The ultra-performance liquid chromatography with tandem mass spectrometry (UPLC-MS/MS) detection method was developed for the residues of 10 NSAIDs (salicylic acid, acetylsalicylic acid, acetaminophen, diclofenac, tolfenamic acid, antipyrine, flunixin meglumine, aminophenazone, meloxicam, metamizole sodium) in swine muscle, liver, kidney, and fat. Swine tissue samples were extracted by phosphorylated acetonitrile with the addition of an appropriate amount of internal standard working solution, defatted with acetonitrile-saturated n -hexane, and purified by Hydrophile-Lipophile Balance (HLB) solid-phase extraction column, then separated by UPLC BEH shield RP 18 column with 0.1% formic acid in water/0.1% formic acid in acetonitrile with gradient elution, which was detected in the multiple reaction monitoring (MRM) modes. The correlation coefficient of the standard curve equation is greater than 0.99, and the coefficient of variation within and between batches is less than 14.4%. We evaluated the analytical method using two green assessment tools. The method established in this study met the requirements of NSAID residue analysis and provides analytical tools for determining and confirming NSAIDs in swine tissue samples. This is the first report on the simultaneous determination of 10 NSAIDs in four swine tissues by the UPLC-MS/MS method and accurate quantification using deuterated internal standards.

Metamizole easily decomposes in the body and has a short action time and low bioavailability. Hence, frequent injection administrations are needed to maintain its plasma concentration. This study aimed to design and develop an in-situ gel based on poloxamer 407 and 188 to assess its long-acting antipyretic effects. The in-situ gel-forming systep00m with optimum sol–gel transition temperature of 35.9 °C to 36.3 °C could be formed using a combination of P407 at a ratio of 21–23% (w/v) and P188 at a ratio of 2–4% (w/v). In vitro erosion test showed that the in-situ gel’s erosion curve and the metamizole release rate both reached about 90% at 6 h, revealing a good linear relationship between the in-situ gel erosion and the drug release. In vitro release test with dialysis tube showed that the release of metamizole from the in-situ gel was remarkably slower than that from the metamizole solution. Approximately 85% of metamizole was released in the dialysis tube within 7 h, implying a good sustained release effect. Pharmacodynamic study showed that the in-situ gel injection extended the action time of metamizole relative to that when using the metamizole solution. Pharmacokinetic study revealed that the in-situ gel significantly increased the blood serum half-life and area under the curve), contributing to a sustained release and improved bioavailability. This study demonstrated that in-situ gel injection could prolong the action of metamizole in the body to reduce the number of administration times and has good clinical application. Dipyrone (DIP) is a commonly used non-steroidal anti-inflammatory drug with strong analgesic, antipyretic, and spasmolytic properties. Following oral administration, DIP is rapidly degraded into four major metabolites: 4-methylaminoantipyrine (MAA), 4-formylaminoantipyrine (FAA), 4-aminoantipyrine (AA), and 4-acetylaminoantipyrine (AAA). In this study, we produced a monoclonal antibody (mAb) and developed a lateral flow immunoassay (LFIA) for the simultaneous detection of the four DIP metabolites in milk samples. By comparing four haptens, a highly sensitive and specific mAb (3C7) was produced with 50% inhibitory concentrations of 8.67, 8.08, 0.98, and 0.82 ng mL−1 for AA, MAA, FAA, and AAA, respectively. Based on 3C7, an LFIA was successfully developed for the determination of DIP metabolites in spiked milk samples with visual limits of detection of 5, 5, 1, and 0.5 ng mL−1 for AA, MAA, FAA, and AAA, respectively, with recoveries (mean ± SD) ranging from 83.0 ± 3.3% to 103.8 ± 3.8%. Our findings revealed that the LFIA we developed is a potential method for the rapid and sensitive detection of DIP metabolites.
