Degradation kinetics of rutin encapsulated in oil-in-water emulsions: impact of particle size

Yuteng Fu,David Julian McClements,Shunjing Luo,Jiangping Ye,Chengmei Liu

BACKGROUND Rutin is a natural bioactive flavonoid that is poor in water solubility and chemical stability. Encapsulation can be used to protect bioactive molecules from chemical or physical decomposition during food processing and storage. Thus, the effect of initial particle size on the ability of oil-in-water emulsions to retain rutin during storage was investigated. RESULTS Rutin was encapsulated in oil-in-water emulsions with different mean surface-weighted diameters: d 3,2 = 0.56 μm (small), 0.73 μm (medium), and 2.32 μm (large). As expected, the resistance of the emulsions to coalescence and creaming during storage increased as the particle size decreased due to weakening of the colloidal and gravitational forces acting on the droplets. The concentration of rutin in the emulsions decreased during storage (28 days), which was mainly attributed to photodegradation of the flavonoid. The loss of rutin from the emulsions during storage was fitted using a second-order equation. The rutin degradation rate constant k decreased and the half-life t 1/2 increased with decreasing droplet size, which was attributed to the stronger encapsulation and light scattering by smaller oil droplets reducing the amount of light that can penetrate into the emulsions. CONCLUSION This study has important implications for the design of more efficacious emulsion-based delivery systems for incorporating health-promoting nutraceuticals into foods. © 2022 Society of Chemical Industry.

Gut-Associated Bacteria of Dendroctonus valens and their Involvement in Verbenone Production

Xu Letian,Lou Qiaozhe,Cheng Chihang,Lu Min,Sun Jianghua

Bark beetles are the most important mortality agent in coniferous forests, and pheromones play important roles in their management. Dendroctonus valens LeConte was introduced from North America to China and has killed millions of healthy pines there. Trapping with semiochemicals and pheromones was deployed in D. valens management in the last decade, but little is known about the ability of gut bacteria to produce the pheromone. In this study, we analyzed the volatiles in D. valens guts and frass after antibiotic treatment versus control. Then, we isolated and identified the bacteria in D. valens guts and frass, examined verbenone (a multifunctional pheromone of D. valens ) production by 16 gut bacterial isolates from the precursor cis -verbenol at three concentrations, and further compared the cytotoxicities between the cis -verbenol and verbenone to the bacterial isolates. cis -Verbenol was not detected in the frass in the control group, but it was in the antibiotic treatment. The amount of verbenone was significantly suppressed in D. valens guts after antibiotic treatment versus control. Thirteen out of 16 gut bacterial isolates were capable of cis -verbenol to verbenone conversion, and cis -verbenol had stronger cytotoxicities than verbenone to all tested gut bacterial isolates. The bacterial species capable of verbenone production largely exists in D. valens guts and frass, suggesting that gut-associated bacteria may help the bark beetle produce the pheromone verbenone in guts and frass. The bacteria may benefit from the conversion due to the reduced cytotoxicity from the precursor to the beetle pheromone.

Comparative studies on the stabilization of pea protein dispersions by using various polysaccharides

Yue Wei,Zhixiang Cai,Min Wu,Yalong Guo,Ran Tao,Ruiqi Li,Pengguang Wang,Aiqin Ma,Hongbin Zhang

The utilization of pea protein as an alternative to animal proteins for the production protein fortified foods and beverage products has recently gained tremendous interest due to the quality and health benefits associated with pea proteins. However, its use in food systems is often limited by technical issues such as the stability of pea protein dispersions (PPD). The objective of this study was to improve the physical stability of PPD by using corn fiber gum (CFG), high-methoxy pectin (HMP), carboxymethyl cellulose (CMC), and konjac glucomannan (KGM). The stabilization mechanism of PPD by these four different polysaccharides was investigated at native pH (roughly 5.5–7.5) and acidified pH (ca. 3.5) using particle size, zeta potential , rheological and physical stability evaluations. All four polysaccharides effectively improved the stability of PPD at various levels, but the physical stability of PPD was highly dependent on the type and concentration of the polysaccharides as well as the pH. Electrostatic repulsion, steric hindrance and the high viscosity of continuous phase were the main contributions to the improvement in PPD stability. Overall, these results have significant implications for the development of protein-based beverages with improved physical stability. The findings derived from this research can provide valuable information that may increase the utilization of pea protein in the food industry.

