Apple shrinkage correlates positively with fructose, glucose, and succinic acid and not with citric acid

Yuanqing Li,Zhuan Peng,Lingling Liu,Junbo Wang,Yi Zhu,Yizhou Gao,Branimir Pavlić,Jian Lou,Zhaojun Ban,Yunbin Jiang

Both pericarp shrinkage and the sugar-acid ratio are vital factors influencing consumer preference in the apple industry. Our study examines the relationship between extrinsic appearance and intrinsic properties by investigating the effects of sugar-acid content on shrinkage, apple fruit development and shrinkage presence were investigated while the fruit was stored at 5 °C with 60 % relative humidity for up to 70 days, which categorized by packaging in microporous plastic bags. Pericarp shrinkage was assessed by exploring fruit sensory preferences, variations in structural differences, and physiological compositional status, and correlations were analyzed. Shrinkage significantly reduced water loss rate (4.44 %), accelerated water core dissipation and microcrack growth, and correlated with total sugar (r = 0.59) and total acid (r = 0.75). Summarily, pericarp shrinkage correlated positively with fructose (r = 0.37), glucose (r = 0.56), and succinic acid (r = 0.55) accumulation, but negatively with citric acid (r = -30) accumulation. The present study serves as a novel exploration of how pericarp shrinkage and sugar-acid biosynthesis and distribution in apples, losses due to cosmetic defects can be reduced for further research.

Therapeutic potential of Rosa rugosa polysaccharide and its nanofiber membrane in psoriasis via PI3K-AKT/mTOR pathway inhibition

Mengxiang Chen,Yi Peng,Rongtao Zhu,Xiaoting Luo,Xuemin Yang,Jiazhi Chen,Haiyan Chen,Wei Zhou,Zhiyun Du

Psoriasis is a chronic inflammatory skin disease with significant impacts on quality of life. Current treatments face challenges like high costs and side effects. This study explores the therapeutic potential of Rosa rugosa polysaccharide (RRPS) and its nanofiber membrane in psoriasis treatment. RRPS was extracted using a yeast fermentation enzyme-assisted method and characterized. In vitro studies on RAW264.7 macrophages showed that RRPS significantly reduced nitric oxide (NO) production, indicating anti-inflammatory effects. The nanofiber membrane, fabricated via electrospinning, has high porosity and sustained-release properties, enhancing drug delivery and skin barrier repair. In vivo studies using an imiquimod-induced psoriasis mouse model demonstrated that both RRPS and its nanofiber membrane improved psoriasis symptoms, reduced transepidermal water loss (TEWL), and inhibited pro-inflammatory cytokines (e.g., TNF-α, IL-6, IL-17A). Mechanistically, they suppressed the PI3K-AKT/mTOR pathway. The nanofiber membrane showed superior efficacy compared to RRPS alone, highlighting the benefits of electrospinning. This study suggests that Rosa rugosa polysaccharide and its nanofiber membrane could be a novel, effective, and safe alternative for psoriasis treatment by targeting the PI3K-AKT/mTOR pathway.

Novel skin permeation enhancers based on amino acid ester ionic liquid: Design and permeation mechanism

Luyao Zheng,Zhiyuan Zhao,Ye Yang,Yaming Li,Chengxiao Wang

This study developed novel ionic liquids (ILs) based on amino acids. We first screened 15 methyl amino acid ester hydrochlorides ([AAC1]Cl) for their skin permeation enhancements using 5-Fluorouracil (5-Fu) and Hydrocortisone (HC) as model drugs. Glycine methyl ester hydrochloride ([GlyC1]Cl), L-proline methyl ester hydrochloride ([L-ProC1]Cl), and L-leucine methyl ester hydrochloride ([L-LeuC1]Cl) were selected, and their ester sites were modified with different carbon chains (C8 and C12). The resulting ILs showed improved permeation to the two drugs. TEWL and CLSM assays revealed the moderation effects of the modified ILs on skin barrier function, whereas L-proline dodecyl ester hydrochloride ([ProC12]Cl) and L-leucine dodecyl ester hydrochloride ([L-LeuC12]Cl) exhibited the strongest activities. Permeation mechanisms were further investigated by ATR-FTIR, solid-NMR, SEM, and TEM analyses. The results suggested that [L-ProC12]Cl and [L-LeuC12]Cl combined the advantages of amino acid esters and IL solvent and could interact with the intercellular lipid domain by the multi-functions of lipid fluidization and lipid extraction, which were observed in a dosage- and time-dependent manner. Additionally, pathological examination suggested that the amino acid ester-based ILs (AAE-ILs) had good biocompatibility . In conclusion, this study has generated novel chemical penetration enhancers (CPEs) based on AAE-ILs and may be potentially utilized in drug transdermal delivery systems (TDDSs).
