Photo-and thermo-regulation by photonic crystals for extended longevity of C. elegans

Shuzhen Yu,Yating Shao,Qineng Qiu,Yu Cheng,Renkun Qing,Cai-Feng Wang,Su Chen,Chen Xu

Methods allowing light energy to be modulated in a controllable fashion are potentially important for finding the correlation between light-related environmental factors and aging-related lifespan. Here, we report photo- and thermo-regulation based on photonic crystals (PCs) for extended longevity of C. elegans . We show that PCs can function as a regulator of visible spectrum to tune photonic energy received by C.elegans . We provide direct evidence that lifespan depends on photonic energy, and the use of PCs reflecting blue light (440–537 nm) gives 8.3 % increasement in lifespan. We demonstrate that the exposure to modulated light alleviates photo-oxidative stress and unfolded-protein response. We realize reflective passive cooling temperature using PCs, and favorable low temperature could be created for worms to extend lifespan. This work offers a new path based on PCs to resist negative effects light and temperature for longevity, provides an available platform for studying the role of light in aging.

Paroxetine promotes longevity via ser-7-dop-4-IIS axis in Caenorhabditis elegans

Zhou Yiming,Chen Lijuan,Wang Meijing,Yang Yang,Hu Bin,Li Guolin,Wei Fang

Paroxetine, a selective serotonin reuptake inhibitor, is widely used in the clinical treatment of depression. While several antidepressants show promise as geroprotectors, the role of paroxetine in aging remains unclear. In this study, we evaluated the lifespan extension effect of paroxetine in Caenorhabditis elegans ( C. elegans ) and elucidated the underlying mechanisms. The results showed that paroxetine can prolong lifespan concomitant extension of healthspan as indicated by increasing mobility and reducing lipofuscin accumulation, as well as confer protection to nematodes against different abiotic stresses. Paroxetine upregulated ser-7 expression and downregulated dop-4 expression. dop-4 RNA interference (RNAi) mimicked the beneficial effect of paroxetine on lifespan. Conversely, ser-7 RNAi abolished paroxetine-induced lifespan extension and the expression changes of dop-4 and genes related to insulin/insulin-like growth factor 1 signaling (IIS). Moreover, paroxetine exhibited a comparable lifespan extension effect to that observed in daf-2 or age-1 mutants; however, this effect was abolished in daf-16 mutant. Taken together, these results suggest that paroxetine promotes health and longevity in C. elegans through the ser-7 - dop-4 -IIS pathway, underscoring its potential as a geroprotector.

Immobilization of Superoxide Dismutase in Mesoporous Silica and its Applications in Strengthening the Lifespan and Healthspan of Caenorhabditis elegans

Yang Yiling,Wang Wenbin,Liu Kefeng,Zhao Jie

Senescence is a major inductive factor of aging-related diseases in connection with an accumulation of reactive oxygen species (ROS). Therefore, it is important to maintain ROS at an appropriate level to keep homeostasis in organisms. Superoxide dismutase (SOD) is a vital enzyme in defending against oxidative damage in vivo. Because of the defects in the direct application of SOD and SOD mimics, mounting delivery systems have been developed for the efficient applications of SOD to realize antioxidant treatment. Among these systems, mesoporous silica nanoparticles (MSNs) have been widely studied because of various advantages such as desirable stability, low toxicity, and adjustable particle sizes. Herein, SOD was immobilized on MSNs using a physical absorption strategy to construct the nanosystem SOD@MSN. The nematode Caenorhabditis elegans (C. elegans) was selected as the model organism for the subsequent antioxidant and anti-aging studies. The research results suggested the nanosystem could not only be effectively internalized by C. elegans but could also protect the nematode against external stress, thus extending the lifespan and healthspan of C. elegans. Therefore, SOD@MSN could be applied as a promising medicine in anti-aging therapeutics.

