Screening Sugars Can Benefit the Parasitoid Cotesia chilonis (Hymenoptera: Braconidae) Without Benefiting Its Host, Chilo suppressalis (Lepidoptera: Crambidae)

Tian Jun-Ce,Chen Yang,Shelton Anthony M,Zheng Xu-Song,Xu Hong-Xing,Lu Zhong-Xian

Most adult parasitoids depend on carbohydrate-rich food as an energy source for longevity, fecundity, and mobility. Thus, providing sugars has been proposed as a technique to maximize the biological control efficacy of parasitoids. However, the sugars provided for parasitoids need to be carefully selected because herbivore hosts might also benefit. Here we explore the effects of 12 naturally occurring sugars on the longevity and fecundity of the rice pest, Chilo suppressalis, and the longevity of its parasitoid, Cotesia chilonis, as well as the effect of sugars on sugar consumption of C. chilonis. Results showed that none of the tested sugars significantly impacted the longevity of C. suppressalis, but fructose, glucose, maltose, and sucrose significantly increased its fecundity. The longevity of C. chilonis was significantly increased when fed fructose, glucose, sucrose, maltose, trehalose, and melezitose. Our data suggest that C. chilonis consumed larger quantities of glucose, fructose, and sucrose followed by maltose, melezitose, and trehalose and the longevity of C. chilonis was positively correlated to sugar consumption. Our data also suggest that the herbivore C. suppressalis and its parasitoid C. chilonis responded differently to trehalose and melezitose. Although additional studies are needed, our data suggest that these sugars could be considered as candidate components for sugar sprays to enhance the activity and efficacy of C. chilonis, but without benefiting its pest host.

Bt rice does not disrupt the host-searching behavior of the parasitoid Cotesia chilonis

Liu Qingsong,Romeis Jörg,Yu Huilin,Zhang Yongjun,Li Yunhe,Peng Yufa

We determined whether plant volatiles help explain why Cotesia chilonis (a parasitoid of the target pest Chilo suppressalis ) is less abundant in Bt than in non- Bt rice fields. Olfactometer studies revealed that C. chilonis females responded similarly to undamaged Bt and non- Bt rice plants. Parasitoids preferred rice plants damaged by 3 rd -instar larvae of C. suppressalis, but did not differentiate between caterpillar-infested Bt and non- Bt plants. According to GC-MS analyses of rice plant volatiles, undamaged Bt and non- Bt rice plants emitted the same number of volatile compounds and there were no significant differences in the quantity of each volatile compound between the treatments. When plants were infested with and damaged by C. suppressalis larvae, both Bt and non- Bt rice plants emitted higher numbers and larger amounts of volatile compounds than undamaged plants, but there were no significant differences between Bt and non- Bt plants. These results demonstrate that the volatile-mediated interactions of rice plants with the parasitoid C. chilonis were not disrupted by the genetic engineering of the plants. We infer that parasitoid numbers are lower in Bt than in non- Bt fields because damage and volatile induction by C. suppressalis larvae are greatly reduced in Bt fields.

Parasitoids as taxonomists: how does the parasitoid Chouioia cunea distinguish between a host and a non-host?

Lina Pan,Wenfang Gao,Xinyu Liu,Dongyu Qin,Tiantian Zhang,Rui Ren,Weiyi Zhang,Meidi Sun,Cuiqing Gao,Penghua Bai,Wei You,Gengping Zhu,Min Li

