Construction of Dual-Functional Emulsion Micelle Interface With Ionic and Reactive Groups

Ziyun Li,Qiwei Zheng,Menghan Zhong,Chao Wang,Shengwei Fang,Renfeng Zhou,Min Xia,Yanrong Jia,Juan Zhang,Peng Ye

Internal emulsification emulsions are environmentally friendly and do not emit volatile organic compounds (VOCs). However, their micelle interface, dominated by ionic hydrophilic groups, hinders cross-linking reactions and affects performance. We proposed a dual-functional micellar interface (DFMI) that distributes both ionic and reactive hydrophilic groups, enhancing cross-linking performance without significantly compromising emulsification properties. In this study, a series of sugar-based hyperbranched waterborne polyurethane emulsions were synthesized with 2,2-dimethylolpropionic acid (DMPA) as the ionic hydrophilic moiety. Dopamine (DA) was introduced at varying levels as the reactive hydrophilic moiety for terminal modification. The results showed that monomolecular micelles and vesicular micelles could be formed by symmetric and asymmetric modification of terminal DA. Symmetric modification tended to form unimolecular micelles, while asymmetric modification tended to form vesicular structures. Adhesion tests conducted after cross-linking with gelatin showed that the asymmetrically modified adhesives with seven-layer structured DA exhibited better cross-linking degrees. Adhesion performance and swelling tests confirmed that the asymmetric structure exposed more terminal reactive groups, enhancing the utilization of the modified groups, as indicated by dynamic light scattering (DLS) for emulsion particle size and surface zeta potential. This study successfully prepared hyperbranched polyurethane emulsions with tunable micelle morphology, balancing emulsification ability, and cross-linking degree, supporting the controlled emulsification of hyperbranched polymers and the development of bio-based adhesives.

Aptamer and molecularly imprinted polymer: Synergistic recognition and sensing of dopamine

Mingmei Shen,Xianwen Kan

Specifically sensitive determination of dopamine (DA) from complex real samples is very important since DA plays a major role in the function of central nervous, which presents the necessary to develop an effective sensor for the assay of DA. In this work, a dual recognition strategy was developed through introducing DA-aptamer and molecularly imprinted polymer (MIP) on gold nanoparticles (AuNPs) and reduced graphene oxide (rGO) modified electrode. The selectivity and sensitivity of the prepared sensor were investigated upon the current response of DA itself to better represent the performances of the sensor. Compared with the single recognition element (MIP or aptamer) based sensors, the prepared dual recognition sensor displayed the highest selectivity to DA, which facilitated the proposed sensor for the detection of DA in real samples. Owing to the high surface and good conductivity of the modified AuNPs and rGO, the sensor showed a wide linear range and a low limit of detection of 5.0 × 10 -8 – 1.0 × 10 -5  mol/L and 4.7 × 10 -8  mol/L for the detection of DA, respectively. The proposed strategy of aptamer and MIP based sensor for electro-active target assay may be used as a general way for the development of other dual recognition based sensors.

A bioinspired Janus polyurethane membrane for potential periodontal tissue regeneration

Yushui He,Xiao Wang,Yuanyuan He,Xin Zhao,Jingjing Lin,Yuan Feng,Jie Chen,Feng Luo,Zhen Li,Jiehua Li,Hong Tan

Guided tissue regeneration (GTR) is the main therapeutic method for periodontal tissue regeneration. The key to the GTR strategy is the membrane which can assist the reconstruction of bone tissue in the periodontal defect and prevent the migration of epithelium and fibroblasts to the defect. However, the existing periodontal membrane cannot effectively promote periodontal tissue regeneration due to the limited bioactivity and physicochemical function. Here, we developed a bioinspired degradable polyurethane membrane with Janus surface morphology by integrating bioactive dopamine (DA) and an antibacterial Gemini quaternary ammonium salt (QAS). The Janus surface of the membrane is fabricated through spontaneous microphase separation, resulting from the different migration of functional segments between the air-contact upper surface with enriched antibacterial QAS and the substrate-contact bottom with enriched bioactive DA. The smooth surface of the upper membrane used to face the soft tissues can reduce cell adhesion to suppress the migration of fibroblasts, while the rough surface with a topological micro-pit structure of the bottom side facing the bone has excellent function of autonomic mineralization and cell adhesion to promote bone tissue reconstruction. In addition, the membrane containing the antibacterial QAS shows excellent antibacterial effect on common oral pathogens, such as S. aureus and S. mutans. Moreover, the specific dopamine group also endows the membrane with excellent antioxidant efficiency. In vivo research shows that this Janus polyurethane membrane can effectively promote periodontal tissue regeneration in a rat periodontal defect model. Combined with its excellent mechanical properties and biocompatibility, the polyurethane membrane is a promising material for potential periodontal tissue regeneration.