A Mechanistic Analysis of Metformin's Biphasic Effects on Lifespan and Healthspan in C. elegans: Elixir in Youth, Poison in Elder

Zhen-Zhen Yu,Jia-Jun Tu,Mei-Ling Ou,Jin-Xiong Cen,Kun Xue,Shao-Jun Li,Jing Zhou,Guo-Dong Lu

Aging, a complex biological process influenced by genetic, environmental, and pharmacological factors, presents a significant challenge in understanding its underlying mechanisms. In this study, we explored the divergent impacts of metformin treatment on the lifespan and healthspan of young and old C. elegans , demonstrating a intriguing “elixir in youth, poison in elder” phenomenon. By scrutinizing the gene expression changes in response to metformin in young (day 1 of adulthood) and old (days 8) groups, we identified nhr-57 and C46G7.1 as potential modulators of age-specific responses. Notably, nhr-57 and C46G7.1 exhibit contrasting regulation patterns, being up-regulated in young worms but down-regulated in old counterparts following metformin treatment. Functional studies employing knockdown approaches targeting nhr-57 , a gene under the control of hif-1 with a documented protective function against pore-forming toxins in C. elegans , and C46G7.1 , unveiled their critical roles in modulating lifespan and healthspan, as well as in mediating the biphasic effects of metformin. Furthermore, deletion of hif-1 retarded the influence of metformin, implicating the involvement of hif-1 / nhr-57 in age-specific drug responses. These findings underscored the necessity of deciphering the mechanisms governing age-related susceptibility to pharmacological agents to tailor interventions for promoting successful aging.

Polysaccharide from Ganoderma atrum delay senescence and enhance stress resistance through modulating IIS and MAPK pathway in Caenorhabditis elegans

Jiayan Xie,Mengni Shu,Xinyi Chen,Jianhua Xie,Yi Chen,Xiaobo Hu,Chunchao Nie,Danyan Liu,Qiang Yu

Ganoderma atrum is an edible and medicinal fungi in oriental countries with various bioactivities, such as antioxidant, anti-inflammatory and anti-aging properties. The study was to evaluate the effects of polysaccharide from Ganoderma atrum (PSG) on lifespan, healthspan (locomotion, pumping, lipofuscin, ROS, chemotaxis and intestinal permeability) and defense system (stress resistance, antioxidant enzymes activities), as well as to investigate its anti-aging mechanism in Caenorhabditis elegans . The findings indicated that PSG prolonged the lifespan (optimal concentration at 400 μg/mL, with an increased average lifespan by 23.10 %) and improved senescence-related physiological indicators of worms. Furthermore, the lipid accumulation was reduced, while stress resistance and antioxidant enzymes activities were enhanced after PSG intervention. Meanwhile, studies with mutants demonstrated that DAF-16/FOXO and SKN-1/Nrf-2 were required for lifespan-extension, which might be attributed to the IIS and MAPK pathways. Overall, these demonstrated that PSG was a potential dietary supplement to promote health and ameliorate aging-associated pathologies.

Kaempferol Extends Male Lifespan Under Blue Light Irradiation in Drosophila

Qimeng He,Hui Jin,Jie Shen

Short-wavelength blue light is ubiquitous in daily life and has a lasting destructive influence. Its potential harm to biological health is significant. This study used Drosophila as a model organism to investigate the protective effects of kaempferol, a flavonoid, against the toxicity of blue light. It also examined its physiological effects on Drosophila under blue light irradiation. In this experiment, fruit flies were fed with three different concentrations of kaempferol solutions (0.1, 0.01, and 0.001 mol/L) dissolved in food. The survival rate and physiological indexes of Drosophila were investigated under blue light irradiation of 2500 lux. The results showed that 0.1 mol/L kaempferol increased the activity of male flies during the day and significantly extended the male survival time under blue light irradiation. However, the study found that kaempferol did not significantly prolong the survival time of Drosophila in the oxidative stress experiment, and no significant difference was observed in the feeding experiment. In summary, our research found that kaempferol, at the concentration of 0.1 mol/L, has a protective effect on Drosophila under blue light irradiation, potentially achieved through alterations in circadian rhythm. Summary Kaempferol protects Drosophila from blue light damage. The effect of kaempferol on Drosophila exhibits sex-specific responses. Kaempferol enhances daytime activity and extends the lifespan of male Drosophila .

Enhancing the operational stability of OLED devices through the utilization of deuterated TTU host materials

Cangyu Wang,Xiaoge Song,Sinyeong Jung,Wansi Li,Maggie Ng,Season Si Chen,Man-Chung Tang

Recent research on anthracene-based hydrocarbon (PNA) as the emissive layer host found that deuteration mediated the formation of these adduct species, presumably quenchers, and thus improved the device lifetime. Meanwhile, studies on deuterium in PNA host materials with stepwise deuteration are scarce. In this work, we report a systematic study of isotopic effects through independent deuteration of the phenyl or/and the dibenzofuran unit on a series of PNA materials polyaromatic dibenzofuran ( PAF , PAF- d 5 , PAF- d 7 and PAF- d 12 ). We fabricated fluorescent OLED based on this triplet-triplet up-conversion active host materials for conventional MR-TADF emitter. A short-lived excited delayed lifetime of less than 2 μs has been observed in their electroluminescence decay studies. In addition, LT 80 of up to 12.7 h has been observed in PAF- d 12 at the brightness of 1000 cd m −2 , corresponding to a 4-fold enhancement of OLED stability compared to the non-deuterated counterpart. We hope the current research can provide insights for further device improvement.

