Quality Assessment of Corydalis saxicola Bunting Using Quantitative Analysis of Multi-Components by Single Marker and Fingerprint Analysis

Yuanle Shen,Xu Yang,Hongli Feng,Huini Ruan,Yuan Zhou,Zihong Li,Jilang Li,Peng Zhang,Kaitong Li,Honglan Wang,Shen Liu,Xiaoxiu Shi,Jielan Huang,Jinyan Xian,Qiujie Chen,Zhenjie Liu,Jianfang Feng

Yan Huanglian is the whole grass and rhizome of Corydalis saxicola Bunting (CSB) of the Papaveraceae family. CSB has various pharmacological effects and is a promising ethnopharmaceutical. The chromatographic fingerprint of 16 batches of CSB samples was established in this study. Quantitative analysis of multi-components by single marker and the external standard method (ESM) was used to quantitatively assess seven constituents of CSB. Because of its high stability and availability, dehydrocavidine was used as a reference to determine the relative correction factors (RCFs) of columbamine, epiberberine, coptisine hydrochloride, palmatine chloride, berberine hydrochloride, and chelerythrine based on high-performance liquid chromatography. The calculated quantitative analysis of multi-components by single marker values is consistent with those obtained from the ESM, and the repeatability of the RCFs was demonstrated. For the first time, the quantitative analysis of multi-components by the single marker method has been established for the simultaneous determination of seven components, and it is demonstrated to be feasible and accurate for the quality evaluation of CSB combined with chromatographic fingerprint analysis. The combination of these methods could be used as a reference for quality control in Chinese medicine.

A series of strategies for solving the shortage of reference standards for multi-components determination of traditional Chinese medicine, Mahoniae Caulis as a case

Wenguang Wang,Xiaoli Ma,Xiaoyu Guo,Mingbo Zhao,Pengfei Tu,Yong Jiang

In order to solve the bottleneck of reference standards shortage for comprehensive quality control of traditional Chinese medicines (TCMs), a series of strategies, including one single reference standard to determine multi-compounds (SSDMC), quantitative analysis by standardized reference extract (QASRE), and quantitative nuclear magnetic resonance spectroscopy (qNMR) were proposed, and Mahoniae Caulis was selected as an example to develop and validate these methods for simultaneous determination of four alkaloids, columbamine, jatrorrhizine, palmatine, and berberine. Comprehensive comparisons among these methods and with the conventional external standard method (ESM) were carried out. The relative expanded uncertainty of measurement was firstly used to compare their credibility. The results showed that all these three new developed methods can accurately accomplish the quantification by using only one purified reference standard, but each of them has its own advantages and disadvantages as well as the specific application scope, which were also discussed in detail in this paper.

FAST CHROMATOGRAPHIC FINGERPRINT ANALYSIS OF GLYCYRRHIZAE RADIX BY ULTRA FAST LIQUID CHROMATOGRAPHY

Erqun Song,Li Xu

In the present study, a method based on ultra-fast liquid chromatography (UFLC) was successfully developed for chromatographic fingerprint (CFP) analysis of Glycyrrhizae radix. The CFP analysis was performed on an Agilent ZorBax XDB-C18 column (2.1 × 100 mm, 1.8 µm) using a gradient elution of methanol and water (containing 1% acetic acid) within a runtime of 30 min. The method was validated by eleven common peaks that were shown in all samples. The results revealed that the method was precise, stable, and reproducible. Using the established method, three Glycyrrhizae radix samples from different sources were evaluated for their similarity. Compared to the conventional high performance liquid chromatography, UFLC provided many advantages, such as time saving, less solvent consumption and better separation efficiency. It was a reliable and powerful method for quality control of traditional Chinese medicines with multi-components.

Effect and mechanism of Magnolia officinalis in colorectal cancer: Multi-component-multi-target approach

