Accurate and timely detection of ions and metabolites in urine is essential for health monitoring. However, traditional methods face challenges in integrating multiplexity, signal stability, and portability. To address these issues, this study developed a 3D-printed portable dual-mode gel microspheres kit. The hydrogel microspheres were prepared by encapsulating the respective indicators: Urine pH Indicator (UpHI) for pH detection, Urine Calcium Ion Indicator (UCaI) for Ca 2+ detection, and Urine Creatinine Indicator (UCrI) for creatinine detection. Each indicator was optimized for its specific target to ensure high sensitivity and selectivity. The system employs dual-mode UV and visible light analysis, combined with smartphone imaging and RGB data processing, ensuring rapid, intelligent, and high-throughput detection. The integration of gel microspheres with a 3D-printed microdevice enhances the portability and user-friendliness of the detection system. This design facilitates long-term storage and transportation while ensuring usability for non-technical individuals in home settings. The kit demonstrated excellent encoding capacity, maintained stable signals for up to a week, and achieved integrated detection, data acquisition, and analysis within approximately 6 min. The cost per microsphere is under $0.20, making it a cost-effective solution. In conclusion, this method offers an effective solution for portable and timely urine health monitoring in home settings and provides a promising approach for the future development of portable kits with expanded detection pathways.

In order to prevent and control the effects of pesticide residues on human health and the ecological environment, the rapid, highly sensitive, and selective detection of multiple pesticide residues has become an urgent problem to be solved. Herein, a lab-on-a-molecule probe based on a host–guest complex (ThT@Q[8] probe) has been developed to simultaneously analyze multiple aromatic pesticides under single wavelength excitation, such as fuberidazole, thiabendazole, carbendazim, thidiazuron, and tricyclazole. The fluorescence titration spectra of the ThT@Q[8] probe with the five pesticides mentioned above showed that the fluorescence intensity exhibited a good linear correlation with the pesticide concentration and the limit of detection was as low as 10–7 M. Because the ThT@Q[8] probe exhibits diverse fluorescence color changes to the five pesticides studied under a 365 nm ultraviolet lamp, we fabricated a single probe used to detect multiple analytes in the RGB triple channel by extracting the RGB variations. Principal component analysis and linear discriminant analysis proved that the ThT@Q[8] probe can recognize and distinguish five pesticides and can be applied at different concentrations. In real samples, the ThT@Q[8] probe recognized and distinguished five pesticides in tap water and Huaxi River water. The 1H NMR spectra results proved that a charge-transfer complex of ThT and pesticides in the Q[8] cavity may be formed. Moreover, we selected a test strip as a carrier to detect pesticides. The results indicate it can be used to quickly and conveniently detect different pesticides due to the rapid color change. Besides, the ThT@Q[8] probe has good cell permeability and can be used to detect pesticide residues in living cells. This work has laid the foundation for the qualitative and quantitative multitarget detection of pesticide residues.

Summary Quantitative fluorescence immunoassay is a vital and convenient method in point-of-care (POC) testing for food safety and clinical healthcare. However, current marking materials, from organic luciferin to quantum dots, still lack the requirement of increasing need for multiple targets of interest because of their too broad emission. Here, a new label material, perovskite nanocrystals (Pe-NCs), with a narrower photoluminescence spectrum is reported, and a sensitive and quantitative immunoassay method named the perovskite nanocrystal fluorescence-linked immunosorbent assay is preliminarily demonstrated. The key point of realizing water-dispersed and stable Pe-NCs was addressed by functionalizing with hydroxyl groups. The fluorescent probes (perovskite antibody) were constructed via electrostatic adsorption with narrow full-width at half-maximum (∼18 nm), laying the foundation for future study of multi-target detection. Both indirect competitive immunity and sandwich immunoassay were successfully demonstrated. This work provides a possible label material for POC tests and may open up a promising avenue of next-generation immunoassay.

