Pharmacokinetics and acetylation of sulfamethoxazole in turbot Scophthalmus maximus after intravascular administration

Chang Zhiqiang,Liu Fei,Lian Chun’ang,Zhai Qianqian,Li Jian

The pharmacokinetic profiles and sulfamethoxazole (SMX) acetylation process in turbot reared at 18°C were investigated. Either SMX (parent drug) or its acetylized metabolite, N 4 -acetylsulfamethoxazole (AcSMX), was administered intravascularly to turbot at a dosage of 50 mg/kg BW. Serum concentrations of the parent drug and its metabolite were both measured by HPLC, and the changes in concentration over time were analyzed in two- and non-compartment models because SMX treatment produced multiple peaks. The results demonstrated that the elimination half-life of the parent drugs, SMX and AcSMX, were 159.2 and 5.9 h, respectively. The apparent volume of distribution was 0.2 and 0.8 L/kg, and the clearance was 0.038 and 0.222 L/(h·kg), for SMX and AcSMX, respectively. SMX acetylation in turbot was 2.8%, and the deacetylation of AcSMX was 0.2%. These findings may be useful in optimizing SMX dosage regimens in turbot aquaculture.

Dietary supplementation of propolis enhanced the innate immune response against Edwardsiella piscicida challenge in turbot (Scophthalmus maximus)

Di Mu,Yu Jiang,Jing He,Yuanxing Zhang,Dahai Yang,Qin Liu,Zhuang Wang

Propolis is non-hazardous resinous substance mixture containing bioactive ingredients such as polyphenols, flavonoids and organic acid. It has been widely used as food supplement and immune adjuvant due to its benefits in anti-microbial and immunomodulation . Edwardsiella piscicida is a kind of threatening pathogen which could cause high mortality in turbot . However, whether propolis could enhance the innate immune response against E. piscicida infection in turbot remains unknown. In this study, we found dietary propolis addition could improve the expression of anti-oxidative stress related enzymes, e.g., SOD , CAT and GPT , and relieved the histopathological changes of juvenile turbot after E. piscicida infection. Moreover, propolis addition increased the expression of cytokines such as il-1β , il-6 and tnf-α in different organs of juvenile turbot. Importantly, rescued survival and decreased bacterial loads were observed in propolis feeding group. Taken together, these findings suggest that the important roles of propolis in protecting juvenile turbot from E. piscicida infection, indicating propolis might be applied as a promising immunopotentiator candidate in aquaculture.

Feeding with poly(I:C) induced long-term immune responses against bacterial infection in turbot (Scophthalmus maximus)

Jing He,Zhuang Wang,Yanbo Zhao,Jin Yang,Yuanxing Zhang,Qin Liu,Dahai Yang

Poly(I:C) is a kind of chemosynthetic double-stranded RNA (dsRNA) analogue which could act as TLR3 agonist and induce IFN production. It is widely applied in anti-virus treatment and immunoregulation, as well as vaccine adjuvant in farmed animals. However, whether poly(I:C) could activate innate immune response to defense against bacterial infection remains unclear. In this study, we established a feeding trial model with different dose of poly(I:C) in turbot larvae, then challenged with Edwardsiella piscicida after 3–7 weeks resting period. The results show that feeding turbot with poly(I:C) exhibited a stronger inflammatory response and antioxidant stress ability, and significantly elevated the survival rate within the decreased bacterial loads. Importantly, the bacterial infection-induced white feces in hindgut of turbot were significantly alleviated after poly(I:C) feeding, and this administration induced protection could last for about 7 weeks. Taken together, these findings indicate that feeding turbot with poly(I:C) could enhance a long-term intestinal mucosal immunity in response to bacterial infection, suggesting that poly(I:C) might be a promising immunostimulant in aquaculture.

