Flower-Like Multicompartment Micelles with Janus-Core Self-Assembled from Fluorocarbon-Terminated Pluronics

Hengchang Liu,Yujun Feng

Multicompartment micelles (MCMs), whose cores have at least two compartments, show potential applications in various areas, but the synthesis of polymers for preparing MCMs is usually tedious and time-consuming. In this work, two well-defined telechelic fluorocarbon-terminated triblock copolymers, F 8 -PEO 100 -PPO 65 -PEO 100 -F 8 (F 8 -F127-F 8 ) and F 8 -PEO 132 -PPO 50 -PEO 132 -F 8 (F 8 -F108-F 8 ), are synthesized via a single-step coupling reaction of Pluronics F127 or F108 with perfluoro-1-octanesulfonyl fluoride and characterized by Fourier-transform infrared and NMR spectroscopies, as well as gel permeation chromatography and surface tensiometry. Both of these fluorocarbon-terminated Pluronics can self-assemble into spherical MCMs with Janus-core in aqueous solution, as evidenced by transmission electron microscopy imaging. Since the lipophilic block (PPO) and fluorophilic segments (F 8 ) are separated by the hydrophilic blocks (PEO), these fluorocarbon-terminated Pluronics will loop to give rise to flower-like MCMs, and the calculations based on thermodynamics and dynamics support the formation of such unique aggregates. A “pre-self-assembly” mechanism is proposed to explain the formation process of flower-like MCMs with Janus-core prepared by these telechelic fluorocarbon-terminated triblock copolymers.

Dynamically Reconfigurable Complex Emulsion Droplets as Intelligent Microreactors

Duo Wei,Nuoqing Yin,Dehua Xu,Lingling Ge,Rong Guo

Multicompartments in complex emulsions, which mimic the organelles of living cells, can spontaneously sequester reactants/products and confine spatially incompatible compounds. This provides opportunities to control spatiotemporal reaction processes and to be a sustainable and high-efficiency platform for biphasic reactions even under stirring-free and ambient temperatures. We introduce a new multicompartment droplet microreactor based on one-step prepared complex emulsions. An organic/aqueous addition reaction between oil-soluble stilbene and water-soluble N-bromosuccinimide is employed as a model biphasic reaction. The conversion in the proposed complex droplet microreactor achieves a rate about 25 times higher than that in a traditional triphasic batch system. Over a 10 times threshold of the reaction rate is finely tuned by rationally engineering diverse droplet morphologies. Reversible in situ control of the reaction kinetics is achieved based on the reversible topology transition manipulated by the surfactant composition and temperature, with no need for adding any extra substance. Furthermore, reaction kinetics are precisely regulated by the droplet size and the shape of the two inner lobes. After rapid demulsification, over 86% of the target product is spontaneously enriched in one separated phase with a maximum purity of 98% following solvent removal. The byproduct succinimide and the solvent fluorocarbon phase are fully recycled to avoid potential pollution problems.

Design and Synthesis of Multi-compartment Microcapsules via Pickering Emulsion Polymerization for Infrared Stealth and Adaptive Camouflage Applications

Kunlin Chen,Jingyu Chen,Changyue Xu,Haie Zhu,Jing Hu,Kejing Yu

High-performance color-changing compounds, recognized as prominent smart materials, dynamically alter their color in response to external environmental stimuli. However, existing compounds exhibit limited responsiveness and color diversity, presenting challenges in the development of textiles responsive to multiple stimuli. This research introduces a novel design for dual-responsive color-changing microcapsules, employing a Pickering emulsion template method. The larger compartment encloses photosensitive dyes, whereas the smaller one contains thermochromic phase-change colorants. Adjusting the density of nanocapsules in the smaller compartment on the microcapsule surface enables a spectrum of colors, including red, yellow, blue, and green, triggered by light and heat. When incorporated into textiles, these microcapsules bestow adaptive color-changing attributes and infrared stealth capabilities onto the fabrics. Additionally, by modulating the color via surface micro/nanostructures, textile surfaces can exhibit hydrophobic and oleophobic properties. Such enhancements extend the textiles’ potential applications in areas like anti-counterfeiting and military operations.

Non-concentric multi-compartment fibers fabricated using a modified nozzle in single-step electrospinning

Baolin Wang,Meidi Wang,Ming-Wei Chang,Zesshan Ahmad,Jie Huang,Jing-song Li

Multi-compartment fibers with well-defined regions and functional media hosting potential provide co-encapsulation opportunities for a variety of bio-molecules, drugs and even nutrients. In this research, fibers possessing multiple compartments were prepared using the single-step electrospinning method and a modified non-concentric multi-needle. Polyvinyl pyrrolidone (PVP) was used as the fiber shell material, while ketoconazole (KCZ, model drug) and Sudan Red (model probe) were encapsulated as two separate segments running along the fiber length. Multi-compartment fiber morphology and structure were examined using optical and electron microscopy. The effect of flow rate on fiber morphology was also investigated and the release of encapsulated KCZ and Sudan Red was examined using UV spectroscopy. The results present an efficient and promising method to engineer multi-compartment fibers in a single step for several biomedical applications in lieu .

