Natural ingredient paeoniflorin could be a lead compound against white spot syndrome virus infection in Litopenaeus vannamei

Yang Hu,Lei Liu,Li-peng Shan,Jiong Chen

White spot syndrome virus (WSSV) is an important pathogen causing high mortality in the shrimp industry in aquaculture, yet there is no treatment available to date. In order to find a treatment against WSSV infection, this study examined the anti-WSSV activity of eight natural compounds using shrimp larvae as a model. Among the eight compounds, paeoniflorin showed the most obvious anti-WSSV effect, with a maximum protection efficiency of WSSV-infected shrimp >60% at 100 μM. Furthermore, pretreatment and post-treatment experiments revealed that paeoniflorin could prevent and treat WSSV infection in shrimp. The antiviral activity of paeoniflorin in aquaculture water decreased rapidly with time, and the results showed that the stable anti-WSSV activity of paeoniflorin could only remain in water for 1 day. Thus, the dosing pattern of continuous medication changes was evaluated. Obviously, in the model of continuous change of paeoniflorin, WSSV copy numbers in the virus-treated shrimp group still progressively increased, while the virus content in WSSV paeoniflorin -treated group continued to decrease. Interestingly, paeoniflorin inhibited horizontal transmission of WSSV to a certain extent. Notably, paeoniflorin significantly increased the expression of antimicrobial peptides of shrimp to resist WSSV. In conclusion, paeoniflorin has the potential to protect shrimp against WSSV.

RNA-seq revealed the signatures of immunity and metabolism in the Litopenaeus vannamei intestine in response to dietary succinate

Yafei Duan,Yun Wang,Dalin Xiong,Jiasong Zhang

The intestine is important for nutrition, metabolism and immunity. Succinate (SA) plays a vital role in the physiological homeostasis of animal intestines. However, the effects of dietary SA on the intestinal immunity and metabolism in shrimp are not clear. In this study, we investigated the immune and metabolic responses in the intestine of Litopenaeus vannamei that were fed diets consisting of different levels of SA: 0 g/kg (Con) and 10 g/kg (SA) for 56 days. The results from a RNA-seq analysis identified 6005 differentially expressed genes (DEGs), including 2728 upregulated genes and 3277 downregulated genes, which were grouped into 312 pathways. The DEGs were most enriched in pathways related to protein synthesis and amino acid metabolism , including “ribosome”, “aminoacyl-tRNA biosynthesis”, “pyrimidine metabolism”, and “arginine and proline metabolism”; additionally, carbohydrate and lipid metabolism pathways were also activated. A large number of immune-related genes were associated with mucus barrier modification, antimicrobial activity , pathogen attachment and recognition, antioxidant activity , and apoptosis. The expression patterns of several candidate genes involved in the immune response and nutrition metabolism were detected by qPCR. This study provides insight into the transcriptomic modulating mechanisms associated with intestinal immunity and the metabolism of L. vannamei in response to the intake of dietary SA.

Effects of hypoxic stress on gastric sieve (GS) filtering function of Litopenaeus vannamei

Letian Li,Yuan Wang,Laizhuang Jin,Lei Feng,Qianhui Song,Di Fang,Na Ying,Xincang Li,Wenhong Fang

The gastric sieve (GS) is a filtering structure found in the posterior stomach chamber (PSC) of Litopenaeus vannamei , which allows small particles to enter the hepatopancreas directly. However, it remains unclear if pathogens can also enter the hepatopancreas through this pathway. In this study, we examined the internal structural changes of the PSC via histological and ultra-structure analysis. We also orally administered fluorescent microspheres and bioparticles ( Vibrio parahaemolyticus and WSSV) of varying sizes to shrimp to determine their passage under hypoxia stress. The results showed that the PSC could be divided into four segments based on structural changes, and the GS performed its filtering function through two kinds of setae with different morphologies. Under normal dissolved oxygen levels (approximately 7.6 mg/L), V. parahaemolyticus and 1 μm fluorescent microspheres were unable to enter the hepatopancreas via GS. In contrast, under hypoxia stress (1.5–2 mg/L), both vibrio and 1 μm fluorescent microspheres could enter the hepatopancreas via GS in a short timeframe. WSSV and 0.1 μm fluorescent microspheres could enter the hepatopancreas via GS irrespective of hypoxia stress, while 5 μm fluorescent microspheres could not enter the hepatopancreas via GS even under hypoxia. These results suggest that GS functions as a particle diameter-dependent filter, with its filtering function influenced by hypoxia stress. Pathogens around 1 μm in size can invade the hepatopancreas through this pathway under hypoxia stress, providing novel insights into the mechanisms of pathogen invasion in shrimp.

