Cinnamaldehyde Acts as a Fungistat by Disrupting the Integrity of Fusarium oxysporum Fox-1 Cell Membranes

Li-Rong Zhou,Hai-Jun Hu,Jie Wang,Yong-Xing Zhu,Xue-Dong Zhu,Jia-Wei Ma,Yi-Qing Liu

To counter the harmful impacts of agricultural chemicals on the environment and human health, there is an increasing demand for safe, eco-friendly, and potent plant-based biopesticides. In this study, we aimed to investigate the antimicrobial effects of ginger essential oil and selected volatile compounds (linalool, eugenol, citral, and cinnamaldehyde [CA]) againstFusarium oxysporumFOX-1. Minimum inhibitory concentrations (MICs) were determined using the mycelium growth inhibition method. The compound CA exhibited the most potent antifungal effect againstF. oxysporumFOX-1 and was selected for further investigation. After treatment with CA at 1/2 MIC or MIC, the spore germination ofF. oxysporumFOX-1 was significantly inhibited at 12 h. Furthermore, microscopic observation revealed that CA treatment resulted in the morphological degradation ofF. oxysporumFOX-1. CA destroyed the cell membrane integrity ofF. oxysporumFOX-1, increasing the relative conductivity and the leakage of intracellular protein, nucleic acids, and malondialdehyde, affecting the integrity and metabolism of the cell membrane. The effects were positively related to CA concentration. Additionally, in vivo experiments with rhizome sections showed that CA significantly reduced the pathogenicity ofF. oxysporumFOX-1. Overall, these findings provide evidence for the potential of using ginger essential oil components as fungicides, offering a basis for future research to develop robust and eco-friendly plant-derived fungicides that serve as a sustainable means to reduce fungus-driven agricultural losses.

Essential oils of Zingiber officinale: Chemical composition, in vivo alleviation effects on TPA induced ear swelling in mice and in vitro bioactivities

Zhang Silu,Zhang Lijun,Yu Mengjie,Luo Donghui,Chen Shuai,Liu Weifeng,Zhang Yehui,Zhang Lanyue,Zhao Tiantian

Zingiber officinale (ZO) is a traditional food condiment. The essential oils of Z. officinale (ZOEOs) are known to have multiple bioactivities. In this study, gas chromatography mass spectrometer (GC-MS) analytical method was used to identify active ingredient present in ZOEOs. A total of 41 compounds were identified in ZOEOs. Major components in ZOEOs were zingiberene (19.71%), (+)-β-cedrene (12.85%), farnesene (12.17%), α-curcumene (10.18%) and β-elemene (3.54%). Experimental results of 12-O-tetradecanoylphorbol-13 acetate (TPA) induced ear swelling validation mice model showed that ZOEOs treatment has better anti-inflammatory effect compared with ibuprofen (positive control) at high concentrations. Histological and immunohistochemical analysis showed that ZOEOs significantly decreased COX-2, IL-6 and NF-κB expression in a dose dependent manner. The mRNA levels of COX-2 and NF-κB were also down regulated by the application of ZOEOs. This indicated that ZOEOs exhibited positive effects in ear skin protection. Antibacterial experimental results showed that EOZOs had anti-bacterial effects on Escherichia coli, Pseudomonas aeruginosa, and Staphylococcus aureus. DPPH radical scavenging, A549 cell line and LNCaP cell line inhibition results indicated that ZOEOs exhibited potential antioxidant and anti-tumor properties. The findings of these study provide scientific basis on therapeutic use of ZO in food, cosmetic and pharmaceutical industries.

Preparation of active films with antioxidant and antimicrobial properties by combining ginger essential oil nanoemulsion with xylan and polyvinyl alcohol

Junhan Liu,Marie-Laure Fauconnier,Aurore Richel,Yuhong Jin

Due to the environmental challenges of petroleum-based packaging, new biodegradable and active food packaging has garnered significant attention. In this work, active films were generated with xylan/polyvinyl alcohol (PVA) as the film-forming matrix, combined with ginger essential oil nanoemulsions (GEO-NEs) at varying concentrations (2.0 %, 4.0 %, 6.0 %, and 8.0 % w /w). The GEO-NEs, produced via ultrasound, had a mean particle size measuring 176.4 ± 1.2 nm and demonstrated excellent stability for up to 28 d. FTIR and XRD analyses revealed that interactions between GEO-NEs and the film matrix occurred through hydrogen bonding, indicating good compatibility between the components. Incorporating GEO-NEs significantly enhanced the UV shielding performance and mechanical characteristics of the composite films, achieving mechanical characteristics comparable to those of commercial packaging materials such as high-density polyethylene (HDPE). Additionally, composite films with 2 % and 4 % GEO-NEs exhibited lower water vapor permeability (WVP) than the control film, indicating improved water barrier performance. GEO-NEs also significantly improved the antioxidant activity of the composite films and imparted certain antimicrobial properties. As a result, these films hold promise for applications in active food packaging.

