Single component Pluronic F127-lipoic acid hydrogels with self-healing and multi-responsive properties

Li Song,Bao Zhang,Ge Gao,Chunsheng Xiao,Gao Li

Herein, Pluronic F127 (PEO 99 -PPO 65 -PEO 99 ) end functionalized with α-lipoic acid (LA) (F127-LA) was synthesized and used for preparation of “single-component, multi-responsive” hydrogels through the photo cross-linking of LA components. The F127-LA hydrogels not only possessed thermo- and reduction- double responsiveness, but also demonstrated self-healing properties under UV irradiation due to the existence of dynamic disulfide bonds. The rheological and mechanical properties of the hydrogels can be adjusted by their concentration. The F127-LA hydrogels presented a swelling ratio decrease from 25 °C to 37 °C due to a lower critical solution temperature (LCST)-type thermo-responsive volume phase transition. The bovine serum albumin (BSA) was loaded into F127-LA hydrogels, and their drug release behavior can be regulated by surrounding temperature and reducing agent concentration. The volume shrinkage of the hydrogels promoted the sustained release of BSA above volume phase transition temperature. Cytocompatibility assay of L929 mouse fibroblasts showed that cellular metabolic activity were merely affected by F127-LA, even at concentrations up to 1000 mg L −1 . This study thus provided a very promising route for the preparation of multifunctional hydrogels.

Preparation and Properties of a Self-Healing, Multiresponsive Color-Change Hydrogel

Qiangwang Geng,Chong Zhang,Kaiwen Zheng,Junying Zhang,Jue Cheng,Wei Yang

Hydrogels are smart materials with a high moisture content, elasticity, and stimuli to pH and temperature and are widely used in drug delivery, wound dressings, tissue engineering, parting materials, and sensors. However, a hydrogel with a multiresponsive feature is desired and challenging to produce. Here, a novel multiresponsive color-change hydrogel, including pH response, light response, and alcohol response, was successfully fabricated by introducing Michler’s hydrol blue (MHB) into a semicrystalline hydrophobically associated hydrogel. The color-change function is obtained by a transformation between MHB and protonated Michler’s hydrol blue (PMHB) deduced by nucleophilic attack processes. An excellent swelling performance of 2010% of the hydrogel was confirmed by swelling, time-dependent rewetting data, and differential scanning calorimetry (DSC) measurements. Its color change was obvious and fast, within 1 min, when exposed to ultraviolet (UV) light, alcohol, or pH change. In addition, the tensile strength of the hydrogel reached up to 0.9 MPa with loading MHB by 2.0%. The fluorescence and self-healing performances were verified to be good due to the presence of a large delocalized π-bond and hydrogen bonds. This material fabricated by a facile process might have potential applications in visual fluorescence sensors.

Layered-Responsive Multivalent Tetrahedral DNA Framework-Decorated CRISPR-Cas12a Nanocapsule Enables Precise and Enhanced Tumor Chemotherapy

Ye Zhang,Wenbin Li,Siting Chen,Yu Zhang,Yitong Zhu,Fei Lan,Huijun Du,Rui Fan,Junfang Zhu,Weilun Pan,Bo Situ,Lei Zheng,Shihua Luo,Xiaohui Yan

The lack of selective tumor targeting and the high toxicity of conventional chemotherapy treatments remain major challenges in cancer therapy. Here, we develop a self-controlled DNA nanostructure-CRISPR-12a system, a triple-locked cascade tumor therapy nanocapsule (Tatna), for efficient and targeted tumor treatment. Tatna integrates structural DNA tetrahedrons (DTs) with high drug-loading capacity, Cas12a/crRNA ribonucleoprotein (Cas12a RNP), and doxorubicin (DOX) to enable multisite response for precise drug delivery and augmented tumor treatment. By incorporation of a nucleolin-targeting aptamer, Tatna achieves selective targeting and efficient tumor cell internalization. Encapsulation in pH-responsive poly l-lactic-co-glycolic acid (PLGA) nanocapsule ensures stable circulation and controlled release of both DOX and Cas12a until tumor-specific activation in the acidic microenvironment. The Cas12a RNP, triggered by APE1 mRNA overexpression in tumor cells, induces trans-cleavage of DTs, releasing DOX and Cas12a to transport into the nucleus and induce enhanced cell apoptosis. This self-regulating and multifunctional approach enhances the efficacy of chemotherapy while reducing off-target effects. Tatna’s programmable, tumor-specific delivery system represents a powerful strategy for advancing precision medicine and personalized cancer treatment.

