Citric acid-loaded W1/O/W2 multiple emulsions efficiently remove colonic ammonia both in vitro and in vivo

Tengfei Bian,Xiujuan Zhang,Gaohong He,Zhijun Duan,Chunxu Dong,Xiangcun Li,Nana Guo

Hepatic encephalopathy (HE) is a neuropsychiatric disorder caused by chronic and acute liver diseases. It is believed that ammonia played an important role on the pathogenesis of the disease. Herein, acid-loaded water-in-oil-in-water (W1/O/W2) multiple emulsions with over 95% encapsulation efficiency were prepared and used to absorb colonic ammonia. The study of citric acid release from W1 to W2 in bulk deionized water indicated that only 17% acid released in 8 hours, which proved the stability of the multiple emulsions and hence prevented the intestine from being irritated by acid burst release. In vitro, the W1/O/W2 emulsion could remove around 90% ammonia in 1.5 hours from either the 3 mmol/L ammonia solution or the artificial colonic fluid with 1 and 0.5 mmol/L ammonia without acidifying the artificial colonic fluid. In vivo, compared with lactulose, W1/O/W2 emulsions could efficiently reduce the blood ammonia to the similar level in the rat models with HE without increasing the water content in feces. All these results indicated a potential application of W1/O/W2 multiple emulsions for the treatment of HE.

Dual-signal multiplex assays based on non-tandem enzymes for simultaneous detection of multiple pathogenic bacteria

Xiang Tianxin,Wen Jin,Fang Jianhua,Nie Chuang,Cheng Na,Ying Ying,Tan Hongliang

Enzymatic assays represent a promising alternative to conventional multiplex optical methods for multi-target detection due to their exceptional specificity and catalytic versatility. Nevertheless, most existing enzymatic systems are limited to single-target detection. In this work, we developed a dual-signal multiplex assay based on non-tandem enzymes for the simultaneous detection of multiple pathogenic bacteria. This assay utilizes the distinct catalytic activities of horseradish peroxidase (HRP) and invertase (INV) to produce two orthogonal signal outputs. Specifically, HRP catalyzes the oxidation of 3,3′,5,5′-tetramethylbenzidine to produce a blue chromogenic signal measurable by absorbance, while INV hydrolyzes sucrose to yield glucose, which is quantified with a personal glucose meter. To implement this assay, HRP and INV are individually encapsulated within zeolitic imidazolate framework-90, and Fe 3 O 4 nanoparticles functionalized with phenylboronic acid are employed for dual-recognition of target pathogens. By integrating spatiotemporal control of enzymatic reactions with distinct signal separation, the assay effectively minimizes crosstalk and enhances detection accuracy and reliability. This assay demonstrates excellent sensitivity and selectivity in milk samples, highlighting its potential for practical applications. We believe this work establishes a versatile and robust platform for multiplex pathogen detection, with significant implications for food safety, clinical diagnostics, and environmental monitoring. Fabrication of multifunction polyurethane with robust, self-healing and anti-corrosion performance based multiple dynamic bonds strategy

Weixiang Xu,Hao Liu,Jianmin He,Bingtao Bao,Jia Jiang,HanZhong Ren,Yiting Xu,Birong Zeng,Conghui Yuan,Lizong Dai

Polyurethanes (PUs) containing multiple dynamically have been extensively studied, however combining their mechanical, self-healing and anti-corrosion ability remains a significant challenge. Therefore, this paper successfully prepared PU coating (PU-SS-Salen-Ni) by introducing Ni 2+ coordinated Salen and bis(4-hydroxyphenyl) disulfide chain extenders . PU-SS-Salen-Ni with quadruple dynamic bonding exhibited excellent tensile strength (15.4 MPa), toughness (72 MJ·m −3 ) and superior self-healing efficiency (86.8 %) after being held at 80 °C for 4 h. Meantime, the |Z| f=0.01Hz of PU-SS-Salen-Ni coating reached at 9.35 × 10 6  Ω·cm 2 at the beginning of immersion, and even maintained at 6.99 × 10 6  Ω·cm 2 after 28 days immersion due to the synergistic effect of multiple dynamic bonds. This study provides insight into the design of PU elastomers with sustainable self-healing, excellent mechanical properties and corrosion protection .

