High-performance piezoelectric materials are widely used in electromechanical applications such as sensors, actuators, and transducers. Herein, improvement in electrical properties of Pb(Ni 1/3 Nb 2/3 )O 3 −Pb(In 1/2 Nb 1/2 )O 3 −PbTiO 3 relaxor ferroelectrics by modulating the polymorphism of nano-scale domain is reported, and piezoelectric coefficients d 33 and d 33 * as high as 948 pC/N and 1108 pm/V, respectively, are achieved. The high piezoelectric response is elucidated by combining cryogenic dielectric , Rayleigh analysis, and scanning electron microscopy with piezoresponse force microscope. During the transition from the tetragonal to rhombohedral phase, the morphology of the ferroelectric domains changes significantly, mesoscopic domain destruction is observed, and thus nano-scale domains appear as an extrinsic contribution of piezoelectricity. In addition, nano-scale domains promote polarization rotation, thus, improving the piezoelectric response. The improved piezoelectric performance demonstrates good thermal stability, retaining the inverse piezoelectric coefficient of approximately 1000 pm/V near 70 °C. This work provides a good example of improving ceramics' piezoelectric response by modulating the polymorphism of nanoscale domains.

Despite the enormous efforts devoted to the research of titanium dioxide (TiO2), controlling TiO2 polymorphism, size and morphology still represents an interesting challenge since its stabilities, transitions and properties vary with them. Herein, we report a novel selective-synthesis of high purity anatase (A), rutile (R) and brookite (B) phase TiO2 from solution under mild hydrothermal conditions. By adjusting the concentrations of chloroacetic acid, urea and sodium hydroxide, A-TiO2 nanocrystals, R-TiO2 nanorods and B-TiO2 hollow spheres and nanorods can be easily produced as investigated by XRD and TEM characterisation. Based on time-varied experiments, two distinct phase transitions were observed under different hydrothermal conditions, namely the initially formed A-TiO2 nanocrystals would transform into R-TiO2 nanorods with augmented crystalline size under acidic conditions while they transformed into B-TiO2 hollow spheres under basic conditions. Moreover, polymorph formation and transition mechanism were explored in detail based on systematic investigation into the effects of synthetic parameters on the products. Finally, the spectra and photocatalytic properties of the three polymorphs were investigated and discussed. The synthetic approach reported here allows for full control over polymorph selection in TiO2 crystallization and provides new insights in this area.

We propose a novel and cyclic synthetic approach for controlling crystal polymorphs of CaCO3 by using green CO2-responsive switchable solvents which acted as both the CO2 capturer for the carbonate source and the polymorphisms director. Five solvents were employed, and various reaction conditions such as calcium resources, calcium concentration, reaction temperature, and reaction time were investigated. Results show that this developed framework permits producing any crystalline CaCO3 phases including metastable vaterite and aragonite in pure phases by selecting a suitable solvent and adjustment of the reaction conditions. Furtherly, the mechanism study demonstrates that the solvents attach on the surface of the primary nanoparticles to selectively control and direct the growth of any specific polymorph phases. Eventually, the nuclei are self-assembled into an oriented geometry, allowing the growth and stability of specific crystals; as such, spherical vaterite, rods, and shuttle-like aragonite crystals can be obtained. This new configuration would be an appropriate and an efficient method to apply to large-scale production, therefore, a promising process attributed to complete solvent recovery and regeneration of the initial reactants, thus being an environmentally risk-free route.

