Fusarium oxysporumis a widespread soil-borne fungal pathogen that can infect various plants, causing wilt and root rot diseases. The root rot disease ofAtractylodes macrocephalacaused byF. oxysporumis among the most serious diseases associated with continuous cropping, significantly hindering its sustainable development. In this study, we aimed to investigate the effect of exogenous application of double-stranded RNA (dsRNA) on silencing theF. oxysporum Tup1gene to reduce its virulence and to evaluate its potential application in controlling root rot disease inA. macrocephala. TheTup1gene was amplified from theF. oxysporumgenome, and different lengths ofTup1-dsRNA were designed and synthesized. The uptake of dsRNA by the fungus was verified usingTup1-dsRNA labeled with fluorescein, and in vitro dsRNA treatment experiments were conducted to assess its impact on the growth and virulence ofF. oxysporum. Additionally,Tup1-dsRNA was applied to the roots ofA. macrocephalato evaluate its effectiveness in controlling root rot disease. The experimental results showed thatF. oxysporumcould effectively uptake exogenously appliedTup1-dsRNA, significantly reducingTup1gene expression. All lengths ofTup1-dsRNA inhibited fungal growth and caused morphological changes in the fungal hyphae. Further plant experiments and Reverse Transcription Quantitative Polymerase Chain Reaction (RT-qPCR) analysis indicated thatTup1-dsRNA treatment significantly reduced the incidence of root rot disease inA. macrocephala, which was supported by the reduction in peroxidase (POD) and catalase (CAT) enzyme activities, malondialdehyde (MDA) content, and proline (Pro) levels in treated root tissues. This study demonstrated that exogenous dsRNA could reduce the virulence ofF. oxysporumby silencing theTup1gene and effectively mitigate the root rot disease it causes inA. macrocephala. The successful application ofTup1-dsRNA provided strong evidence for the potential of RNA interference (RNAi) technology in plant disease control. Future research could further optimize the design and application of dsRNA to enhance its practical value in agriculture.

Panama disease, or Fusarium wilt, the most serious disease in banana cultivation, is caused byFusarium oxysporum f.sp.cubense(FOC) and has led to great economic losses worldwide. One effective way to combat this disease is by enhancing host plant resistance. The cerato-platanin protein (CPP) family is a group of small secreted cysteine-rich proteins in filamentous fungi. CPPs as elicitors can trigger the immune system resulting in defense responses in plants. In this study, we characterized a novel cerato-platanin-like protein in the secretome ofFusarium oxysporum f.sp.cubenserace 4 (FOC4), named FocCP1. In tobacco, the purified recombinant FocCP1 protein caused accumulation of reactive oxygen species (ROS), formation of necrotic reaction, deposition of callose, expression of defense-related genes, and accumulation of salicylic acid (SA) and jasmonic acid (JA) in tobacco. These results indicated that FocCP1 triggered a hypersensitive response (HR) and systemic acquired resistance (SAR) in tobacco. Furthermore, FocCP1 enhanced resistance tobacco mosaic virus (TMV) disease andPseudomonas syringaepv.tabaci6605 (Pst.6605) infection in tobacco and improved banana seedling resistance to FOC4. All results provide the possibility of further research on immune mechanisms of plant and pathogen interactions, and lay a foundation for a new biological strategy of banana wilt control in the future. Keywords:Fusarium oxysporum;cerato-platanin;elicitor;FocCP1;immune response;systemic acquired resistance

Enzymes that degrade fungal cell walls and the resulting oligosaccharides are promising weapons to combat plant fungal disease. In this study, we identified a novel endo-chitosanase, AqCoA, from Aquabacterium sp. A7-Y. The enzyme showed a specific activity of 18 U/mg toward 95% deacetylated chitosan at pH 5.0 and 40 °C. AqCoA also showed activity toward sodium carboxymethylcellulose, indicating substrate promiscuity. AqCoA hydrolyzed chitosan into chitooligosaccharides (CoA-COSs) with degrees of polymerization (DPs) of 3–5 but showed no activity toward CoA-COSs with DPs <6, indicating an endo-type activity. At 2.5 μg/mL, AqCoA inhibited appressorium formation of Magnaporthe oryzae; the produced CoA-COSs also inhibited the growth of M. oryzae and Fusarium oxysporum. Furthermore, CoA-COSs acted as immune elicitors in rice by inducing the reactive oxygen species burst and the expression of defense genes. These results demonstrated that AqCoA and its resulting CoA-COSs might be effective tools for protecting plants against pathogenic fungi.