BACKGROUND How parasitoids distinguish hosts from non-hosts remains an unknown question. Chouioia cunea Yang (Eulophidae) is an important fall webworm parasitoid that attacks many forest and agricultural pests. To study the differences in the chemical clues used by C. cunea to distinguish host and non-host plants, we used gas chromatography–mass spectrometry (GC–MS) to identify volatile compounds of two C. cunea hosts ( Hyphantria cunea and Helicoverpa armigera ) and two non-hosts ( Spodoptera exigua and Spodoptera frugiperda ). Additionally, we used behavioral assays to compare the attraction of C. cunea to various compounds. RESULTS The two natural host species were more attractive than the two non-host species, in the following order: Hyphantria cunea > Helicoverpa armigera > S. exigua = S. frugiperda . The pupae of the natural hosts contained 1-dodecene, which was not produced by the two natural non-hosts. When the ‘attractants’ based on the difference between the species-specific blend emitted by pupae and the optimal blend were sprayed onto the natural non-host pupae, they significantly improved the attraction of C. cunea to the non-host pupae. CONCLUSION These results revealed that specific host-produced volatile compounds guide C. cunea to distinguish between natural hosts and non-hosts. Overall, this study provides a foundation for developing a behavior-modifying strategy to re-direct C. cunea attacks to control important non-host pests. © 2023 Society of Chemical Industry.

Isolation and identification of attractants from the pupae of three lepidopteran species for the parasitoid Chouioia cunea Yang

Min Li,Yixin Yang,Yonghong Yao,Weifang Xiang,Jiayi Han,Yonghui Wang,Penghua Bai,Jing Wang,Gengping Zhu,Liang Man,Fan Zhang,Lina Pan

BACKGROUND Chouioia cunea Yang (Hymenoptera: Eulophidae) is a parasitic wasp and natural enemy of several lepidopteran pests during their pupal stage. The volatiles from pupae of three hosts, Hyphantria cunea (Arctiidae), Antheraea pernyi (Saturniidae) and Lymantria dispar (Erebidae), were analyzed and compared to elucidate the chemical cues used by C. cunea to locate its hosts. RESULTS The attraction of C. cunea to H. cunea pupae has no obvious association with the types of plant leaves consumed by H. cunea before pupation. C. cunea exhibited the strongest attraction to the pupae of H. cunea , followed by those of A. pernyi and L. dispar based on behavioral experiments. Gas chromatography-mass spectrometry and GC-electroantennography (GC-EAD) analyses showed that these three host pupae consisted of essentially the same active volatile components but at different relative amounts. Active components derived from these pupae by GC-EAD were alkanes from C12 to C27, and C. cunea showed different levels of attraction to different single compounds. CONCLUSION Host location by C. cunea primarily depends on common compounds emanating from the pupae of several host species. The relative amount of each component varies across host species, guiding host preferences by C. cunea . Optimal blends of several components were identified. Understanding the chemical cues used by C. cunea to locate its host could increase the possibility of developing attractants for parasitic wasps and subsequently increasing the parasitism rate of C. cunea on various hosts. © 2019 Society of Chemical Industry

Behavioral responses of Anagrus nilaparvatae to common terpenoids, aromatic compounds, and fatty acid derivatives from rice plants

Guofeng Mao,Jiaxin Tian,Ting Li,Hatem Fouad,Hassan Ga'al,Jianchu Mo

The attraction of parasitoids to their hosts is a key issue in efforts to enhance biological control efficiency. This study was conducted to identify which rice plant volatiles are attractive to females of the rice planthopper egg parasitoid Anagrus nilaparvatae Pang et Wang (Hymenoptera: Mymaridae) by using Y-tube olfactometer behavior assays. Our results demonstrated that 10 of 17 selected volatiles – that is, 3-carene, linalool oxide, linalool, benzyl alcohol, benzaldehyde, methyl salicylate, ( E )-2-hexenal, ( E )-2-hexen-1-ol, ( E )-3-hexen-1-ol, and hexanal – from three major groups of rice plant volatiles – terpenoids, aromatic compounds, fatty acid derivatives – attracted female A. nilaparvatae at various doses. Two full-component mixtures of 10 behaviorally active compounds at different ratios were more attractive to wasps compared with individual volatiles. Blends composed of the 10 behaviorally active compounds in their optimally attractive doses (i.e., each in the dose that elicited the strongest response in the wasps) or at equal mass ratio may be considered as attractants for A. nilaparvatae and thus help to control rice planthoppers.