Characterization of active films based on chitosan/polyvinyl alcohol integrated with ginger essential oil-loaded bacterial cellulose and application in sea bass (Lateolabrax japonicas) packaging

Xuening Chen,Weiqing Lan,Jing Xie

The poor mechanical properties, low water-resistance, and limited antimicrobial activity of chitosan (CS)/polyvinyl alcohol (PVA) based film limited its application in aquatic product preservation. Herein, bacterial cellulose (BC) was used to load ginger essential oil (GEO). The effects of the addition of BC and different concentrations of GEO on the physicochemical and antimicrobial activities of films were systematically evaluated. Finally, the application of sea bass fillets was investigated. Fourier transform infrared spectroscopy (FTIR) and X-ray diffraction analysis (XRD) analysis indicated dense networks were formed, which was verified by enhanced physical properties. The mechanical properties, barrier properties, and antimicrobial activities enhanced as GEO concentration increased. CPB 0.8 (0.8 % GEO) film had better tensile strength (TS) and barrier performance, improved the quality, and extended the shelf-life of sea bass for another 6 days at least. Overall, active films are potential packaging materials for aquatic products.

Wei Zhou,Yunxia He,Fei Liu,Liangkun Liao,Xiaobing Huang,Ruyi Li,Ying Zou,Lei Zhou,Liqiang Zou,Yuhuan Liu,Roger Ruan,Jihua Li

This study aimed to develop an active edible film based on carboxymethyl chitosan (CMCS) and pullulan (Pul) incorporated with galangal essential oil (GEO) by the casting method. And their physical properties, structural and preservation effect on mangoes were characterized. The CMCS/Pul ratio was determined to be 2.5:2.5 after the optimization of physical properties, mechanical properties and barrier properties of the blend film. The results of FT-IR and XRD showed that hydroxyl groups of Pul interacted with the carboxyl groups of CMCS and the blend films had good compatibility. Good thermal stability of CMCS/Pul-GEO films was further proven by TGA curves. The CMCS/Pul-8 %GEO film showed effective preservations on mango fruits during 15 days of storage at 25 ± 1 °C, based on the characterization by fruits weight loss, firmness, titratable acidity, soluble solids. Consequently, CMCS/Pul-GEO blend films may be a promising eco-friendly packaging material for the industrial application of fruit preservation.

Effects of active film based on chitosan/polyvinyl alcohol on the quality of refrigerated sea bass (Lateolabrax Japonicus) fillets

Xuening Chen,Weiqing Lan,Zhenfei Xu,Huanyu Li,Jing Xie

There has been a growing interest in employing active films for fish preservation due to the environmental concern of plastic packaging and the high perishability of fish. This work aimed to develop active films (CPB 0.8 ) based on chitosan/polyvinyl alcohol integrated with ginger essential oil (GEO) -loaded bacterial cellulose, and to evaluate the preservation effect of the films on refrigerated sea bass fillets. Different active films were obtained by the solution blending method. It was evaluated by measuring microbiological indicators, physicochemical indexes, and water loss of sea bass fillets. The sample wrapped by CPB 0.8 film had a lower microbial load after 12 days of storage. The results showed that the sample wrapped by CPB 0.8 film had a lower microbial load after 12 days of storage compared to the active film sample without GEO and the control sample. Besides, the reduction of hardness and springiness of fillets packaged with CPB 0.8 film was markedly inhibited. Additionally, the fillets wrapped by CPB 0.8 film indicated a lower K-value (56.54%) under the rejection limit (60%) on the 10th day, which confirmed that CPB 0.8 films extended the shelf-life of fillets to 10 days. Also, the active packaging could preserve fish quality by controlling water migration and lipid oxidation. Overall, these findings indicate that CPB 0.8 films can be useful for the preservation of fish and effectively reduce petroleum-based plastics.