Diseases caused by tuber fungi severely disrupt the storage life and quality of post-harvest Gastrodia elata ( G. elata ). Herein, it was found that the decay of G. elata tubers is primarily attributed to Fusarium solani and Penicillium crustosum . To inhibit postharvest rotting in G. elata tubers, microwave-treatment (MT) was employed as a disinfection method. However, they could not be completely inhibited by a single MT in vivo test, which were at various power levels (300, 500, 700 W) and durations (20, 30, 40 s). Hence, MT (700w, 30s) combined with modified atmosphere packaging (MAP) was employed, which were polyethylene films of varying thickness (20, 30, 40 μm), and its efficiency was validated. Results indicate that the combined treatment improved the quality of infected G. elata tubers. In addition, it decreased 79.81% in decay volume of G. elata tubers compared with the control group. Further results indicate that the reactive oxygen species and energy metabolism pathways of G. elata tubers were activated by the combined treatment, activating the enzyme activity and relative expression of superoxide dismutase, catalase and peroxidase and maintaining the levels of ATP, ADP and energy charge. Taken together, these results indicate that MT combined with MAP is a new and effective method for the post-harvest preservation of tubers and root crops.

The soft rot disease caused by Fusarium oxysporum leads to a significant reduction in the yield of Dendrobium officinale . However, research into the synergistic inhibitory effect of essential oils in D. officinale is extremely limited. In this study, we systematically investigated the direct and indirect inhibitory activity of carvacrol, citral and their combination against F. oxysporum , and their synergistic inhibitory mechanism. Carvacrol and citral exhibited significant direct and indirect inhibitory activity against F. oxysporum with EC 50 values of 54.37 mg/L and 119.32 mg/L (direct), 18.01 μL/(L•air) and 48.70 μL/(L•air) (indirect). Synergistic analysis revealed that the optimal synergistic toxicity of carvacrol and citral combination (Ca•Ci) against F. oxysporum was 10:1, with co-toxicity coefficient (CTC) of 131.57 and EC 50 value of 44.24 mg/L. Microscopy confirmed that the Ca•Ci led to more significant tip constriction, uneven surfaces and serious rupture of F. oxysporum mycelia than single compound. Transmission electron microscopy (TEM) showed that Ca•Ci also caused significant ultrastructural alterations to F. oxysporum , manifesting as cytoplasmic disorganization and partial organellar disintegration. Moreover, Ca•Ci dramatically increased the sensitivity of F. oxysporum to calcofluor white compared with a single compound. Ca•Ci also considerably upregulated the expression of chitinase-related gene ( FOXG _12882) and β -1,3-glucanase-related gene ( FOXG _10637) in F. oxysporum , resulting in higher chitinase and β -1,3-glucanase activity. However, it should be noted that carvacrol exerted a greater contribution than citral. In conclusion, the combination of carvacrol and citral greatly disrupted the cell wall integrity of F. oxysporum , thereby exhibiting a synergistic effect.

