Photothermal Effect and Multi-Modality Imaging of Up-Conversion Nanomaterial Doped with Gold Nanoparticles

Wei Zhang,Yang Zang,Yanli Lu,Jinhui Han,Qingyun Xiong,Jinping Xiong

Two key concerns exist in contemporary cancer chemotherapy in clinics: limited therapeutic efficiency and substantial side effects in patients. In recent years, researchers have been investigating revolutionary cancer treatment techniques and photo-thermal therapy (PTT) has been proposed by many scholars. A drug for photothermal cancer treatment was synthesized using the hydrothermal method, which has a high light-to-heat conversion efficiency. It may also be utilized as a clear multi-modality bioimaging platform for photoacoustic imaging (PAI), computed tomography (CT), and magnetic resonance imaging (MRI). When compared to single-modality imaging, multi-modality imaging delivers far more thorough and precise details for cancer diagnosis. Furthermore, gold-doped upconverting nanoparticles (UCNPs) have an exceptionally high target recognition for tumor cells. The gold-doped UCNPs, in particular, are non-toxic to normal tissues, endowing the as-prepared medications with outstanding therapeutic efficacy but exceptionally low side effects. These findings may encourage the creation of fresh effective imaging-guided approaches to meet the goal of photothermal cancer therapy.Keywords:up-conversion;nanomaterials;photothermal;multi-modality imaging

A ternary doped single matrix material with dual functions of bone repair and multimodal tracking for applications in orthopedics and dentistry

Xiyu Li,Qin Zou,Li Chen,Wei Li

Hydroxyapatite (HA) has been broadly used for the repair of human hard tissues due to its bioactivity and similarity to the mineral of bones and teeth. We prepared a fluorine (F), ytterbium (Yb) and holmium (Ho) ternary doped HA (HYH-F) material and investigated its dual functions of bone repair and multimodal tracking. The results showed that the HYH-F material could display clear X-ray micro-computed tomography (Micro-CT) images based on Yb ions, and Ho ions due to its superparamagnetic properties for magnetic resonance imaging (MRI), while the F ions could enhance the upconversion (UC) fluorescence of the Yb/Ho combination. The ternary doped HYH-F material shows good compatibility with cells and bone tissue. A joint usage of Micro-CT imaging and UC fluorescence imaging distinctly and synergistically demonstrated the distribution state and degradation of the HYH-F material during new bone reconstruction. The single matrix HA material with ternary doping will be beneficial for future biomedical investigation and applications, and it can not only be a bone repair biomaterial, but also provide lifelong multimodal tracking efficacy.

Ultrasensitive Biomimetic Skin with Multimodal and Photoelectric Dual-Signal Sensing

Han Gao,Wenshan Cai,Aotian Li,Yike Du,Ji-Liang Zhu,Zhicheng Ye

Mimicking biological skin enabling direct, intelligent interaction between users and devices, multimodal sensing with optical/electrical (OE) output signals is urgently required. Owing to this, this work aims to logically design a stretchable OE biomimetic skin (OE skin), which can sensitively sense complex external stimuli of pressure, strain, temperature, and localization. The OE skin consists of elastic thin polymer-stabilized cholesteric liquid crystal films, an ion-conductive hydrogel layer, and an elastic protective membrane formed with thin polydimethylsiloxane. The as-designed OE skin exhibits customizable structural color on demand, good thermochromism, and excellent mechanochromism, with the ability to extend the full visible spectrum, a good linearity of over 0.99, fast response speed of 93 ms, and wide temperature range of 119 °C. In addition, the conduction resistance variation of ion-conductive hydrogel exhibits excellent sensing capabilities under pressure, stretch, and temperature, endowing a good linearity of 0.99998 (stretching from 0 to 150%) and high thermal sensitivity of 0.86% per °C. Such an outstanding OE skin provides design concepts for the development of multifunctional biomimetic skin used in human–machine interaction and can find wide applications in intelligent wearable devices and human–machine interactions.