Effects of the Exopolysaccharide from Lactiplantibacillus plantarum HMX2 on the Growth Performance, Immune Response, and Intestinal Microbiota of Juvenile Turbot, Scophthalmus maximus

Gege Hu,Yihui Wang,Rui Xue,Tongji Liu,Zengjia Zhou,Zhennai Yang

In this study, the exopolysaccharide (EPS) fromLactiplantibacillus plantarum(HMX2) was isolated from Chinese Northeast Sauerkraut. Its effects on juvenile turbot were investigated by adding different concentrations of HMX2-EPS (C: 0 mg/kg, H1: 100 mg/kg, H2: 500 mg/kg) to the feed. Compared with the control group, HMX2-EPS significantly improved the growth performance of juvenile turbot. The activities of antioxidant enzymes, digestive enzymes, and immune-related enzymes were significantly increased. HMX2-EPS could also increase the secretion of inflammatory factors and enhance the immune response of turbot by regulating the IFN signal transduction pathway and exhibit stronger survival rates after theA. hydrophilachallenge. Moreover, HMX2-EPS could improve the diversity of intestinal microbiota in juvenile fish, increase the abundance of potential probiotics, and reduce the abundance of pathogenic bacteria. The function of gut microbes in metabolism and the immune system could also be improved. All results showed better effects with high concentrations of HMX2-EPS. These results indicated that HMX2-EPS supplementation in the diet could promote growth, improve antioxidant activity, digestive capacity, and immunity capacity, and actively regulate the intestinal microbiota of juvenile turbot. In conclusion, this study might provide basic technical and scientific support for the application ofL. plantarumin aquatic feed. Keywords:Lactiplantibacillus plantarum;exopolysaccharide (EPS);immunity;intestinal health;Scophthalmus maximus

Evaluation on Activity of Cytochrome P450 Enzymes in Turbot Via a Probe Drug Cocktail

Zhi-Qiang Chang,Jian Li,Qian-Qian Zhai

Cytochrome P450s (CYPs) are the main catalytic enzymes for metabolism by a variety of endogenous and exogenous substrates in mammals, fish, insects, etc. We evaluated the application of a multidrug cocktail on changes in CYP1, CYP2, and CYP3 activity in Turbot Scophthalmus maximus. The probe drugs were a combination of caffeine (5 mg/kg body weight), dapsone (5 mg/kg), and chlorzoxazone (10 mg/kg). After a single intraperitoneal injection of the cocktail, the concentration of all three probe drugs in the plasma increased quickly to a peak and then decreased gradually over 24 h. Pharmacokinetic profiles of the three probe drugs were determined using a noncompartmental analysis, and the typical parameters were calculated. In the assay for CYP induction, pretreatment with rifampicin significantly reduced the typical pharmacokinetic metrics for caffeine and chlorzoxazone, but not dapsone, indicating that the activity of CYP1 and CYP2 in turbot were induced by rifampicin.Received November 25, 2013; accepted May 29, 2014.

Effect of allicin and its mechanism of action in purine removal in turbot

Tingting Li,Likun Ren,Dangfeng Wang,Minjie Song,Qiuying Li,Jianrong Li

Low-purine food is not only the focus of gout patients, but also the focus of contemporary green diet development. Fish are usually considered as high-purine foodstuff because of the high nutritional value and high purine content. Therefore, it is necessary to reduce purine content in fish to ensure that they are suitable for patients with hyperuricemia or gout. In this study, the effect of allicin on purine reduction in turbot during cooking was investigated using high-performance liquid chromatography, and the change of xanthine oxidase (XO) activity was also studied. Molecular docking analysis was further performed to elucidate the mechanism of purine reduction by allicin. The results revealed that in the step of soaking, allicin could reduce purine content in fish by slightly enhancing XO activity, promoting hypoxanthine transformation into xanthine. The removal of total purines in experimental and control group reached 70.45% and 57.20%, respectively. Moreover, allicin could change the thermal stability of xanthine by providing an acidic environment, resulting in the rapid decrease of xanthine and hypoxanthine levels by boiling. Thus, this study provides a simple method to decrease purine levels, suggesting a possibility that allicin can function as a purine remover in food.