Phenolics from the rhizosphere soil are a key factor that leads to obstacles in the continuous cropping system. While we tested the phenolics in the rhizosphere soil of Lanzhou lily ( Lilium davidii var . unicolor ), a high relative content 4-Vinyl Guaiacol (4-VG) was firstly detected. Therefore, we attempted to explore the role of 4-VG stress in Lanzhou lily physiology and disease, and the potential enhanced effects of pathogenicity in Fusarium oxysporum B4 ( F. oxysporum B4). The effects of different concentrations of 4-VG (0, 0.01, 0.05, 0.1, 0.2, 0.5, 1 mM) on the growth of F. oxysporum B4 and the production of fusaric acid were examined. The result indicated that 4-VG inhibited the growth of F. oxysporum B4 hypha, promoted spore germination, and increased fusaric acid content. Furthermore, transcriptome sequencing of F. oxysporum B4 hypha after stress mediated by 0.5 mM 4-VG was performed to help characterize the response mechanism to 4-VG. Compared with the control group, differentially expressed genes (DEGs) were mainly related to the growth and fusaric acid synthesis of F. oxysporum B4. Except that, the increase of infection area formed after F. oxysporum B4 infected the squama of Lanzhou lily under the high concentration of 4-VG was measured and indicated that 4-VG played a role in the F. oxysporum B4 infection. The exogenous addition of 4-VG significantly inhibited the growth and health of Lanzhou lily. Under the dual stress of F. oxysporum B4 and different concentrations of 4-VG, it significantly enhanced the incidence and disease index of Lanzhou lily. These results suggest the key role of 4-VG from rhizosphere soil in the occurrence of Lanzhou lily Fusarium wilt.

Watermelon ( Citrullus lanatus ), a key global crop in the Cucurbitaceae family, is significantly impacted by wilt disease caused by Fusarium oxysporum . This study used suspension-cultured watermelon cells to examine the physiological and molecular effects of fusaric acid (FA), a major phytotoxin produced by the pathogen, at final concentrations of 0, 50, 100, 200, and 300 μM. The 100 μM FA treatment, used for most assays, reduced fresh cell weight by ∼52 %, decreased packed cell volume by ∼70 %, and more than doubled malondialdehyde (MDA) content, indicating severe lipid peroxidation. Antioxidant enzyme activities showed differential responses: superoxide dismutase (SOD) activity declined significantly at both 2 h and 24 h, peroxidase (POD) remained largely unchanged, and catalase (CAT) showed no significant variation. FA exposure also caused a progressive increase in extracellular pH and conductivity, reflecting loss of membrane integrity. Pretreatment with exogenous plant hormones (IAA, MeJA, ABA, SA; 100 μM each for 12 h) mitigated several FA-induced effects, with SA showing the greatest recovery in fresh weight and most pronounced reduction of POD activity. Transcriptome profiling revealed thousands of FA-responsive genes within 2–24 h, and inhibition of DNA methylation with 5-azacytidine altered the expression of multiple hormone-related genes. These results demonstrate that FA rapidly impairs growth, membrane stability, and antioxidant defense in watermelon cells, and that targeted hormonal and epigenetic interventions can partially counteract these effects. The findings provide a mechanistic basis for developing priming strategies to enhance watermelon resistance to Fusarium wilt.

In this paper, we synthesized amphiphilic chitosan using one-pot method with the goal to develop delivery system for the natural pesticide spinosad . Chitosan-Isoleucine-Polyethylene glycol methyl ether (CIP) copolymers based nanomicelles (CIPNMs) were prepared by self-assembly with the assistance of ultrasonic treatment and then the spinosad was loaded on the micelles . Sphere-shaped spinosad-loaded CIPNMs (SSD@CIPNMs) with the particle sizes of approximately 454.9 nm were obtained with the CIP co -polymers. The shell of CIPNMs could effectively prevent the photodegradation of spinosad and played a role of protective barrier. In addition, the prepared SSD@CIPNMs not only delayed the release of spinosad but also had a property of pH-responsive. The release rate of SSD@CIPNMs at pH = 6.4 was greater than that of at pH = 7.0. And, the contact angle of SSD@CIPNMs on banana leaves surface was 60.47°, which meant SSD@CIPNMs had a good affinity with banana leaves. Furthermore, SSD@CIPNMs showed higher antifungal activity against Fusarium oxysporum compared with the unencapsulated spinosad at the same drug dose. This work was contribution to the development of nano-drug carrier systems of natural polymer materials, with a great potential for targeted treatment of banana wilt.

