Findings on prenatal polyunsaturated fatty acids (PUFA) and offspring allergies have been inconsistent, and the majority of studies have focused on Western populations. This study aimed to investigate the associations between maternal erythrocyte PUFA and offspring allergies in the first 2 years in the Chinese population. We included 573 mother–infant pairs from a birth cohort. Based on the outpatient medical records, we identified the diagnosis and time of offspring allergic disease onset. We measured erythrocyte fatty acids by gas chromatography. Associations were examined using Cox regression. We found that higher maternal total PUFA levels (HR= 0.80; 95%CI: 0.68, 0.94), especially of arachidonic acid (AA) (HR= 0.79; 95%CI: 0.65, 0.97) andn-3 PUFA (HR= 0.77; 95%CI: 0.62, 0.97), were associated with reduced risk of offspring allergies. Similar results were found for eczema. Compared with children without a maternal allergy history, the associations of total PUFA (p= 0.028) andn-6 PUFA (p= 0.013) with offspring allergies were stronger in those with a maternal allergy history. Maternal erythrocyte total PUFA, especially AA, andn-3 PUFA were inversely associated with offspring allergies within 2 years of age. There was a significant interaction between maternal allergy history and maternal PUFA in offspring allergies. Keywords:allergic disease;polyunsaturated fatty acids;offspring;pregnancy

T-2 toxin is a kind of group A trichothecenes mycotoxins , frequently detected in various foods and feeds, having high toxic effects on both humans and animals. The present study aims to investigate the toxic effects of T-2 toxin exposure to ICR mice during pregnancy and lactation on liver glycolipid metabolism of young mice. The pregnant mice were given 0, 0.005 and 0.05 mg of T-2 toxin/kg bw daily through oral gavage from late gestation (GD 14) to the lactation (LD 21). Liver and serum samples of the young mice were collected on postnatal day 21 (PND 21), PND 28 and PND 56. The results showed that T-2 toxin increased the contents of triglycerides (TG), total cholesterol (T-CHO) and glucose in serum of young mice on PND 21 and PND 28. In addition, obvious lipid droplets of liver in T-2 toxin treatment groups were observed, especially in 0.05 mg group of PND 21and PND 28. Compared with the control group, T-2 treatment also increased the expressions of genes associated with liver glycolipid metabolism, such as PEPCK , Glut2 , Fas , Acox1 , Hmgcr , PPARα , Srebp1 and CD36 . These results demonstrated T-2 toxin exposure to pregnant mice could cause liver glycolipid metabolism disruption in the young mice and the toxic effects weakened on PND 56.

Aims Omega-3 (n-3) and omega-6 (n-6) polyunsaturated fatty acids (PUFAs) are indispensable for human health. Notably, imbalanced levels of maternal PUFAs during pregnancy can exert lasting impacts on the developing fetus. There is an urgent need for a robust and rapid method that can simultaneously quantify multiple n-3 and n-6 PUFAs in plasma and adapt to large-scale clinical screening. Methods Sample preparative process consisted of non-derivatized alkali hydrolyzing and liquid–liquid extraction followed by reconstituting after being blown dry with nitrogen, and the total chromatographic run time was only 5.6 min. Results The LC-MS/MS method was validated according to current national and international guidelines. Acceptable reproducibility and accuracy were shown based on the intra-day precision was 1.79%–7.02%, the inter-day precision was 0.01%–5.63%, and the accuracy was 87.42%–102.78%. The correlation coefficients of linearity were >0.9930. All measured plasma individual PUFAs levels in pregnant women significantly increased, apart from eicosapentaenoic acid (EPA, C20:5n3) and n-6/n-3 PUFA ratio levels decreased were observed. The analysis performance is satisfactory and suitable for clinical routine detection. Conclusion This method could be a valuable tool for 10 kinds of n-3 and n-6 PUFAs nutritional assessment, supplementary guidance, and intervention during pregnancy.

Ursodeoxycholic acid (UDCA) has been widely used for treatment of biliary diseases with high incidence among pregnant women. However, the effect of maternal UDCA exposure on offspring metabolism particularly on hepatic lipid metabolism has not been reported. Seventeen pregnant Sprague-Dawley rats were administered with low-dosage UDCA (L-UDCA, 250 mg/kg, n = 6), high-dosage UDCA (H-UDCA, 500 mg/kg, n = 6) or phosphate buffered saline (CON, n = 5) during pregnancy. After parturition, pups from each litter were sacrificed for blood and hepatic tissues sampling at 21 days of age. Maternal exposure to H-UDCA increased body weight and liver weight of the offspring before weaning. The levels of triglyceride (TG) in both plasma and liver of offspring were increased in maternal UDCA exposure and led to a greater hepatic lipid deposition in offspring. UDCA treatment predominantly promoted the d e novo lipogenesis (DNL) in offspring liver by upregulating the gene expression of ATP citrate lyase ( Acly ), acetyl-coA carboxylases ( Acc ), and fatty acid synthase ( Fasn ), as well as protein expression of FASN and ACC. The level of fibroblast growth factor 15 (FGF15) in plasma of offspring was increased in UDCA treatment compared to control. Moreover, protein expression of DNA methyltransferase 1 (DNMT1) in the liver of offspring was decreased in a dose-dependent manner and the CpG methylation level in the Fasn promoter region was decreased. This study provides insights into maternal UDCA exposure promoting offspring hepatic lipid deposition, which is associated with hypomethylation modification of cytosine-phosphate-guanine (CpG) islands on the promoter of Fasn gene.