Quantitative detection of multiple biological small molecules is critical for health evaluation and disease diagnosis. In this study, a microarray chip featuring a bienzyme-immobilized polyaniline nanowire forest on fluorine-doped tin oxide (bienzyme-PANI/FTO) is developed for this purpose. On such a chip, the target molecules are oxidized under the catalysis of their attached oxidases to produce hydrogen peroxide, which further induces the partial oxidation of local PANI nanowires in the presence of horseradish peroxidase (HRP) enzyme. The redox state change of PANI nanowires is monitored by the oblique incident reflectivity difference (OIRD) technique in a real-time and wireless manner, thus allowing for quantitative analysis of the target molecules. As typical model targets, hydrogen peroxide, glucose, lactic acid, and cholesterol are successfully detected with low detection limits, excellent specificities, and broad detection ranges, all of which fully meet the requirements for clinical analysis of human serum samples. Simultaneous detection of multiple targets on an individual chip is further demonstrated using the OIRD scanning mode. Meanwhile, by simple electrochemical reduction of the PANI nanowires, the chip is reusable for more than eight detection cycles without evident decay in its performance. The detection principle of this chip is also universal to other small molecules, and thus, it shows great promise as a valuable device to analyze biological small molecules.

Inflammatory responses and tumor developments are closely related, with interleukin-6 (IL-6) playing important roles in both processes. IL-6 has been extensively identified as a potential tumor biomarker. This study developed an isotope dilution mass spectrometry (IDMS) method for quantifying IL-6 based on signature peptides. These peptides were screened by excluding those with missed cleavage or post-translational modification. The method’s accuracy was verified using amino acid-based IDMS, in which purified IL-6 protein samples were quantified after hydrolyzing them into amino acids, and no significant difference was observed (p-value < 0.05). The method demonstrated good linearity and sensitivity upon testing. The specificity and matrix effect of the method were verified, and a precision study showed that the coefficient of variation was less than 5% for both the intra-day and inter-day tests. Compared to immunoassays, this method offers distinct advantages, such as the facilitation of multi-target analysis. Furthermore, the peptides used in this study are much more convenient for storage and operation than the antibodies or purified proteins typically used in immunoassays.

Biological staining is essential for imaging, functional investigation, and disease diagnostics. In this research, innovative green-emissive carbon dots (G-CDs) were hydrothermally prepared from 1,2,4-triaminobenzene and ascorbic acid (AA) and further explored for versatile imaging of mammalian cells, plant cells, and microorganisms. The G-CDs, exhibiting fluorescence enhancement, outperform conventional dyes in terms of staining efficiency and specificity across various biological systems. For mammalian cells (SH-SY5Y, Hep3B, and HEK293), the G-CDs can stain both the nuclei and nucleoli in 10 min via a micropinocytic endocytosis approach, with the latter brighter. Fluorescence enhancement was greater for RNA than for DNA. Interestingly, for plant cells, only cell walls were stained, because positive charges on G-CDs enable strong adsorption onto the negatively charged cell walls, thereby hindering their further entry. G-CDs can stain cell walls and nuclei for fungi (including Aspergillus flavus and Aspergillus niger ), and cell walls and nucleoids for bacteria ( Bacillus subtilis and Escherichia coli ). The targeted staining arises from both the binding of G-CDs to DNA, RNA, pectin, peptidoglycan, and cellulose, and fluorescence enhancement due to surface passivation inhibiting rotation and vibration of functional groups. Electrostatic interactions, coordination, hydrogen bonding, and amidation collectively account for the interactions between G-CDs and these biomolecules . Finally, their practical applications in leucorrhea examinations demonstrated their capability to differentiate between bacterial and fungal vaginosis, highlighting their potential in clinical diagnostics.