Ring-Size Effects on the Stability and Spectral Shifts of Hydrogen Bonded Cyclic Ethers Complexes

Tang Shanshan,Tsona Narcisse T.,Du Lin

In order to investigate ring-size effects on the stability and spectral shifts of hydrogen bonded cyclic ethers complexes, the strength of hydrogen bonds in gas phase complexes formed between 2,2,2-trifluoroethanol (TFE) and selected cyclic ethers were examined using FTIR spectroscopy. TFE was chosen as hydrogen bond donor in these complexes, while trimethylene oxide (TMO), tetrahydrofuran (THF) and tetrahydropyran (THP) were selected as hydrogen bond acceptors. Comparable OH-stretching red shifts were observed in the three kinds of complexes. The difference of red shifts is so small (<7 cm −1 ) for TFE−TMO/THF/THP complexes that one can conclude that their stabilities and the strength of the hydrogen bonds are nearly similar and do not show any marked dependence with the ring size of the hydrogen bond acceptor. The equilibrium constants for the complexation were determined, and atoms-in-molecules (AIM) and natural bond orbital (NBO) analyses were performed to further investigate the intermolecular interactions. Regardless of the ring size, hydrogen bonds in the complexes showed similar strength, in agreement with the observed OH-stretching red shifts.

Batch preparation of homogeneous size hydroxyapatite based composite ceramic microspheres

Zhou Kui,Zou Jiawei,Chi Wenchao,Lin Yanjun,Li Wenchao,Cao Chuanliang

A new and precise control printing method was developed for preparing homogeneous microspheres. Hydroxyapatite-sodium alginate (HA-SA) mixed suspension was used as printing material and CaCl 2 solution was used as solidifying liquid to prepared ceramic microspheres by pneumatic extrusion printing technology. By adjusting the positive pressure, dwell time, and hydroxyapatite concentration, the influence of each parameter on the shape of printed microspheres was studied, and the suitable preparation parameters were determined. Then, the microspheres with uniform particle size distribution were successfully prepared in large quantities by selecting appropriate printing parameters. Then, the microspheres were dried and sintered at different temperatures. During the drying and sintering process, the microspheres showed obvious shrinkage, but still keep good sphericity. The phases of sintered microspheres are hydroxyapatite and calcium oxide. The microstructure shows typical hydroxyapatite grain characteristics.

Nanocellulose and Cellulose Making with Bio-Enzymes from Different Particle Sizes of Neosinocalamus Affinis

Jiaxin Zhao,Xiaoxiao Wu,Xushuo Yuan,Xinjie Yang,Haiyang Guo,Wentao Yao,Decai Ji,Xiaoping Li,Lianpeng Zhang

Cellulose is one of the most abundant, widely distributed and abundant polysaccharides on earth, and is the most valuable natural renewable resource for human beings. In this study, three different particle sizes (250, 178, and 150 μm) of Neosinocalamus affinis cellulose were extracted from Neosinocalamus affinis powder using bio-enzyme digestion and prepared into nanocellulose (CNMs). The cellulose contents of 250, 178, and 150 μm particle sizes were 53.44%, 63.38%, and 74.08%, respectively; the crystallinity was 54.21%, 56.03% and 63.58%, respectively. The thermal stability of cellulose increased gradually with smaller particle sizes. The yields of CNMs for 250, 178, and 150 μm particle sizes were 14.27%, 15.44%, and 16.38%, respectively. The results showed that the Neosinocalamus affinis powder was successfully removed from lignin, hemicellulose, and impurities (pectin, resin, etc.) by the treatment of bio-enzyme A (ligninase:hemicellulose:pectinase = 1:1:1) combined with NH3·H2O and H2O2/CH3COOH. Extraction of cellulose from Neosinocalamus affinis using bio-enzyme A, the smaller the particle size of Neosinocalamus affinis powder, the more cellulose content extracted, the higher the crystallinity, the better the thermal stability, and the higher the purity. Subsequently, nanocellulose (CNMs) were prepared by using bio-enzyme B (cellulase:pectinase = 1:1). The CNMs prepared by bio-enzyme B showed a network structure and fibrous bundle shape. Therefore, the ones prepared in this study belong to cellulose nanofibrils (CNFs). This study provides a reference in the extraction of cellulose from bamboo using bio-enzymes and the preparation of nanocellulose. To a certain extent, the utilization of bamboo as a biomass material was improved. Keywords:Neosinocalamus affinis;cellulose;nanocellulose;bio-enzymes;particle size