Glutamate enhances shrimp antibacterial ability by boosting its cellular immunity

Lei Ma,Wenyong Jiang,Zhiyao Lu,Yanmei Tong,Qingyun Liu,Zhonghua He,Xiuli Chen,Yongzhen Zhao,Fan Wang

Glutamate has been widely used in the feed industry to promote animal growth and enhance meat quality. However, its application in crustacean aquaculture remains relatively underexplored. In this study, we demonstrate a correlation between plasma glutamate increased the total hemocyte counts in shrimp within two hours, an effect that was reversed by co-injection with a Grik2 inhibitor. Subsequent feeding trials showed that incorporating 0.25 % glutamate into shrimp diets significantly raised the glutamate concentration in muscle tissue, resulting in a 7 % increase in glutamate content. Moreover, feeding experiments combined with differential transcriptome analysis revealed that shrimp fed a diet supplemented with 0.25 % glutamate exhibited enhanced resistance to Vibrio infections, likely through the upregulation of immune-related genes. These findings suggest that glutamate not only improves the anti- Vibrio defenses of shrimp but also enhances their flavor profile. Therefore, glutamate could serve as a beneficial feed supplement to promote disease resistance in shrimp aquaculture.

Clostridium autoethanogenum protein substitution and bile acids addition altered intestinal health and transcriptome profiles of hepatopancreas in Litopenaeus vannamei

Jia Xu,Chaozhong Zheng,Shuyan Chi,Shuang Zhang,Junming Cao,Beiping Tan,Shiwei Xie

Dietary Clostridium autoethanogenum protein (CAP) has the potential to replace fishmeal (FM), but excessive CAP substitutions will affect gut health. Bile acids (BAs) have multiple benefits, but little information is available on the effects of BAs on Pacific white shrimp ( Litopenaeus vannamei ) fed a CAP diet (CAP replacing FM). An 8-week feeding trial were used to investigate the impacts of CAP substitution and BAs addition on structural changes of intestine, apoptosis, inflammatory cytokine and NFκB pathway of intestine, and transcriptomic profiles of hepatopancreas in Pacific white shrimp. Compared to PC (control) treatment, the NC (CAP50, CAP replacing 50% FM) treatment significantly decreased the fold height (FH) and villus length (VL) of midgut, and impaired the mitochondrial cristae. Compared to NC group, the FH was increased in BA1 (CAP50 with 0.03% BAs) and BA2 (CAP50 with 0.06% BAs) groups, the muscle layer thickness (MT) was decreased in BA2 and BA3 (CAP50 with 0.09% BAs) groups, the VL was increased in BA1 group, and the mitochondrial cristae was normal and well-defined in BA1 and BA2 groups (all p  < 0.05). The NC increased the genes expression of apoptosis and NFκB signalling pathway, while the BA2 decreased the genes expression of apoptosis, inflammatory cytokine and NFκB signalling pathway. In transcriptomics analysis, a total of 171 differential expression genes (DEGs) were identified in NC group and 50 DEGs in BA2 group. There was 13 DEGs was observed both in NC and BA2 groups. The 5 KEGG pathways were statistically enriched in all treatments, including “Carbon metabolism”, “One carbon pool by folate”, “Glycine, serine and threonine metabolism”, “Amino sugar and nucleotide sugar metabolism”, and “PPAR signaling pathway”. In conclusion, CAP50 substitution impaired the intestinal health and 0.06% BAs addition improved it, both treatments altered the transcriptomics profiles of hepatopancreas in Pacific white shrimp.

