Insecticidal activities of Salvia hispanica L. essential oil and combinations of their main compounds against the beet armyworm Spodoptera exigua

Yijuan Chen,Jiaxuan Luo,Nan Zhang,Wenjuan Yu,Jiexian Jiang,Guanghui Dai

Plant essential oils are the powerful candidates to develop natural insect management agents. Their chemical compounds along with their biological activity are often affected by many factors. However, the relationships between the composition and bioactivity of the compounds are not well understood. In this study, eight plant essential oils were screened, and the main compounds from the most effective oil showing insecticidal properties against third-instar larvae of the beet armyworm ( Spodoptera exigua Hübner) were evaluated for comparative and synergistic interactions by topical application. The results indicated that Salvia hispanica L. essential oil showed significant insecticidal activity against S. exigua compared with the other seven essential oils. Fifteen major compounds were identified from S. hispanica via gas chromatography-mass spectrometry, with the most abundant compound (α-thujone) possessing the highest insecticidal activity. Several combinations of the four major compounds (α-thujone, (+)-camphor, 1,8-cineole, and α-caryophyllene) from S. hispanica exhibited synergistic insecticidal activities. This study demonstrates the potential of S. hispanica essential oil and α-thujone as effective botanical insecticides in the control of beet armyworm and provides a better understanding of essential oil compound-pest interactions.

Repellence or attraction: secondary metabolites in pepper mediate attraction and defense against Spodoptera litura

Ge-Ge Yuan,Lin-Chao Zhao,Yuan-Wen Du,Huan Yu,Xiao-Bin Shi,Wen-Chao Chen,Gong Chen

BACKGROUND Resistance to insect pests is an important self-defense characteristic of pepper plants. However, the resistance of different pepper cultivars to Spodoptera litura larvae, one of the main insect pest species on pepper, is not well understood. RESULTS Among seven pepper cultivars evaluated, cayenne pepper ‘FXBX’ showed the highest repellency to third instar S. litura larvae, Chao tian chili pepper ‘BLTY2’ showed the lowest repellency. Plant volatiles (1-hexene, hexanal, β -ionone, ( E , E )-2,6-nonadienal, and methyl salicylate) affected host selection by S. litura . Among these, 1-hexene, hexanal, and β -ionone at concentrations naturally-released by pepper leaves were found to repel S . litura . Interestingly, S. litura larvae fed on the larva-attracting pepper cultivar, (BLTY2) had an extended developmental period, which was about 13 days longer than larvae fed on FXBX. Besides, the survival rate of larvae fed on BLTY2 was 22.5 ± 0.0%, indicating that the leaves of BLTY2 can kill S. litura larvae. Correlation analysis showed that larval survival rate, emergence rate, female adult longevity, and pupal weight were positively correlated with the vitamin C, amino acids, protein, cellulose, and soluble sugar contents, but were negatively correlated with wax and flavonoids contents. CONCLUSION We identified two different modes of direct defense exhibited by pepper cultivars against S. litura . One involves the release of repellent volatiles to avoid been fed on (FXBX cultivar). The other involves the inhibition of the growth and development or the direct killing of S. litura larvae which feeds on it (BLTY2 cultivar). © 2022 Society of Chemical Industry.

Down-Regulation of P450 Genes Enhances Susceptibility to Indoxacarb and Alters Physiology and Development of Fall Armyworm, Spodoptera frugipreda (Lepidoptera: Noctuidae)

Hafeez Muhammad,Li Xiaowei,Ullah Farman,Zhang Zhijun,Zhang Jinming,Huang Jun,Fernández-Grandon G. Mandela,Khan Muhammad Musa,Siddiqui Junaid Ali,Chen Limin,Ren Xiao Yun,Zhou Shuxing,Lou Yonggen,Lu Yaobin