Fusarium wilt is one of the major constraints on global watermelon production, and Fusarium oxysporum f. sp. niveum (Fon) is the causative agent of Fusarium wilt in watermelon and results in severe yield and quality losses worldwide. The enhancement of antifungal activity from antagonistic bacteria against Fon is highly practical for managing Fusarium wilt in watermelon. The aim of this study was to maximize the antifungal activity of Bacillus velezensis LZN01 by optimizing fermentation conditions and analysing its regulatory mechanism via transcriptome sequencing. The culture and fermentation conditions for strain LZN01 were optimized by single-factor and response surface experiments. The optimum culture conditions for this strain were as follows: the addition of D-fructose at 35 g/L and NH 4 Cl at 5 g/L in LB medium, pH 7, and incubation at 30°C for 72 h. The fungal inhibition rate for strain LZN01 reached 71.1%. The improvement of inhibition rate for strain LZN01 in optimization fermentation was supported by transcriptomic analysis; a total of 491 genes were upregulated, while 736 genes were downregulated. Transcriptome analysis revealed that some differentially expressed genes involved in carbon and nitrogen metabolism, oxidation–reduction, fatty acid and secondary metabolism; This optimization process could potentially lead to significant alterations in the production levels and types of antimicrobial compounds by the strain. Metabolomics and UPLC/Q-Exactive Orbitrap MS analysis revealed that the production yields of antimicrobial compounds, such as surfactin, fengycin, shikimic acid, and myriocin, increased or were detected in the cell-free supernatant (CFS) after the fermentation optimization process. Our results indicate that fermentation optimization enhances the antifungal activity of the LZN01 strain by influencing the expression of genes responsible for the synthesis of antimicrobial compounds. Graphical The yields of antimicrobial compounds, such as surfactin, fengycin, shikimic acid, and myriocin, increased or were observed in the optimized fermentation CFS. In conclusion, fermentation conditions improved the antifungal activity of the LZN01 strain and influenced the expression of genes involved in the synthesis of antimicrobial compounds.

GH19 (glycoside hydrolase 19) chitinases play crucial roles in the enzymatic conversion of chitin and biocontrol of phytopathogenic fungi. Herein, a novel multifunctional chitinase of GH19 (CaChi19A), which contains three chitin-binding domains (ChBDs), was successfully cloned from Chitinilyticum aquatile CSC-1 and heterologously expressed in Escherichia coli. We also generated truncated mutants of CaChi19A_ΔI, CaChi19A_ΔIΔII, and CaChi19A_CatD consisting of two ChBDs and a catalytic domain, one ChBD and a catalytic domain, and only a catalytic domain, respectively. CaChi19A, CaChi19A_ΔI, CaChi19A_ΔIΔII, and CaChi19A_CatD exhibited cold adaptation, as their relative enzyme activities at 5 °C were 40.7, 51.6, 66.2, and 82.6%, respectively. Compared with CaChi19A and other variants, CaChi19A_ΔIΔII demonstrated a higher level of stability below 50 °C and retained relatively high activity over a wide pH range of 5–12. Analysis of the hydrolysis products revealed that CaChi19A and CaChi19A_ΔIΔII exhibit exoacting, endoacting, and N-acetyl-β-d-glucosaminidase activities toward colloidal chitin. Furthermore, CaChi19A and CaChi19A_ΔIΔII exhibited inhibitory effects on the hyphal growth of Fusarium oxysporum, Fusarium redolens, Fusarium fujikuroi, Fusarium solani, and Coniothyrium diplodiella, thereby illustrating effective biocontrol activity. These results indicated that CaChi19A and CaChi19A_ΔIΔII show advantages in some applications where low temperatures were demanded in industries as well as the biocontrol of fungal diseases in agriculture.