New insight into the molecular basis of chromium exposure of Litopenaeus vannamei by transcriptome analysis

Lefei Jiao,Tianmeng Dai,Tinglan Cao,Min Jin,Peng Sun,Qicun Zhou

Heavy metal pollution arising from agricultural and industrial activities poses a significant threat to the aquatic environment, especially the increasing levels of chromium (Cr) that is exacerbating marine pollution. Given the economic importance of the Pacific white shrimp Litopenaeus vannamei ( L. vannamei ), understanding the impact of marine Cr pollution is deemed to be significant. In this study, we used the transcriptome sequencing (RNA-seq) technique to characterize the molecular mechanism of Cr exposure in L. vannamei . Gene ontology enrichment analysis showed substrate-specific and ion transport-related functions were mainly influenced by Cr exposure. We further identified genes involved in protein digestion and absorption (PEPT1, BAT1, MDU1), chemical carcinogenesis (GST and UGTs), ABC transporters (ABCC2), apoptosis (CAPN1, CASP10, PARP), implying the potentially Cr disintoxication mechanisms in L. vannamei . Genes within pancreatic secretion (ALT, LDH), lysosome (CTSL and HEXB), and peroxisome (ACOX1, ECI2, NUDT12) pathways implied the potentially Cr toxicity mechanisms in L. vannamei .

Effect of glycine supplementation on growth performance, body composition and salinity stress of juvenile Pacific white shrimp, Litopenaeus vannamei fed low fishmeal diet

Shi-wei Xie,Li-xia Tian,Yan Jin,Hui-jun Yang,Gui-ying Liang,Yong-jian Liu

An 8-week feeding trial was conducted to evaluate the optimum dietary glycine level in low fishmeal diet for juvenile Pacific white shrimp , Litopenaeus vannamei . Six practical diets (44.4% protein, 8.8% lipid) were formulated to contain graded levels (2.26, 2.33, 2.44, 2.58, 2.67 and 2.74%) of l -glycine. Each diet was randomly assigned to triplicate groups of 30 shrimps (approximately 0.61   g) and the shrimps were fed 4 times a day to apparent satiation. The results indicated that the weight gain and specific growth rate were significantly increased with the supplementation of glycine. The highest weight gain and specific growth rate were observed at 2.56% dietary glycine, and then decrease with a further increase in dietary glycine. Hepatosomatic index , protein productive value and condition factor were also significantly influenced by the dietary glycine levels and show a similar trend to those of growth performance. There were no significant differences observed in feed efficiency , protein efficiency ratio and survival among all treatments. Mg, Ca and Fe concentrations in whole body were significantly increased with the supplementation of glycine. Non-essential amino acid, essential amino acid and arginine ratio in muscle were significantly affected by the dietary glycine levels. Dietary glycine levels higher than 2.44% significantly increased the survival of shrimps after 2   h and 3   h of acute salinity change, and T-SOD activity and Na + /K + -ATPase activity in hematological and hepatopancreas before and after acute salinity change are also significantly affected by the dietary glycine levels. Analysis by second-order regression of weight gain indicated that the optimal dietary glycine level was 2.54% of the dry diet (5.7% of the dietary protein and 6.66% of the dietary amino acids).

Dietary effects of succinic acid on the growth, digestive enzymes, immune response and resistance to ammonia stress of Litopenaeus vannamei