The fall armyworm (FAW), Spodoptera frugiperda (J.E. Smith), is a pest of many important crops globally. Effective control is challenging, with the pest exhibiting resistance to different synthetic pesticides across various groups. However, the mechanisms employed by resistant insects for overexpression of relevant detoxification genes remain unclear. The activity of detoxification enzymes was investigated in this study. Additionally, using RNA interference (RNAi), a functional analysis was completed of two P450s genes in an indoxacarb resistant population of fall armyworms. Elevated resistance levels (resistance ratio = 31.37-fold) in indoxacarb-selected populations of FAW were observed after 14 generations. The qRT-PCR showed higher expression of two cytochrome P450 genes, CYP321A7 and CYP6AE43, in this selected population compared to the control population. RNAi was applied to knock down the P450 dsCYP321A7 and dsCYP6AE43 genes in the FAW larvae. Droplet feeding of the dsRNAs (CYP321A7 and CYP6AE43) via an artificial diet significantly increased mortality rates in the indoxacarb treated population. A shorter larval developmental time of FAW was detected in all dsRNAs-fed larvae. Correspondingly, larval mass was reduced by dsRNAs in indoxacarb resistant populations of fall armyworm. Larval feeding assays demonstrate that dsRNAs targeting, specifically of CYP321A7 and CYP6AE43 enzymes, could be a beneficial technique in the management of indoxacarb resistant populations. Further study on the potential use of dsRNA and its application should be conducted in efforts to counter the development of resistance in FAW against various insecticides in the field.

Structural design and insecticidal activity of 1,3,4-oxadiazole-ring containing pyridylpyrazole-4-carboxamides

Abdalla Khallaf,Ping Wang,Shuping Zhuo,Hongjun Zhu,Hui Liu

The insecticidal activity of 1,3,4-oxadiazole-ring containing pyridylpyrazole-4-carboxamides against diamondback ( Plutella xylostella ) and noctuid ( Spodoptera frugiperda ) moths were evaluated. Only substitutions on the 1,3,4-oxadiazole moiety dictated bioreactivity, with 8q2 m -CF 3 Ph being the most effective against Plutella xylostella, showing 80%–93% mortality at a concentration of 0.5–500 μg ml −1 while the others remained inactive or less active (highest mortality being 63%). The larvicidal activity against Spodoptera frugiperda was inferior compared with that of Plutella xylostella , with the highest mortality being 33%. A crystal of 8q5 was obtained and analyzed, revealing the effects of intramolecular H-bonds and intermolecular π  π interactions.

Characterization of Indoxacarb Resistance in the Fall Armyworm: Selection, Inheritance, Cross-Resistance, Possible Biochemical Mechanisms, and Fitness Costs

Muhammad Hafeez,Xiaowei Li,Farman Ullah,Zhijun Zhang,Jinming Zhang,Jun Huang,Limin Chen,Junaid Ali Siddiqui,Xiaoyun Ren,Shuxing Zhou,Muhammad Imran,Mohammed A. Assiri,Myron P. Zalucki,Yonggen Lou,Yaobin Lu