A multi-responsive bidirectional bending actuator based on polypyrrole and agar nanocomposites

Taoping Wang,Mingtong Li,Hui Zhang,Yunyu Sun,Bin Dong

Smart actuators have recently been widely studied due to their potential applications in the fields of wearable devices, artificial muscles and microrobots. However, it is still challenging to develop smart actuators with a fast response time, large deformation, multi-stimuli response ability and programmable shape-changing capability. Here, we report a novel smart actuator, which consists of polypyrrole and agar. Due to the water absorbing ability, the photothermal effect and the protonation behavior originating from polypyrrole and agar, the resulting actuator is responsive to a variety of stimuli, including humidity, light, temperature, HCl and NH3 gas. In addition, the actuator exhibits fast responsiveness, controllable actuation and large bending. Interestingly, two external stimuli can be combined to activate the actuator, leading to continuous shape changes in a programmable fashion. Multi-stimuli responsive actuators have opened up great opportunities in a variety of applications. As a proof-of-concept, we have demonstrated their application as a walking device with cargo transportation and delivery capabilities.

Tough and multi-responsive hydrogels based on core-shell structured macro-crosslinkers

Du Gao-lai,Cong Yang,Chen Long,Chen Jing,Fu Jun

Novel hydrogels based on core-shell structured macro-crosslinkers were synthesized, which exhibited high toughness and multiple responsiveness. Sodium dodecyl sulfate (SDS) micelles mediated by NaCl were used to encapsulate hydrophobic stearyl methacrylate (C18) in the core, and hydrophilic 2-acrylamido-2-methyl-1-propanesulfonic (AMPS) monomers in the corona. Such core-shell micelles were simultaneously copolymerized with acrylamide monomers through free radical polymerization. As a result, hydrogels crosslinked by amphiphilic “poly(C18)-PAMPS” macro-crosslinkers were obtained. These hydrogels showed excellent tensile and compression strength and toughness. Cyclic compression loading-unloading tests demonstrated that the hydrogels were of outstanding fatigue resistance, and showed partial damage of energy dissipation mechanism. The damaged energy dissipation mechanism could be recovered at room temperature and the recovery could be accelerated at elevated temperatures. The hydrogels were sensitive to the change in pH and ion strength, showing reversible swelling/deswelling behaviors.

Remotely Controlled Light/Electric/Magnetic Multiresponsive Hydrogel for Fast Actuations

Xianshuo Wei,Qijun Wu,Lian Chen,Ye Sun,Lin Chen,Chunmei Zhang,Shanshan Li,Chunxin Ma,Shaohua Jiang

As a kind of soft smart material, hydrogel actuators have extensive development prospects, but it is still difficult for these actuators to integrate multiresponsiveness, multiple remote actuation, high strength, fast responsiveness, and programmable complex deformation. Herein, we have explored an anisotropic bilayer hydrogel actuator with an Fe3O4/co-poly(isopropylacrylamide-4-benzoylphenyl acrylate) [Fe3O4/P(NIPAM-ABP)] active layer and an isotropic conductive adhesive (ICAs) passive layer based on the layer-by-layer method. Benefiting from the fibrosis and porosity of the Fe3O4/P(NIPAM-ABP) hydrogel, the ICAs-Fe3O4/P(NIPAM-ABP) hydrogel actuator has excellent mechanical strength (tensile strength of 3.1 ± 0.3 MPa) and response speed (temperature (45 °C): bending speed of 2400.3°/s; near-infrared (NIR) light: bending speed of 356.4°/s; electricity (2 V): bending speed of 180°/s; water (10 °C): recovery speed of 30.0°/s). In addition, the good photothermal properties and magnetic conductivity of Fe3O4 nanoparticles provide precise remotely controllable light- and magnetic-actuated properties for the hydrogel actuator. The Ag microsheets with excellent conductivity (1.4 × 104 S/cm) provide remotely controllable electrical-actuated property for the hydrogel actuator. Combined with the responsiveness of P(NIPAM-ABP), the actuator can achieve short-range actuation including temperature-, ethanol-, and salt-responses. More importantly, it can achieve remote actuation including light, electrical, and magnetic responses. Finally, the Fe3O4/P(NIPAM-ABP) fibers can provide excellent anisotropic structures for the actuator to achieve precise deformational programmability. Inspired by some phenomena in nature, several actuating devices with the above characteristics have been successfully developed. This study can provide a general method for multifunctional anisotropic hydrogel actuators and will provide a new strategy for exploring smart materials suitable for complex bioinspired systems.