Yafei Duan,Yun Wang,Jiasong Zhang,Yongxu Sun,Jun Wang

Organic acids acts as an growth promoter and antimicrobial agent in aquaculture. The present study investigated the effects of a natural organic acid - succinic acid (SA) on the growth, digestive enzymes , immune response and resistance to ammonia stress of Litopenaeus vannamei . The shrimps were firstly fed with diets containing different levels of SA: 0% (Control), 0.25% (SA1), 0.50% (SA2), and 1.0% (SA3) ( w / w ) for 56 days, followed by an acute ammonia stress for 48 h. The results indicated that dietary of SA improved the growth of shrimp, and increased the survival rate of shrimp after ammonia stress for 48 h. The amylase , lipase and pepsin activity increased in hepatopancreas in three SA group, while trypsin activity was only increased in the SA1 and SA2 groups. At 56 d, T-NOS activity, proPO and HSP70 gene expression level increased in the three SA group, PO activity increased in the SA1 and SA2 groups, T-AOC content and Toll gene expression level increased in the SA2 and SA3 groups, Trx and SOD gene expression level increased in the SA2 group, while Imd , GS and GDH gene expression level was no changes. After exposure to ammonia stress for 48 h, immune biochemical parameters (T-AOC and PO) and genes ( proPO , HSP70 , Trx and GDH ) expression level increased in the three SA group, T-NOS activity, Toll , Imd and GS gene expression level increased in the SA2 and SA3 groups, while SOD gene expression level increased in the SA1 and SA2 groups. These results indicated that SA improved growth, enhanced digestive and immune capacities of L. vannamei against ammonia stress, and may be a potential feed additive for shrimp. The optimal dietary supplementation dosage is 0.50% ( w / w ) in diet.

Cultured shrimp Penaeus vanmamei from eastern China accumulates PAHs and PYRs through different pathways: Water and food sources matter

Juan-ying Li,Yuxiao Tong,Qi Gong,Yaxiong Tian,Yanhao Yang,Luting Wen,Hongjie Chen,Qian Wang

Eastern China is an important area for aquaculture, but the aquaculture environment, as well as the aquatic product quality, are threatened by environmental pollutants such as polycyclic aromatic hydrocarbon (PAHs) and pyrethroid pesticides (PYRs). A three-month monitoring program for PAHs and PYRs throughout the whole culture processes of the shrimp Penaeus vanmamei was carried out. Specifically, concentrations of PAHs and PYRs ranged from 339 ± 19.5–631 ± 75.6 ng·L −1 and 65.3 ± 6.74–93.5 ± 10.8 ng·L −1 in water, 183 ± 33.8–369 ± 25.1 ng·g −1 dw and 441 ± 132–521 ± 62.8 ng·g −1 dw in sediments, while 416 ± 84.3–646 ± 19.7 and 483 ± 23.3–513 ± 22.1 ng·g −1 dw in suspended solids. Mean concentrations of PAHs and PYRs in the adult shrimp were 634 ± 82.3 ng·g −1 ww and 665 ± 58.1 ng·g −1 ww, respectively. A health threat would arise if men consume more than 28.8 g and women consume more than 24.4 g each day. PAHs in the aquaculture environment were generally from the influent water and rainfall, while PYRs were mainly introduced by feed. After entering the culture environment, PAHs primarily increased the water load, leading to more significant bioaccumulation of the shrimp from the water phase (72–95 %). For PYRs, the main sources were feed (44.4 %) and influent water (20.6 %), and they were accumulated in shrimp through feed assimilation directly (15.0 %–32.0 %). Additionally, feed waste containing PYRs increased the contamination burden of sediments and suspended solids, leading to bioaccumulation through assimilation (4.00 %–15.0 %) as well. Although the contribution of influent water to PYRs in the culture environment was lower than that of feed, the contribution of aqueous exposure to the enrichment of PYRs in the shrimp was higher (53.0 %–81.0 %) than that of food (feed, sediments, and suspended solid) intake (19.0 %–47.0 %). The hydrophobicity of PYRs led to an easier bioaccumulation from freely dissolved phase to biota. Therefore, it is important to pay attention to the water quality during the whole culture period, as well as feed quality, to ultimately improve the quality of aquaculture products. In summary, different pollutants in the aquaculture environment and products were from different sources and exposure pathways. Water quality monitoring and improvement are more efficient in decreasing PAHs in the shrimp, while PYRs reduction should be assisted with feed quality control.