Quorum sensing system and influence on food spoilage in Pseudomonas fluorescens from turbot

Li Tingting,Yang Bing,Li Xuepeng,Li Jianrong,Zhao Guohua,Kan Jianquan

The spoilage of aquatic products is mainly caused by the bacterial growth, and the specific spoilage organism (SSO) plays an important role. Quorum sensing (QS) is a microbial cell–cell communication system which is coordinated with the population density, and is controlled by N -acyl-homoserine lactone (AHLs) as the Gram-negative bacteria communication signals. In this study, the SSO was Pseudomonas fluorescens (PF-04), isolated from the turbot ( Scophthalmus maximus L. ) during aerobically refrigerated storage. The supernatant extract of PF-04 tested the AHLs activities utilizing biosensor Chromobacterium violaceum CV026. AHL production was influenced by the environment temperature, and AHL production reduced obviously at 10 °C compare with 25 °C. In Luria-Bertani (LB) supplemented with 0.5–1.0% NaCl, AHL production reached the maximum. The AHL production was also regulated by pH of culture medium, acidic condition was conducive to persistent existence of the AHL molecules, but the alkaline environment would cause chemically unstable of AHL molecules. QS system in P. flurosecens played an imperative role in biofilm formation, protease and siderophore production. AHLs could regulate above three factors in PF-04. In summary, this study showed that (1) the influence of different environmental conditions (temperature, NaCl and pH) on AHL production revealed the correlation of QS in foods and (2) that proved the effect of external AHLs to regulate the biofilm formation, protease and siderophore production in PF-04.

Effects of Tetraselmis subcordiformis (Wille) Butcher with antimicrobial peptides as feed additives on gut microbial community and immunity of turbot (Scophthalmus maximus L.)

Zhu Bingkui,Sun Fengjie,Gao Zhengquan,Yao Ting,Han Subing,Zhang Hao,Ma Yuyang,Wu Yuyong,Du Ran,Meng Chunxiao,Cui Yulin

As harmless alternative of antibiotics, two antimicrobial peptides (AMPs), including anti-lipopolysaccharide factor isoform 3 (ALFPm3) and cathelicidin (CATHPb1), were co-expressed in the common fish food Tetraselmis subcordiformis. Lysates from T. subcordiformis transformant were able to inhibit the bacterial growth of Staphylococcus aureus, Vibrio parahaemolyticus, and Vibrio splendidus. To test the effects of AMPs in vivo, turbot ( Scophthalmus maximus ) underwent three dietary treatments, including commercial feeds (CON group), commercial feeds mixed with wild-type T. subcordiformis (WT group), and commercial feeds mixed with a mutant strain of T. subcordiformis (CSD group). Compared to CON group , both CSD and WT groups showed an increase in both the relative abundance and diversity of beneficial bacteria. The content of malondialdehyde was reduced and the enzymatic activities of lysozyme were increased in plasma. Comparisons of the biochemical factors in CSD group with those in CON group revealed that the relative abundance of Vibrio was decreased from 92.7% to 75.1%, and the number of bacteria containing the potentially pathogenic phenotypes was reduced. In the CSD group, the enzymatic activities of catalase and the expression levels of IgM, NKEF, IFNγ, MHC I, and IL-8R were higher compared to the WT group, whereas the levels of both TNFα and IL-1β were reduced. These results indicated that as an oral delivery vehicle for the AMPs, T. subcordiformis transformant improved the immunity, inflammatory response, and gut microbiome community of turbot.

Taurocholate supplementation attenuates the changes in growth performance, feed utilization, lipid digestion, liver abnormality and sterol metabolism in turbot (Scophthalmus maximus) fed high level of plant protein

