Effects of β-cyclodextrin on the enzymatic hydrolysis of hemp seed oil by lipase Candida sp.99–125

Pei Wang,Zhaodi Ke,Juanjuan Yi,Xin Liu,Limin Hao,Qiaozhen Kang,Jike Lu

To enhance the stability of lipases, the additive method is attractive because of its high efficiency and simplicity, of which β-cyclodextrin (β-CD), as an additive has attracted attention. In the present study, the effects of β-CD on enzymatic hydrolysis of hemp seed oil by lipases were investigated. The stability, spectroscopy, and reusability of C andida sp.99–125 were compared by the addition of β-CD, respectively. The results showed that Candida sp.99–125 could produce the highest yield of α-linolenic acid (ALA, 18.27%, w/w) in the presence of β-CD after 24h, and the thermal stability of lipase was also improved. The UV spectroscopy showed that the absorbance of lipase decreased with increasing concentrations of β-CD and the fluorescence result was similar. Moreover, the reusability of lipase with β-CD was better than free lipase. This study revealed that β-CD could increase hydrolysis activity and stability of the lipase, thereby improving the hydrolytic process of hemp seed oil by Candida sp.99–125.

Formation, Stability and Properties of Hemp Seed Oil Emulsions for Application in the Cosmetics Industry

Liujun Pei,Yuni Luo,Xiaomin Gu,Jiping Wang

Hemp seed oil has good sun protection, repair, anti-allergy and anti-aging effects, and is a high quality raw material for the production of skincare products. However, the influence of surfactants and co-surfactants on the emulsification of hemp seed oil in aqueous solution has been rarely investigated. In this investigation, the surfactants and co-surfactants which were suitable for hemp seed oil emulsion were optimized, and the factors which influence the particle size and the stability of the emulsion were studied. Moreover, the application of hemp seed emulsion in different cosmetics was also further investigated. It was found that sorbitan monooleate/polysorbate-80 and glyceryl stearate were the most effective surfactant and co-surfactant in the emulsification of hemp seed oil. After three months the hemp seed oil emulsion had no layered separation and remained homogeneous at -48C, 258C and 408C. The water-resistant sunscreens and the hemp seed oil sunscreens were homogeneous pastes, had a soft texture and were stable at higher temperatures. No microorganisms were detected in these cosmetic creams. Therefore, hemp seed oil is suitable for sunscreen cosmetic materials.

Characterization of Key Odorants in Hemp Seed Oil Extracted from the Hemp Seeds Roasted Under Various Conditions

Yuchen Bai,Ningke You,Hongyu Tian,Xuebing Zhao

Hemp seed oil is nutritious. Besides cannabinoids, it has a higher Omega-3 fatty acid content, making it an ideal and healthy edible oil. Roasting is a key factor affecting the flavor of hemp seed oil. In this study, solvent-assisted flavor evaporation (SAFE) combined with gas chromatography–mass spectrometry (GC-O-MS) was used to analyze the volatile compounds of hemp seed oil after the hemp seeds were roasted at different temperatures (120 °C, 140 °C, 160 °C, 180 °C). α-pinene, (1S)-(1)-β-pinene, myrcene, and (E)-β-ocimene, which are typical odorants for herbs and fresh flavors, had an active odor value (OAV) ≥ 1. The sweet, nutty, and toasty flavors became more pronounced as the temperature increased. 2-pentylfuran provided the caramel flavor, and 2,5-dimethylpyrazine, 2-ethyl-6-methylpyrazine, and 2-ethyl-5-methylpyrazine provided the toasty flavor. The combination of sensory evaluation and data analysis was effective in differentiating the odors of hemp seed oil extracted at different temperatures. Reconstitution and deletion experiments showed that these odorants could mimic the overall aroma profile. This article provides a theoretical basis for the effect of over-roasting on the flavor of hemp seed oil.