Stretchable Multimodal Photonic Sensor for Wearable Multiparameter Health Monitoring

Jingjing Guo,Jialin Tuo,Jiangtao Sun,Zhuozhou Li,Xiaoyan Guo,Yanyan Chen,Rong Cai,Jing Zhong,Lijun Xu

Stretchable sensors that can conformally interface with the skins for wearable and real-time monitoring of skin deformations, temperature, and sweat biomarkers offer critical insights for early disease prediction and diagnosis. Integration of multiple modalities in a single stretchable sensor to simultaneously detect these stimuli would provide a more comprehensive understanding of human physiology, which, however, has yet to be achieved. Here, this work reports, for the first time, a stretchable multimodal photonic sensor capable of simultaneously detecting and discriminating strain deformations, temperature, and sweat pH. The multimodal sensing abilities are enabled by realization of multiple sensing mechanisms in a hydrogel-coated polydimethylsiloxane (PDMS) optical fiber (HPOF), featured with high flexibility, stretchability, and biocompatibility. The integrated mechanisms are designed to operate at distinct wavelengths to facilitate stimuli decoupling and employ a ratiometric detection strategy for improved robustness and accuracy. To simplify sensor interrogation, spectrally-resolved multiband emissions are generated upon the excitation of a single-wavelength laser, utilizing upconversion luminescence (UCL) and radiative energy transfer (RET) processes. As proof of concept, this work demonstrates the feasibility of simultaneous monitoring of the heartbeat, respiration, body temperature, and sweat pH of a person in real-time, with only a single sensor.

Controllable fabrication of multi-modal porous PLGA scaffolds with different sizes of SPIONs using supercritical CO2 foaming

Jinjing Wang,Yi Zhang,Jianfei Sun,Zhen Jiao

Multi-modal porous scaffolds can promote bone regeneration. In the present study, they were successfully fabricated from poly (lactic-co-glycolic acid) (PLGA)/superparamagnetic iron oxide nanoparticles (SPIONs) by supercritical CO 2 foaming, where SPIONs was employed as heterogeneous nucleation agent. The effects of SPION size on pore nucleation in PLGA/SPIONs nanocomposites were investigated. It was found that the addition of smaller SPIONs size caused the reduction of pore size. Multi-modal porous scaffolds could be available by controlling the SPIONs content, soaking temperature, pressure, and depressurization rate. The porosity of the PLGA/SPIONs scaffolds ranged from (58.68 ± 0.62)% to (94.97 ± 0.14)% could be obtained via adjusting the supercritical CO 2 foaming conditions. Moreover, it was found that the compressive modulus and porosity of the scaffolds were strongly associated with the porous structure. This study provided a new green preparation method for magnetic scaffolds to realize the controllable adjustment of the multi-mode porous scaffolds.

Ultrastretchable Composite Organohydrogels with Dual Cross-Links Enabling Multimodal Sensing

Chenguang Jiang,Xuexue Ding,Wenyuan Xie,Defeng Wu

Building multiple cross-links or networks is a favorable way of diversifying applications of the hydrogels, which is also available for the organohydrogels prepared via the solvent replacement way. However, the situations become more complicated for organohydrogels due to the presence of replaced solvents. Therefore, the correlations between the multiple cross-links and final performance need to be better understood for the organohydrogels, which is vital for tailoring their inherent properties to expand final application scenarios. Polyacrylamide (PAM)/poly(vinyl alcohol) (PVA)/MXene composite organohydrogels with dual cross-links, namely, the covalently cross-linked PAM chains as the primary network and the physically cross-linked PVA/PAM chains with MXene particles as the secondary cross-links, were developed here for the study. The occurrence of the secondary cross-links plays multiple roles as sacrificial units endowing the system with ultrastretchability with an excellent strain–resistance effect and as temperature-sensitive units endowing the system with thermosensation ability with an outstanding temperature coefficient of resistance. Thus, the optimized sample can be used as a strain sensor with excellent environmental tolerance for detecting human motion as a pressure sensor to probe compression with weak deformation and as a thermal sensor to capture environmental temperature changes. This work provides valuable information on developing organohydrogels with superior performance for multimodal sensors.

Magnetic Targeting Enhanced Theranostic Strategy Based on Multimodal Imaging for Selective Ablation of Cancer

Zhiwei Li,Shengnan Yin,Liang Cheng,Kai Yang,Yonggang Li,Zhuang Liu

The booming development of nanomedicine offers great opportunities for cancer diagnostics and therapeutics. Herein, a magnetic targeting-enhanced cancer theranostic strategy using a multifunctional magnetic-plasmonic nano-agent is developed, and a highly effective in vivo tumor photothermal therapy, which is carefully planed based on magnetic resonance (MR)/photoacoustic (PA) multimodal imaging, is realized. By applying an external magnetic field (MF) focused on the targeted tumor, a magnetic targeting mediated enhanced permeability and retention (MT-EPR) effect is observed. While MR scanning provides tumor localization and reveals time-dependent tumor homing of nanoparticles for therapeutic planning, photoacoustic imaging with higher spatial resolution allows noninvasive fine tumor margin delineation and vivid visualization of three dimensional distributions of theranostic nanoparticles inside the tumor. Utilizing the near-infrared (NIR) plasmonic absorbance of those nanoparticles, selective photothermal tumor ablation, whose efficacy is predicted by real-time infrared thermal imaging intra-therapeutically, is carried out and then monitored by MR imaging for post-treatment prognosis. Overall, this study illustrates the concept of imaging-guided MF-targeted photothermal therapy based on a multifunctional nano-agent, aiming at optimizing therapeutic planning to achieve the most efficient cancer therapy.