Simple SummaryThe fall armyworm (FAW),Spodoptera frugiperda(J.E. Smith), is a voracious insect pest native to the Western Hemisphere, particularly in South America. The polyphagous fall armyworm feeds on more than 350 plants in such families, including Poaceae, Asteraceae, and Fabaceae. Transgenic plants that expressBacillus thuringiensisproteins (Bt plants) and synthetic insecticides are the main tactics to controlSpodoptera frugiperda, although widespread usage of synthetic chemicals has resulted in the emergence of resistance. We assessed cross-resistance, resistance mechanism, and fitness costs based on the life history traits ofSpodoptera frugiperda. Results indicated that after 24 generations of selection, the resistance to indoxacarb was increased by 472.67-fold as compared to the Ind-UNSEL. Significantly longer developmental time of larvae extended pupal duration, shorter adult longevity, and lower fecundity were observed in Ind-SEL as compared with the Ind-UNSEL population. Butoxide synergist increased susceptibility to indoxacarb, indicating that P450 enzymes may be involved in indoxacarb resistance. Therefore, it is crucial that we comprehend how insecticides work and how resistance develops in order to develop techniques for managing resistance. These data are valuable to understand the indoxacarb resistance mechanism and provides vital information for scientific-based guidance of pest management decisions.AbstractThe fall armyworm,Spodoptera frugiperda(J.E. Smith), is a voracious insect pest that is difficult to control due to resistance to insecticides and Bt proteins. We assessed cross-resistance, resistance mechanism, and fitness costs based on the life history traits ofS. frugiperda. We established anS. frugiperdastrain selected for resistance to indoxacarb (Ind-SEL) from a field-collected population and an unselected strain, Ind-UNSEL. Results indicated that after 24 generations of selection, the resistance to indoxacarb was increased by 472.67-fold as compared to the Ind-UNSEL. There was high cross-resistance to deltamethrin (31.23-fold) with very low or negligible cross-resistance to chlorantraniliprole, emamectin benzoate, and/or methoxyfenozide in the Ind-SEL population. Butoxide synergist increased susceptibility to indoxacarb, indicating that P450 enzymes may be involved in indoxacarb resistance. Significantly longer developmental time of larvae extended pupal duration, shorter adult longevity, and lower fecundity were observed in Ind-SEL as compared with the Ind-UNSEL population. The Net reproductive rate (R0) was the only growth parameter that differs between crosses of Ind-SEL♂ × Ind-UNSEL♀ (176 ± 46) and Ind-SEL♀ × Ind-UNSEL♂ (328 ± 57). On the other hand, all population growth parameters differ between Ind-SEL and Ind-UNSEL strains. Our work contributes to the growing body of research that demonstrates the importance of strain genetics in fitness cost experiments and helps resistance management programs make decisions. Keywords:Spodoptera frugiperda;indoxacarb selection;cross-resistance;inheritance of resistance;fitness cost

Indoxacarb resistance-associated mutation of Liriomyza trifolii in Hainan, China

Fen Li,Xueyan Gong,Linlin Yuan,Xuelian Pan,Haifeng Jin,Rongcai Lu,Shaoying Wu

Liriomyza trifolii , which has been recently prevalent in China, harms more than 300 plant species, especially cowpea in Hainan. This pest also affects the quality and production of vegetables in winter. Indoxacarb is the first commercial oxadiazine pesticide , which is a new efficient insecticide used to control pests of Diptera, including L. trifolii . The unique mechanism of indoxacarb is that indenyl is transformed into N-demethoxycarbonyl metabolite (DCJW) in insects and acts on inactivated sodium channel; DCJW could then destroy the conduction of nerve impulses , which leads to movement disorders, feeding stoppage, paralysis, and eventually the death of pests. The field population of L. trifolii developed resistance by 769 times higher than the sensitive population in Sanya, Hainan. Results revealed the existence of a mutation (i.e., V1848I) in the sixth transmembrane segment of Domain IV of the sodium channel in the field population. The homozygous resistant genotype frequency for the V1848I mutation was 10–15% among the three field-collected populations. This paper reports for the first time the presence of the kdr mutation V1848I in resistant populations of L. trifolii to indoxacarb. The present study will contribute to the understanding of the evolution of indoxacarb resistance and contribute to the development of resistance management practices for winter vegetables in Hainan.

Preparation of a Chlorantraniliprole–Thiamethoxam Ultralow-Volume Spray and Application in the Control of Spodoptera frugiperda

Kun Wei,Weiming Xu,Qin Liu,Liyun Yang,Zhuo Chen

Traditional pesticide spraying methods such as knapsack sprayers are inefficient. Ground application equipment is restricted by the terrain and damages the crops. Pesticides cannot be sprayed over a large area in a short time; thus, when there are concentrated outbreaks of pests with strong migration capacity, such as Spodoptera frugiperda, pests could not be controlled in a small area. Therefore, it is difficult to achieve a satisfactory control effect. Plant protection unmanned aerial vehicles (UAVs) have high efficiency and spray pesticides at low altitude and can suitably prevent and control the outbreak of pests. However, pesticide formulations suitable for UAVs are rare. In this study, we developed ultralow-volume (ULV) sprays suitable for plant protection UAVs. We studied their properties and found that the ULV sprays have good wettability and can be fully spread. The minimum contact angle on corn leaves is up to 19°, and the maximum retention is 2.7 times that of the reference product. They also have superior resistance to rain washout and can effectively extend the duration and utilization of pesticides. When sprayed by plant protection UAVs, the decrease rate of the ULV spray at 3 L/ha to S. frugiperda larvae is 81.31% and the control effect is 83.01%, both higher than those of the reference product 40% chlorantraniliprole–thiamethoxam water-dispersible granules (decrease rate 67.39%, control effect 70.35%), sprayed with a knapsack sprayer.