The volatile compounds produced by plants play an important role in plant growth, plant communication, and resistance to biological and abiotic stresses.Astragalus membranaceusvar.mongholicus(AM) is a perennial herbaceous plant (Leguminosae) that is widely cultivated in northwest China. The bioactive compounds in its root have shown various pharmacological activities. Root rot disease caused byFusariumspp. often occurs inAMplanting with increasing severity in continuous monoculture. It is currently still unclear what are the effects of the volatile compounds produced by freshAMon itself, other crops cultivated on the same field afterAM, pathogen, and rhizobia. In this study, we found that seed germination and seedling growth ofAM, lettuce (Lactuca sativaL.), and wheat (Triticum aestivumL.) could be affected if they were in an enclosed space with freshAMtissue. Additionally, 90 volatile compounds were identified by SPME-GC-MS from wholeAMplant during the vegetative growth, 36 of which were specific to aerial parts ofAM(stems and leaves, AMA), 17 to roots (AMR), and 37 were found in both AMA and AMR. To further identify the allelopathic effects of these volatile compounds, five compounds (1-hexanol, (E)-2-hexenal, (E,E)-2,4-decadienal, hexanal, and eugenol) with relatively high content inAMwere tested on three receptor plants and two microorganisms. We found that (E,E)-2,4-decadienal and (E)-2-hexenal showed significant inhibitory effects on the growth ofAMand lettuce. One-hexanol and hexanal suppressed the growth of wheat, while eugenol showed a similar effect on all three plant species. Moreover, the activities of these compounds were dose dependent. Notably, we discovered that (E)-2-hexenal and eugenol also inhibited the growth of the pathogenFusarium solaniby as high as 100%. Meanwhile, all five compounds tested suppressed the rhizobiaSinorhizobium fredii. In summary, this study furthered our understanding of the comprehensive allelopathic effects of the main volatile components ofAM.

In agriculture, soil-borne fungal pathogens, especiallyFusarium oxysporumstrains, are posing a serious threat to efforts to achieve global food security. In the search for safer agrochemicals, silica nanoparticles (SiO2NPs) have recently been proposed as a new tool to alleviate pathogen damage including Fusarium wilt. Hollow mesoporous silica nanoparticles (HMSNs), a unique class of SiO2NPs, have been widely accepted as desirable carriers for pesticides. However, their roles in enhancing disease resistance in plants and the specific mechanism remain unknown. In this study, three sizes of HMSNs (19, 96, and 406 nm as HMSNs-19, HMSNs-96, and HMSNs-406, respectively) were synthesized and characterized to determine their effects on seed germination, seedling growth, andFusarium oxysporumf. sp.phaseoli(FOP) suppression. The three HMSNs exhibited no side effects on cowpea seed germination and seedling growth at concentrations ranging from 100 to 1500 mg/L. The inhibitory effects of the three HMSNs on FOP mycelial growth were very weak, showing inhibition ratios of less than 20% even at 2000 mg/L. Foliar application of HMSNs, however, was demonstrated to reduce the FOP severity in cowpea roots in a size- and concentration-dependent manner. The three HMSNs at a low concentration of 100 mg/L, as well as HMSNs-19 at a high concentration of 1000 mg/L, were observed to have little effect on alleviating the disease incidence. HMSNs-406 were most effective at a concentration of 1000 mg/L, showing an up to 40.00% decline in the disease severity with significant growth-promoting effects on cowpea plants. Moreover, foliar application of HMSNs-406 (1000 mg/L) increased the salicylic acid (SA) content in cowpea roots by 4.3-fold, as well as the expression levels of SA marker genes ofPR-1(by 1.97-fold) andPR-5(by 9.38-fold), and its receptor gene ofNPR-1(by 1.62-fold), as compared with the FOP infected control plants. Meanwhile, another resistance-related gene ofPALwas also upregulated by 8.54-fold. Three defense-responsive enzymes of POD, PAL, and PPO were also involved in the HMSNs-enhanced disease resistance in cowpea roots, with varying degrees of reduction in activity. These results provide substantial evidence that HMSNs exert their Fusarium wilt suppression in cowpea plants by activating SA-dependent SAR (systemic acquired resistance) responses rather than directly suppressing FOP growth. Overall, for the first time, our results indicate a new role of HMSNs as a potent resistance inducer to serve as a low-cost, highly efficient, safe and sustainable alternative for plant disease protection.