Lignocellulose and polydopamine composite superhydrophobic and photothermal membrane: A green innovative solution for oil/water separation and high-viscosity oil adsorption

Dongsheng Song,Congcong An,Zhe Zhang,Yu Qiu,Zhimin Zhao,Dingqiang Zheng,Ming Zhang

A superhydrophobic CNF/PDA composite membrane has been developed to revolutionize oily wastewater treatment for achieving high efficiency, cost-effectiveness, and environmental sustainability. Inspired by the adhesive properties of mussels and the self-cleaning mechanism of lotus leaves, this innovative material is fabricated through a novel lyophilization and spray modification process using cellulose nanofibers (CNF), dopamine hydrochloride (DA), and octadecylamine (ODA). The membrane is synthesized via an aerobic, weakly alkaline self-polymerization process, which facilitates the formation of robust covalent and non-covalent bonds between the catecholamine structure of DA and CNF. ODA is integrated through hydrogen bonding and Schiff base reactions with polydopamine (PDA), enhancing structural stability. The resulting membrane demonstrates a tensile strength of 2.9 MPa, alongside exceptional separation efficiency (98.5 %) and flux rates (3745 L·m −2 ·h −1 ). Even after five consecutive water-in-oil (W/O) emulsion separation cycles, a separation efficiency exceeding 91.8 % is maintained, with hydrophobicity and structural integrity preserved. Durability tests confirm its robustness, with 98.5 % of initial capacity retained after ten adsorption-desorption cycles. The membrane's porous architecture and photothermal brown-black hue synergistically boost light-to-heat conversion, facilitating swift thermal response and efficient high-viscosity oil uptake. This demonstrates viable applications as a sustainable solution for oily wastewater treatment, promoting greener industrial application.

Bisubstrate multi-colorimetric assay based on the peroxidase-like activity of Cu2+-triethylamine complex for copper ion detection

Hang-Yu Zhou,Tong-Qing Chai,Li-Jing Peng,Wei-Yi Zhang,Tao Tian,Hao Zhang,Feng-Qing Yang

A bisubstrate colorimetric system was developed for rapid and multi-color assay of copper ion (Cu 2+ ). Cu 2+ -triethylamine (Cu 2+ -TEA) complex with the peroxidase-like activity can catalyze the 3,3′,5,5′-tetramethylbenzidine (TMB) to produce oxidized TMB (TMB + , color in blue) and dopamine (DA) to oxidized DA (oxDA, color in orange) in the presence of hydrogen peroxide, which can obtain a three-color colorimetric result due to the couple of complementary colors of blue and orange. The determination of Cu 2+ is achieved based on the absorbance value at 460 nm of the composite peak of TMB + and oxDA. After systematically optimizing the reaction conditions, the developed bisubstrate colorimetric method exhibits high sensitivity for the detection of Cu 2+ (limit of quantitation (LOQ) is 0.0625 μM) with a wide linear range of 0.0625–8.0 μM (R 2  = 0.991). Meanwhile, the reaction solution shows an orange, brown, and green color change with the increase in Cu 2+ concentration, which can realize the semi-quantitatively detection by the naked eyes. Furthermore, TMB was also used as a single substrate for Cu 2+ detection with a linear range of 0.5–9.0 μM (R 2  = 0.994), and the LOQ is determined to be 0.5 μM. The LOQ of bisubstrate system is 8 times lower than that of single substrate system, indicating a higher sensitivity for the detection of Cu 2+ using the bisubstrate colorimetric system. In addition, the developed bisubstrate colorimetric method exhibits excellent selectivity for Cu 2+ detection, which was applied in the determination of Cu 2+ in lake water and soil samples, and satisfactory spiked recoveries were obtained.