Molecular mechanism underlying the effects of temperature and pH on the size and surface charge of octenylsuccinated oat β-glucan aggregates

Zhen Wu,Chenyang Zhao,Yongxia Huang,Fayin Ye,Guohua Zhao

Environmental temperature and pH induced significant changes in the size and surface charge ( ζ ) of octenylsuccinated oat β -glucan aggregates. The underlying mechanisms were explored by using 1 H-NMR, fluorescence spectra, thermodynamic analysis, and SAXS. At pH 6.5, the size decreased with temperature while ζ continuously increased. With increasing pH at 293 K, parabolic and U-shaped trends were observed in the size and ζ , peaking at pH 8.5 and 6.5, respectively. At any tested pH, the size decreased with temperature. Overall, ζ significantly increased with temperature at each pH. As temperature increased, the compactness of hydrophobic-domains increased while the compactness of hydrophilic-domains decreased. In an acidic environment, both the compactness increased with decreasing pH, but in an alkaline environment, they decreased with pH. The compactness changes were co-driven by enthalpy and entropy and corresponded to changes in the hydrophobic interactions in hydrophobic-domains, hydrogen bonds in hydrophilic-domains and electrostatic repulsions among octenylsuccinate molecules.

Size control of ropivacaine nano/micro-particles by soft-coating with peptide nanosheets for long-acting analgesia

Jing Liu,Weiwei Wu,Fei Peng,Deying Gong,Yi Kang,Yujun Zhang,Congyan Liu,Yuncheng Li,Guoyan Zhao,Feng Qiu,Wensheng Zhang

Rationale: To meet the need of long-acting analgesia in postoperative pain management, slow-releasing formulations of local anesthetics (LAs) have been extensively investigated. However, challenges still remain in obtaining such formulations in a facile and cost-effective way, and a mechanism for controlling the release rate to achieve an optimal duration is still missing. Methods: In this study, nanosheets formed by a self-assembling peptide were used to encapsulate ropivacaine in a soft-coating manner. By adjusting the ratio between the peptide and ropivacaine, ropivacaine particles with different size were prepared. Releasing profile of particles with different size were studied in vitro and in vivo . The influence of particle size and ropivacaine concentration on effective duration and toxicity were evaluated in rat models. Results: Our results showed that drug release rate became slower as the particle size increased, with particles of medium size (2.96 ± 0.04 μm) exhibiting a moderate release rate and generating an optimal anesthetic duration. Based on this size, formulations at different ropivacaine concentrations generated anesthetic effect with different durations in rat sciatic nerve block model, with the 6% formulation generated anesthetic duration of over 35 h. Long-acting analgesia up to 48 h of this formulation was also confirmed in a rat total knee arthroplasty model. Conclusion: This study provided a facile strategy to prepare LA particles of different size and revealed the relationship between particle size, release rate and anesthetic duration, which provided both technical and theoretical supports for developing long-acting LA formulations with promising clinical application.

Enhanced adsorption performance of varying-length mRNA on Oligo dT affinity resins through optimal pore size and grafting

Yuan-Zhi Tan,Liang-Zhi Qiao,Shan-Shan Wang,Juan Zhang,Jing Qian,Min Zhu,Shan-Jing Yao,Dong-Qiang Lin

The enormous potential of mRNA-based therapeutics in vaccines and medicine has led to the consideration of various mRNA sizes, ranging from ∼1000 to 10,000 nucleotides (nt). However, the commonly used Oligo-deoxythymidine (Oligo dT) affinity resins are not initially designed for such a wide range of sizes, and present challenges in achieving optimal performance for the separation of a given mRNA. In this paper, two factors of the resin beads, pore size and grafting were focused. The effects of varying pore sizes, ranging from 150 to 850 nm, on both static and dynamic adsorption for varying-length mRNA (1000–8500 nt) were investigated. It was found that static adsorption capacity and uptake kinetics were correlated with the mRNA length. Interestingly, dynamic binding capacities (DBCs) showed an increase-then-decrease trend with increasing pore size. An optimal pore size of about 350 nm could lead to the highest DBCs for all tested mRNA. Additionally, the effects of polymer grafting on the adsorption performance were studied. The results revealed that polymer-grafted resin exhibited 24–55% higher DBCs than non-grafted resin. The enhanced DBCs could be attributed to the improved uptake rate facilitated by the polymer grafted. These investigations emphasize the importance of optimizing pore size and grafting for Oligo dT affinity resins, providing valuable guidance for selecting and designing new resins for different mRNA products.