Amphipathic Peptide Antibiotics with Potent Activity against Multidrug-Resistant Pathogens

Jingru Shi,Chen Chen,Dejuan Wang,Ziwen Tong,Zhiqiang Wang,Yuan Liu

The emergence and prevalence of multidrug-resistant (MDR) bacteria have posed a serious threat to public health. Of particular concern are methicillin-resistantStaphylococcus aureus(MRSA) andblaNDM,mcr-1andtet(X)-positive Gram-negative pathogens. The fact that few new antibiotics have been approved in recent years exacerbates this global crisis, thus, new alternatives are urgently needed. Antimicrobial peptides (AMPs) originated from host defense peptides with a wide range of sources and multiple functions, are less prone to achieve resistance. All these characteristics laid the foundation for AMPs to become potential antibiotic candidates. In this study, we revealed that peptide WW307 displayed potent antibacterial and bactericidal activity against MDR bacteria, including MRSA and Gram-negative bacteria carryingblaNDM-5,mcr-1ortet(X4). In addition, WW307 exhibited great biofilm inhibition and eradication activity. Safety and stability experiments showed that WW307 had a strong resistance against various physiological conditions and displayed relatively low toxicity. Mechanistic experiments showed that WW307 resulted in membrane damage by selectively targeting bacterial membrane-specific components, including lipopolysaccharide (LPS), phosphatidylglycerol (PG), and cardiolipin (CL). Moreover, WW307 dissipated membrane potential and triggered the production of reactive oxygen species (ROS). Collectively, these results demonstrated that WW307 represents a promising candidate for combating MDR pathogens. Keywords:antimicrobial peptide;antibiotic resistance;multidrug-resistant bacteria;membrane damage

Highly Stretchable, Ultratough, and Strong Polyesters with Improved Postcrystallization Optical Property Enabled by Dynamic Multiple Hydrogen Bonds

Shuai Sun,Yijiao Xue,Xiaodong Xu,Liping Ding,Zhen Jiang,Linghui Meng,Pingan Song,Yongping Bai

It has been highly desirable to develop high-performance thermoplastic polyesters that are strong, tough, and highly stretchable, in addition to high glass transition and excellent optical properties. The performance portfolio is essential for the practical applications of polyesters in high-end areas of aerospace, energy, medical sterilization, and optics. However, current material design strategies have failed to endow polyesters with such integrated performances. Herein, we report the facile synthesis of modified copolyesters with multiple hydrogen bonds (H-bonds) on the main chains via condensed polymerization. The resultant copolyester records a break strain as high as 438% and a large toughness of 106.7 MJ/m3, representing the most stretchable and toughest thermoplastic polyesters so far, while exhibiting a high tensile strength of 59.6 MPa because of intermolecular multiple H-bonding. In addition to achieving an enhanced glass transition temperature (∼85.9 °C), the copolyester films exhibit improved postcrystallization optical transparence and good flexibility due to the grain refinement effect. This work provides an innovative design concept to prepare a new class of advanced polyesters with outstanding mechanical, thermal, and optical properties, thus holding great promise for many potential industrial applications.

A molecular beacon based on DNA-templated silver nanoclusters for the highly sensitive and selective multiplexed detection of virulence genes

Dan Han,Chunying Wei

In this work, we develop a fluorescent molecular beacon based on the DNA-templated silver nanoclusters (DNA-Ag NCs). The skillfully designed molecular beacon can be conveniently used for detection of diverse virulence genes as long as the corresponding recognition sequences are embedded. Importantly, the constructed detection system allows simultaneous detection of multiple nucleic acids , which is attributed to non-overlapping emission spectra of the as-synthesized silver nanoclusters. Based on the target-induced fluorescence enhancement, three infectious disease-related genes HIV, H1N1, and H5N1 are detected, and the corresponding detection limits are 3.53, 0.12 and 3.95   nM, respectively. This design allows specific, versatile and simultaneous detection of diverse targets with easy operation and low cost.