He Zhang,Jun Tang,Huiliang Cao,Chenguang Wang,Chong Shen,Jun Liu

Ethnopharmacological relevance Colorectal cancer (CRC) is a prevalent malignant tumor of the digestive tract. Traditional Chinese medicine (TCM) has a long history of treating CRC, with advantages such as effectiveness, multi-target, multi-pathway, and minimal side effects. TCM Magnolia officinalis (M. officinalis) refers to the dried bark, root bark, and branch bark of either Magnolia officinalis Rehd.et Wils. or Magnolia officinalis Rehd.et Wils. var. biloba Rehd.et Wils . It is commonly utilized to alleviate the side effects of chemotherapy for CRC, owing to its anti-inflammatory and anti-tumor properties. However, current research primarily focuses on the individual components and does not take into consideration the characteristics of multi-component-multi-target action. Aim of the study Our aim is to study the new action characteristics of M. officinalis in the treatment of CRC. Materials and methods Utilizing network pharmacology to identify potential active ingredients, key targets, and main signaling pathways of M. officinalis for the treatment of CRC. The binding effect was further validated through molecular docking analysis. Furthermore, the aforementioned components were identified using liquid chromatography-mass spectrometry (LC-MS), and the cleavage pathways of the main components were analyzed. Subsequently, both in vitro and in vivo experiments were carried out to investigate the anti-CRC effect of the active ingredients of M. officinalis and its potential mechanism. Results Network pharmacology and Molecular docking identified 5 main active ingredients and 6 core targets of M. officinalis for the treatment of CRC. Then, LC-MS identified the active components of M. officinalis. At the same time, both in vitro and in vivo experiments have confirmed the ability of Eucalyptol (Euc) and Obovatol (Obo)to inhibit inflammation and tumor cell proliferation. The possible mechanism involved is that Euc and Obo counteract CRC by inhibiting the over-activation of NF-κBp65/JAK and Bcl-2/Caspase signaling pathways, respectively. They also play a role in the anti-CRC effect of M. officinalis. Conclusion Magnolol (MAG), Honokiol (HK), Euc, Obo, and Neohesperidin (NHP) in M. officinalis may be the pharmacological substance basis for its anti-cancer effect on CRC. The treatment of CRC with M. officinalis is characterized by its multi-component, multi-target, and multi-pathway approach. These findings provide a theoretical basis for further inspiring the clinical application of M. officinalis and the development of efficacy targets.

Huang Lanyi,Liu Qing,Zhang Wenfang,Lin Bishan,Gao Yongjian,Deng Hong,Zhang Shu

Danggui–Jianzhong decoction (DGJZ) is a famous classical traditional Chinese medicine formula, which ingredients are complex and the quality is difficult to control. Our study aimed to identify the overall chemical profile of DGJZ qualitatively by ultra-high performance liquid chromatography with quadrupole time-of-flight mass spectrometry (UPLC-Q-TOF-MS) and UPLC. A total of 77 components, including terpenoids, flavonoids, phenolic acids, gingerols and other components, were firstly detected and characterized by UPLC-Q-TOF-MS and 18 peaks marked after analyzing the UPLC fingerprint. Finally, paeoniflorin, liquiritin, ferulic acid, cinnamic acid, glycyrrhizic acid and 6-gingerol were quantified, which was validated in terms of linearity, precision, accuracy, repeatability and recovery. Taken together, the chemical constitutes of DGJZ were systematically identified and a reliable quantitative method coupled with fingerprint analysis was successfully employed for evaluating the holistic quality, which will provide a robust foundation for the quality control of DGJZ.

Development of a Quality Evaluation Method for Allii Macrostemonis Bulbus Based on Solid-Phase Extraction–High-Performance Liquid Chromatography–Evaporative Light Scattering Detection Chromatographic Fingerprinting, Chemometrics, and Quantitative Analysis of Multi-Components via a Single-Marker Method

Jianfa Wu,Lulu Wang,Ying Cui,Chang Liu,Weixing Ding,Shen Ren,Rui Dong,Jing Zhang

As a traditional Chinese medicine (TCM), Allii Macrostemonis Bulbus (AMB) is a key herb for the treatment of thoracic paralytic cardiac pain, but its quality evaluation method has not yet been fully clarified. In this study, chromatographic fingerprints of AMB were developed using solid-phase extraction–high-performance liquid chromatography–evaporative light scattering detection (SPE-HPLC-ELSD) to evaluate the quality of AMB from various origins and processing methods. This was achieved by employing chemical pattern recognition techniques and verifying the feasibility and applicability of the quality evaluation of AMB through the quantitative analysis of multi-components via a single-marker (QAMS) method. Through the analysis of the fingerprints of 18 batches of AMB, 30 common peaks were screened, and 6 components (adenosine, syringin, macrostemonoside T, macrostemonoside A, macrostemonoside U, and macrostemonoside V) were identified. Moreover, three differential markers (macrostemonoside A, macrostemonoside T, and macrostemonoside U) were screened out using chemometrics techniques, including principal component analysis (PCA), hierarchical cluster analysis (HCA), and orthogonal partial least squares discriminant analysis (OPLS-DA). Subsequently, a QAMS method was established for macrostemonoside T and macrostemonoside U using macrostemonoside A as an internal reference. The results demonstrate the method’s accuracy, reproducibility, and stability, rendering it suitable for the quality evaluation of AMB. This study provides a theoretical basis for drug quality control and the discovery of quality markers for AMB.