Effect of temperature fluctuation on the freshness, water migration and quality of cold-storage Penaeus vannamei

Shouchun Liu,Luyao Zhang,Zhuyi Li,Meijiao Liu,Jing Chen,Pengzhi Hong,Saiyi Zhong,Jiamin Huang

This study aimed to simulate cold chain sales temperatures to predict the effects of temperature fluctuations on the physicochemical properties, moisture distribution, and flavor of vacuum-packed Penaeus vannamei . The results showed that the increase in freeze-thaw cycles, the pH, total volatile base nitrogen (TVB-N) and total plate count (TPC) of shrimp increased significantly. Meanwhile, sensory evaluation scores, hardness, elasticity and chewiness decreased. Low-frequency nuclear magnetic resonance (LF-NMR) analysis showed that the water content of shrimp continued to decline during storage as the immobilized water balance fell and the free water ratio increased in the F-T groups. In addition, W1S, W1W, W2S, and W6S in the electronic nose, which were characteristic sensors of whole shrimp odor, could distinguish samples with different sensory freshness, TPC, and TVB-N contents and rapidly detect the occurrence of shrimp quality deterioration. This study provided an experimental basis for the quality control of Penaeus vannamei during storage, transportation, and sales. It can further provide a reference for studying the cold chain preservation of aquatic products.

Effect of Freeze–Thaw Cycles on the Freshness of Prepackaged Penaeus vannamei

Shouchun Liu,Luyao Zhang,Jing Chen,Zhuyi Li,Meijiao Liu,Pengzhi Hong,Saiyi Zhong,Haifeng Li

The effect of temperature fluctuations on the freshness of shrimp in simulated trays was investigated by setting a freeze–thaw (F-T) cycle of 12 h after freezing at −20 °C and thawing at 1 °C under refrigeration. The results showed that the shrimp’s physicochemical properties deteriorated to different extents with the increase in F-T cycles. The total colony count of shrimp was 6.07 lg CFU/g after 21 cycles, and the volatile saline nitrogen content reached 30.36 mg/100 g, which exceeded the edible standard. In addition, the sensory quality and textural properties (hardness, elasticity, chewiness, and adhesion) declined to different degrees with increased F-T cycles. LF-NMR and protein property measurements showed that F-T cycles resulted in reduced water holding capacity and protein denaturation, which were the main factors leading to the deterioration of shrimp quality. Furthermore, flavor changes were analyzed using an electronic nose sensor to establish a freshness model. The W1W, W1S, W2S, and W5S sensors were correlated with the quality changes in shrimp and used as the main sensors for detecting the freshness ofPenaeus vannamei. As a result, to better maintain the overall freshness, temperature fluctuations should be minimized in sales and storage, and fewer than 8 F-T cycles should be performed.

Exploring the impact of nonylphenol exposure on Litopenaeus vannamei at the histological and molecular levels

Xianbin Su,Teng Li,Xiaowen Zhu,Peihua Zheng,Huakang Pan,Hui Guo

Nonylphenol (NP) is one of the common pollutants in the environment that have toxic effects on aquatic animals. Nevertheless, little is known about the possible toxicity mechanism of NP on the hepatopancreas of Litopenaeus vannamei . In the present study, the detrimental effects of NP on the hepatopancreas of the L. vannamei were explored at the histological and transcriptomic levels. The findings indicated that after NP exposed for 3, 12, and 48 h, the hepatopancreas histology was changed significantly. Transcriptomic analysis showed that a total of 4302, 3651, and 4830 differentially expressed genes (DEGs) were identified at 3, 12, and 48 h following NP exposure. All these DEGs were classified into 12 clusters according to the expression patterns at different time points. GO and KEGG enrichment analyses of DEGs were also performed, immunological, metabolic, and inflammatory related pathways, including arachidonic acid metabolism (ko00590), the PPAR signaling pathway (ko03320), and the regulation of TRP channels by inflammatory mediators (ko04750) were significantly enriched. Six DEGs were selected for validation by quantitative real-time PCR (qRT-PCR) and the results confirmed the reliability of transcriptome data. All results indicated that NP is toxic to L. vannamei by damaging the histopathological structure and disrupting the biological function. The findings would provide a theoretical framework for lowering or limiting the detrimental impacts of NP on aquaculture and help us to further study the molecular toxicity of NP in crustaceans.