Preparation and insecticidal activities of betulin-cinnamic acid-related hybrid compounds and insights into the stress response of Plutella xylostella L.

Xiaobo Huang,Nan Hao,Lili Shu,Zhenyang Wei,Jiawei Shi,Yuee Tian,Genqiang Chen,Xinling Yang,Zhiping Che

BACKGROUND The emergence and increasing intensification of insect pest resistance greatly shortens the service life of commercial insecticides. Hence, the continuous development of novel insecticides is extremely necessary. Discovering new insecticide molecules based on natural products with insecticidal activity has been a popular technique and has made outstanding contributions in this field. RESULTS A series of betulin-cinnamic acid-related hybrid compounds was prepared by molecular hybridization through the bridge of acyloxy groups and their chemical structures were unambiguously confirmed. Against A. pisum , compounds 2e , 3a-3e , 3g , 3i , 3j , and 3m showed complete antifeedant activity at 2 mg/mL; compound 5b exhibited 2.7 times the aphicidal activity of the precursor betulin. Against P. xylostella , compound 2l displayed 3.9 times the insecticidal activity of betulin. Meanwhile, some interesting structure–activity relationships were obtained. Furthermore, compound 2l could destroy the ultrastructure of midgut cells and significantly inhibit the activity of α-amylase in P. xylostella larvae. Molecular docking indicated that the lupane skeleton and p -nitrocinnamoyloxy groups played a key role in the binding of compound 2l to α-amylase. CONCLUSION Compounds 5b and 2l could be further optimized structurally as the secondary lead compounds for the discovery of novel insecticide molecules. © 2025 Society of Chemical Industry.

Susceptibility of fall armyworm, Spodoptera frugiperda (J.E. Smith), to eight insecticides in China, with special reference to lambda-cyhalothrin

Yun-Xia Zhao,Jing-Mei Huang,Huan Ni,Di Guo,Feng-Xia Yang,Xin Wang,Shun-Fan Wu,Cong-Fen Gao

Fall armyworm (FAW), Spodoptera frugiperda (J.E. Smith), is the main destructive insect pest of grain crops that occurs in all maize growing regions of the Americas. It has rapidly invaded the Southern China since January 2019. However, the current status of insecticide resistance in S. frugiperda has not been reported in China. In this study, we determined the susceptibility of eight populations of FAW to eight insecticides by an artificial diet incorporation method. The results showed that among eight insecticides, emamectin benzoate, spinetoram, chlorantraniliprole, chlorfenapyr, and lufenuron showed higher toxicity to this pest, while lambda-cyhalothrin and azadirachtin exhibited lower toxicity. Susceptibility of S. frugiperda to indoxacarb was significantly different (10.0-fold for LC 50 ) across the various geographic populations. To investigate the biochemical mechanism of FAW to lambda-cyhalothrin, we performed the synergism tests and the results showed that piperonyl butoxide (PBO) and triphenyl phosphate (TPP) produced a high synergism of lambda-cyhalothrin effects in the two field populations. Sequencing of the gene encoding the acetylcholinesterase (AChE) gene in the two field populations identified two amino acid mutations, all of which have been shown previously to confer resistance to organophosphates (OPs) in several arthropod species. The results of this study provided valuable information for choosing alternative insecticides and for insecticide resistance management of S. frugiperda .

