PPARs Activity Affects the Hatchability Through Lipid Metabolism Regulation in Silkworm, Bombyx mori L.

Xia Xu,Chunguang Cui,Xin Du,Jine Chen,Xiuling He,Linbao Zhu,Chengjie Hu,Fang Xu,Chenkai Ma,Shaofang Yu,Xingjian He,Houhui Song,Yongqiang Wang

Lipid metabolism serves as the primary energy source for organisms. Silkworm eggs for spring use are divided into two types: autumn-produced eggs for next spring rearing (AS) and spring-produced eggs for next spring rearing (SS). Production practice revealed significant differences in hatching rates between these two types of silkworm production strain QiufengA. In this study, we identified differentially expressed genes (DEGs) primarily enriched in energy metabolism pathways. In particular, the PPARs are involved in energy regulation through lipid metabolism. Furthermore, both AS and SS contained the same eight long-chain fatty acids but in different amounts. Interference with PPARs activity in silkworm eggs disrupted the expression of key genes in this pathway, resulting in a significant decrease in hatching rate. Additionally, knockdown of the pathway key geneBmPlin4led to the reduction in lipid droplets. In conclusion, PPARs regulates the hatching rate of silkworms mainly by affecting lipid metabolism. This study proved the importance of PPARs for hatching and identifies them as potential target genes for population control.

Alcalase treatment for removal of adhesiveness in greenfin horse-faced filefish (Thamnaconus septentrionalis) eggs

Li Bian,Fenghui Li,Pengyan Zhang,Qing Chang,Changlin Liu,Jianlong Ge,Jie Tan,Shengnong Zhang,Siqing Chen

The greenfin horse-faced filefish Thamnaconus septentrionalis is a new marine culture species in East Asia in recent years. It has demersal and adhesive eggs. The artificial fertilization technique of T. septentrionalis has not been established, and commercial hatcheries currently rely on the collection of naturally fertilized eggs. It is impracticable to separate dead eggs from the egg mass , and a large number of waste and faecal matter adhere to the surface of fertilized eggs, causing excess bacterial and fungal loading in eggs incubation tanks. This study aimed to develop an effective method to remove the adhesiveness of fertilized eggs of T. septentrionalis . The effectiveness of tannic acid (0.5%, 0.2% and 0.1%), sodium hypochlorite (0.05%, 0.025% and 0.01%) and alcalase enzyme (4%, 2%, 1% and 0.5%) to eliminate the adhesiveness of T. septentrionalis eggs was evaluated. Eggs were exposed to different deadhesion solutions for up to 20 min, and the number of deadhesive eggs was counted. The results showed that only the alcalase enzyme could effectively remove the adhesiveness of eggs, and the deadhesion efficiency was positively associated with enzyme concentration. The deadhesion rate of the 0.5% alcalase enzyme treatment was more than 70%, while that of the 4%, 2% and 1% alcalase enzyme treatments were more than 90%. The hatching rates were not affected by alcalase enzyme treatment. The hatching rates of the 2% and 1% treatments were higher than those of the control groups. Ultrastructural analysis showed that the surface of eggs had a pore-canal structure, and the zona radiata externa was gradually degraded and finally peeled off during the deadhesion process. Similar to freshwater fishes with adhesive eggs, biochemical analysis showed that the adhesiveness of T. septentrionalis eggs was related to the presence of glycoproteins and mucopolysaccharides in the zona radiata externa. Sodium dodecyl sulphate-polyacrylamide gel electrophoresis (SDS-PAGE) of zona radiata externa proteins showed that the nonwater-activated eggs, water-activated eggs and adhesive fertilized eggs had same protein pattern, and only small peptide fragments were detected in the deadhesive fertilized eggs. This study provides valuble theoretical and technical support for improvement of hatching technology of filefish.

Dose-dependent inhibitory effects of glyphosate on invasive Pomacea canaliculata reproductive and developmental growth under oxidative deposition

Dongxia Liang,Yinshi Li,Shuoyu Li,Dele Meng,Fengchun Li,Siying Huang,Maojian Gong,Junhao Qin,Huashou Li

Glyphosate (GLY) is the most widely used herbicide worldwide, and its effects on animals and plants have attracted increasing attention. In this study, we explored the following: (1) the effects of multigenerational chronic exposure to GLY and H 2 O 2, alone or in combination, on the egg hatching rate and individual morphology of Pomacea canaliculata ; and (2) the effects of short-term chronic exposure to GLY and H 2 O 2 , alone or in combination, on the reproductive system of P. canaliculata . The results showed that H 2 O 2 and GLY exposure had distinct inhibitory effects on the hatching rate and individual growth indices with a substantial dose effect, and the F1 generation had the lowest resistance. In addition, with the prolongation of exposure time, the ovarian tissue was damaged, and the fecundity decreased; however, the snails could still lay eggs. In conclusion, these results suggest that P. canaliculata can tolerate low concentrations of pollution and in addition to drug dosage, the control should focus on two time points, the juvenile and early stage of spawning.

