Ganoderma lucidum spore oil (GLSO) is a dietary supplement, with glycerides (GLs) recognized as its important active component. However, comprehensive profiling and accurate structural characterization of GLs in GLSO remain underexplored. In this study, 59 GLs from GLSO were identified by ultra-performance liquid chromatography-quadrupole time-of-flight mass spectrometry (UPLC-Q-TOF MS) and molecular networking (MN). The double bond isomers of these compounds were further resolved by the Paterno-Büchi (PB) reaction coupled with UPLC-Q-TOF MS, resulting in the identification of 36 unsaturated GLs, including 5 pairs of positional isomers. In summary, 64 GLs were characterized, consisting of 9 diacylglycerols (DGs) and 55 triacylglycerols (TGs). Additionally, the compositional variations, antioxidant activities, and relative isomer ratios of C C positional isomers of GLSO from eight different manufacturers were revealed, with 11 GLs correlating with antioxidant activity. This study enhances the understanding of the nutritional value of GLSO and lays a foundation for future quality standard formulation.

Ganoderma lucidum (G. lucidum) spore oil, an important health supplement, has been found to frequently contain the excessive levels of phthalate plasticizers (PAEs), which may cause the safety and quality risks. This study used supercritical fluid extraction (SFE) with 95 % ethanol as the entrainer, added at ratios of 0 % to 25 % of the spore powder mass, to investigate the effects of different extraction stages and entrainer on the reduces of PAEs content, physicochemical properties, and active components. Results showed that the acid value, ergosterol content, and PAEs content of the spore oil in the first stage were significantly higher than those in subsequent stages. Peroxide values were increased but remained below 0.22 g/100 g, while the total triterpene content fluctuated. The main fatty acids were oleic, palmitic, linoleic, and stearic acids. At a 10 % entrainer ratio, the extraction yield increased to 32.30 ± 0.60 %, with the highest ergosterol content, the lowest DINP content, and achieved a 40.73 % reduction in PAEs content. This ratio did not affect the fatty acid composition, indicating it enhanced the oil yield and reduced PAEs without impacting the physicochemical properties and active ingredients. Therefore, the optimal entrainer ratio was determined to be 10 %, crucial for improving G. lucidum spore oil quality and safety in industrial production.

Background: Ganoderma lucidum ( G . lucidum ) is a traditional Chinese herbal medicine that has shown potential as an alternative adjuvant therapy for cancer patients. However, the mechanisms and adjuvant therapeutic effects of G . lucidum in cancer treatment remain unclear. Methods: In this work, G . lucidum spore oil (GanoOil), a newly developed oily G . lucidum spore extract was used to investigate the mechanisms and adjuvant therapeutic effects of GanoOil in conjunction with the chemotherapeutic drug cyclophosphamide (CTX) for preventing breast cancer metastasis. Results: In the model of lung metastasis, orally administered GanoOil increased the population of CD8 + T cells and IL-6 cytokine levels in mouse blood, while also enhancing the activity of natural killer cells in the spleen. Furthermore, the combination of GanoOil and CTX effectively suppressed the lung metastasis of circulating breast cancer cells, alleviated CTX-induced weight loss, and reduced the ratio of lung and spleen weight to body weight in mice. Moreover, high concentrations of GanoOil exhibited no significant toxicity or side effects in both in vitro and in vivo experiments. Conclusion: In conclusion, GanoOil is a safe drug that can enhance immune activity in mice to achieve therapeutic effects on cancer, and can also synergistically inhibit tumor metastasis with CTX.

The incidence of acute liver injury is increasing and poses a significant threat to human health. Ganoderma lucidum spore oil (GLSO), a lipid substance extracted from Ganoderma lucidum spore powder using supercritical CO 2 technology, has been investigated for its potential to prevent acute liver injury. However, the specific mechanism underlying the protective effects of GLSO remains incompletely understood. In this study, we investigated the preventive effect of GLSO on acute liver injury in rats, focusing on the gut microbiome and serum metabolomics. GLSO effectively alleviated liver dysfunction and reduced inflammation, leading to the prevention of acute liver injury in rats. Serum metabolomics analysis revealed that GLSO primarily modulated lipid metabolic pathways related to glycerophospholipid metabolism and sphingolipid metabolism. Specifically, GLSO decreased the levels of metabolites such as lysophosphatidylcholine (LPC), glycerophosphatidylcholine (GPC), and sphinganine 1-phosphate (SA1P), while increasing the levels of phosphatidylglycerol (PG) and digalactosylceramide (DGC). Gut microbiomics data indicated that GLSO effectively regulated the composition of the gut microbiota in rats with acute liver injury. Specifically, it increased the abundance of Firmicutes and decreased the abundance of Proteobacteria . Mantel test correlation analysis revealed a close relationship between gut microbial Burkholderiales and lipid metabolites in GLSO-mediated prevention of acute liver injury. GLSO exerts its preventive effects on acute liver injury by remodeling the gut microbiota and regulating lipid metabolism. These findings provide novel insights and potential directions for the development of new drugs targeting acute liver injury.