Quantitative studies of rhubarb using quantitative analysis of multicomponents by single marker and response surface methodology

Jiachen Sun,Yueting Wu,Shengjie Dong,Xia Li,Wenyuan Gao

In this work, we developed a novel approach to evaluate the contents of bioactive components in rhubarb. The present method was based on the quantitative analysis of multicomponents by a single-marker and response surface methodology approaches. The quantitative analysis of multicomponents by a single-marker method based on high-performance liquid chromatography coupled with photodiode array detection was developed and applied to determine the contents of 12 bioactive components in rhubarb. No significant differences were found in the results from the quantitative analysis of multicomponents by a single-marker and the external standard method. In order to maximize the extraction of 12 bioactive compounds in rhubarb, the ultrasonic-assisted extraction conditions were obtained by the response surface methodology coupled with Box–Behnken design. According to the obtained results, we showed that the optimal conditions would be as follows: proportion of ethanol/water 74.39%, solvent-to-solid ratio 24.07:1 v/w, extraction time 51.13 min, and extraction temperature 63.61°C. The analytical scheme established in this research should be a reliable, convenient, and appropriate method for quantitative determination of bioactive compounds in rhubarb.

A single, multi-faceted, enhanced strategy to quantify the chromatographically diverse constituents in the roots of Euphorbia kansui

Jin-Jun Hou,Wan-Ying Wu,Jian Liang,Zhou Yang,Hua-Li Long,Lu-Ying Cai,Lin Fang,Dan-Dan Wang,Shuai Yao,Xuan Liu,Bao-Hong Jiang,De-An Guo

Kansui radix is a famous poisonous traditional Chinese medicine. However, due to its different types of constituents with broad polarity, a variety of UV absorptions and lack of the reference standards, it was difficult to simultaneously determine the main component in kanui radix. A single, multi-faceted, enhanced strategy, exogenous reference standard – single standard to determine multi-components method (ERS-SSDMC), was proposed. Thirteen major components of kansui radix, including three jatrophane diterpenoids, eight ingenane diterpenoids and two triterpenes, among which there were three pairs of isomers, were simultaneously assayed. A C8 column, packed with 2.7   μm core–shell particles, was optimized to separate these constituents in 25   min on HPLC instrument detected at a program wavelength. Ethyl benzoate employed as single exogenous reference standard. The method was fully validated with respect to linearity ( r 2   >   0.9995), LOQs (0.1–0.4   μg/mL), precision, accuracy (92–114%, RSD   <   4.4%) and stability. The robustness of the method was performed by Plackett–Burmantest tests which eight primary chromatographic parameters were investigated. It was found that the two factors, wavelength and flow rate, should be strictly controlled. A total of 75 batches of kansui radix and its three different processing products were successfully analyzed and discriminated by applying the proposed method. This work demonstrates an effective strategy for the SSDMC method making the simultaneous assay of complex multi-component TCM system achievable.

Establishment of Dissolution Test Method for Multi-Components in Traditional Chinese Medicine Preparations Based on In Vitro–In Vivo Correlation

Pengcheng Guo,Qizheng Wang,Xiaoqiang Xiang,Yufeng Zhang,Yue Pan,Zhong Zuo,Jianxin Wang

In this study, a multi-component integrated dissolution evaluation system of Yuanhu Zhitong tablets (YZTs) was established based on in vitro and in vivo correlation (IVIVC). The dissolution tests of five quality markers (Q-markers), including tetrahydropalmatine, α-allocryptopine, protopine, corydaline, and byakangelicin, in YZTs were conducted under different dissolution conditions, and pharmacokinetic studies were performed in beagle dogs to construct a correlation model using numerical deconvolution. The data of the five ingredients were integrated in vitro and in vivo according to the biopharmaceutical classification system (BCS) to establish an IVIVC integrating multiple Q-markers. The dissolution media with the best correlation of components were obtained and validated. The results showed that all five components were classified as BCS I compounds, and α-allocryptopine, byakangelicin, tetrahydropalmatine, and corydaline showed good correlation in the paddle method, 75 rpm, with dissolution media of artificial gastric fluid, acetate buffer, acetate buffer and 0.1 M HCl, respectively. Protopine showed good correlation in the paddle method, 100 rpm, with dissolution media of 0.1 M HCl. The integrated BCS I Q-markers showed the best correlation in the medium of acetate buffer. The multi-component integrated dissolution evaluation system established in this experiment accurately predicted the pharmacokinetic data of YZTs by verifying the media, which can be used for the quality control of YZTs. The present study provides an effective and promising strategy for the dissolution evaluation for traditional Chinese medicine preparations.