Graphical The extraordinary coordination versatility of Th 4+ cation and the unexpected bridging role of formate allow polymorphism control of thorium-organic frameworks (TOFs) with five distinct phases, wherein the highest degree of interpenetration and porosity for TOFs have been achieved. The resulting TOFs exhibit considerably different adsorption behaviors toward organic dyes, thus suggesting that TOFs represent an excellent platform for structure-property relationship study. Polymorphism control of metal-organic frameworks is highly desired for elucidating structure-property relationships, but remains an empirical process and is usually done in a trial-and-error approach. We adopted the rarely used actinide cation Th 4+ and a ditopic linker to construct a series of thorium-organic frameworks (TOFs) with a range of polymorphs. The extraordinary coordination versatility of Th 4+ cations and clusters, coupled with synthetic modulation, gives five distinct phases, wherein the highest degree of interpenetration (threefold) and porosity (75.9 %) of TOFs have been achieved. Notably, the O atom on the capping site of the nine-coordinated Th 4+ cation can function as a bridging unit to interconnect neighboring secondary building units (SBUs), affording topologies that are undocumented for other tetravalent-metal-containing MOFs. Furthermore, for the first time HCOOH has been demonstrated as a bridging unit of SBUs to further induce structural complexity. The resulting TOFs exhibit considerably different adsorption behaviors toward organic dyes, thus suggesting that TOFs represent an exceptional and promising platform for structure-property relationship study.

Relaxor antiferroelectrics are considered promising candidate materials for achieving excellent energy storage capabilities. However, the trade-off between high recoverable energy density and high efficiency remains a major challenge in relaxor antiferroelectrics for practical applications. Herein, guided by phase-field simulation, we propose a strategy of designing polymorphic heterogeneous shell in core-shell dual-phase dielectrics to synergistically control micro and local heterostructures, resulting in comprehensive improvements in breakdown electric field, polarization fluctuation and saturation behaviors. Leveraging the core-shell effect and polarization heterogeneity, an ultrahigh recoverable energy density of 12.7 J cm -3 and an impressive efficiency of 87.2% are achieved in lead-free relaxor antiferroelectrics, making a performance breakthrough in core-shell dielectrics. This work opens up a new avenue to efficiently develop high-performance energy storage dielectrics and is expected to be popularized in other fields.

Introduction Glioblastoma multiforme (GBM), a highly invasive and prognostically challenging brain cancer, poses a significant hurdle for current treatments due to the existence of the blood-brain barrier (BBB) and the difficulty to maintain an effective drug accumulation in deep GBM lesions. Methods We present a biomimetic nanoplatform with angiopep-2-modified macrophage membrane, loaded with indocyanine green (ICG) templated self-assembly of SN38 (AM-NP), facilitating active tumor targeting and effective blood-brain barrier penetration through specific ligand-receptor interaction. Results Upon accumulation at tumor sites, these nanoparticles achieved high drug concentrations. Subsequent combination of laser irradiation and release of chemotherapy agent SN38 induced a synergistic chemo-photothermal therapy. Compared to bare nanoparticles (NPs) lacking cell membrane encapsulation, AM-NPs significantly suppressed tumor growth, markedly enhanced survival rates, and exhibited excellent biocompatibility with minimal side effects. Conclusion This NIR-activatable biomimetic camouflaging macrophage membrane-based nanoparticles enhanced drug delivery targeting ability through modifications of macrophage membranes and specific ligands. It simultaneously achieved synergistic chemo-photothermal therapy, enhancing treatment effectiveness. Compared to traditional treatment modalities, it provided a precise, efficient, and synergistic method that might have contributed to advancements in glioblastoma therapy.