Biomimetic Activator of Sonodynamic Ferroptosis Amplifies Inherent Peroxidation for Improving the Treatment of Breast Cancer

Anwei Zhou,Tianliang Fang,Kerong Chen,Yurui Xu,Zhuo Chen,Xinghai Ning

Ferroptosis is a type of nonapoptotic cell death and is gradually emerging as an important anticancer treatment. However, its therapeutic efficacy is impaired by low intracellular levels of reactive oxygen species (ROS) and long-chain polyunsaturated fatty acids, significantly limiting its therapeutic potential. Herein, a multimodal strategy to improve ferroptosis is presented, in which a state-of-art engineered erythrocyte, termed as sonodynamic amplified ferroptosis erythrocyte (SAFE), is developed for simultaneously activating ferroptosis and oxygen-riched sonodynamic therapy (SDT). SAFE is composed of internalizing RGD peptide and red blood cell membrane hybrid camouflaged nanocomplex of hemoglobin, perfluorocarbon, ferroptosis activator (dihomo-γ-linolenic acid, DGLA), and sonosensitizer verteporfin. It is identified that SAFE, under ultrasound stimulation, can not only substantially supply oxygen to overcome tumor hypoxia associated therapeutic resistance, but effectively activate ferroptosis through the coeffect of SDT triggered ROS production and DGLA mediated lipid peroxidation. In vivo studies reveal that SAFE selectively accumulates in tumor tissues and induces desirable anticancer effects under mild ultrasound stimulation. Importantly, SAFE can effectively inhibit tumor growth with minimal invasiveness, resulting in a prolonged survival period of mice. Therefore, a multimodal ferroptosis therapy driven by oxygen-riched sonodynamic peroxidation of lipids, significantly advancing synergistic cancer treatment, is presented.

Skin somatosensory system inspired gelatin-based organohydrogel electronic skin for infant hazard alarms

Yubin Feng,Yukai Guo,Weihang Zhu,Xinting Yang,Chenke Wei,Zhe Yang,Andrew K. Whittaker,Chun Shen,Yue Zhao,Bai Yang,Quan Lin

Multi-functional electronic skin (e-skin) demonstrates immense potential for applications in physiological monitoring and human-computer interaction. However, achieving reliable multi-modal sensing in complex environments and hazard alarms for infants remains a significant challenge. Drawing inspiration from the skin's somatosensory system, we herein present an innovative multi-modal sensing organohydrogel, fabricated by incorporating gelatin, laponite, and LiCl into a copolymer network within a glycerol/water binary solvent. The designed organohydrogel exhibits skin-like composition and modulus. Furthermore, the low-temperature induced triple-helix structure of gelatin and the presence of binary solvents, imparting excellent stretchability, thermosensitivity, and outstanding environmental tolerance. Remarkably, this organohydrogel is well-suited for gelatin-based e-skin applications, mimicking the skin's sensory functions to detect strain, pressure, temperature, and humidity. Furthermore, the gelatin-based e-skin can be integrated with a custom-designed data recognition and analysis system, enabling real-time monitoring of an infant's physiological and movement states. This capability proves particularly valuable for providing timely alerts in hazardous situations, including asphyxiation caused by chest pressure, fever, or excessive sweating. This work paves the way for the next generation of e-skin for human health monitoring and hazard detection.