Quality assessment of Compound Yuxingcao Mixture produced by different manufacturers using high performance liquid chromatography and near infrared spectroscopy combined with multivariate algorithms

Ye Jin,Junjie Pan,Kejun Cheng

A comprehensive strategy based on high performance liquid chromatography (HPLC) and near infrared (NIR) spectroscopy was developed to assess the quality consistency of Compound Yuxingcao Mixture (CYM) from different manufacturers. Simultaneous determination of 10 marker components (neochlorogenic acid, chlorogenic acid, cryptochlorogenic acid, caffeic acid, acteoside, forsythoside A, quercitrin, baicalin, wogonoside and wogonin) in CYM and 7 marker components (neochlorogenic acid, chlorogenic acid, cryptochlorogenic acid, hyperoside, isoquercitrin, quercitrin and quercetin) in Houttuyniae Herba was carried out. Similarity analysis using chromatographic fingerprints and principal component analysis (PCA) were also performed to assess the quality consistency of CYM from different manufacturers. NIR spectroscopy combined with partial least squares (PLS), feed-forward back-propagation network (BP-ANN), and particle swarm optimization based least square support vector machine (PSO-LS-SVM) algorithms were employed for rapid determination of higher-concentration marker components (baicalin, wogonoside and chlorogenic acid) and rapid identification of product manufacturers. The comparative results showed that the PSO-LS-SVM models exhibited more satisfactory fitting results and predictive abilities. This study demonstrated that the combination of multi-component simultaneous determination and fingerprint analysis can help to gain an in-depth and comprehensive understanding of the quality of CYM; NIR technique combined with chemometric methods is useful for the rapid determination, identity and consistency evaluation of CYM.

YangHong Li,Bao Yu,Wei Zhao,Xiyu Pu,Xue Zhong,Xingbao Tao,Yunhong Wang,Weiguo Cao,Dan Zhang

Research background: YiQing granules (YQGs) are a patented drug commonly used in clinics in China. However, the quality control index of this preparation is relatively limited, which does not effectively ensure the quality of the product. Thus, a comprehensive quality evaluation of YQGs is lacking. Objective: The aim of this study was to establish a method for quantitative and qualitative analysis of YQGs based on ultra-high performance liquid chromatography-photodiode array detector (UPLC-PAD) fingerprint combined with quantitative analysis of multicomponents by a single marker (QAMS).Methods: We established and verified the comprehensive evaluation method that UPLC fingerprint combined with QAMS and stoichiometric method, to assess the overall quality of YQGs produced by different manufacturers. Berberine was selected as an internal reference, and the relative correction factors of coptisine, epiberberine, baicalin, berberine, palmatine, wogonoside, baicalein, wogonin, aloe-emodin, rhein, emodin, and chrysophanol were established.Study results: Results showed that the experimental time of the fingerprint was significantly reduced to approximately 0.5 h using the UPLC–PAD method. A total of 32 common peaks with similarity greater than 0.9 were identified. The accuracy of QAMS was compared with the external standard method, with no significant difference between the two methods. Conclusion: The UPLC fingerprint combined with QAMS method established in this study is feasible and reliable; it can be used for the comprehensive quality evaluation of YQGs and can provide a reference for the quality evaluation of other traditional Chinese medicine preparations.

Novel application of neural network modelling for multicomponent herbal medicine optimization

Ren Yong-Shen,Lei Lei,Deng Xin,Zheng Yao,Li Yan,Li Jun,Mei Zhi-Nan

The conventional method for effective or toxic chemical substance identification of multicomponent herbal medicine is based on single component separation, which is time-consuming, labor intensive, inefficient, and neglects the interaction and integrity among the components; therefore, it is necessary to find an alternative routine to evaluate the components more efficiently and scientifically. In this study, sodium aescinate injection (SAI), obtained from different manufacturers and prepared as “components knockout” samples, was chosen as the case study. The chemical fingerprints of SAI were obtained by high-performance liquid chromatography to provide the chemical information. The effectiveness and irritation of each sample were evaluated using anti-inflammatory and irritation tests, and then “Gray correlation” analysis (GCA) was applied to rank the effectiveness and irritability of each component to provide a preliminary judgment for product optimization. The prediction model of the proportions of the expected components was constructed using the artificial neural network. The results of the GCA showed that the irritation sorting of each SAI component was in the order of B > A > G > J > I > H > D > F > E > C and the effectiveness sorting of SAI components was in the order of D > C > B > A > F > E > H > I > G > J; the predictive proportion of SAI was optimized by the BP neural network as A: B: C: D: E: F = 0.7526: 0.5005: 5.4565: 1.4149: 0.8113: 1.0642. This study provided a scientific, accurate, reliable, and efficient approach for the proportion optimization of multicomponent drugs, which has a good prospect of popularization and application in product upgrading and development of herbal medicine.
