Three Tomicus bark beetles ( T. yunnanensis , T. brevipilosus and T. minor ) coexist in the trunks of Pinus yunnanensis but possess relatively independent spatio-temporal ecological niches. This makes them become a good case for addressing functional differentiation of chemosensory-related proteins among closely related species in odorant recognition and insecticide sequestering. In this study, we used odorant binding protein (OBP) orthologues highly expressed in antennae to illustrate this question. We first identified 33, 35 and 34 OBPs from T. yunnanensis , T. brevipilosus and T. minor , respectively, with 33 groups of OBP orthologues. Sequence and phylogenetic analyses revealed the conservation and differences of OBP orthologues across three Tomicus beetles. Expression profiles revealed that the majority of Tomicus OBPs were abundantly expressed in antennae where 11 groups of OBP orthologues shared a significantly antenna-enriched distribution. Four groups of antennal OBP orthologues (OBP2, OBP9, OBP10 and OBP16 totaling 12 genes) presented diverse ligand-binding properties, responding differently to five classes of ecologically relevant odorants as well as various insecticides. In particular, OBP orthologues were tuned differentially to host and non-host odorants, and chlorpyrifos was identified as the best ligand for 12 Tomicus OBPs. Our study thus sheds light on functional conservation and divergence of OBP orthologues among three sympatric Tomicus bark beetles, possibly as an implication for the early or late of host colonization, ecological niche differences and the strong habitat adaptation.

Understanding the mechanism of insect resistance to toxic effects of phytochemicals can provide an insight into plant–herbivore interactions. Monochamus alternatus Hope, a main vector of pine wood nematode ( Bursaphelenchus xylophilus ), prefers to infest masson pine ( Pinus massoniana Lamb) and causes huge economic and environmental losses. α-Pinene is the primary component of resin produced by masson pine and plays a crucial role in defending against herbivorous insects, but its defensive effects on M . alternatus larvae are barely known. Here, we explored the physiological metabolism in M . alternatus larvae that were fumigated with (+)-α-pinene, (-)-α-pinene, β-pinene, (R)-(+)-limonene, myrcene and (-)-α-phellandrene. (+)-α-Pinene showed the highest fumigant toxicity to M . alternatus larvae among these monoterpenes, and the fourth instar larvae possessed the highest resistance to this compound at the larval stage. Comparative transcriptome analysis detected 423 up-regulated and 311 down-regulated genes in the larvae fumigated with (+)-α-pinene, which were mainly related to detoxification, energy and protein metabolism. Among the differentially expressed genes encoding for detoxification enzymes, 14 P450s, 13 UGTs, 1GSTs and 3ABCs were dramatically up-regulated in the larvae fumigated with (+)-α-pinene. Silencing of P450s via RNA interference led to an increased mortality in the larval fumigated with (+)-α-pinene. Piperonyl butoxide exposure significantly increased the mortality and inhibited the expressions of P450s, UGTs and ABCs in the larvae fumigated with (+)-α-pinene. These results suggested an important role of P450s in the resistance of M . alternatus larvae to (+)-α-pinene fumigation, which may facilitate the understanding of terpenoid detoxification in M . alternatus larvae.

The red-necked longhorn beetle, Aromia bungii , is an invasive wood-boring insect that has recently invaded Europe and Japan, seriously damaging stone fruit trees in both native and invaded regions. Rose oxide, including both cis - and trans -isomers, was previously found in the volatiles produced by A. bungii adults, while the inter- and intra-specific behavioral functions of this compound remain unclear. By manually shaking the beetles, a much greater amount of rose oxide was released from the beetles, compared to that released from nondisturbed beetles. Rose oxide was further determined to be original from a white-colored secretion which was immediately sprayed by the beetles that were hand captured or squeezed by tweezers. These findings indicate that this pungent compound putatively has a role as an allomone component released from the defensive secretion by A. bungii adults when threatened, which may be an adaptive defensive strategy against the predation of natural enemies such as insectivorous birds. Y-tube olfactory assays revealed that rose oxide, at a high concentration, was significantly repellent to both sexes of A. bungii adults, suggesting that rose oxide may also have a function in the intraspecific communication of A. bungii . Considering the low price of the commercially available products, this repellent effect of rose oxide may facilitate the development of novel control strategies, such as push–pull strategy, for A. bungii .