Facile synthesis of ZIF-67 derived dodecahedral C/NiCO2S4 with broadband microwave absorption performance

Congyu Zhou,Zhengjun Yao,Bo Wei,Wenying Li,Zhejia Li,Xuewei Tao,Jintang Zhou

The increasing hazard of electromagnetic radiation prompts people to pursue absorbing materials with better performance. However, absorbing materials with a single loss mechanism usually is unable to obtain better absorbing performance due to low impedance matching or high filling ratio. Therefore, this work proposes a C/NiCo2S4 (CNCS) material with both dielectric loss/magnetic loss to achieve efficient absorption of electromagnetic waves. The simple preparation of CNCS materials was achieved through the etching of the ZIF-67 template by nickel nitrate and the subsequent hydrothermal vulcanization process. Its unique prismatic dodecahedron hollow structure promotes multiple scattering of electromagnetic waves. The attachment of the magnetic NiCo2S4 particles on the surface of the carbon template further promotes the interface polarization and dipole polarization, which is equivalent to the formation of a resistance-rich microcircuit and enhances the effect of the conductance loss on electromagnetic waves. At 2–18 GHz, the CNCS-2 with 30% paraffin addition achieves an effective bandwidth of 5.54 GHz at a matching thickness of 1.7 mm, and has a maximum reflection loss of −36.44 dB at 1.5 mm. By adjusting the thickness of the material matching layer (1–3 mm), an effective bandwidth of up to 13.48 GHz can be achieved, perfectly covering the X-band and Ku-band. Based on the simple preparation process of the material, the special hollow structure and the multiple loss mechanisms for electromagnetic waves, we believe that CNCS can become a strong competitor for high-efficiency broadband absorbers.

Enhanced antibacterial activity of nano-conjugates of cephradine against multidrug-resistant bacteria: Experimental and theoretical insights

Ali Shujat,Perveen Samina,Shah Muhammad R.,Hassan Ali,Ali Mumtaz

Background: Currently, bacteria are getting resistant to antibiotics which pose a serious threat to human health. In this scenario, new nanoantibiotics have the potential to avoid the emerging bacterial resistance mechanisms. Hence, this study attempts to unveil the activity enhancement of Cephradine (Cfdn) by using experimental and theoretical investigations. Methods: Silver (Ag) and gold (Au) nanoparticles (NPs) stabilized with Cfdn (Cfdn-AgNPs and Cfdn-AuNPs) were synthesized using a one-pot reduction method, and their characteristics were analyzed using atomic force microscope (AFM), scanning electron microscope (SEM), Fouriertransform infrared (FT-IR) spectroscopy, UV-visible spectroscopy. The stability of the NPs was evaluated under different physiological conditions. The antimicrobial action of Cfdn, when conjugated with the Ag and Au NPs, was demonstrated by conducting experiments with multidrugresistant Staphylococcus aureus (S. aureus) cell cultures. AFM images were used to observe changes in S. aureus cell morphology before and after treatment with free and conjugated Cfdn. Moreover, theoretical analysis was carried out to further validate the enhanced activities of Cfdn-AgNPs and Cfdn-AuNPs.The spherical (20-55 nm) Cfdn-AgNPs and Cfdn-AuNPs demonstrated good stability under saline, pH, and thermal conditions. Interestingly, a significant improvement was achieved in the antibacterial activity, with Cfdn-AgNPs and Cfdn-AuNPs exhibiting activity enhancements of up to 2 and 6 times, respectively. AFM images confirmed improved kinetics of the antibiotic upon conjugation, where Cfdn alone did not cause sufficient damage to bacterial membranes even after a 4-hour treatment, whereas treatment with Cfdn-AgNPs and Cfdn-AuNPs resulted in effective cell membrane damage and complete disruption of cell morphology within the same timeframe. Moreover, theoretical simulation employing density functional theory (DFT) validated the enhanced antibacterial potential of Cfdn-AgNPs and Cfdn-AuNPs. Interestingly, cytotoxicity analysis revealed improved cytocompatibility of the NPs after conjugation with Cfdn.We established a systematic strategy for the development of innovative nanoantibiotics based on experimental and theoretical evidence, which can be exploited in the battle against bacterial resistance.