Protective effects of (-)-epigallocatechin gallate and curcumin against acrylamide toxicity

Shanshan Wang,Hufei Wang,Yanqiu Chen,Jilai Liu,Xiaozhen He,Da Huang,Yuanzi Wu,Yonghai Chen,Zuquan Weng

Human lymphocytes as well as zebrafish embryos were used to study the toxicity of acrylamide, and the intervention effects of the dietary antioxidants (-)-epigallocatechin gallate and curcumin have been examined. The alkaline comet assay revealed that acrylamide (1, 5, 10 mmol/L) had dose-response effects on basic DNA damage and oxidative DNA damage in human lymphocytes. Exposure of zebrafish embryos to acrylamide resulted in decrease of hatching rate, and in increases of mortality and teratogenicity. (-)-Epigallocatechin gallate and curcumin reduced DNA damage and protected zebrafish embryos in the growth and development.

Sub-lethal Camphor Exposure Triggers Oxidative Stress, Cardiotoxicity, and Cardiac Physiology Alterations in Zebrafish Embryos

Du Zheng-Cai,Xia Zhong-Shang,Zhang Ming-Zhe,Wei Yan-Ting,Malhotra Nemi,Saputra Ferry,Audira Gilbert,Roldan Marri Jmelou M.,Hsiao Chung-Der,Hao Er-Wei,Hou Xiao-Tao,Deng Jia-Gang

Camphor is a terpene ketone with aromatic and volatile properties in nature derived from the bark of Cinnamomum camphora or synthesized from turpentine. Camphor exhibits various biological properties such as anti-microbial, anti-viral, anti-coccidial, and anti-cancer. It is also used as a form of topical medication for skin irritation, joint pain, and as a relief for itching from insect bites. However, even though the high dose of camphor has been documented to be toxic/lethal in humans in different studies, camphor’s developmental toxicity has not yet been explored, and its extensive mechanism of action is still unclear. In the present study, we aimed to assess the toxic effects of camphor in zebrafish embryos in the initial developmental stages. The obtained results demonstrated that a sub-lethal dose of camphor caused a decrease in hatching rate, body length, and substantial elevation in malformation rate on zebrafish embryos. On further observation, in the following time frame, curved body and pericardial edema of zebrafish were also observed. Furthermore, exposure to a sub-lethal dose of camphor was also able to trigger cardiotoxicity in zebrafish larvae. Later, on subsequent biochemical analysis, it was found that the antioxidant capacity inhibition and oxidative stress elevation that occurred after camphor exposure might be associated with the inhibition of total superoxide dismutase (SOD) activity and an increase in reactive oxygen species (ROS) and malondialdehyde (MDA) concentration. In addition, compared to the control group, several apoptotic cells in treated zebrafish were also found to be elevated. Finally, after further investigation on marker gene expressions, we conclude that the developmental toxicity of camphor exposure might be associated with apoptosis elevation and oxidative stress. Taken together, the current study provides a better understanding of the developmental toxicity of camphor on zebrafish, a promising alternative animal model to assess the developmental toxicity of chemical compounds.

Environmentally relevant concentrations of antidepressant mirtazapine impair the neurodevelopment of zebrafish (Danio rerio)

Jie Zhou,Yanbin Zhao,Jiayin Dai,Kun Zhang

Mirtazapine is a commonly prescribed antidepressant and has been found widespread in aquatic environments. However, its toxicities to aquatic organisms has rarely been explored. Herein, we conducted a comprehensive study on the developmental effects of mirtazapine on early life stages of zebrafish at environmentally relevant concentrations (3.9 ng/L and 43.5 ng/L). Out of the endpoints measured, spontaneous contraction of embryos at 24 h post fertilization (hpf) and hatching rate and heart rate of embryos at 50 hpf and 56 hpf, respectively, were significantly affected. In light-dark transition behavior test, mirtazapine significantly reduced the swimming frequency and swimming speed of embryos at both concentrations of 3.9 ng/L and 43.5 ng/L. Furthermore, the total swimming distances in dark conditions were also significantly reduced. Transcriptomic analysis was further conducted. It demonstrated that the decreased neural activities in embryos may be associated with altered epinephrine and neuregulin signaling. The present results fill a data gap regarding the exposure of fish to mirtazapine at environmentally relevant concentrations and provide new insights into the neurotoxic mechanisms of mirtazapine exposure.