Synthesis and luminescence properties of novel red phosphors LiRGe2O6:Mn4+ (R = Al or Ga)

Renping Cao,Wenjie Luo,Qingqiang Xiong,Aihui Liang,Shenhua Jiang,Yongchun Xu

LiRGe 2 O 6 :xMn 4+ (R = Al or Ga) (0 ≤ x ≤ 1.0 mol%) phosphors are synthesized by solid-state reaction method in air, and their crystal structures, luminescence properties, and fluorescence lifetimes are investigated. Their photoluminescence (PL) bands peaking at ∼ 670 nm owing to the 2 E →  4 A 2 transition of Mn 4+ ion cover the region from 620 to 750 nm (λex = 289 nm), and photoluminescence excitation (PLE) bands (λem ≈ 670 nm) are observed in the range of 200–550 nm. With excitation 289 nm, PL intensity of LiAlGe 2 O 6 :0.6%Mn 4+ phosphor is far stronger than that of LiGaGe 2 O 6 :0.6%Mn 4+ phosphor owing to the influence of crystal field. The optimal Mn 4+ ions concentration is ∼0.6 mol%. The fluorescence lifetime and quantum efficiency of LiAlGe 2 O 6 :0.6%Mn 4+ and LiGaGe 2 O 6 :0.6%Mn 4+ phosphors are ∼1.55 ms, 68.5 μs, 32%, and 15%, respectively. Luminous mechanism, the temperature quenching mechanism, and the influence of crystal field to luminous properties are analyzed and discussed, respectively. The experiment results are helpful to develop other novel Mn 4+ -doped phosphors.

Ginsenoside Prolongs the Lifespan of C. elegans via Lipid Metabolism and Activating the Stress Response Signaling Pathway

Xiaoxuan Yu,Hui Li,Dongfa Lin,Weizhuo Guo,Zhihao Xu,Liping Wang,Shuwen Guan

Panax ginsengis a valuable traditional Chinese medicine in Northeast China. Ginsenoside, the active component of ginseng, has not been investigated much for its effects on aging and its underlying mechanism(s) of action. Here, we investigated the effects of total ginsenoside (TG), a mixture of the primary active ginsenosides fromPanax ginseng, on the lifespan ofCaenorhabditis elegans(C. elegans). We found that TG extended the lifespan ofC. elegansand reduced lipofuscin accumulation. Moreover, TG increased the survival ofC. elegansin response to heat and oxidative stress via the reduction of ROS. Next, we used RNA-seq to fully define the antiaging mechanism(s) of TG. The KEGG pathway analysis showed that TG can prolong the lifespan and is involved in the longevity regulating pathway. qPCR showed that TG upregulated the expression ofnrh-80,daf-12,daf-16,hsf-1and their downstream genes. TG also reduced the fat accumulation and promoted lipid metabolism. Moreover, TG failed to extend the lifespan ofdaf-16andhsf-1mutants, highlighting their role in the antiaging effects of TG inC. elegans. The four main constitution of TG were then confirmed by HPLC and included ginsenoside Re, Rg1, Rg2and Rd. Of the ginsenosides, only ginsenoside Rd prolonged the lifespan ofC. elegansto levels comparable to TG. These findings provided mechanistic insight into the antiaging effects of ginsenoside inC. elegans. Keywords:Panax ginseng;lifespan;lipid metabolism;stress-resistant;Caenorhabditis elegans

An Investigation into the Performance and Mechanisms of Soymilk-Sized Handmade Xuan Paper at Different Concentrations of Soymilk

Chunfang Wu,Yangyang Liu,Yanxiao Hu,Ming Ding,Xiang Cui,Yixin Liu,Peng Liu,Hongbin Zhang,Yuliang Yang,Hongdong Zhang