Background Glioblastoma multiforme (GBM) is a highly aggressive brain tumor with a poor prognosis. Current treatment options are limited and often ineffective. CAR T cell therapy has shown success in treating hematologic malignancies, and there is growing interest in its potential application in solid tumors, including GBM. However, current CAR T therapy lacks clinical efficacy against GBM due to tumor-related resistance mechanisms and CAR T cell deficiencies. Therefore, there is a need to improve CAR T cell therapy efficacy in GBM. Methods We conducted large-scale CRISPR interference (CRISPRi) screens in GBM cell line U87 MG cells co-cultured with B7-H3 targeting CAR T cells to identify genetic modifiers that can enhance CAR T cell-mediated tumor killing. Flow cytometry-based tumor killing assay and CAR T cell activation assay were performed to validate screening hits. Bioinformatic analyses on bulk and single-cell RNA sequencing data and the TCGA database were employed to elucidate the mechanism underlying enhanced CAR T efficacy upon knocking down the selected screening hits in U87 MG cells. Results We established B7-H3 as a targetable antigen for CAR T therapy in GBM. Through large-scale CRISPRi screening, we discovered genetic modifiers in GBM cells, including ARPC4 , PI4KA , ATP6V1A , UBA1 , and NDUFV1 , that regulated the efficacy of CAR T cell-mediated tumor killing. Furthermore, we discovered that TNFSF15 was upregulated in both ARPC4 and NDUFV1 knockdown GBM cells and revealed an immunostimulatory role of TNFSF15 in modulating tumor-CAR T interaction to enhance CAR T cell efficacy. Conclusions Our study highlights the power of CRISPR-based genetic screening in investigating tumor-CAR T interaction and identifies potential druggable targets in tumor cells that confer resistance to CAR T cell killing. Furthermore, we devised targeted strategies that synergize with CAR T therapy against GBM. These findings shed light on the development of novel combinatorial strategies for effective immunotherapy of GBM and other solid tumors.

Radiation therapy (RT) is one of the most effective and widely used treatment methods for glioblastoma multiforme (GBM). However, its efficacy is often compromised by the inherent radioresistance of tumor cells, while the restrictive nature of the blood-brain barrier (BBB) specifically impedes the delivery of radiosensitizer. Thus, we constructed and characterized polyethylene glycol (PEG)-functionalized silver-gold core-shell nanoparticles (PSGNPs) targeting both BBB (TfRA4) and GBM (DNA1) (TDSGNPs). Afterwards, studies conducted both in vitro and in vivo were employed to assess the BBB penetration capabilities, abilities of GBM targeting and radiosensitization effect. Transmission electron microscope images of PSGNPs showed a core-shell structure, and the results of ultraviolet-visible absorption spectroscopy and dynamic light scattering displayed that TDSGNPs were successfully constructed with excellent dispersion properties. TDSGNPs could be specifically taken up by U87MG cells and the uptake peaked at 24 h. TDSGNPs combined with RT obviously increased the apoptosis proportion of the cells. It was shown by the in vitro and in vivo investigations that TDSGNPs could target U87MG cells after crossing the BBB, and further study revealed that TDSGNPs showed an uptake peak in the tumor sites after 3 h intravenous injection. The radiosensitization of TDSGNPs was better than that of the nanoparticles modified with single aptamers and the median survival of tumor-bearing mice was greatly extended. This study demonstrated that TDSGNPs could penetrate BBB to target GBM, functioning as a promising radiosensitizer for the targeted therapy of GBM.

Improvement of the treatment for Glioblastoma multiforme (GBM) especially the development of in situ controllable drug release is still a major concern. In this study, we developed waterborne biodegradable polyurethane (WBPU) scaffolds incorporated with redox-sensitive and RGD-decorated paclitaxel (PTX) polymer-drug conjugates (PDCs) for targeted GBM therapy in situ . The drug scaffolds could be implanted at residual GBM site post-operation. Dual-targeting PTX-PDCs were obtained through step-by-step conjugation of disulfide linked PTX, poly(ethylene glycol) (PEG), and arginine-glycine-aspartic acid (RGD). The RGD-modified PTX-PDCs were spherical nanoparticles (NPs) that would be released from scaffolds and identified GBM cells actively. Internalized redox-sensitive PTX-PDCs would be decomposed and release PTX inside GBM cells under the circumstances of glutathione (GSH). The release profiles of PTX from the scaffolds with/without GSH were investigated. In vitro cytotoxicity assay revealed that the dual-targeting PTX-PDCs from scaffolds could specifically kill GBM cells and protect normal cells, suggesting that dual-targeting PTX-PDC-loaded scaffolds may have the potential to repair tumor-induced brain injury. In vivo anti-recurrence assay indicated that the PTX-PDC-scaffolds could deliver PTX-PDCs to the GBM cells followed by inhibiting tumor growth and inducing apoptosis. In general, the PTX prodrug-loaded devices exhibited significant anti-GBM effects and normal tissue protection simultaneously, indicating that the WBPU scaffolds incorporated with dual-targeting PTX-PDCs may be a promising strategy for local therapy of GBM.