Cytochrome P450s Are Essential for Insecticide Tolerance in the Endoparasitoid Wasp Meteorus pulchricornis (Hymenoptera: Braconidae)

Xiaorong Xing,Mengwen Yan,Huilin Pang,Fu’an Wu,Jun Wang,Sheng Sheng

Simple SummaryP450s are involved in the detoxification process in insects; however, less attention has been paid for their roles in parasitoid wasps. Here, we identified 28 P450s from the previous constructed transcriptome dataset ofMeteorus pulchricornisand validated their expression patterns under insecticides exposure. Furthermore, we silencedCYP369B3in adult wasps and demonstrated that the knocking downCYP369B3significantly increased of the mortality of adult wasps when they were exposed to each chemical insecticide, phoxim, cypermethrin, and chlorfenapyr, respectively. The present study provides a theoretical base for functional research on P450 genes in parasitoid wasps.AbstractWith the widespread application of insecticides, parasitoid wasps may also be under risk when exposed to insecticides directly at their free-living stages. The endoparasitoid waspMeteorus pulchricornisis the predominant natural enemy of many lepidopteran pests, such asSpodoptera lituraandHelicoverpa armigera. The cytochrome P450 monooxygenases constitute a ubiquitous and complex superfamily of hydrophobic, haem-containing enzymes. P450s are involved in the detoxification of many xenobiotics. However, their exact roles in the tolerance mechanism in parasitoids toward insecticides has received less attention. Here, 28 P450 genes inM. pulchricorniswere identified from a previously constructed transcriptome dataset. These P450 genes belonged to CYP2, -3, and -4, and mitochondrial clans. Subsequently, eight candidateMpulCYPswere selected from four CYP clans to validate their expression patterns under phoxim, cypermethrin, and chlorfenapyr exposure by qRT-PCR. The results showed that all three insecticides had significant effects on the expression ofMpulCYPs. To further study the function of P450s,CYP369B3was silenced, and its expression levels ofCYP369B3were significantly decreased. Survival analysis indicated that after dsRNA injection, the mortality rate of wasps was significantly increased whenM. pulchricornisfemales were exposed to insecticides compared to control groups. Our findings provide a theoretical base for elucidating the mechanism of insecticide tolerance and promote functional research on P450 genes in parasitoid wasps. Keywords:Meteorus pulchricornis;cytochrome P450s;detoxification;insecticide tolerance

Caterpillar-induced rice volatiles provide enemy-free space for the offspring of the brown planthopper

Xiaoyun Hu,Shuangli Su,Qingsong Liu,Yaoyu Jiao,Yufa Peng,Yunhe Li,Ted CJ Turlings

Plants typically release large quantities of volatiles in response to herbivory by insects. This benefits the plants by, for instance, attracting the natural enemies of the herbivores. We show that the brown planthopper (BPH) has cleverly turned this around by exploiting herbivore-induced plant volatiles (HIPVs) that provide safe havens for its offspring. BPH females preferentially oviposit on rice plants already infested by the rice striped stem borer (SSB), which are avoided by the egg parasitoid Anagrus nilaparvatae, the most important natural enemy of BPH. Using synthetic versions of volatiles identified from plants infested by BPH and/or SSB, we demonstrate the role of HIPVs in these interactions. Moreover, greenhouse and field cage experiments confirm the adaptiveness of the BPH oviposition strategy, resulting in 80% lower parasitism rates of its eggs. Besides revealing a novel exploitation of HIPVs, these findings may lead to novel control strategies against an exceedingly important rice pest.

