A novel chiral sensing platform, employing a biomolecule-based nanocomposite prepared by calf thymus double stranded DNA, methylene blue and multiwall carbon nanotubes (DNA–MB–MWNTs), was utilized for the discrimination of quinine (QN) and quinidine (QD). The DNA-based nanocomposite, which could be used as an electrochemical sensing unit and chiral probe, was characterized by transmission electron microscopy (TEM), ultraviolet-visible spectroscopy (UV-Vis) and cyclic voltammetry (CV). After the proposed sensing platform interacted with QN and QD, a larger electrochemical signal was obtained from QD. The comparative experiments indicated that the proposed strategy not only simplified the fabrication processes but also enhanced the enantioselective interaction in chiral analysis. In addition, experimental factors such as acidity, interaction time and the concentration of enantiomers were investigated in regards to the effect on enantioselective interaction.

An O -9-( tert -butylcarbamoyl) quinidine (t-BuCQD) functionalized polymeric monolithic capillary column was prepared by the in situ copolymerization method. The physicochemical properties of the optimized monolithic column were characterized by scanning electron microscopy and micro-LC. Satisfactory column permeability, efficiency, stability and reproducibility were obtained for this monolithic column. The chiral recognition ability of the resulting monolith was also evaluated using 47 N -derivatized amino acids, eight N -derivatized dipeptides, and two herbicides. Under the selected conditions, the enantiomers of all chiral analytes were baseline separated with exceptionally high selectivity and resolution using micro-LC. It is worth noting that this chiral stationary phase (CSP) containing quinidine with a tert -butyl carbamate residue as chiral selector exhibits much higher enantioselectivity and diastereoselectivity than the previously developed O -9-[2-(methacryloyloxy)-ethylcarbamoyl]-10,11-dihydroquinidine (MQD) based CSP for N -derivatized amino acids and dipeptides. These results indicate that this novel quinidine-based polymeric monolith can be used as an effective tool for the enantioseparation of chiral acidic compounds.

The use of agrochemicals as pure enantiomers avoids the presence of non-active pesticide compounds in the environment. The quality control of these agrochemicals requires the development of new chiral methodologies. In this work, the performance of the monolithic column named poly(O-9-[2-(methacryloyloxy)-ethylcarbamoyl]-10,11-dihydroquinidine-co-2-hydroxyethyl methacrylate-co-ethylene dimethacrylate) (poly(MQD- co -HEMA- co -EDMA)) was assessed as stationary phase for the enantiomeric separation of seven phenoxy acid herbicides (dichlorprop, 2-(4-chlorophenoxy)propionic acid, 2-(3-chlorophenoxy)propionic acid, mecoprop , fenoprop, 2-phenoxypropionic acid, and fenoxaprop) in a nano-liquid chromatography (nano-LC) system. Under optimized conditions (60/40 ( v/v ) ACN/0.1 M ammonium acetate (pH 6.0) at a flow rate of 20 μL/min), it was possible to obtain baseline separation for most of the studied compounds. The analysis of an enantiomerically pure mecoprop commercial herbicide sample which also contained three achiral phenoxy acid herbicides (MCPA, 2,4-D and dicamba) commonly present in these kinds of formulations showed that the use of a 80/20 ( v/v ) ACN/0.1 M ammonium acetate (pH 5.3) enabled the simultaneous separation of these compounds and mecoprop enantiomers in less than 8 min. The analytical characteristics of the methodology were evaluated in terms of selectivity, linearity, precision, accuracy, LOD, and LOQ. Results demonstrated that determined amounts of R-mecoprop in commercial formulations were in agreement with the labeled content and the enantiomeric impurity was below the LOQ. The poly(MQD- co -HEMA- co -EDMA) capillary monolithic column exhibits great potential as a useful and environmental-friendly tool for the quality control of agrochemicals by nano-LC.

Dehydrocorydaline (DHC), one of the main active components of Corydalis yanhusuo, is an important remedy for the treatment of coronary heart disease. Our previous study revealed a higher unbound concentration of DHC in the heart than plasma of mice after oral administration of C. yanhusuo extract or DHC, but the underlying uptake mechanism remains unelucidated. In our investigations, we studied the transport mechanism of DHC in transgenic cells, primary neonatal rat cardiomyocytes, and animal experiments. Using quantitative real-time polymerase chain reaction and Western blotting, we found that uptake transporters expressed in the mouse heart include organic cation transporter 1/3 (OCT1/3) and carnitine/organic cation transporter 1/2 (OCTN1/2). The accumulation experiments in transfected cells showed that DHC was a substrate of OCT1 and OCT3, with Km of 11.29 ± 3.3 and 8.96 ± 3.7 μM, respectively, but not a substrate of OCTN1/2. Additionally, a higher efflux level (1.71-fold of MDCK-mock) of DHC was observed in MDCK-MDR1 cells than in MDCK-mock cells. Therefore, DHC is a weak substrate for MDR1. Studies using primary neonatal rat cardiomyocytes showed that OCT1/3 inhibitors (quinidine, decynium-22, and levo-tetrahydropalmatine) prevented the accumulation of DHC, whereas OCTN2 inhibitors (mildronate and l-carnitine) did not affect its accumulation. Moreover, the coadministration of OCT1/3 inhibitors (levo-tetrahydropalmatine, THP) decreased the concentration of DHC in the mouse heart. Based on these findings, DHC may be accumulated partly by OCT1/3 transporters and excreted by MDR1 in the heart. THP could alter the distribution of DHC in the mouse heart.