Oriented immobilization of enzyme–DNA conjugates on magnetic Janus particles for constructing a multicompartment multienzyme system with high activity and stability

Hao Shen,Xuelian Zheng,Zixin Zhou,Wenting He,Mengqi Li,Ping Su,Jiayi Song,Yi Yang

Various organelles (e.g., mitochondria and chloroplasts) have a multicompartment structure, providing superior function of material transformation, selective segregation and energy conversion. Enlightened by the elegant evolution of nature, intended isolation of the biochemical process by cooperative multicompartments in cells has become an appealing blueprint to construct bioreactors. In this study, we develop a “soft separation” way to establish a delicate multicompartment multienzyme system (MMS) with polyphenol-encapsulated enzyme–DNA conjugates, which are anchored on magnetic Janus particles, providing a biomimetic catalysis network with the model cascade reactions in confinement. The well-designed MMS exhibits preferable bioactivity benefitting from the dependable DNA bridges and the oriented immobilization of enzymes, while the polyphenol shell further protects the anchored enzymes from exterior attacks, such as heat and enzymatic degradation. Moreover, by applying the MMS as nanomotors, the asymmetrical distribution of enzymes on Janus particles is found to improve mutual elevation between the self-driven locomotion and enzyme-mediated reactions, delivering enhanced dispersal ability and bioactivity. Owing to the excellent enzymatic activity, promoted stability and satisfying biocompatibility, the assembled MMS is proved to be promising for the in vitro and intracellular sensing of glucose, showing significant potential for biochemical analysis applications.

Interfacial-Design Mediated Construction of Hybrid Multicompartment Architectures with Layered Physicochemical Barriers for Separately Sequestration of Oxidation–Reduction Molecules and Their Redox Reaction Regulation

Hai Chen,Xueer Han,Junlu Gao,Xiaoyi Tan,Hongjie Dai,Liang Ma,Yuhao Zhang

Hybrid multicompartment artificial architectures, inherited from different compartmental systems, possess separate microenvironments that can perform different functions. Herein, a hybrid compartmentalized architecture via hybridizing ferritin nanocage (Fn) with non-aqueous droplets using aminated-modified amphiphilic gelatin (AGEL) is proposed, which enables the generation of compartmentalized emulsions with hybrid multicompartments. The resulting compartmentalized emulsions are termed “hybrasome”. Specifically, by chemically attaching ethylenediamine to gelatin, the programmed noncovalent docking of Fn-AGEL nanoparticles is implemented and their interfacial self-rearrangement generates hybrasome with layered physicochemical barriers. Confocal Laser Scanning Microscopy images show that Fn nanocages are deposited on the non-aqueous droplets, separated by gelatin layers. Interfacial adsorption kinetics reveal that lower permeation and rearrangement rates of Fn are responsible for their double-layered structure formation. By choosing oxidized iron nanoparticles and reductant carnosic acid (CA) as models, these two molecules are co-encapsulated separately within the hybrasome, resulting in significant inhibition of the redox reaction. After structural destruction in the intestine, a redox reaction is triggered and leads to the Fe 2+ redox products release, which generates a suitable valence state of iron element for cell absorption. Overall, this approach may open up an avenue for facile construction of hybrid compartmentalized architectures used to co-encapsulate incompatible compounds separately and control the sequential release of targeted components.

Enzyme Cascade Reaction-Propelled Multicompartmental Colloidal Motors

Qihan Zhang,Jun Liu,Yingjie Wu

Graphical An enzyme-driven multicompartmental colloidal motor was synthesized by using a facile microfluidic-based microdroplet technology. The colloidal motor could trigger enzyme cascade reaction in biocompatible fuel to generate local concentration gradient to provide net propulsion force for autonomous movement. Due to the advantages of the multicompartment, the colloidal motor has great potential for co-encapsulation of active species in biomedical applications. Compartmentalization is a crucial natural methodology to enable multiple biocatalytic transformations to proceed efficiently. Herein, we report a biocompatible multicompartmental colloidal motor that can achieve autonomous movement in the biological environment through two-enzyme cascade reactions of immobilized enzymes. The colloidal motors with the heterogeneous multicompartment structure were prepared in one step by microfluidic technology, and the compartmentalized encapsulation of glucose oxidase (GOD) and catalase (CAT) was realized. The fabricated colloidal motor was size controllable by tuning the flow rates of the microfluidic system, and its autonomous movement can be triggered by good responsiveness to the alkaline environment. In glucose medium of pH 7.5, the pH-responsive alginate cores of the colloidal motor swell to facilitate fuel penetration and enzyme-catalyzed reactions. The enzyme cascade between GOD and CAT immobilized in the colloidal motor chamber results in the self-propulsion of the colloid motor in glucose medium. The compartmentalized encapsulation of immobilized enzyme improves the stability of the enzyme and enables multicompartmental colloidal motors to self-propel in an alkaline intestinal environment through an enzyme cascade reaction. These features indicate that such multicompartmental colloidal motors actuated by enzyme cascade reaction in biocompatible fuel have great potential for co-encapsulation and autonomous movement in different applications.