Background Ganoderma lucidum ( G. lucidum ), has been documented as a medicinal herb in classical texts and officially recognized in both Eastern and Western pharmacopeias. G. lucidum spore oil (GLSO), a lipid substance extracted from sporoderm-broken spores, has shown potential in enhancing immune function and prolonging the survival of tumor patients. However, the mechanisms underlying GLSO’s immunomodulatory effects remain poorly unknown. Methods The effect of psychological stress on tumor progression and macrophage phagocytosis was analyzed by an in vivo small animal imaging system and flow cytometry. The effect of psychological stress on phospholipid composition in mice was investigated by LC–MS/MS based lipidomic analysis. The effectiveness of GLSO in tumor-bearing mice subjected to restraint stress was observed by tumor burden and phagocytosis of macrophages. Finally, the underlying mechanism of GLSO on macrophage phagocytosis in mice subjected to psychological stress was explored by RNA-seq, and the FcγR/SYK-mediated macrophage phagocytosis pathway was confirmed by qPCR, Western blotting, and confocal laser technology. Results Our study discovered that psychological stress-triggered tumor progression is contributed to by liposoluble components-impaired macrophage phagocytosis. Lipidomics analysis further identified lysophosphatidylinositol [LPI (18:0)] as a key factor suppressing macrophage phagocytic capacity under psychological stress. GLSO was shown to mitigate psychological stress-evoked tumor progression by enhancing macrophage-mediated phagocytosis of tumor cells in vivo . Mechanistically, transcriptomics analysis revealed that the LPI-mediated FcγR phagocytosis pathway is a crucial axis driving the therapeutic effect of GLSO under psychological stress. Conclusion Our findings illustrate that psychological stress-promoted cancer progression is contributed by the critical liposoluble components LPI (18:0)-mediated FcγR phagocytosis signaling inhibition. GLSO alleviates the dampened phagocytosis of macrophages caused by stress through regulating LPI/FcγR-mediated phagocytosis-related pathways, underscoring its potential as a therapeutic intervention for stress-related tumor progression. Solid self-microemulsifying drug delivery systems (SMEDDS) offer unique advantages over conventional liquid formulations, including enhanced physical stability, simplified storage/transportation, and improved drug delivery efficiency. In this study, we developed a paclitaxel (PTX)-loaded SMEDDS using Ganoderma lucidum spore oil (GLSO) as a multifunctional oil phase. Through formulation screening and optimization, the PTX-GLSO-SMEDDS was successfully converted into a solid powder. Solid-state analyses confirmed the amorphous dispersion of PTX within the matrix, while the powder exhibited favorable fluidity, stability, and rapid in vitro drug release kinetics. PTX-GLSO-SMEDDS demonstrated potent antitumor effects by inducing apoptosis and suppressing metastasis in colorectal cancer (CRC) cells. In vivo studies revealed dual therapeutic advantages: enhanced tumor suppression and synergistic immunomodulation via GLSO-mediated activation of tumor-associated cytotoxic T lymphocytes regulation. Notably, the formulation reduced systemic toxicity compared to conventional PTX injections. By integrating GLSO's inherent bioactivity with SMEDDS technology, this work establishes a robust platform that synergizes chemotherapy and immunotherapy, offering a clinically translatable strategy for CRC treatment with improved safety and therapeutical outcomes. The findings underscore GLSO's potential as a natural, multifunctional excipient for advanced drug delivery systems.

Ganoderma lucidum spore oil (GLSO) has been long practiced and widely recognized as a healthy food composite performing potential anticancer and antioxidant activity. However, its further application might be limited by the native poorly soluble and instable properties. Herein, size-tunable GLSO nanosystems have been designed and tailored to three sizes of particles at 20 nm, 40 nm and 80 nm by high-pressure homogenization, for achieving robust therapeutic effect against cancer cells. As expected, toxicity of biocompatible different-sized [email protected] on MGC803 cancer cells significantly enhanced, thanks to the improved cellular uptake and retention efficacy of [email protected] In addition, middle-sized 40 [email protected] performed superior permeability across Caco-2 intestinal absorption barrier to improve internalized quantity of GLSO. More importantly, [email protected] could effectively prevent cell migration and invasion, simultaneously activating caspase activity to trigger cell death. Interestingly, middle size of 40 [email protected] performed superior accumulation efficacy in cells. Ultimately, 40 [email protected] showed potent suppression effect on tumors in vivo , effectively reversing liver and kidney damage induced by cancer, which was accompanied with no obvious toxicity and side effects during treatment. Therefore, this study not only provides a simple method for preparation of GLSO-like traditionally functional composite by high-pressure homogenization, but also sheds light on the application in future clinical cancer therapy.