Monochamus alternatus is an important stem-borer that endangers economic and ecological forest, and also the main vector of pine wilt disease, a significant forest quarantine disease, which caused seriously losses for economic and ecological services. At present, M. alternatus mainly focuses on chemical control, among which Avermectin (AVM) is widely used as a high-performance spectrum insecticide. However, there are some problems, such as short duration, and environmental pressure caused by excessive use, which limit its application in pest control. In this paper, a drug delivery system based on dendritic mesoporous silica (DMSNs) was constructed to deliver biological pesticide AVM, so as to develop a new type of nano-pesticide, which could effectively control the stem borer M. alternatus and control Bursaphelenchus xylophilus . The [email protected] NPs composite was prepared by simple physical adsorption, and the loading rate was 85.73%, which significantly improved the utilization rate of AVM. The local application of AVM optimized by DMSNs showed that the contact toxicity (LC 50 = 14.27 μg/mL) and stomach toxicity (LC 50 = 10.90 μg/mL) of M. alternatus larvae were higher than those of AVM/EC and AVM treatment, which was based on the good permeability and adhesion of DMSNs. At the same time, DMSNs had better degradation ability in the natural environment: in PBS buffer solutions with different pH values, DMSNs completely degraded in 28 days. In addition, [email protected] NPs has a good absorption and transmission effect in trees, which indicates that nanoparticle-mediated pesticide delivery system will be a significant progress in the control of forest and fruit pests.

Pine wilt disease is a major forestry quarantine disease caused by pinewood nematode and spread by the vector insect Monochamus alternatus (MA), which has brought destructive harm to pine trees. Avermectin (AVM) is a broad-spectrum fungicide, insecticide, and nematicide. However, AVM easily degrades under ultraviolet light and is almost insoluble in water, which seriously affect its control effect on the pine wilt disease. In this study, an original nanopesticide was designed to improve the bioactivity and utilization of AVMactive ingredients, with antiphotolysis and slow release properties especially. Bovine serum albumin (BSA) was dissolved in water, taking advantage of the oleophobic properties of the protein to wrap AVM in an oil phase by self-assembly. The obtained AVM BSA nanoparticles (AVM@BSA·NPs) had preferable sustained-release performance, the cumulative release reached 90.2%, the nanoparticles could be released continuously for about 20 days, the particle size of the AVM@BSA·NPs was about 65 nm, and the loading efficiency reached 78%. The nanodelivery system could effectively protect AVM from photolysis, and the retention of AVM@BSA·NPs was close to 80 and 75% after 48 and 72 h under ultraviolet light, respectively. In addition, the nanopesticides were prepared by labeling AVM@BSA·NPs with fluorescein isothiocyanate (FITC) to research the mechanism of contact and stomach toxicity of AVM using the probe function of FITC. Compared with AVM original, the stomach toxicity and contact toxicity of AVM@BSA·NPs increased by 35.3 and 19.6%, respectively. There are broad application prospects of AVM@BSA·NPs for the prevention and control of forest diseases and other pests.

The pine-forest guardian Dastarcus helophoroides mainly rely on olfaction to locate its host accurately and interact socially. Odorant binding proteins of D. helophoroides play an important role in olfactory recognition and transporting odors to olfactory receptors to trigger signal transduction. However, some compounds affect olfactory recognition of D. helophoroides by binding to odorant binding proteins. In this study, DhelOBP4 and DhelOBP21, which are involved in the recognition of plant volatiles, were expressed to explore compounds that affect olfactory recognition. The affinity of 24 compounds with DhelOBP4 and DhelOBP21 was investigated by vitro fluorescence competitive binding assays. It was found that cyetpyrafen, chlorfenapyr and spirodiclofen had high affinity with DhelOBP4 and DhelOBP21. At the same time, we found that these three pesticides had a good evasive effect on the D. helophoroides and elicited EAG response in a dose-dependent manner. Besides, it was found that silencing DhelOBP4 and DhelOBP21 would affect D. helophoroides identifying these three pesticides. Through homology modeling and molecular docking, we identified key amino acid sites involved in the binding of DhelOBP4 and DhelOBP21 to these three pesticides, which might revealed their specific binding molecular interactions. This study is helpful to provide a valuable addition of OBP-pesticide interactions of D. helophoroides .