Ultrasonic-ethanol pretreatment assisted aqueous enzymatic extraction of hemp seed oil with low Δ9-THC

Wang Zhang,Jiaye Yu,Donghua Wang,Xiaoyu Han,Tong Wang,Dianyu Yu

In this study, ultrasonic-ethanol pretreatment combined with AEE was developed for oil extraction from hemp seeds. The oil yield reached a maximum of 23.32 % at 200 W ultrasonic power and 30 min ultrasonic time, at this point, the degradation rate of Δ 9 -THC was 83.11 %. By determining the composition of hemp seed before and after pretreatment, it was shown that ultrasonic-ethanol pretreatment reduced the protein content of the raw material. An enzyme mixture consisting of pectinase and hemicellulase (1/1/1, w/w/w) was experimentally determined to be used, and the AEE extraction conditions were optimized using the Plackett-Burman design and the Box-Behnken. The optimal conditions were determined to be pH 5, total enzyme activity of 37,800 U/g, liquid–solid ratio of 10.4 mL/g, enzyme digestion temperature of 32 °C, enzymatic time of 189 min, and oil recovery of 88.38 %. The results of confocal laser scanning microscopy (CLSM) and scanning electron microscopy (SEM) showed that the emulsion formed during ultrasonic ethanol pretreatment was not uniformly distributed, and the droplets appeared to be aggregated; and the irregular pores of hemp seed increased after pretreatment. The contents of Δ 9 -THC and CBN in the extracted oil samples were 9.58 mg/kg and 52.45 mg/kg, respectively. Compared with the oil extracted by Soxhlet extraction (SE), the oil extracted by this experimental method was of better quality and similar in fatty acid composition.

CLG from Hemp Seed Inhibits LPS-Stimulated Neuroinflammation in BV2 Microglia by Regulating NF-κB and Nrf-2 Pathways

Shanshan Wang,Qian Luo,Yuefang Zhou,Peihong Fan

The healthy benefits of hemp (Cannabis sativa L.) seed have often been attributed to its oils and proteins. Recent studies reveal that hemp seed phenylpropionamides could also show various bioactivities. Continuation of our study on hemp seed provided a phenylpropionamide, coumaroylaminobutanol glucopyranoside (CLG). This work investigated the neuroprotective effect of CLG and its underlying mechanism using lipopolysaccharide-induced BV2 microglia. Our study demonstrated that CLG increased adenosine monophosphate-activated protein kinase (AMPK) expression, suppressed the nuclear factor-kappa B (NF-κB) signaling pathway by inhibiting the phosphorylation of IκBα and NF-κB p65 and decreased proinflammatory cytokine levels in a concentration-dependent manner. Furthermore, CLG reduced the production of cellular reactive oxygen species and stimulated the nuclear factor erythroid 2-related factor 2 (Nrf-2) signaling pathway. Collectively, these results suggested that CLG effectively and simultaneously inhibited inflammatory responses and oxidative stress through the NF-κB and Nrf-2 signaling pathways. AMPK was also involved in the anti-inflammatory effect of CLG. This study provides new insights into the diverse bioactive constituents of hemp seed.

Cannabis Seed Oil Alleviates Experimental Atherosclerosis by Ameliorating Vascular Inflammation in Apolipoprotein-E-Deficient Mice

Wenjing Huang,Zhujun Zeng,Yan Lang,Xia Xiang,Gaofu Qi,Gan Lu,Xiangliang Yang

In recent decades, epidemiological, clinical, and experimental studies have demonstrated that a diet with antioxidant or anti-inflammatory function plays a central role in the prevention of atherosclerosis (AS). The purpose of this study was to explore the effects of Cannabis seed oil (CO) administration on in vitro antioxidant capacity as well as blood lipid profiles, lipid peroxidation, inflammatory response, and endothelial cell integrity. Female ApoE–/– mice were fed a high-cholesterol diet and administrated with CO or phosphate-buffered saline (PBS) and seal oil by gavage for 8 weeks. The results show that CO administration reduced the levels of serum triglycerides and low-density lipoprotein cholesterol at week 6. Additionally, a decrease in serum tumor necrosis factor α and nitric oxide was also observed. Moreover, results from CD31 staining and scanning electron microscopy revealed that CO treatment alleviated the endothelial cell damage and lipid deposition induced by a high-cholesterol diet. The ratio of lesion area to the total aorta area was 19.57% for the CO group, which was lower than the PBS control group (24.67%). Collectively, CO exerted anti-atherosclerotic effects by modulating serum lipid profiles and inflammatory responses and improving endothelial cell integrity and arterial lipid deposition. The results provide a promising preventive strategy for the early progression of AS.