Limited research on the toxicokinetics of nanoparticles (NPs) in pregnant rats hinders our understanding of the potential risks they pose to the pregnant population. This study examined how exposure dose and size of NPs impacted their specific distribution in pregnant rats under repeated dosing. Results revealed that Au NPs mainly accumulated in the spleen and liver, followed by the uterus, while the heart, placenta, and fetus showed the least accumulation. The decrease of exposure size induced faster elimination of Au NPs in serum and organs. The increase of exposure dose induced faster elimination in serum but slower elimination in organs, higher accumulation, and larger size of Au NPs in vivo. The accumulation and biodistribution of Au NPs varied across different organs depending on the exposure size. Particularly, Au NPs with an in vivo size of 40 nm were shown to transfer the placenta and accumulate in the fetus, regardless of the exposure dose and size. Such effect was closely related to the transport routes of Au NPs across the placenta, possibly via vesicular transport and the uptake of trophoblast cells. Our study illustrating the specific distribution of NPs in vivo provides important evidence for assessing the health risk of NPs.

Background: Pesticide exposure has the potential for causing acute or chronic health effects at certain developmental stages of life. It can result in irreversible damage to structure and function of an organ. The exposure of females to pesticides before or after pregnancy is a matter of concern because of the possibility of having a child born with congenital disorders and developmental defects. Prenatal exposure of any toxicants can program the developing foetus to develop diseases that manifest in later life. Very few reports are available related to the pesticide exposure induced morphological changes in kidney which has prompted us to undertake this research study. Aim: The aim of this study is to evaluate the effect of pesticide, chlorpyrifos on the kidney development of rat neonates after being exposed prenatally to pesticides throughout their gestational period. Materials and Methods: Virgin female wistar rats weighing 200-250 gms were mated with adult males. It consisted of twelve, divided into two sub groups six rats per each were treated with selected pesticides, chlorpyrifos (insecticide) readily available commercially for various agricultural and non-agricultural settings. Insecticide chlorpyrifos was given at a dose of 10mg/kg per orally to the female rats throughout their gestation period. After gestation period rat neonates were sacrificed and their kidney section was processed for histopathological analysis. The SEM study of chlorpyrifos exposed kidney section of rat neonates was performed, investigating changes in morphometric and ultra-structural features of kidney in rat neonates. Results: The histopathological analysis shows morphological changes in the kidneys of prenatally pesticides exposed rat neonates markedly different from control group. Chlorpyrifos exposed neonates kidney shows damaged kidney corpuscles and glomeruli. Conclusion: This study concluded that pesticide exposure during gestation period to pregnant rats causes significant renal toxicity in their neonates.

Maternal health, specifically changes in the gut microbiota, can profoundly impact offspring health; however, our understanding of how gut microbiota alterations during the preconception period influence the offspring remains limited. In this study, we investigated the impact and mechanisms of preconception maternal gut dysbiosis on the development of the enteric nervous system (ENS) in mice. We found that preconception maternal exposure to antibiotics led to the abnormal development of the ENS in offspring, increasing their susceptibility to water avoidance stress at the adult stage. Metagenomic, targeted metabolomic, and transcriptomic analyses revealed that preconception antibiotic exposure disrupted the expression of genes crucial for embryonic ENS development by altering maternal gut microbiota composition. Multi-omics analysis combined with Limosilactobacillus reuteri and propionate gestational supplementation demonstrated that the maternal gut microbiota and metabolites may influence embryonic ENS development via the GPR41–GDNF/RET/SOX10 signaling pathway. Our findings highlight the critical importance of maintaining a healthy maternal gut microbiota before conception to support normal ENS development in offspring. Graphical Preconception administration of antibiotics to female mice reduces the abundance of Limosilactobacillus reuteri in the maternal gut microbiota during pregnancy, subsequently affecting propionate levels. Decreased propionate levels downregulated the expression of Gdnf / Ret / Sox10 mediated by GPR41, leading to abnormal development of the enteric nervous system during the embryonic period. This dysplasia results in colonic dysmotility, impaired colonic epithelium, and increased susceptibility to water-avoidance stress in offspring. Highlights Preconception maternal antibiotic exposure results in the abnormal development of the enteric nervous system (ENS) and intestinal mucosa in mouse offspring, thereby increasing their susceptibility to water avoidance stress in adulthood. Preconception maternal antibiotic exposure alters the composition of the maternal gut microbiota, leading to abnormal embryonic ENS development via the modulation of gene expression. Multi-omics analysis and supplementation with Limosilactobacillus reuteri and propionate during pregnancy indicate that the maternal gut microbiota and metabolites affect embryonic ENS development through the GPR41–GDNF/RET/SOX10 signaling pathway.

Choline in human breast milk is crucial for infant neurodevelopment. However, choline concentration variation in human breast milk remains understudied. In this work, a colorimetric biosensor of bovine serum albumin@Hemin@Ag nanozyme was developed, leveraging its peroxidase-like activity to oxidize 2,2′-azino-bis(3ethylbenzo-thiazoline-6-sulfonic acid) diammonium salt in the presence of hydrogen peroxide generated from choline oxidation by choline oxidase. Under the optimal conditions, a wide linear range (1–500 μM) and a low limit of detection (0.55 μM) were obtained for choline quantification. Longitudinal analysis of paired human breast milk samples revealed a significant decline in choline level from 15- to 90-d postpartum, influenced by maternal diet during pregnancy and pre-pregnancy body mass index. This research not only provides a robust tool for evaluating breast milk's quality, but also underscores the importance of diet during pregnancy and body mass index management for lactating mothers.
