Low swelling hyperbranched poly(amine-ester) hydrogels for pH-modulated differential release of anticancer drugs

Hongbin Zhang,Yourui Dong,Lei Wang,Guojie Wang,Jian Wu,Yudong Zheng,Huai Yang,Siquan Zhu

Hydrogels composed of hyperbranched poly(amine-ester) (HPAE) macromers with 36% of terminal CC groups were prepared as a pH-sensitive multi-drug release system with low swelling ratios (4.2% for the lowest one). Anticancer drugs including doxorubicin hydrochloride (Dox), 5-fluorouracil (5FU) and hydroxycamptothecin (HPT) were separately encapsulated in HPAE macromers before hydrogel formation and their release can be controlled by the conformational change of HPAE macromers under different pH conditions, fast when pH > 7 and slow when pH < 7. Furthermore, this system allows a combination release of multiple drugs and the release behavior of each drug was relatively independent with no interference from each other. This is different from traditional pH-sensitive hydrogel systems which control the drug release by a swelling-shrinking mechanism. HPAE hydrogels exhibited pH-controllable drug release properties with a relatively constant volume, because the swelling of hydrogel is limited due to the unique globe like structure of HPAE molecules. These findings show the great promise of HPAE hydrogels as a smart release system for controlled delivery of single or multiple drugs in combined therapy, and where a constant volume of the release system is required.

Sterically stabilized spongosomes for multidrug delivery of anticancer nanomedicines

Yiyin Chen,Angelina Angelova,Borislav Angelov,Markus Drechsler,Vasil M. Garamus,Regine Willumeit-Römer,Aihua Zou

Multidrug delivery devices are designed to take advantage of the synergistic effects of anticancer agents in combination therapies. Here we report novel liquid crystalline self-assembled nanocarriers enhancing the activity of the phytochemical anticancer agent baicalin (BAI) in combination with Brucea javanica oil (BJO), which ensures safe formulations for clinical applications. Small-angle X-ray scattering (SAXS) and cryogenic transmission electron microscopy (cryo-TEM) evidenced the multicompartment, sponge-type nano-organization of the blank and multidrug-loaded liquid crystalline carriers. Physico-chemical stability of the sponge nanoparticles was achieved through PEGylation of the lipid membranes, which make up the drug nanocarriers. The proposed green nanotechnology for nanocarrier preparation by supramolecular self-assembly provided a multidrug encapsulation efficiency as high as 75%. The apoptosis study with the human lung carcinoma cell line A549 demonstrated improved efficacy of the multidrug delivery nanocarriers in comparison to the single-drug reservoirs. The obtained results evidenced the synergistic anticancer apoptotic effects of the multidrug-loaded nanosponge carriers and suggested the opportunity for in vivo translation towards the treatment of lung, gastrointestinal, and ovarian cancers.

Sialic acid–targeted drug delivery and imaging system for pH- and glutathione-triggered multiple anticancer drug release and enhanced oxidative stress

Licong Peng,Xianwu Zeng,Qianqian Qi,Hailiang Zhang,Jinping Fu,Miao Zhou,Jianchao Yuan

The emergence of multiple drug delivery systems can solve the disadvantages of single-drug therapy, such as high dose and easy generation of drug resistance. Here, we designed a sialic acid–targeted dextran-mercaptopurine prodrug linked by carbonyl vinyl sulfide for coordinate ZnO quantum dots to achieve multiple drug delivery (doxorubicin, 5-fluorouracil, 6-mercaptopurine), which can be released under the trigger of pH and glutathione. To enhance the antitumor effect, we used inorganic photosensitizer CdSe quantum dots to achieve photodynamic therapy, which can produce cytotoxic reactive oxygen species (hydroxyl radicals) under light conditions. Notably, we found that glutathione is consumed by the delivery of 6-mercaptopurine. It is able to efficiently amplify intracellular oxidative stress via increasing •OH generation. After chelating 99mTc4+ radioisotopes by diethylenetriamine pentaacetic acid, the drug delivery system could be tracked under in vivo single-photon emission computed tomography imaging. The results showed that the phenylboronic acid targeting substance can specifically recognize sialic acid, so that the drug system has a good accumulation in the tumor site, which can better increase the therapeutic effect. Compared to free doxorubicin, the drug system can reduce the IC50 value of cells 4.4-fold under light conditions and significantly inhibit tumor growth in vivo. These data indicate that the sialic acid–targeted nanomedicine system has achieved ideal antitumor effects and apparent photodynamic therapy effects and has broad application prospects.

Combating Biofilm Associated Infection In Vivo: Integration of Quorum Sensing Inhibition and Photodynamic Treatment based on Multidrug Delivered Hollow Carbon Nitride Sphere

Yuhuan Sun,Hongshuang Qin,Zhengqing Yan,Chuanqi Zhao,Jinsong Ren,Xiaogang Qu

Recently, quorum sensing (QS) inhibitors (QSIs) have been combined with antibiotics to enhance antibiofilm efficacy in vitro and in vivo. However, targeting QS signals alone is not enough to prevent bacterial infections. Drug resistance and recurrence of biofilms makes it difficult to eradicate. Herein, photodynamic therapy (PDT) is selected to unite QSIs and antibiotics. A synergistically antibiofilm system, which combines QSIs, antibiotics, and PDT based on hollow carbon nitride spheres (HCNSs) is envisaged. First, HCNS provides the multidrug delivering ability, enabling QSIs and antibiotics to be released in sequence. Subsequently, multistage releases sensitize bacteria effectively, potentiating the chemotherapeutic effects of the antibiotics. Finally, the integration of QSIs and PDT not only minimizes the possibility of drug resistance, but also overcomes the problem of limited mass and extension of PDT. Even after 48 h of incubation, the bacterial biofilm is obviously inhibited. And its biofilm disperse efficiency exceeds 48% (compared with QSI-potentiated chemotherapy group) and 40% (compared with PDT group). Besides, the inhibition of the QS system influences phenotypes related to virulence factor production and surface hydrophobicity, which weaken biofilm invasion and formation. Eventually, this system is applied to disperse bacterial biofilm in vivo. Overall, PDT and QS modulation are devoted to eradicate drug resistance and recurrence of the biofilm.

Building Micelle Analog Nanoparticle for Multidrug Delivery: Dual-Polymer Nanoparticles with Hydrophilic Shell and Double Hydrophobic Layers

Zhengzhong Wu,Yanjun Liu,Jun Cao,Lei Huang,Xianglin Luo

Multidrug delivery systems are capturing much attention while ingenious drug carriers are drug delivery researchers' pursuit. A method building dual-polymer nanoparticles (DPNs), one analogous micelle nanoparticle, with double hydrophobic core layers and a hydrophilic shell, is developed by combining co-axial injection and solvent evaporation using a hydrophobic and an amphiphilic polymer. Hydrophobic drugs can be efficiently and zonally encapsulated in core and/or outer hydrophobic zone of DPN. As a result, DPN can sequentially release drugs and appears stronger growth inhibition to Hela cells and cell uptake than normal drug-loaded micelles. Moreover, this method is universal to other material combinations of hydrophobic/amphiphilic polymers. Therefore, DPN has potential as one multidrug delivery system.