A multi-stimuli-responsive nanochannel inspired by biological disulfide bond

Xiaofang Wang,Huiming Wang,Meining Zhang

Disulfide bonds exist widely in channel protein and play an essential role in matter exchange and signal transduction (e.g., rhodopsin, canonical transient receptor potential 5 (TRPC5)). The research on disulfide bond in nanochannel is significant for the cognition of their biological functions. However, the fragility of biological channel limits the in-situ study and practical application. Herein, an innovative biologically-inspired artificial nanochannel based on disulfide bond (NCDS) with excellent durability, adjustable surface property is proposed. The constructed NCDS has a multi-response to UV-light, thiol (e.g., cysteine (Cys)) or pH stimulation, and can obtain reversibility after regulation by hydrogen peroxide (H 2 O 2 ) or H + . The biomimetic NCDS shows great potential in biosensor and intelligent response design. This study also shines new light to channel protein based on disulfide bond that despite the nanochannel has specificity, it will be modulated by the change of nature environment, such as UV-light and chemical microenvironment (e.g., redox state and pH), which might be the reason of some disease.

Synthesis and multiple responsiveness study of Poly(N-isopropylacrylamide) containing azopyridine

Xiaoqiang Xue,Yangjing Chen,Xuepei Miao,Jian Liu,Yi Ding,Yuheng Zhang,Yunkai Sun,Wenyan Huang,Qimin Jiang,Hongting Pu

Environmentally sensitive polymer materials exhibit diverse properties due to their ability to respond to external environmental changes such as light, temperature, pH, and et al. These responses lead to corresponding alterations in their structure or state, resulting in unexpected performance. In this study, azo pyridine groups were introduced into the end groups and side chains of PNIPAM to synthesize two distinct polymer structures (PNIPAM-AZO and P (NIPAM- co -PAZO- co -OEGMA). This integration enables the polymers to respond to multiple stimuli, including temperature, light, pH, and gas (CO 2 ). A comprehensive investigation of the properties of these multi-responsive polymer materials was conducted. The findings reveal that the presence of azo pyridine units enables both polymers to modulate the (lower critical solution temperature) LCST of PNIPAM through UV, pH, and CO 2 , with a maximum temperature adjustment of 13 °C when CO 2 is introduced. This modulation range is influenced by the azo pyridine content in the polymer. Notably, when azo pyridine is present only at the polymer end group, the maximum LCST regulation range is limited to 3 °C. Moreover, the LCST of the two polymers can also be adjusted by pH. In the presence of carbonate ions, the protonation of azo pyridine by carbonic acid increases the solubility of the polymer and enhances the LCST. Introducing argon gas removes CO 2 , thereby restoring the polymer's solubility and LCST to their neutral condition levels. By alternately introducing CO 2 and argon gas, the LCST of the polymer exhibits cyclic changes.

A size-tunable and multi-responsive nanoplatform for deep tumor penetration and targeted combinatorial radio-/chemotherapy

Fatima Zohra Dahmani,Danni Zhong,Yuchen Qi,Aboubaker El G. Dahmani,Tingting Xie,Bo Zhou,Wanli Li,Ke Yao,Lei Li,Min Zhou

Nowadays, the design of multistimuli-responsive and deep penetrating nanotherapeutics remains an auspicious means for improving the efficiency of anticancer therapeutics. Herein, we develop a targeted size-tunable nanosystem that would concurrently and uniformly release its therapeutic cargo into tumor cells by exploiting both extracellular and intracellular signals. This nanosystem (HPDAu) is composed of self-assembled pH-responsive DOX–PAMAM (PD) conjugates and ultrasmall PAMAM-stabilized gold nanoparticles (AuNPs), incorporated into a hyaluronidase-responsive hyaluronic acid nanoshell. HPDAu nanoparticles with an initial particle size of ∼100 nm could disassemble into tiny cationic nanostructures (∼5 nm, PD and AuNPs) upon incubation with HAase, and showed a burst drug-release under tumor microenvironment-mimicking conditions (HAase and acidic conditions). Such characteristics resulted in significantly improved tumor penetration both in vitro and in vivo, along with a higher cellular uptake efficiency and tumor accumulation as compared to free drug solutions. Taking advantage of these features along with the chemotherapeutic effect (DOX) and AuNP/DOX-induced radiosensitization, HPDAu plus radiotherapy (RT) treatment resulted in a substantial increase of apoptotic and cytotoxic effects against 4T1 cells. More importantly, the radio-/chemotherapeutic feasibility of HPDAu was further validated in a 4T1 orthotropic model, revealing a prominent antitumor efficacy and reduced side-toxicity compared to monotherapy and free drug solutions. Therefore, this versatile nanoplatform with active targeting, size tunability and radio-/chemotherapeutic features could be a promising tool for tumor combinatorial therapy.