Chitosan/polyvinyl alcohol-based active film maintain the quality of farmed sea bass (Lateolabrax maculatus) during refrigerated storage: Inhibit lipid oxidation and maintain myofibrillar proteins conformation

Weiqing Lan,Shuting Liu,Xuening Chen,Jing Xie

In this study, the effects of chitosan (CS)/polyvinyl alcohol (PVA)-based active film on lipid oxidation and myofibrillar proteins (MPs) oxidation and conformational changes in refrigerated (4 °C) sea bass ( Lateolabrax maculatus ) fillets were analyzed. The CS/PVA matrix combined with bacterial cellulose (BC) and 0.8 % (v/v) ginger essential oil (GEO), made the mixture form the film solution (CPB 0.8 ) by desiccation. Sea bass fillets coated with CS/PVA-based films were evaluated for lipid and protein oxidation at 3-day intervals. Ca 2+ -ATPase, total sulfhydryl, myofibril fragmentation index (MFI), and the secondary structure content were investigated to assess the oxidation degree and conformational changes in MPs. Malondialdehyde (MDA) and hydrogen peroxide (H 2 O 2 ) content were determined to evaluate the oxidation degree of sea bass fillets. Compared with other films, the results demonstrated that CPB 0.8 film could inhibit the oxidation and degradation of MPs after 18 days, which has the lowest MDA (86.44 nmol/g), Ca 2+ -ATPase (0.1567 U/mg), total sulfhydryl (53.7667 μmol/g), MFI (57.2). In addition, the conformational analysis of MPs showed that CPB 0.8 film could better maintain the secondary and tertiary structure of MPs, which effectively delay the destruction of protein conformation and maintain the microstructure of muscle fibers in fish. Therefore, CPB 0.8 film will be a hopeful strategy of keeping the quality of sea bass and have a long-term preservation effect.

Characterisation of microemulsion nanofilms based on Tilapia fish skin gelatine and ZnO nanoparticles incorporated with ginger essential oil: meat packaging application

Le Zhang,Anjun Liu,Wenhang Wang,Ran Ye,Yaowei Liu,Jindong Xiao,Kun Wang

Summary Microemulsion nanofilms based on fish skin gelatine (FSG) and 3% (w/w, gelatine) ZnO nanoparticles (ZnONP) incorporated with ginger essential oil (GEO) at different concentration (10%, 20%, 40%, 80%, w/w, gelatine) were developed and characterised. Film thickness and elongation increased, while film tensile strength and gel strength decreased as GEO level increased ( P < 0.05). The water vapour permeability was reduced ( P < 0.05) by GEO. Microstructural study revealed a dispersion system with GEO lipid droplet embedded into FSG matrix and some aggregation. The developed films showed strong antibacterial activity against food spoilage bacterial of psychrotrophs, mesophiles and lactobacillus spp. (LAB) in situ , and food pathogenic bacteria of Escherichia coli and Listeria monocytogenes in vitro . Together with the antioxidant activity towards lipid oxidation, the release of total volatile bases nitrogen (TVB-N) was subsequently reduced during fresh meat storage. In conclusion, the incorporation of GEO provided the potential to make FSG-ZnONP films more available for meat storage.

Flavor, antimicrobial activity, and physical properties of composite film prepared with different surfactants

Xin Li,Zong-Cai Tu,Xiao-Mei Sha,Yun-Hua Ye,Zhong-Ying Li

Different surfactants (lecithin, Tween-20, and Tween-80) were added in composite film during the preparation. Flavor, antimicrobial activity, and physical properties of ginger essential oil -gelatin film were investigated, in order to study the effect of surfactants on the properties of film. The flavor of GEO was not detected in the film prepared with Tween-20 and film prepared with Tween-80, and these two films exhibited stronger antimicrobial activity; film prepared with lecithin possessed higher value in thickness, elongation at break, water solubility, Δ E and opacity, lower value in water vapor property, and tensile strength; attenuated total reflectance-Fourier transform infrared spectrum results suggested, Tween-20 and Tween-80 enhanced the strength of covalent bond, and lecithin weakened the strength of hydrogen bond; and the result of scanning electron microscope showed that Tween-20 and Tween-80 improved the dispersion of oil droplets in film. Therefore, this study suggested that surfactants had an influence on the physical properties and molecular structure of a resulting film; in addition, Tween-20 and Tween-80 could reduce the flavor of GEO in film, improving the antimicrobial activity of film.