Programmable strand displacement-based magnetic separation for simultaneous amplified detection of multiplex microRNAs by chemiluminescence imaging array

Shuzhen Yue,Tingting Zhao,Sai Bi,Zhipeng Zhang

High throughput analysis of miRNAs is of great significance to clinical and biomedical applications . In this work, we have developed a highly sensitive and selective chemiluminescence imaging array (CLIA) for simultaneous detection of three miRNAs with high throughput, easy operation and low cost. In this assay, three kinds of hairpins that can specially recognize respective miRNAs (miR-155, miR-let-7a and miR-141) are parallel modified on the magnetic beads (MBs). The employment of MBs enables quick sorting of multiplex targets in high yield and purity with high throughput. Upon introduction of the target miRNAs, they hybridize to the corresponding hairpins and initiate the DNA machines with the assistance of Klenow fragment exo- DNA polymerase and Nb.BbvCI NEase , achieving exponential amplification of targets. After that, the toehold-mediated strand displacement (TMSD) reactions are performed through sequential addition of displacement probes under magnetic separation, resulting in the release of HRP-tagged DNA hybrids in solution for CL imaging and recovery of MBs for repetitive use. This CLIA method demonstrates ultrahigh sensitivity with detection limits down to fM level, wide linear range over 5 orders of magnitude, excellent selectivity to distinguish one-base mismatched target miRNA, and good performance in real sample analysis. Together with the advantages of high throughput, easy operation, acceptable accuracy, and good recyclability and reproducibility, the proposed multiplex CLIA method holds great potential in practical applications, such as profiling the pattern of miRNA expression, disease screening, biomedical research, and so on.

Multiplex digital PCR with digital melting curve analysis on a self-partitioning SlipChip

Yan Yu,Ziqing Yu,Xufeng Pan,Lei Xu,Rui Guo,Xiaohua Qian,Feng Shen

Digital polymerase chain reaction (digital PCR) can provide absolute quantification of target nucleic acids with high sensitivity, excellent precision, and superior resolution. Digital PCR has broad applications in both life science research and clinical molecular diagnostics. However, limited by current fluorescence imaging methods, parallel quantification of multiple target molecules in a single digital PCR remains challenging. Here, we present a multiplex digital PCR method using digital melting curve analysis (digital MCA) with a SlipChip microfluidic system. The self-partitioning SlipChip (sp-SlipChip) can generate an array of nanoliter microdroplets with trackable physical positions using a simple loading-and-slipping operation. A fluorescence imaging adaptor and an in situ thermal cycler can be used to perform digital PCR and digital MCA on the sp-SlipChip. The unique signature melting temperature (Tm) designed for amplification products can be used as a fingerprint to further classify the positive amplification partitions into different subgroups. Amplicons with Tm differences as low as 1.5 degrees celsius were clearly separated, and multiple amplicons in the same partition could also be distinguished by digital MCA. We further demonstrated this digital MCA method with simultaneous digital quantification of five common respiratory pathogens, including Staphylococcus aureus, Acinetobacter baumannii, Streptococcus pneumoniae, Hemophilus influenzae, and Klebsiella pneumoniae. Since digital MCA only requires an intercalation dye instead of sequence-specific hydrolysis probes to perform multiplex digital PCR analysis, it can be less expensive and not limited to the number of fluorescence channels.