Electrochemical redox-active species play crucial role in electrochemically multiplexed immunoassays. A one-pot method for synthesizing four kinds of new electrochemical redox-active species was reported using HAuCl 4 and Na 2 PdCl 4 as dual oxidating agents and aniline derivatives as monomers. The synthesized polyaniline derivative-Au/Pd composites, namely poly(N-methyl-o-benzenediamine)-Au/Pd, poly(N-phenyl-o-phenylenediamine)-Au/Pd, poly(N-phenyl-p-phenylenediamine)-Au/Pd and poly(3,3’,5,5’-tetramethylbenzidine)-Au/Pd, exhibited electrochemical redox activity at −0.65 V, −0.3 V, 0.12 V and 0.5 V, respectively. Meanwhile, these composites showed high H 2 O 2 electrocatalytic activity because of the presence of Au/Pd. The as-prepared composites were used as electrochemical immunoprobes in simultaneous detection of four tumor biomarkers (carcinoembryonic antigen (CEA), carbohydrate antigen 19-9 (CA199), carbohydrate antigen 72-4 (CA724) and alpha fetoprotein (AFP)). This immunoassay shed light on potential applications in simultaneous gastric cancer (related biomarkers: CEA, CA199, CA724) and liver cancer diagnosis (related biomarkers: CEA, CA199, AFP). The present strategy to the synthesize redox species could be easily extended to other polymers such as polypyrrole derivatives and polythiophene derivatives. This would be of great significance in the electrochemical detection of more analytes.

Botanical pesticides are promising alternatives for sustainable pest management due to their environmental compatibility, biodegradability, and multitarget mechanisms that help delay pest resistance. Unfortunately, most botanical pesticides face challenges such as limited water solubility and poor foliar adhesion, thereby limiting their large-scale field application. Herein, the star polycation (SPc) nanocarrier and X-100 surfactant were introduced to successfully develop an efficient nanodelivery platform for veratramine (VAM). The SPc could spontaneously assemble with VAM via a hydrogen bond and van der Waals force, and the integration of Triton X-100 stabilized the nanoscale size (84 nm) to prepare a VAM nanopesticide. Interestingly, this optimized system reduced the contact angle and increased the retention of VAM on plant leaves while remarkably facilitating its systemic transport through roots and leaves. Compared to commercial VAM, the VAM nanopesticide showed stronger contact and stomach toxicity against multiple pest species such as aphids, thrips, and mites, which exhibited fast-acting properties in the field with high control efficacy even at 7 d after the spraying. Subsequently, the RNA-seq and biological experiments demonstrated that the application of VAM nanopesticide influenced various pathways in aphids, inducing starch and sucrose metabolism, protein digestion and absorption, etc., with inhibitory effects on the biosynthesis of trehalose and protein. Importantly, the application of the VAM nanopesticide did not adversely influence the predatory lady beetles or cowpea seedlings, confirming its safety in actual applications. Overall, our work addressed the formulation instability and environmental loss of VAM while ensuring its ecological safety, which offered a scalable nanoplatform to optimize botanical pesticides for enhanced efficacy and minimized ecological risks.

To enhance the precision and reliability of early disease detection, especially in malignancies, an exhaustive investigation of multi-target biomarkers is essential. In this study, an advanced integrated electrochemical biosensor array that demonstrates exceptional performance was constructed. This biosensor was developed through a controllable porous-size mechanism and in-situ modification of carbon nanotubes (CNTs) to quantify multiplex biomarkers—specifically, C-reaction protein (CRP), carbohydrate antigen 125 (CA125), and carcinoembryonic antigen (CEA)—in human serum plasma. The fabrication process involved creating a highly ordered three-dimensional inverse-opal structure with the CNTs ( p CNTs) modifier through microdroplet-based microfluidics, confined spatial self-assembly of nanoparticles, and chemical wet-etching. This innovative approach allowed for direct in-situ modification of nanomaterial onto the surface of electrode array, eliminating secondary transfer and providing exceptional control over structure and stability. The outstanding electrochemical performance was achieved through the synergistic effect of the p CNTs nanomaterial, aptamer, and horseradish peroxidase-labeled (HRP-) antibody. Additionally, the integrated biosensor array platform comprised multiple individually addressable electrode units ( n = 11), enabling simultaneous multi-parallel/target testing, thereby ensuring accuracy and high throughput. Crucially, this integrated biosensor array accurately quantified multiplex biomarkers in human serum, yielding results comparable to commercial methods. This integrated technology holds promise for point-of-care testing (POCT) in early disease diagnosis and biological analysis.