pH-responsive liposomes for controlled release of Alpinia galanga essential oil: Investigating characteristics, stability, control release behavior, and functionality

Wei Liang,Xiangzhen Ge,Qian Lin,Wenqing Zhao,Li Niu,Marat Muratkhan,Wenhao Li

The development of intelligent packaging materials has received more attention. This study designed nanoliposomes encapsulating Alpinia galanga essential oil with γ-polyglutamic acid as a pH trigger switch and evaluated its characteristics, storage stability, controlled release behavior, and functional properties. The characteristic peaks in the ultraviolet and infrared spectrums, and the morphological observation by transmission electron microscopy indicated that the essential oils were successfully encapsulated with pH-responsive liposomes (Lip-PHEO). Also, the Lip-PHEO exhibited uniform particle size (246.50 nm), stable PDI value (0.24) and ζ-potential (33.51 mV), as well as good storage stability at 4 °C. Compared to the control, Lip-PHEO exhibited intelligent controlled release behavior, i.e., it can promote essential oil component release in response to pH changes, resulting in enhanced antioxidant scavenging levels (ABTS + and DPPH free radicals) and antibacterial performance ( Escherichia coli and Staphylococcus aureus ). The findings provide insights for exploiting intelligent response packaging materials and expanding the application range of essential oil-based materials.

Chitosan/gelatin coating loaded with ginger essential oil/β-cyclodextrin inclusion complex on quality and shelf life of blueberries

Md Alomgir Hossen,Islam Md Shimul,Dur E. Sameen,Zainab Rasheed,Wuxia Tanga,Mingrui Chen,Yaowen Liu

Blueberries are highly susceptible to fungal pathogens and oxidative deterioration due to their thin epidermal layer, which can be mitigated by applying a natural polymer-based antimicrobial coating to their surface. This study aimed to develop a chitosan/gelatin-based antimicrobial coating utilizing ginger essential oil (GEO) to extend the postharvest quality of blueberries. To ensure GEO's stability within the coating, it was initially encapsulated with β-cyclodextrin (β-CD) using the inclusion complexation technique. The GEO/β-CD inclusion complex (IC) formed rhomboidal shapes with high encapsulation efficiency and small particle sizes. When the optimized GEO/β-CD IC was incorporated into the chitosan/gelatin polymer solution, it significantly increased surface hydrophobicity and free radical scavenging activity, and suppressed the growth of three selected fungi, namely Botrytis cinerea, Penicillium italicum and Alternaria alternaria . The results of postharvest storage quality revealed that blueberry samples coated with CH/Gel-GEO/β-CD IC-5 effectively maintained the quality of blueberries by decreasing weight loss and decay incidence, and regulating anthocyanin and other oxidation-related enzyme activities compared to the control group during 16 days at 25 °C and 40 days at 4 °C storages. In conclusion, it can be stated that CH/Gel-GEO/β-CD composite coating can be a promising technology to address the drawbacks of blueberry preservation.

Chitosan-ginger essential oil nanoemulsions loaded gelatin films: A biodegradable material for food preservation

Xiaohan Wang,Zhaohui Xue,Yijie Sun,Bo Peng,Caie Wu,Xiaohong Kou

The alarming issue of food waste, coupled with the potential risks posed by petroleum-based plastic preservation materials to both the environment and human health necessitate innovative solutions. In this study, we prepared nanoemulsions (NEs) of chitosan (CS) and ginger essential oil (GEO) and systematically evaluated the effects of varying NEs concentrations (0, 10 %, 30 %, 50 %) on the physicochemical properties and biological activities of gelatin films. These films were subsequently applied to blueberry preservation. The scanning electron microscopy confirmed that the NEs were well-integrated with the Gel matrix, significantly enhancing the performance of the Gel films, including improvements of mechanical properties (tensile strength from 7.71 to 19.92 MPa; elongation at break from 38.55 to 113.65 %), thermal, and barrier properties (water vapor permeability from 1.52 × 10 −9 to 6.54 × 10 −10  g·m/Pa·s·m 2 ). The films exhibited notable antibacterial and antioxidant activities due to the gradual release of GEO, thereby extending the storage life of blueberries. Moreover, the prepared composite films demonstrated excellent biodegradability and environmental friendliness, with the majority of the material decomposing within 30 days under soil microbial action. In conclusion, the active films loaded with NEs exhibit superior performance and hold significant potential for developing biodegradable materials for food preservation.