Development of novel broad-spectrum amphipathic antimicrobial peptides against multidrug-resistant bacteria through a rational combination strategy

Jing Zhang,Liang Luan,Youdong Xu,Shuyuan Jiang,Wenpeng Zhang,Long Tian,Weifeng Ye,Jiaqi Han,Changhao Zhang,Taoran Wang,Qingbing Meng

Introduction In recent years, cationic amphipathic antimicrobial peptides (AMPs) have shown great promise in combating antibiotic resistance on account of their distinctive membrane-disruptive mechanism. However, the clinical application of AMPs is restricted by their unsatisfactory stability and safety. Although attempts have been made to improve the stability and safety of AMPs, many of them are accompanied by a decline in their antimicrobial activity and bacterial selectivity. Objectives To develop AMPs with excellent and balanced antimicrobial activity, stability, and safety using a combination strategy. Methods A series of sC18 4b -derived peptide analogues were designed by a combination strategy of subtly adjusting the charges, hydrophobic properties, and introducing specific unnatural amino acids in a well-balanced manner. The antimicrobial activity, cytotoxicity, hemolytic activity, stability, anti-biofilm activity, mechanism of action, synergistic effects, in vivo efficacy, and pharmacokinetics of the analogues were evaluated. Results Among these analogues, P-α-02-B stood out for its broad-spectrum and potent antimicrobial activity, anti-biofilm activity, desirable bacterial selectivity, high plasma stability, and synergistic effect with antibiotic levofloxacin. P-α-02-B exhibited strong membrane disturbance effect, which could be explained by its rigid α-helical structure revealed by molecular dynamics simulations. More importantly, P-α-02-B showed favorable therapeutic efficacy in vivo , whether used alone or in combination with levofloxacin. Conclusion P-α-02-B is a promising antimicrobial agent for MDR bacterial infections, demonstrating the effectiveness of the combination strategy for AMP development.

The Antimicrobial Peptide D-CONGA-Q7 Eradicates Drug-Resistant E. coli by Disrupting Bacterial Cell Membranes

Zonghan Jiang,Leisheng Sun,Yuanyuan Li,Haoyu Li,Yu Fu,Jiyun Li,Zhiliang Sun

Simple SummaryEscherichia coliis a ubiquitous pathogen, with certain strains causing severe diarrhea and intestinal illnesses, which pose significant health risks to both children and animals. As antimicrobial resistance continues to rise, traditional antibiotics are increasingly ineffective, highlighting the urgent need for novel therapeutic strategies. In this study, we investigated the antimicrobial peptide D-CONGA-Q7, aiming to evaluate its efficacy and underlying mechanism against drug-resistantE. colistrains. Our results demonstrate that D-CONGA-Q7 effectively disrupts bacterial cell membranes, leading to bacterial death, and exhibits comparable activity against polymyxin- and meropenem-resistant strains. In vivo experiments revealed that D-CONGA-Q7 significantly alleviated intestinal inflammation, reduced inflammatory cytokine levels, and promoted the repair of damaged intestinal tissues. Furthermore, D-CONGA-Q7 displayed good stability. These findings suggest that D-CONGA-Q7 not only represents a promising new approach to combat drug-resistant bacterial infections but also holds potential for clinical application.Escherichia coli(E. coli) is a zoonotic bacterium widespread in the environment, highly transmissible, and responsible for significant economic losses and millions of cases of illness annually. The rise of multidrug-resistant (MDR) strains has rendered last-line antibiotics such as polymyxin and meropenem ineffective, making the development of new antibiotics urgent. Although D-CONGA-Q7 has broad-spectrum bactericidal activity, its underlying mechanism remains poorly understood. In this study, we used in vitro and in vivo experiments to demonstrate that D-CONGA-Q7 effectively kills both antibiotic-sensitive and multidrug-resistant strains ofE. coli. D-CONGA-Q7 disrupts the cell membranes of Gram-negative bacteria, and the treatment ofE. colistrainLN175with D-CONGA-Q7 resulted in a significant up-regulation of theMlacgene, suggesting that D-CONGA-Q7 may interact with phospholipids in the cell membrane. Furthermore, in treatingK88-induced bacterial enteritis in the small intestine, D-CONGA-Q7 significantly reduced intestinal inflammation. In conclusion, this study provides a novel approach to combat drug-resistantE. coli.