Janus functional electrospun polyurethane fibrous membranes for periodontal tissue regeneration

Jingjing Lin,Yushui He,Yuanyuan He,Yuan Feng,Xiao Wang,Lei Yuan,Yanchao Wang,Jie Chen,Feng Luo,Zhen Li,Jiehua Li,Hong Tan

Guided tissue regeneration (GTR) technique with GTR membranes is an efficient method for repairing periodontal defects. Conventional periodontal membranes are just as physical barriers in resistance to the growth of fibroblasts, epithelial cells, and connective tissue, but they lack the capacity to facilitate the regeneration of periodontal tissue. To address this issue, the exploitation of novel GTR membranes with bioactive functions based on therapeutic requirements is critical. Here, we exploited a biodegradable bilayer polyurethane fibrous membrane by uniaxial electrostatic spinning to construct two sides with Janus properties by integrating the bioactive molecules dopamine (DA) and antimicrobial Gemini quaternary ammonium salt (QAS). The DA-containing side, located on the inside of the injury, can effectively promote cell adhesion and mesenchymal stem cell growth, as well as support mineralization and antioxidant properties, which are beneficial for bone regeneration. The QAS-containing side, located on the outer surface of the injury, endows antibacterial properties and limits fibroblast adhesion and growth on its surface due to its strong hydrophilicity. In vivo study demonstrates that the Janus polyurethane fibrous membrane can significantly promote the regeneration of periodontal defects in rats. The combination of its superior mechanical properties and biocompatibility, this polyurethane fibrous membrane has potential applications in the field of periodontal regeneration.

Liangxue Tongyu Prescription exerts neuroprotection by regulating the microbiota-gut-brain axis of rats with acute intracerebral hemorrhage

Yingying Sun,Dandan Zhou,Anlan Liu,Yu Zhou,Yang Zhao,Yuan Yuan,Weifeng Guo,Jianxiang Li

Liangxue Tongyu Prescription (LTP) is a classic herbal formula for treating acute intracerebral hemorrhage (AICH) in China. Previous studies have shown that LTP significantly ameliorates neurological impairments and gastrointestinal dysfunction in patients with AICH. However, the underlying molecular mechanism remains unclear. The aim of this study is to investigate whether LTP exerts its neuroprotective effect on AICH rats through the microbiota-gut-brain axis and explore its potential underlying mechanism. In the current study, AICH models were established by injecting autologous whole blood into the right caudate nucleus of rats. Behavioural and pathological evaluations demonstrated that LTP ameliorated neuronal and intestinal damage in AICH rats. Analysis via western blot, quantitative real-time PCR, immunohistochemistry (IHC) and tunel staining indicated that LTP upregulated the expression of brain-derived neurotrophic factor (BDNF) and nerve growth factor(NGF) and reduced neuronal cell apoptosis. Additionally, 16S rDNA sequencing revealed that LTP mitigated dysbiosis of intestinal microbiota in AICH rats. LTP increased the levels of noradrenaline (NA), dopamine (DA), glutamate (GLU) and modulated brain-gut peptides such as gastrin (GAS), motilin (MTL), ghrelin in AICH rats. Furthermore, LTP enhanced vagus nerve discharge. In summary, this research provides evidence suggesting that LTP’s influence on AICH may involve modulation of the microbiota-gut-brain axis, offering a potential scientific rationale for its therapeutic efficacy in improving outcomes of AICH.

Guo Zhi-Yong,Yao Qiu-Hong,Zheng Wen-Hui,Jiao Rui-wen,Zhang Chen,Zhang Li,Ye Ting-Xiu,Chen Xi

New approaches for the engineering of well-defined, pore modality, and multi-chemical functionality nanocomposites are crucial to generate the next generation of functional materials with recoverable and easy preparation properties. Here, a catalyst and heat free polymerization reaction is exploited and fabricated zwitterionic system around magnetic nanoparticles. N-aminoethyl piperazine propane sulfonate (AEPPS) and dopamine (DA) are introduced as the zwitterionic system, which provided abundant zwitterionic groups (NH 2 , SO 3 − , N + ) and strong adhesion and various oxidation state properties. And that, the zwitterionic engineering will assemble between AEPPS and DA whereby Schiff base formation or Michael type addition. Whereafter, a series of sophisticated array of microscopic, spectroscopic, and structure techniques verify the formation of highly crosslinking internal zwitterionic architectures, well-defined core–shell structure, and better porosity. The zwitterionic structure–function relationships and striking porous structure are explored in a multi-interaction adsorption assay. The adsorption capacity of the magnetic nanocomposites was 1065.8 mg/g. And that, the system exhibited with hydrophilic-hydrophobic activity towards glycoprotein and better performance to bioactive protein (Ig-G) isolation form human whole blood sample. The synergistic enhancement interaction in hydrophilic target enrichment, easy preparation, and soft substrate properties of the AEPPS-DA zwitterionic materials make them intriguing candidates for sustainable biomedical loading and chromatographic separation.