Anti-Thrombotic Effect of Hemp Seed Peptides in Zebrafish by Dual Regulation of vegfr1 Signaling and Caspase-3-Mediated Apoptosis

Jie Shi,Jingyi Ge,Zhenghai Zhang,Lianhui Wei,Yan Dong,Guowei Li,Qingli Yang,Jing Pan,Yanru Ji

This study aimed to investigate the antithrombotic activity of hemp seed peptides derived from trypsin hydrolysates and elucidate their molecular mechanisms using a ponatinib-induced zebrafish ( Danio rerio ) thrombosis model. Through ultrafiltration and LC–MS/MS analysis, we identified 1741 peptide sequences (< 3 kDa), with 67% distributed in the 0.5–1.5 kDa range. In vivo experiments demonstrated that 125 μg/mL hemp seed peptides significantly ameliorated ponatinib-induced thrombosis by increasing 16.55% cardiac erythrocyte staining intensity, enhancing 54.8% blood flow velocity, and restoring 11.2% intersegmental blood vessel diameters. In addition, the observed anticoagulant effect may be partially attributed to upregulated expression levels of vascular endothelial growth factor receptor 1 ( vegfr1 ) and down-regulated expression levels of cysteinyl aspartate-specific proteinase-3 ( caspase-3 ) at the mRNA level. These findings propose hemp seed peptides as novel dietary antithrombotic agents with potential applications in functional food development.

A study on pH-mediated variations and differences in the structure and function of hemp seed albumin, globulin, and protein isolate

Peng-Wei Xu,Xiao-Fan Yuan,Bing Zhao

In this comprehensive study, hemp seed protein isolate (HPI), albumin (ALB), and globulin (GLB) were prepared from hemp seed kernels. ALB was identified as a potential glycoprotein , distinguishing itself with a higher count of free sulfhydryl groups, fewer disulfide bonds , and a smaller molecular weight distribution. Conversely, GLB demonstrated fewer free sulfhydryl groups, a larger number of disulfide bonds, and a larger molecular weight distribution. HPI predominantly consisted of globulin with minor albumin. Varying pH from 3.0 to 9.0 considerably changed the structure and conformation of GLB and HPI but had minimal impact on ALB, according to surface hydrophobic, circular dichroism , and fluorescence spectrum assays. The pH-induced structural difference and variations in ζ-potential and solubility had synergistic effects on the emulsifying properties of GLB and HPI, while ALB's emulsifying properties were primarily associated with changes in ζ-potential and solubility. GLB and HPI demonstrated superior solubility and emulsification ability at pH 3. In contrast, ALB exhibited better functionality at pH 9. Understanding the pH-dependent properties of hemp seed proteins is crucial for optimizing their application in diverse fields.

Polysaccharides from Hemp Seed Protect against Cyclophosphamide-Induced Intestinal Oxidative Damage via Nrf2-Keap1 Signaling Pathway in Mice

Ran Xue,Ming Du,Tian-Yi Zhou,Wan-Zheng Ai,Zhong-Shan Zhang,Xing-Wei Xiang,Yu-Fang Zhou,Zheng-Shun Wen

Hemp seed has been used as a traditional oriental medicine and health food in China for centuries. Polysaccharides from hemp seed (HSP) exhibit important properties of intestinal protection, but there are limited data on the specific underlying mechanism. The primary objective of this study was to investigate the protective effect of HSP on intestinal oxidative damage induced by cyclophosphamide (Cy) in mice. The results showed that pretreatment with HSP significantly increased the average daily gain, thymus index, spleen index, superoxide dismutase (SOD), catalase (CAT), and glutathione peroxidase (GSH-Px) activity in serum and ileal homogenate and significantly reduced malondialdehyde (MDA) content in ileal homogenate. In addition, the expression levels of SOD, GSH-Px, Nrf2, heme oxidase-1 (HO-1), and quinoneoxidoreductase-1 (NQO1) mRNA in ileal homogenate were significantly increased. Western blot results showed that HSP significantly upregulated the expression of Nrf2 protein and downregulated the expression of Keap1 protein in the ileum. Collectively, our findings indicated that HSP had protective effects on intestinal oxidative damage induced by Cy in mice, and its mechanism might be related to the activation of Nrf2-Keap1 signaling pathway.
