Background Pregnancy is a critical period during which women usually do not prefer taking medication. Therefore, non-pharmacological and safe methods are needed to improve sleep quality during pregnancy. This study aims to identify whether an educational program on sleep enhancement for pregnant women has any effect on maternal sleep quality. Method This was a randomized-controlled trial. Pregnant women ( N = 181) were divided into two groups, an intervention group ( n = 90) and a control group ( n = 91). A two-session educational program for the enhancement of sleep quality was provided to the intervention group, whereas solely the routine practices of the hospital were put in place for the control group. Results The intervention group had a significantly lower mean Pittsburgh Sleep Quality Index score, and accordingly, better sleep quality than the control group. Participation in the education program was significantly related to sleep quality and accounted for 6% of the total variance in sleep quality ( R 2 = 0.055) ( p < 0.001). Conclusion It was concluded that the educational program on sleep enhancement implemented to improve the sleep quality of pregnant women enhanced maternal sleep quality. Background: Polycyclic aromatic hydrocarbons (PAHs) are a class of large organic compounds that persist in the environment and are classified as an important subset of persistent organic pollutants (POPs). This study aimed to assess PAH exposure in pregnant women and newborns in Haikou City, China, and evaluate their impact on glycolipid metabolism. Methods: A total of 300 pregnant women and their newborns were selected for the study between May 2022 and December 2023. Data on lifestyle and dietary habits were collected, and PAH levels in venous blood and umbilical cord blood were measured using gas chromatography-mass spectrometry. Glycolipid metabolism indicators, including fasting plasma glucose (FPG), triglycerides (TG), total cholesterol (TC), high-density lipoprotein (HDL), and low-density lipoprotein (LDL), were also measured. Correlation and regression analyses were conducted to explore the relationships between PAH exposure and metabolic indicators. Results: Thirteen PAHs were detected in both maternal and cord blood. The median concentrations of ΣPAHs (the total concentration of the 16 detected PAHs) were 11.211 μg/L in maternal blood and 10.921 μg/L in newborns. Significant correlations were observed between PAH exposure and glycolipid metabolism, with PAH exposure linked to reduced levels of TC and TG. Factors such as proximity to roads, cooking fuel type, and dietary habits influenced PAH levels. Higher education levels were associated with lower PAH concentrations, whereas living near roads and using gas as cooking fuel increased exposure. Conclusion: There is a notable risk of PAH exposure in pregnant women and newborns in Haikou, influenced by environmental and lifestyle factors. PAH exposure during pregnancy may affect glycolipid metabolism in both mothers and newborns, highlighting the need for interventions to reduce exposure.

Perfluorooctanoic acid (PFOA) is a novel type of persistent synthetic organic pollutant, and its exposure on pregnant women can cause some adverse effects, such as pregnancy-induced hypertension, gestational diabetes mellitus, and preeclampsia. Therefore, understanding the metabolic changes caused by PFOA exposure is of great significance to protect pregnant women from its adverse effects. In this study, the metabolomes from the urine samples of pregnant women exposure to PFOA at different degrees were analyzed by GC-MS and LC-MS. The samples in different groups were distinguished and the differential metabolites were screened based on the VIP value, FC, and P-value of each comparison group through multivariate statistical analysis. The pathways related to differential metabolites were searched to reveal the effects of PFOA exposure on metabolic changes in pregnant women at different degrees. Finally, the ROC of differential metabolites was performed, and the differential metabolites with large area under the curve (AUC) values were selected and compared to identify the mutually differential metabolites. Meanwhile, these metabolites were fitted with a multivariable to explore if they could be used to distinguish different groups. The quantitative comparison of mutually differential metabolites revealed that the levels of L-cysteine, glycine, and 5-aminovaleric acid were positively correlated with the degree of PFOA exposure, indicating that different degrees of PFOA exposure could affect the synthesis or degradation of GSH and change the metabolism of oral or intestinal microbiota. Additionally, they may cause oxidative stress and abnormal fat metabolism in pregnant women.