Stability and rheological behaviors of different oil/water emulsions stabilized by natural silk fibroin

Xiuying Qiao,Lijun Wang,Zhengzhong Shao,Kang Sun,Reinhard Miller

In this study, oil/water emulsions stabilized by natural silk fibroin were prepared, and the effects of silk fibroin concentration ( φ SF ), oil volume fraction ( ϕ o ) and oil type on the stability and rheological behavior investigated. After creaming and reaching a stable state, the residual emulsion fraction increases with an increase in φ SF , ϕ o and oil polarity, while the size of emulsion droplets decreases with an increase in φ SF and oil polarity but a decrease in ϕ o . Due to the colloidal networks formed by the dispersed oil droplets, the emulsions show a predominantly elastic behavior with a frequency independent modulus and low phase angle. However, the droplet networks are not very strong, and a remarkable shear-thinning behavior occurs for all emulsions during steady shear flow. Moreover, stress overshoot also appears for some emulsions with low φ SF and polar oil during transient shear flows, due to the deformation of droplets before the breakage of their associates. The modulus, viscosity and yield stress become larger with the increase of φ SF , ϕ o and oil polarity, which is very beneficial for strengthening the creaming stability of the emulsions.

Gelatin-Based Nanocomplex-Stabilized Pickering Emulsions: Regulating Droplet Size and Wettability through Assembly with Glucomannan

Weiping Jin,Jieyu Zhu,Yike Jiang,Ping Shao,Bin Li,Qingrong Huang

Particle size and surface wettability play leading roles in the distribution of particles on the oil–water interface and the stability of emulsions. This work utilized nanocomplexes assembled from gelatin and tannic acid to stabilize Pickering emulsions. The sizes and surface wettability of particles were further regulated by using a polysaccharide. The sizes of nanocomplexes ranged from 205.8 to 422.2 nm and increased with the addition of polysaccharide. Their contact angles decreased from 84.1° to 59.3°, revealing their hydrophilic nature. Results of fluorescence microscopy and cryo-scanning electron microscopy indicated that nanocomplexes were located at the oil–water interface. Interfacial shear and dilatational rheological data revealed a fast and irreversible adsorption behavior, which differed from rearrangement of gelatin molecules at the oil–water interface. The minimal concentration of nanocomplexes required to stabilize emulsions was 0.1 wt %. Our results demonstrated that protein–polyphenol–polysaccharide nanocomplexes had the potential to be applied to form stable surfactant-free food emulsions for the delivery of nutraceuticals.

Influence of Nanoemulsion Droplet Size of Removing Water Blocking Damage in Tight Gas Reservoir

Yuan Li,Fujian Zhou,Jie Wang,Bojun Li,Hang Xu,Erdong Yao,Longhao Zhao

During the production process, water phase incursion into the reservoir causes water blocking damage and seriously affects the production of tight gas reservoirs. Recently, nanoemulsions have been used as highly effective water blocking removing agents in the field, but their mechanism is still unclear. In this research, a series of nanoemulsions with different droplet sizes were synthesized, and their water blocking removing performance was intensively investigated. To begin, the relationship between the droplet size and the chemical composition of the nanoemulsion was determined by dynamic light scattering. Second, the influence of the nanoemulsion droplet size on the surface tension and the contact angle experiments was studied. Finally, NMR and permeability recovery experiments were used to study the relationship between the droplet size and the water locking removing effect of the nanoemulsions. Simultaneously, the surfactant release process was investigated using the static adsorption curves of the nanoemulsions. The experimental results show that the droplet size of nanoemulsion has an exponential relationship with the oil phase content. The surface tension decreases with the increase in droplet size, but the wetting reversal effect decreases with the increase in droplet size. The nanoemulsion with an oil phase content of 5 wt.% has the best water locking removing effect, and the permeability recovery value of the core reaches 59.54%. The adsorption control of the nanoemulsion on the surfactant is the key to its water blocking removing ability. This comprehensive study shows that the nanoemulsion with an oil phase content of 5 wt.% has optimum adsorption control capability. Thus, it can be used as a promising candidate for removing water blocking in tight gas reservoirs.