Multi-responsive nanocomposite hydrogels with high strength and toughness

Jingli Yang,Shuang Liu,Ying Xiao,Guorong Gao,Yuanna Sun,Qingzhong Guo,Jiangyu Wu,Jun Fu

Multi-responsive hydrogels with high strength have great significance for potential applications in smart soft devices. However, it remains a challenge to incorporate multiple responsive moieties with energy dissipation mechanisms. Herein, multi-responsive nanocomposite hydrogels with high compressive strength and toughness were synthesized via in situ copolymerization of N-isopropylacrylamide (NIPAM) and acryloyloxyethyltrimethyl ammonium chloride (DAC) in an aqueous dispersion of exfoliated LAPONITE® RDS with a minute amount of N,N′-methylenebisacrylamide (MBAA) as a crosslinker. The combined use of clay and MBAA is demonstrated to be favorable for the high strength and toughness, and helped in avoiding precipitation of clay nanosheets, which otherwise occurred upon addition of cationic DAC. The effect of the NIPAM/DAC molar ratio, MBAA and clay contents on the properties of the hydrogels has been systematically investigated. Compression tests showed a compressive strength up to 6.2 MPa, with fracture strain higher than 90%. The presence of ionic DAC moieties in the hydrogels rendered a very high swelling ratio up to 40 (g g−1). These hydrogels were responsive to temperature changes due to the presence of NIPAM units, with the transition temperature (Ttrans) dependent on the molar ratio of NIPAM and DAC monomers. The internal electrostatic repulsion of the NIPAM/DAC copolymer network changed upon exposure to solutions with different pH and/or ion strength. Cyclic swelling–shrinking was demonstrated by shuttling the gels between pure water and 0.1 mol L−1 NaCl solution.

Aerogel-Directed Energy-Storage Films with Thermally Stimulant Multiresponsiveness

Jing Lyu,Guangyong Li,Meinan Liu,Xuetong Zhang

Phase change materials offer enormous potential for thermal energy storage due to their high latent heat and chemical stability. Researchers have developed numerous innovative strategies to overcome the leakage of organic phase change materials and enhance thermal performance. However, the manufacture of form-stable, free-standing energy storage films based on phase change materials with high latent heat remains difficult. Therefore, the present study focused on the production of free-standing, form-stable energy storage films with high phase-change enthalpy and thermally stimulant multiresponsiveness from the simple composite of paraffin with a polytetrafluoroethylene/silica (PTFE/SiO2) aerogel framework, where paraffin was effectively confined in PTFE/SiO2. The resulting paraffin/PTFE/SiO2 films exhibited a large phase change enthalpy (128 J/g) at a paraffin content of 62.8 wt %. The temperature-dependent wettability, transmittance, and mechanical properties of this composite film have also been investigated.

Multiresponsive MXene (Ti3C2Tx)-Decorated Textiles for Wearable Thermal Management and Human Motion Monitoring

Chang Ma,Qi Yuan,Haishun Du,Ming-Guo Ma,Chuanling Si,Pengbo Wan

Over the past few years, wearable electronics and smart textiles have seen tremendous growth in both academia and industries. However, it is still a challenge to prepare robust, flexible, wearable, and multiresponsive textile electronics. A newly blooming two-dimensional (2D) transition-metal carbide/nitride (MXene) is regarded as an ideal active material to build multifunctional electronics due to its intriguing properties. Herein, a hydrophobic and multifunctional textile composite (Si-MAP) was prepared by decoration of conductive MXene nanosheets onto air-laid paper, followed by wrapping with poly(dimethylsiloxane) (PDMS). These obtained smart textiles exhibited excellent electronic/photonic/mechanical triresponsive properties: Si-MAPs could reach high equilibrium temperatures (104.9 and 118.7 °C) under quite low power illumination (1.25 W cm–2) and working voltage (4 V). The Si-MAP pressure sensor exhibited high sensitivity and rapid response time (30–40 ms), which can capture a wide range of human movements. Moreover, the thin PDMS layer not only rendered the textile composites hydrophobic but also improved the stability and adaptation for daily use. Remarkably, the hydrophobic Si-MAPs have maintained the advantages of breathability and washability, which make them suitable for wearing. Thus, this smart Si-MAP textile provides a reference for the study of the next generation of light, portable, and wearable textile-based electronic devices.