Synthesis and insecticidal activity of sulfonate derivatives of sesamol against Mythimna separata in vivo

Zhi-Ping Che,Jin-Ming Yang,Xi-Jie Shan,Yue-E Tian,Sheng-Ming Liu,Xiao-Min Lin,Jia Jiang,Mei Hu,Gen-Qiang Chen

A series of sulfonate derivatives of sesamol were synthesized and evaluated for their insecticidal activity against a crop-threatening agricultural pest, the pre-third-instar larvae of Mythimna separata in vivo. Among all the target compounds, compounds 3b, 3g, 3h, and 3p exhibited more promising insecticidal activity than sesamol and toosendanin, and the final mortality rates (FMRs) of 3b, 3g, 3h, 3p, 1, and toosendanin were 60.7%/60.7%/67.9%/53.6%/32.1%/50.0%, respectively. Especially compound 3h exhibited the most potent insecticidal activity with FMRs of 67.9%. This suggested that a 4-fluorophenylsulfonyl group introduced at the hydroxyl position of sesamol was necessary for obtaining the most potent compound.

Self-assembled nano-vesicles based on mPEG-NH2 modified carboxymethyl chitosan-graft-eleostearic acid conjugates for delivery of spinosad for Helicoverpa armigera

Chuang Zhou,Ziming Yang,Li Zhang,Enming Dong,Zuyu He,Xianwu Liu,Chao Wang,Yan Yang,Jing Jiao,Yunhao Liu,Yu Chen,Puwang Li

In this work, carboxymethyl chitosan-graft-eleostearic acid (CMCS-g-EA) was synthesized via the amide reaction between the amino groups of carboxymethyl chitosan and the carboxyl group of eleostearic acid, and then mPEG-NH 2 was grafted to CMCS-g-EA to prepare amphiphilic polymers (mPEG-CMCS-g-EA). The chemical structures of the above conjugates were characterized by FT-IR and 1 H NMR. Both CMCS-g-EA and mPEG-CMCS-g-EA based nano-vesicles were prepared by ultrasonic self-assembly method and they exhibited a low critical aggregation concentration (CAC) of 14.97 μg/mL, 16.82 μg/mL, respectively. The spinosad-loaded mPEG-CMCS-g-EA nano-vesicles (SSD@mPEG-CMCS-g-EA NVs) were spherical in shape with an average diameter of 502.8 nm and the zeta potential of −25.60 mV. The encapsulation efficiency (EE) and drug loading content (LC) of SSD@mPEG-CMCS-g-EA nano-vesicles were 42.00%, 23.07%, respectively. In vitro release revealed that the SSD@mPEG-CMCS-g-EA nano-vesicles exhibited a sustained and pH-responsive drug release property, and could significantly enhance the photostability of spinosad . Furthermore, the toxicological tests demonstrated that the SSD@mPEG-CMCS-g-EA nano-vesicles could efficiently inhibit the growth and development of Helicoverpa armigera . These results indicated that the SSD@mPEG-CMCS-g-EA nano-vesicles were highly potential for the treatment of Helicoverpa armigera .

Effects of azadirachtin on detoxification-related gene expression in the fat bodies of the fall armyworm, Spodoptera frugiperda

Yu Haikuo,Yang Xianmei,Dai Jinghua,Li Yuning,Veeran Sethuraman,Lin Jintian,Shu Benshui

The fall armyworm, Spodoptera frugiperda, has become a worldwide pest and threatens world food production. A previous study indicated that azadirachtin, the most effective botanical insecticide for S. frugiperda , inhibits larval growth of the insect. The effect of azadirachtin on the tissues of the larvae, however, remains to be determined. In this study, the effects of azadirachtin on the structure of fat bodies were analyzed. Comparative transcriptomic analysis was conducted between controls and samples treated with 0.1 μg/g azadirachtin for 7 days to explore potential relevant mechanisms. The expression of 5356 genes was significantly affected after azadirachtin treatment, with 3020 up-regulated and 2336 down-regulated. Among them, 137 encode detoxification enzymes, including 53 P450s , 20 GSTs , 27 CarEs , 16 UGTs , and 12 ABC transporters . Our results indicated that azadirachtin could destroy fat body structure and change the mRNA levels of detoxification-related genes. The up-regulated genes encoding detoxification enzymes might be related to detoxifying azadirachtin. Our results elucidate a preliminary mechanism of azadirachtin detoxification in the fat bodies of S. frugiperda larvae.
