Norfloxacin chitosan/chitosan oligosaccharide microcapsules (NCCM) were prepared by emulsion-chemical crosslinking method. The characteristics of obtained microcapsules were evaluated by scanning electron microscopy, Fourier transform infrared spectroscopy and release experiments. Cumulative release profile of norfloxacin from the chitosan microcapsules in natural seawater was measured and the controlled release of drugs was at a uniform rate in 48 h. The chitosan microcapsules were applied onto the antibacterial study of the shrimp culture in natural seawater. It is observed that the seawater in the NCCM added groups was relatively clear and the biomass of Vibrio increased slowly in contrast to the control and norfloxacin groups. The inhibition rate of Vibrio in norfloxacin groups obvioursly decreased after the 5 th day, whereas, it remained high and stable during experiment period in NCCM groups. The results showed that the chitosan microcapsules as release materials have excellent antibacterial effects on Vibrio in the farming of Penaeus vannamei Boone. The controlled release could obviously reduce dosage of antibiotics and delivery times, and effectively improve the utilization rate of norfloxacin drugs for shrimps

White spot syndrome virus (WSSV) is a pathogenic and threatening virus in shrimp culture for which there is no effective control strategy. Finding antiviral lead compounds for the development of anti-WSSV drugs is urgent and necessary; in this study, esculin from 12 monomeric compounds exhibited an excellent anti-WSSV activity. The results showed that esculin increased the survival rate of WSSV-infected shrimps by 59% and reduced the virus copy number in vivo over 90% at 100 μM. In the pre-treatment and post-treatment experiments, esculin could prevent and treat WSSV infection. Compared with the control group, the virus copy number decreased by 30% after 6 h of esculin pre-incubation with WSSV particles and inhibited horizontal transmission of WSSV to a certain extent. Considering that the antiviral activity of esculin was stable in the aquacultural water for 2 days, we evaluated the dosing pattern of continuous medication changes. Obviously, the survival rate of WSSV-infected shrimps was 0% at 108 h when no esculin exchange was made, while at 120 h the survival rate was over 40% at continuous medicine changes. In addition, esculin significantly increased the expression of antimicrobial peptides and thus improved the ability of shrimp to resist WSSV. Overall, our findings suggest that esculin has the potential to be developed into an anti-WSSV medicine.

As one of the most harmful pathogenic bacteria in shrimp aquaculture, Vibrio parahaemolyticus often causes massive mortality in shrimp. Accurate and rapid detection of V. parahaemolyticus in shrimp farming is essential for avoiding huge economic losses caused by related diseases. In this study, we designed a dual-recognition platform for efficient identification and quantification of V. parahaemolyticus . First, the target bacterium was captured with magnetic beads functionalized by aptamers (Apt-MBs), and then, the broad-spectrum fluorescent probe FcMBL@CdSe-ZnS was used to detect the bacterium based on the interactions between fragment crystallizable mannose-binding lectin (FcMBL) and pathogenic bacteria. The proposed dual-recognition strategy centered around aptamers and FcMBL@CdSe-ZnS was applied to definite quantification of V. parahaemolyticus over a wide range of 10–10 8 CFU/mL with a limit of detection of 4 CFU/mL within 55 min. The feasibility was demonstrated by using the platform to detect V. parahaemolyticus from shrimp intestine, aquaculture water, and seawater. Graphical

White spot syndrome virus (WSSV) is an important pathogen causing high mortality in the shrimp industry in aquaculture, yet there is no treatment available to date. In order to find a treatment against WSSV infection, this study examined the anti-WSSV activity of eight natural compounds using shrimp larvae as a model. Among the eight compounds, paeoniflorin showed the most obvious anti-WSSV effect, with a maximum protection efficiency of WSSV-infected shrimp >60% at 100 μM. Furthermore, pretreatment and post-treatment experiments revealed that paeoniflorin could prevent and treat WSSV infection in shrimp. The antiviral activity of paeoniflorin in aquaculture water decreased rapidly with time, and the results showed that the stable anti-WSSV activity of paeoniflorin could only remain in water for 1 day. Thus, the dosing pattern of continuous medication changes was evaluated. Obviously, in the model of continuous change of paeoniflorin, WSSV copy numbers in the virus-treated shrimp group still progressively increased, while the virus content in WSSV paeoniflorin -treated group continued to decrease. Interestingly, paeoniflorin inhibited horizontal transmission of WSSV to a certain extent. Notably, paeoniflorin significantly increased the expression of antimicrobial peptides of shrimp to resist WSSV. In conclusion, paeoniflorin has the potential to protect shrimp against WSSV.

With the vigorous development of the mariculture industry, the untreated wastewater from mariculture has exerted significant pressure on the water environment. The untreated N and P in the wastewater from mariculture can deteriorate the quality of the mariculture. In this study, a composite in situ treatment system involving macroalgae ( Caulerpa lentillifera / Caulerpa sertularoides f. Longipes ) and immobilized degrading bacteria was established to handle wastewater from shrimp culture. The changes in nutrients in aquaculture wastewater were studied by chemical analysis, and the microbial community structure was analyzed using molecular biology technology and high-throughput sequencing technology. The removal efficacy of nutrients in aquaculture wastewater and the composition of microorganisms in the wastewater were examined, and the primary causes for the alteration of the microbial community were analyzed. The results demonstrated that when the macroalgae in the system were Caulerpa lentillifera ( CL ), the removal efficiencies of TN, PO 4 3- -P, and COD from shrimp culture were 59.04%, 34.26%, and 68.61% respectively. When the macroalgae was Caulerpa sertularoides f. Longipes ( CSF ), the removal efficiencies of TN, PO 4 3- -P, and COD generated by experimental shrimp culture were 51.50%, 33.69%, and 50.88% respectively. The biomass (wet weight) of both macroalgae species also increased, facilitating the removal of nutrients from the wastewater. Additionally, both Proteobacteria and Bacteroidetes were the dominant bacteria in the three samples, and the addition of the composite in-situ treatment system had no impact on the dominant bacteria in the water. The results of FAPROTAX analysis indicated that compared with the untreated samples, the abundances of methyl-functional bacteria and amino acid-functional bacteria in the samples increased due to organic matter such as COD produced during shrimp culture and the addition of feed, suggesting that shrimp culture can influence the abundances of functional bacteria in the water. In conclusion, the combined in situ treatment system can effectively eliminate nutrients from aquaculture wastewater, and the combined effect of macroalgae and immobilized degrading bacteria plays a vital role in this process.