Study of the Neurotransmitter Changes Adjusted by Circadian Rhythm in Depression Based on Liver Transcriptomics and Correlation Analysis

Peng Wang,Xiaoxia Gao,Fang Zhao,Yao Gao,Kexin Wang,Jun-Sheng Tian,Zhenyu Li,Xue-Mei Qin

Depression has drawn increasing attention from the public around the world in recent years. Studies have shown that liver injury caused by chronic stress is relevant to depression and neurotransmitter changes. It is essential to clarify the relationship between neurotransmitter changes and hepatic gene expression in depression. In this study, we used the chronic unpredictable mild stress (CUMS) model combined with UHPLC–MS to explore the changes of neurotransmitters in serum and hippocampus and to decipher the differential gene expression in the liver by using the RNA-Seq combined with multivariate statistical analysis. Compared with the control group, the levels of neurotransmitters including 5-hydroxytryptamine (5-HT), acetylcholine, glutamate (Glu), and dopamine (DA) in the hippocampus and 5-HT, norepinephrine, γ-aminobutyric acid (GABA), and 5-hydroxyindoleacetic acid in serum were significantly changed in the CUMS rats. The results of liver transcriptomic analysis and correlation analysis showed that the Glu, DA, 5-HT, and GABA were impacted by 68 liver genes which were mainly enriched in three pathways including circadian rhythm, serotonergic synapse, and p53 signaling pathway. The expressive levels of clock genes and serotonergic synapse genes were validated by using q-PCR, and the diurnal rhythms of neurotransmitters were validated by in vivo hippocampus microdialysis. The CUMS stressors might cause phase advance of Glu and GABA by adjusting clock genes. The transcriptomic technique combined with correlation analysis and in vivo microdialysis could be used to discover comprehensive pathways of depression. It provides a new strategy for the rational assessment of the mechanism of disease.

Mussel-inspired quaternary composite hydrogels with high strength and high tissue adhesion for transdermal drug delivery: Synergistic hydrogen bonding and drug release mechanism

Yu Cai,Chao Liu,Kaihua Gong,Hui Li,Haoyuan Song,Yang Zhang,Dawei Ding,Jie Liu,Jianpeng Guo,Liang Fang

Low adhesion to skin remains a significant challenge for most hydrogel based transdermal drug delivery systems. Meanwhile, most of the methods to improve bio-adhesion often ignore the effect of the cohesion. Inspired by the adhesion mechanism of natural mussels and the synergistic balance theory based on cohesion and adhesion, we designed a poly (acrylamide- co -hydroxyethyl acrylate)-polydopamine-polyvinylpyrrolidone (PAHDP) quaternary composite hydrogel to enhance the cohesion and adhesion through the synergistic effect of multiple hydrogen bonds to achieve strong bio-adhesion to skin. The structure of PAHDP was characterized by FTIR, 1 H NMR, SEM and TGA . The peeling adhesion force of PAHDP on porcine skin was improved by 9-fold compared with PAH hydrogel. At the same time, the adhesion force of drug-loaded PAHDP was 1.3 times that of Voltaren® EX . The rheological test further demonstrated that the synergistic hydrogen bonding can significantly improve the PAHDP cohesion. In addition, in vitro release, skin permeation and pharmacokinetics test of PAHDP hydrogel containing diclofenac sodium (DIC) showed that monomer dopamine (DA) could promote drug release. FTIR and molecular modeling were utilized to further demonstrate the enhanced drug release mechanism of DA. MTT assay, H&E staining and skin erythema analysis demonstrated that PAHDP was non-irritant to the skin. In conclusion, the innovatively prepared PAHDP hydrogel exhibits high skin adhesion, excellent mechanical properties, non-irritation and high drug release ability. This study also provides a simple and efficient strategy and theoretical basis for the preparation of high adhesion and high drug release hydrogels for transdermal drug delivery system.