Invaluable paper relics that embody a rich traditional culture have suffered damage, requiring urgent restoration. In this context, the utilization of soymilk as a sizing agent holds great significance and reverence. This study investigates the use of soymilk as a sizing agent for Xuan paper and evaluates its effects on various properties and the long-term behavior of the paper. The findings reveal that the application of soymilk as a sizing agent for Xuan paper imparts distinct properties, including hydrophobicity, improved mechanical properties, and unique chromaticity. These characteristics—arising from the papillae on the surface of the Xuan paper, the protein folding of the soy protein, and hydrogen-bonding interactions between the soy protein and paper fibers—play a crucial role in shaping the paper’s unique attributes. From a physicochemical perspective, the aging process leads to multiple changes in paper properties. These changes include acidification, which refers to a decrease in pH, as well as a decline in mechanical strength, an increase in chromaticity, and a decrease in the degree of polymerization (DP) of the paper. The Ekenstam equation is employed to predict the lifespan of the paper, showing longer lifespans for Sheng Xuan paper and a negative correlation between soymilk concentration and lifespan in soymilk-sized paper. Our work provides valuable insights for the preservation and maintenance of paper, highlighting the potential benefits and challenges of using soymilk for surface sizing. Keywords:surface sizing;soymilk;Xuan paper;lifespan;potential mechanism

Bisphenol S decreased lifespan and healthspan via insulin/IGF-1-like signaling-against mitochondrial stress in Caenorhabditis elegans

Wenmiao He,Zhiwei Liu,Hongchao Zhang,Qian Liu,Zhenkun Weng,Dongmei Wang,Wenhui Guo,Jin Xu,Dayong Wang,Zhaoyan Jiang,Aihua Gu

Bisphenol S (BPS) is widely presented and affects aging with unclear mechanisms. Here, we applied C. elegans to evaluate the effects of BPS on lifespan and healthspan and to investigate the underlying mechanisms. Both early-life and whole-life exposure to BPS at environmentally relevant doses (0.6, 6, 60 μg/L) significantly decreased lifespan, and healthspan (body bend, pharyngeal pumping, and lipofuscin accumulation). BPS exposure impaired mitochondrial structure and function, which promoted ROS production to induce oxidative stress. Furthermore, BPS increased expressions of the insulin/IGF-like signaling (IIS). Also, BPS inhibited expression of the IIS transcription factor daf-16 and its downstream anti-oxidative genes. Quercetin effectively improved BPS-induced oxidative stress byreversing BPS-regulated IIS/ daf-16 pathway and anti-oxidative gene expressions. In daf-2 and daf-16 mutants, the effects of BPS and quercetin on lifespan, healthspan, oxidative stress, and anti-oxidative genes expressions were reversed, demonstrating the requirement of IIS/ daf-16 for aging regulation. Molecular docking and molecular dynamics simulations confirmed the stable interaction between DAF-2 and BPS mainly via three residues (VAL1260, GLU1329, and MET1395), which was attenuated by quercetin. Our results highlighted that adverse effects of BPS on impairing lifespan and healthspan by affecting IIS/ daf-16 function against mitochondrial stress, which could be inhibited by quercetin treatment. Thus, we first revealed the underlying mechanisms of BPS-induced aging and the potential treatment.

Revealing the chemistry of an anode-passivating electrolyte salt for high rate and stable sodium metal batteries

Lina Gao,Juner Chen,Yaqin Liu,Yusuke Yamauchi,Zhenguo Huang,Xueqian Kong

Stabilizing the reactive metal anode is a critical challenge for the development of next-generation alkali metal batteries. This work demonstrates that sodium-difluoro(oxalato)borate (NaDFOB)-based carbonate-ester electrolytes have excellent compatibility with the anode in sodium metal batteries, enabling high rate performance and long cycle life. The NaDFOB-based electrolytes possess favourable electrochemical stability and effectively passivate the Na metal anode by forming a compact, robust and conductive solid-electrolyte interphase (SEI) layer. Quantitative nuclear magnetic resonance (NMR) measurements of electrolyte solvents indicated that the SEI layer forms mainly in the initial cycles, and it prevents further solvent degradation. Solid-state NMR and X-ray photoelectron spectroscopy studies revealed the chemical composition of the NaDFOB-derived SEI film, which includes a mixed phase consisting primarily of sodium diborate, tetrafluoroborate and carbonate. The formation of such a composite SEI rich in borate and tetrafluoroborate provides robust structural and chemical stability, and facilitates fast ion transport for uniform Na stripping/plating.