OsRCI-1-Mediated GLVs Enhance Rice Resistance to Brown Planthoppers

Kaiming Mao,Chengzhe Li,Huacai Zhai,Yuying Wang,Yonggen Lou,Wenhua Xue,Guoxin Zhou

Green leaf volatiles (GLVs) play pivotal roles in plant anti-herbivore defense. This study investigated whether the rice 13-lipoxygense geneOsRCI-1is involved in GLV production and plant defense in rice. The overexpression ofOsRCI-1(oeRCIlines) in rice resulted in increased wound-induced levels of two prominent GLVs,cis-3-hexen-1-ol andcis-3-hexenal. In a previous study, we found that the overexpression ofOsRCI-1reduced the colonization by the rice brown planthopper (BPH,Nilaparvata lugens) but increased the attractiveness to the egg parasitoidAnagrus nilaparvataecompared to wild-type (WT) plants. This study found that whencis-3-hexen-1-ol, but notcis-3-hexenal, was added to WT plants, it could change the BPH’s colonization preference, i.e., more BPHs preferred to colonize theoeRCIlines. The exogenous application ofcis-3-hexen-1-ol orcis-3-hexenal to BPH-infested WT plants could weaken or overturn the preference ofA. nilaparvataeforoeRCIlines. However, field experiments revealed that onlycis-3-hexenal was attractive to the parasitoid and increased the parasitism rates of BPH eggs. These results indicate thatOsRCI-1is involved in rice GLV production and therefore modulates both direct and indirect defense in rice.

Olfactory behavioral responses of two Drosophila species and their pupal parasitoid to volatiles from bananas

Yaluan Bi,Xuan Zhang,Xuefei Chang,Jiaxin Li,Shan Xiao,Bo Zhang,Cong Dang,Linlin Sun,Hongwei Yao,Qi Fang,Fang Wang,Gongyin Ye

Background Behavior-based manipulation is an essential part of Drosophila integrated pest management (IPM). Effective compounds are useful for improving the efficiency of baits and the development of attract-and-kill or push–pull strategies to manage Drosophila populations. Here, we investigated the olfactory behavior of two Drosophila species, as well as their pupal parasitoid, to volatiles from bananas, for the identification of effective compounds to control fly populations. Results The results showed that overripe bananas were most attractive to both flies, with a higher attraction index (AI) in Drosophila melanogaster than in Drosophila suzukii . The profiles of volatile organic compounds (VOCs) from bananas with three different ripening stages were compared, and six VOCs were selected for behavioral tests. D. suzukii showed significantly different responses to isoamyl alcohol with D. melanogaster . The effects of ethyl butyrate and ethyl isovalerate on the two flies were dose-dependent, with lower concentrations acting as repellent and higher concentrations acting as attractant. Isoamyl acetate, isoamyl butyrate and isoamyl isovalerate (0.005–0.05% v/v) were attractive to both flies. The parasitoid wasp Pachycrepoidus vindemmiae was repelled by isoamyl alcohol and ethyl butyrate, and attracted by ethyl isovalerate, but showed no significant response to isoamyl acetate, isoamyl butyrate and isoamyl isovalerate at the tested concentration . Conclusion This study indicated that the behavioral response of Drosophila to the odor of chemical compounds is dose-dependent. Isoamyl alcohol and isoamyl acetate at certain concentrations could be used as repellents, whereas ethyl isovalerate as an attractant in control strategy design for D. suzukii. © 2023 Society of Chemical Industry.

An odorant receptor mediates the attractiveness of cis-jasmone to Campoletis chlorideae, the endoparasitoid of Helicoverpa armigera