We simultaneously set up two competitive (direct and indirect) enzyme-linked immunosorbent assays (ELISAs) based on the same antibody for estimation of cotinine (COT) in pregnant women especially and population generally exposed to environmental tobacco smoke. The results show that the limits of detection (LODs) for direct competitive ELISA and indirect competitive ELISA were 0.04 μgL -1 and 0.1 μgL -1 , respectively. Direct competitive ELISA was found to be more sensitive than indirect competitive ELISA. Thereafter, we applied our direct competitive ELISA for the detection of COT from urinary samples taken from 450 volunteers from the Zhejiang Province of China. COT was detected in 100% of participants with concentration ranging from LOD to 5358.0 μgL -1 . The GM and 95th percentile concentration of COT in pregnant women were 6.3 μgL -1 and 57.2 μgL -1 , respectively. Males had statistically higher COT concentrations than females ( P < 0.0001), active smokers had statistically higher COT concentrations than non-smokers ( P < 0.0001), whereas, non-pregnant women were found to have higher COT concentration than pregnant women. We conclude that our developed direct competitive ELISA is useful for detecting the COT in urinary concentration of human. The human urinary data obtained in this study indicated that common people generally and pregnant women especially were highly exposed to COT. Further studies are needed to focus on the sources of exposure, potential health effects and risk assessment of exposure to COT. Copyright © 2014 John Wiley & Sons, Ltd.

The colonization of Enterococcus in the female vagina leads to neonatal and pediatric enterococcal septicemia. Linezolid (LZD) is a kind of mainstream drug for treating multidrug-resistant Gram-positive infections. OptrA is the main LZD-resistance gene at Enterococci in human isolates. It is essential to explore the prevalence of optrA-carrying Enterococcus in vaginal secretions of late pregnant women and the drug resistance of optrA. From May to June 2023, this study recruited 340 volunteers in late pregnancy (35–40 weeks of pregnancy) to provide non-repetitive vaginal discharge samples. Luria-Bertani broth and florfenicol (10 µg/mL) were used to enrich bacteria. Enterococci was identified through time-of-flight mass spectrometry. Additionally, antimicrobial susceptibility, polymerase chain reaction, and next-generation sequencing assays were applied to this study. Fifty-four optrA-carrying Enterococcus strains were obtained, the proportion of the whole vagina of late pregnant women was 15.88% (54 out of 340), and Enterococcus faecalis account the highest proportion. All optrA-carrying Enterococcus were resistant to at least three drugs. Tetracycline, chloramphenicol, erythromycin, and LZD have higher bacterial resistance rates. Genetic environment analysis revealed that IS1216E, fexA, and erm(A) may synergistically exert multidrug resistance with optrA. It is necessary to strengthen the surveillance of optrA-carrying Enterococcus in pregnant women. This study provides scientific support for controlling hospital infections and managing antibiotic-resistant bacteria, and provides a scientific basis for rational clinical medication.

Maternal exposure to benzene and related compounds, trichloroethylene, and polycyclic aromatic hydrocarbons has been suggested as risk factors for reproductive and developmental problem. In different countries and groups, there are differences in socioeconomic circumstances, consumer products, dietary habits, lifestyles, and so on, resulting in different exposure risks from these chemicals. This study investigated the correlation between their metabolite concentrations and socioeconomic, demographic, and dietary factors to explore the possible exposure source of the concerned pollutants in pregnant women. We conducted biological monitoring to assess the exposure of these chemicals using urine samples from 590 to 639 pregnant women during 2nd or 3rd trimester of pregnancy in six cities of China. Socioeconomic and demographic characteristics and dietary habits were collected from questionnaires. The detection rate was over 74% of the urine samples for all metabolites. Compared with the Fourth National Report on Human Exposure to Environmental Chemical concentrations for females (FNRHEEC, the US Centers for Disease Control and Prevention), the metabolite concentrations of benzene compounds and trichloroethylene, excluding MA reported here, were higher. Principal component analysis results showed that SBMA and MHA could be proxy of the principal sources of metabolites for benzene compounds. The concentrations of SBMA and MA were higher in Fuzhou and Wuhan, respectively. The concentration of DCVMA was higher in Shenzhen, Xi’an, and Nanning, and the concentration of PGA was higher in Fuzhou, Wuhan, and Xi’an. Also, the 1-OHPG concentration in Wuhan is higher than that in Fuzhou and Shenzhen. Unhealthy dietary habits, using cosmetics and indoor exposure, contacting chemical solvent during pregnancy were associated with increased benzene compounds, trichloroethylene, and PAH exposure. There were significant positive associations between 1-OHPG level and maternal BMI, low education status, and cooking without a range hood. Pregnant women in China may be at a greater risk of exposure to most of the target compounds than US females, and their exposure levels varied in different regions. Some adverse environmental and behavioral factors may increase the exposure of environmental toxins, which can urge people to take measures to reduce the health risk to pregnant women during pregnancy.

