (T), than major sperm protein (MSP) and vitellogenin (Vn). Moreover, OFL and NOR also showed significant trans-generational reproductive toxicities in T4 and T4′, the great-grand-daughter of F1 and F9. Differences between T4 and T4′ and between OFL and NOR, indicated influences of multi-generational exposure and urged more exploration on different mechanisms between FQs.

Amitriptyline (AMI), one of the widely used tricyclic antidepressants (TCAs), has become a pharmaceutical contaminant frequently detected in aquatic ecosystems. However, the impacts of AMI exposure and underlying mechanisms on fish are still limited. In this study, adult zebrafish (F0) were exposed to AMI at 0 (control), 0.8, and 8 μg/L for 14 days. Subsequently, the exposed zebrafish were paired for spawning, and their offspring (F1) were reared in an AMI-free medium until 5 days post-fertilization (dpf). This study aimed to assess variations in behaviors and neurotransmitter levels in both the F0 (at the end of the 14-day exposure) and F1 generations (at 5 dpf). As a result, waterborne AMI exposure significantly reduced the locomotor activity, frequency of body contact, and duration of chase in F0 zebrafish, and resulted in notable changes in monoamine neurotransmitter levels in their brains. Parental exposure to AMI significantly elevated the heart rate and eye movement but reduced the locomotor activity in the F1 zebrafish, also along with significant changes in monoamine neurotransmitters and acetylcholine. Furthermore, significant correlations between the changes in behavioral traits and neurotransmitter levels were identified in both F0 and F1 generations. Our findings confirm the critical role of monoamine modulation in the neurobehavioral toxicity of AMI on zebrafish and their offspring, and emphasize the importance of paying attention to its multigenerational effects on fish.