Y.-L. Sun,J.-F. Dong,C. Ning,P.-P. Ding,L.-Q. Huang,J.-G. Sun,C.-Z. Wang

Parasitic wasps rely on olfaction to locate their hosts in complex chemical environments. Odorant receptors (ORs) function together with well-conserved odorant coreceptors (ORcos) to determine the sensitivity and specificity of olfactory reception. Campoletis chlorideae (Hymenoptera: Ichneunmonidae) is a solitary larval endoparasitoid of the cotton bollworm, Helicoverpa armigera , and some other noctuid species . To understand the molecular basis of C. chlorideae ’s olfactory reception, we sequenced the transcriptome of adult male and female heads (including antennae) and identified 211 OR transcripts, with 95 being putatively full length. The tissue expression profiles, as assessed by reverse-transcription PCR, showed that seven OR s were expressed only or more highly in female antennae. Their functions were analysed using the Xenopu slaevis oocyte expression system and two-electrode voltage-clamp recordings. CchlOR62 was tuned to cis -jasmone, which was attractive to female C. chlorideae adults and H. armigera larvae in the subsequent behavioural assays. Further bioassays using caged plants showed that the parasitism rate of H. armigera larvae by C. chlorideae on cis -jasmone-treated tobacco plants was higher than on the control plants. Thus, cis -jasmone appears to be an important infochemical involved in the interactions of plants, H. armigera and C. chlorideae , and CchlOR62 mediates the attractiveness of cis -jasmone to C. chlorideae .

Volatiles from Cotton Plants Infested by Agrotis segetum (Lep.: Noctuidae) Attract the Larval Parasitoid Microplitis mediator (Hym.: Braconidae)

Mengyu Li,Shike Xia,Tao Zhang,Livy Williams III,Haijun Xiao,Yanhui Lu

Herbivore-induced plant volatiles (HIPVs), chemicals produced by plants infested by herbivorous insects, can act as kairomones that recruit natural enemies of the pest herbivore.Agrotis segetum(Denis and Schiffermüller) is a common, important pest of seedling cotton in Xinjiang Province, China, and the braconidMicroplitis mediator(Haliday) is an important mortality factor of this pest’s larvae. In olfactometer tests, which included healthy foliage, infested foliage, or infested roots,M. mediatorpreferredA. segetum-infested cotton plants to healthy cotton plants. In GC-MS analyses of plant-emitted volatiles, we found that compounds emitted increased 14.9- and 13.3- fold after leaf infestation and root infestation, respectively, compared to healthy control plants. The volatiles were mainly p-xylene, nonanal, tetradecane, decanal, benzaldehyde, β-caryophyllene, and humulene, while linalool was only present in the leaf-infestation treatment. In addition, principal component analysis indicated that all 18 compounds were associated with the infested plants, especially β-caryophyllene, p-xylene, and decanal. Based on the above studies and previous functional evaluations of the volatile compounds, it can be demonstrated that these compounds play a crucial role in modulating the interactions betweenA. segetumandM. mediatorand regulating parasitoid behavior. It may be possible to enhance the biological control ofA. segetumbyM. mediatorthrough the application of HIPVs.Keywords:turnip moth;parasitoid wasp;herbivore-induced plant volatiles;behavioral response;biological control

Evaluation of floral-derived volatile blend for attracting aphid parasitoids and lady beetles in the tobacco fields

Shaolong Wu,Fengli Liu,Weiai Zeng,Zhipeng Xiao,Jiaying Li,Kai Teng,Qianshaung Guo,Jinhao Zhao,Yongjun Du

In tobacco planting in China, the artificial release of aphid parasitoids Aphidius gifuensis has been applied to control aphids for a decade. However, due to seasonal effects and crop rotation, the sustainability of releasing parasitoids and the aphid control efficacy have not been properly evaluated. In this study, effective semiochemical-based attractants were further screened and optimized in the tobacco fields. Film lures were developed to release the synthetic attractant blends. In tobacco planting fields, the synthetic mixture of β-pinene, methyl salicylate , linalool , and limonene(6:6:2:20 mg) significantly increased the population of natural enemies, including A. gifuensis and ladybeetles ( Coccinella septempunctata , Harmonia axyridis and Hippodamia variegate ) monitored using using malaise traps. The increase in the natural enemy population was related to the season and the lure placement density in the tobacco fields, as well as the distribution of surrounding habitats and flowering plants. A density of 135 lures per hectare significantly increased the number of parasitoids, reduced the population of aphids, elevated the rate of parasitized aphids by 42%, and effectively suppressed the aphid population in the tobacco fields. Combined with the protection of flowering plants around tobacco fields, synthetic lures may provide a technical measure for conservative biological control.