In situ valence-transited arsenic nanosheets for multi-modal therapy of colorectal cancer

Zheng Hongyue,Zhang Ke,Piao Jigang,Mu Chaofeng,Xie Xiaowei,Cheng Mengying,Yue Tianxiang,Sun Jiang,Li Bin,Wei Yinghui,Zheng Hangsheng,Jiang Lai,Habiballah Douae Nihed,Li Fanzhu

Late-stage and advanced colorectal cancer (CRC) often prove to be resistant to current treatment regimens, due to the evolving tumor microenvironment. Chemotherapy-dominated multi-modal therapeutic strategies based on the specific CRC microenvironment open a new horizon for eradicating colorectal tumors. Here, in situ valence-transited arsenic nanosheets are developed as a multi-modal therapeutic platform by responding to the H 2 S-enriched CRC microenvironment. Carrier-free pegylated nanosheets of pentavalent arsenic (As V ), aminooxyacetic acid (AOAA), and copper ion (Cu 2+ ) are innovatively self-assembled via coordination with high loading content and good stability. As V in pegylated arsenic nanosheets (CAA-PEG NSs) is rapidly released and reduced to trivalent arsenic (As III ) to exert its chemotherapy in the local tumor. Furthermore, the immunosuppressive microenvironment is thoroughly remodeled via H 2 S depletion of As V to As III conversion and impairment of H 2 S production by AOAA. Additionally, the in situ produced ultrasmall CuS nanoparticles exhibit photothermal activity against CRC under the guidance of photoacoustic imaging. This multi-modal therapeutic strategy, dominated by chemotherapy, completely inhibits CRC progression and prevents its relapse.

Tumor Microenvironment-Driven Structural Transformation of Vanadium-Based MXenzymes to Amplify Oxidative Stress for Multimodal Tumor Therapy

Hai Zhu,Tinghua Li,Xinhao Peng,Xiaoxian Zhang,Xuequan Zhang,Qiusheng Wang,Lei Lei,Jun Zhang,Bin He,Jun Cao

MXenzymes, a promising class of catalytic therapeutic material, offer great potential for tumor treatment, but they encounter significant obstacles due to suboptimal catalytic efficiency and kinetics in the tumor microenvironment (TME). Herein, this study draws inspiration from the electronic structure of transition metal vanadium, proposing the leverage of TME specific-features to induce structural transformations in sheet-like vanadium carbide MXenzymes (TVMz). These transformations trigger cascading catalytic reactions that amplify oxidative stress, thereby significantly enhancing multimodal tumor therapy. Specifically, the engineered HTVMz, coated with hyaluronic acid, exhibits good stability and generates a thermal effect under NIR-II laser irradiation. The thermal effect, combined with TME characteristics, facilities a structural transformation into ultra-small vanadium oxide nanozymes (VO x ). The enlarged surface area of VO x substantially enhances ROS regeneration and amplifies oxidative stress, which promotes lysosomal permeability and induces endoplasmic reticulum stress. The high-valent vanadium in VO x interacts with intracellular glutathione, disrupting redox homeostasis and intensifying oxidative stress further. These amplifications accelerate tumor apoptosis, induce ferroptosis, and suppress HSP90 expression. Consequently, the heightened thermal sensitivity of HTVMz synergistically promotes tumor cell death via multimodal therapeutic pathways. This study presents an innovative strategy for tumor catalytic therapy by manipulating MXenzymes structures, advancing the field of catalytic therapy.

Motion-Interference Free and Self-Compensated Multi-Receptor Skin with all Gel for Sensory Enhancement

Yibing Luo,Hao Wang,Yuning Liang,Ruijie Xie,Zixuan Wu,Yubin Zhou,Kai Tao,Shaowu Pan,Bo-Ru Yang,Yongqing Fu,Fei Liu,Fengwei Huo,Jin Wu

Stretchable multimodal electronic skin (e-skin) has attracted intensive research interest but faces great challenges related to strain interference, crosstalk issues, and integration of multiple sensitive materials. Herein, a stretchable and strain-isolated multimodal (SSIM) e-skin capable of concurrently and sensitively monitoring temperature, humidity, UV light, and oxygen, while also possessing self-compensation capability is developed. The SSIM sensing platform is created by chemically anchoring polyethylene terephthalate onto polydimethylsiloxane through silane treatment to form island-bridge structures. This method effectively isolates strain and improves interfacial adhesion, achieving a state-of-the-art low strain interference of 0.2% and an adhesion energy exceeding 300 J m − 2 (13.4 times that of the untreated material), ensuring the e-skin's stable operation even under dynamic stretching. To mitigate crosstalk and fabrication complexity, a single hydrogel film is employed to facilitate self-compensating multimodal sensing through various sensing mechanisms and physical isolations. The SSIM e-skin can simultaneously monitor several environmental and physiological signals with minimized crosstalk without interference from body movements. It enables remote respiration monitoring with wireless circuitry, highlighting its substantial potential in health monitoring, medical diagnostics, and neurorehabilitation.