Min Gu,Nan Bai,Trond M Kortner

Inclusion of plant ingredients in carnivorous fish feeds is often associated with decreased lipid digestibility, reduced bile acid levels and hypocholesterolaemia, which may result in lower growth performance and feed utilization. The present study aimed at evaluating the effects of dietary taurocholate supplementation in turbot ( Scophthalmus maximus ) fed high levels of plant protein and gaining further insight into how cholesterol and bile acid metabolism is modulated by taurocholate. A 56   day feeding trial was conducted using juvenile turbot (initial weight 8.5   g) fed one of three isonitrogenous and isolipid diets: a fish meal diet (FM), a plant protein meal based diet (a mixture of 40% soybean meal and 16% wheat gluten meal, named as PP) and a taurocholate supplemented diet (0.5% supplemented in PP, named as C-tau). Compared with fish fed the FM diet, the fish fed the PP diet had significantly lower growth performance, feed utilization, hepatosomatic index (HSI) and more atrophied hepatocytes. PP diet also induced enteritis in the distal intestine. Lipid digestion and metabolism was depressed as indicated by decreased whole body lipid content, lipid digestibility, pancreatic lipase activity and cholesterol and total bile acids (TBA) levels. Except the distal intestinal histology, all these tested parameters of fish fed the C-tau diet were significantly higher than those of fish fed the PP diet. Furthermore, the HSI, whole body lipid content, lipid digestibility and TBA levels of C-tau fed fish were similar to those of FM fed fish. Regardless of taurocholate, dietary plant protein could significantly increase the expression of HMG-CoA reductase ( hmgcr ) and decrease the hepatic cholesterol 7α-hydroxylase ( cyp7a1 ). Compared with the fish fed PP diet, taurocholate supplementation significantly increased the expression of cyp7a1 , but had no effect on hmgcr expression. In conclusion, dietary supplementation with taurocholate attenuated the plant protein meal induced negative effects on growth performance, feed utilization and lipid digestion and metabolism. In addition, the cholesterol and TBA-increasing effects of taurocholate could possibly be attributed to an improved digestion and absorption of cholesterol and TBA synthesis due to the up-regulation of cyp7a1 expression. Statement of relevant Dietary taurocholate is useful for fish meal sparing

Acute low-dose phosphate disrupts glycerophospholipid metabolism and induces stress in juvenile turbot (Scophthalmus maximus)

Ting Qi,Lele Wu,Jiachen Yu,Zongcheng Song,Feng Liu,Jun Li,Xiefa Song,Xian Li

Phosphate, as the main nutrient factor of lake eutrophication brought by pollutants discharged from agriculture and industry, is always considered to be a low-toxicity substance to aquatic animals. But the toxicity mechanism is unclear, and published information is limited. In this study, a 96 h acute stress experiment was conducted on juvenile turbot ( Scophthalmus maximus ) with 0, 10, and 60 mg/L phosphate solutions. Metabonomic analysis revealed that low-dose phosphate (10 mg/L) disrupted glycerophospholipid, purine, and glycolipid metabolism, as well as the tricarboxylic acid (TCA) cycle in juveniles, even at 96 h of stress, which may lead to cell structure damage and signal recognition disorder between cells. Upregulated key genes in the main glycerophospholipid metabolic pathways, which matched the results of the metabolomic study, were detected. Furthermore, low-dose phosphate (10 mg/L) induced oxidative stress and immunotoxicity in fish, resulting in the raising of relevant genes expression such as cat and sod in liver and kidney. In addition, all phosphate-treated groups had induced lesions on gill tissue, as evidenced by pathological observations. In this study on toxic effects on and mechanism of phosphate in aquatic animals using metabolomics, gene expression, and histopathology, we confirm that acute low-dose phosphate could disrupt glycerophospholipid metabolism and induce stress in juvenile turbot. This can provide advice on the amount of phosphate accumulation for marine fish farming and on protecting species diversity and marine ecosystem from the point of view of phosphate toxicity to marine animals.

Characterization of T-cell receptors and immunoglobulin heavy chains loci and identification of T/B cell clusters in teleost

Weijie Chen,Jing Hu,Jianchang Huang,Qin Liu,Qiyao Wang,Yuanxing Zhang,Dahai Yang

Bacterial disease is one of the important factors leading to economic losses in the turbot ( Scophthalmus maximus ) cultivation industry. T lymphocytes are major components of cellular immunity , whereas B lymphocytes produce immunoglobulins (Ig) that are key elements of humoral immune responses against infection. However, the genomic organization of genes encoding T-cell receptors (TCR) and immunoglobulin heavy chains (IgHs) in turbot remains largely unknown. In this study, abundant full-length transcripts of TCRs and IgHs were sequenced by Isoform-sequencing (Iso-seq), and we investigated and annotated the V, D, J and C gene loci of TCRα, TCRβ, IgT, IgM and IgD in turbot. Furthermore, through single-cell RNA sequencing (scRNA-seq) of blood leukocytes, we confirmed that these identified TCRs and IgHs were highly expressed in T/B cell clusters, respectively. Meanwhile, we also identified the IgM + IgD + B and IgT + B cells with differential gene expression profiles and potential functions. Taken together, our results provide a comprehensive understanding of TCRs and IgHs loci in turbot, which will contribute to evolutionary and functional characterization of T and B lymphocytes in teleost.