Salbutamol (SAL) is used to promote animal growth, because it increases feeding efficiency and carcass lean percentage. SAL has been banned in many countries because of the potential hazard to human health. To investigate the SAL residue level in humans, a sensitive direct competitive ELISA was developed. Under the optimal experimental conditions, the limit of detection (LOD) for the assay was 0.01 ng mL−1, and the standard curve was constructed at concentrations of 0.01–100 ng mL−1. We used the developed assay to detect the urinary concentration of SAL in 1648 participants. SAL was detected in 96% of participants with a concentration ranging from the LOD to 30.83 ng mL−1. Pregnant women were found to have a lower SAL concentration than non-pregnant women. Sources of exposure, risk assessment and measures for reducing the exposure level require further investigation.

Tartrazine is commonly used in foodstuffs, its level is controlled by government, however, it is still used in some food products at levels which are not permitted. We aimed to develop a sensitive and simple method to assess the exposure to tartrazine in pregnant women and the general population in China. A competitive enzyme-linked immunosorbent assay (ELISA) was developed based on polyclonal antibodies for tartrazine. Using this assay, we measured the urinary concentrations of tartrazine in the participants. Under optimal experimental conditions, the standard curve was constructed at concentrations of 0.04–1000 ng mL−1 and the limit of detection (LOD) was set at 0.04 ng mL−1. No cross-reactivity of the antibody was observed with seven other artificial dyes based on this assay. Human blank urine was spiked with tartrazine at concentrations of 0.1, 25, 500 ng mL−1 and analyzed by ELISA, the recoveries of tartrazine were from 99.8% to 106.6% and coefficients of variation were from 3.28% to 14.9%. The study comprised of 351 volunteers (age: 19–69 years), of which 30% were male, 16% were nonpregnant women and 54% were pregnant women. The geometric means for tartrazine in three groups were 2.64, 2.44 and 3.21, respectively. No difference was observed in these groups. The values indicated that both the general population and pregnant women are exposed to tartrazine. Sources of exposure, potential health effects and risk assessment require further investigation.

DNA adducts are widely recognized as biomarkers of exposure to environmental carcinogens and associated health effects in toxicological and epidemiological studies. This study presents a targeted and sensitive method for comprehensive DNA adductome analysis using ultra-high-performance liquid chromatography coupled with triple-quadrupole tandem mass spectrometry (UHPLC-QqQ-MS/MS). The method was developed using calf thymus DNA, with careful optimization of mass spectrometric parameters, chromatographic separation conditions, and pretreatment methods. Ultimately, a targeted method was established for 41 DNA adducts, which showed good linearity (R 2 ≥0.992), recovery (80.1–119.4 %), accuracy (81.3–117.8 %), and precision (relative standard deviation <14.2 %). The established method was employed to analyze DNA adducts in peripheral blood cells from pregnant women in Shanxi and Beijing. Up to 23 DNA adducts were successfully detected in samples of varying sizes. From 2 μg of maternal DNA samples, seven specific adducts were identified: 5-methyl-2'-deoxycytidine (5-MedC), 5-hydroxymethyl-2'-deoxycytidine (5-HmdC), N 6 -methyl-2'-deoxyadenosine (N 6 -MedA), 8-hydroxy-2'-deoxyguanosine (8-OHdG), 5-hydroxy-2'-deoxycytidine (5-OHdC), 1,N 6 -etheno-2'-deoxyadenosine (1,N 6 -εdA), and N 2 -methyl-2'-deoxyguanosine (N 2 -MedG). This study reveals that exposure to higher concentrations of ambient air pollutants may elevate the levels of DNA methylation and oxidative damage at different base sites, highlighting the application potential of DNA adducts as sensitive biomarkers of air pollution exposure.