Temozolomide (TMZ) therapy offers minimal clinical benefits in patients with glioblastoma multiforme (GBM) with high EGFR activity, underscoring the need for effective combination therapy. Here, we show that tonicity-responsive enhancer binding protein (NFAT5) lysine methylation, is a determinant of TMZ response. Mechanistically, EGFR activation induces phosphorylated EZH2 (Ser21) binding and triggers NFAT5 methylation at K668. Methylation prevents NFAT5 cytoplasm interaction with E3 ligase TRAF6, thus blocks NFAT5 lysosomal degradation and cytosol localization restriction, which was mediated by TRAF6 induced K63-linked ubiquitination, resulting in NFAT5 protein stabilization, nuclear accumulation and activation. Methylated NFAT5 leads to the upregulation of MGMT, a transcriptional target of NFAT5, which is responsible for unfavorable TMZ response. Inhibition of NFAT5 K668 methylation improved TMZ efficacy in orthotopic xenografts and patient-derived xenografts (PDX) models. Notably, NFAT5 K668 methylation levels are elevated in TMZ-refractory specimens and confer poor prognosis. Our findings suggest targeting NFAT5 methylation is a promising therapeutic strategy to improve TMZ response in tumors with EGFR activation.

Adapting the mechanical strength between the implant materials and the brain tissue is crucial for the postoperative treatment of glioblastoma. However, no related study has been reported. Herein, we report an injectable lipoic acid‑iron (LA-Fe) hydrogel (LFH) that can adapt to the mechanical strength of various brain tissues, including human brain tissue, by coordinating Fe 3+ into a hybrid hydrogel of LA and its sodium salt (LANa). When LFH, which matches the mechanical properties of mouse brain tissue (337 ± 8.06 Pa), was injected into the brain resection cavity, the water content of the brain tissue was maintained at a normal level (77%). Similarly, LFH did not induce the activation or hypertrophy of glial astrocytes, effectively preventing brain edema and scar hyperplasia. Notably, LFH spontaneously degrades in the interstitial fluid, releasing LA and Fe 3+ into tumor cells. The redox couples LA/DHLA (dihydrolipoic acid, reduction form of LA in cells) and Fe 3+ /Fe 2+ would regenerate each other to continuously provide ROS to induce ferroptosis and activate immunogenic cell death. As loaded the anti-PDL1, anti-PDL1@LFH further enhanced the efficacy of tumor-immunotherapy and promoted tumor ferroptosis. The injectable hydrogel that adapted the mechanical strength of tissues shed a new light for the tumor postoperative treatment.

Glioblastoma multiforme (GBM), particularly the deep-seated tumor where surgical removal is not feasible, poses great challenges for clinical treatments due to complicated biological barriers and the risk of damaging healthy brain tissue. Here, we hierarchically engineer a self-adaptive nanoplatform (SAN) that overcomes delivery barriers by dynamically adjusting its structure, surface charge, particle size, and targeting moieties to precisely distinguish between tumor and parenchyma cells. We further devise a SAN-guided intuitive and precision intervention (SGIPi) strategy which obviates the need for sophisticated facilities, skilled operations, and real-time magnetic resonance imaging (MRI) guidance required by current MRI-guided laser or ultrasound interventions. In a preclinical intracranial GBM mouse model, SGIPi-based photodynamic therapy effectively impedes GBM progression with high tumor specificity and significantly extends overall survival. Moreover, the SGIPi potentiates chemotherapy while minimizing adverse effects; it eradicates intracranial GBM lesions in 100% cases solely through Temozolomide chemotherapy. This SGIPi strategy holds potential to improve the clinical management of GBM, with the possibility of extending survival rates and even achieving complete remission, and may inspire research focus from expensive and complex hardware development to simpler, delivery-based GBM interventions.
