Mass Transfer Study of Dehydration by Triethylene Glycol in Rotating Packed Bed for Natural Gas Processing

Shaobo Cao,Ping Liu,Liangliang Zhang,Baochang Sun,Haikui Zou,Guangwen Chu,Yong Luo,Jianfeng Chen

To ensure steady operation of gas transmission lines and meet the utilization specification of downstream processes, gas dehydration is employed in offshore natural gas production, where urgent demands for space and size exist. In this work, a rotating packed bed (RPB) was used to intensify the triethylene glycol (TEG) dehydration process and the effects of operating variables on the overall volumetric mass transfer coefficient (Kya) and height of mass transfer unit (HTU) were investigated. The value of HTU in a RPB was measured to be 4.3–7.9 mm, which was at least an order of magnitude lower than that in conventional columns, indicating the significant reduction in the size of the dehydration apparatus. The predicted Kya matches well with the experimental data, and the mean relative error (MRE) is only 6.68%.

Acid-triggered size reduction of nanomedicines for enhancing tumor therapy efficacy

Chongzhi Wang,YiKai Tang,Cheng Li,Wei Wu,Xiqun Jiang

Nanomedicines whose size can be varied on demand may synchronously achieve excellent tumor accumulation and penetration. In this study, by taking advantage of the pH sensitivity of a boronate ester, we synthesized acid-triggered size-reducing polymer nanoparticles (named PCD) by cross-linking β-cyclodextrin-cored multiarm polymers through the boronate ester. In PCD, the antitumor agent doxorubicin was loaded via the pH-sensitive hydrazone linkage. The in vitro and in vivo properties of PCD were investigated using a non-responsive nanoparticle (named UCD) as a reference. In the neutral condition of the bloodstream, PCD was stable and exhibited a suitable size for long circulation time. Upon entering tumors, PCD decomposed into stable and smaller multiarm polymers, which could deeply penetrate tumors. The high tumor accumulation and penetration of PCD offered significantly better anti-tumor efficacy than UCD.

ROS-Sensitive Polymeric Nanocarriers with Red Light-Activated Size Shrinkage for Remotely Controlled Drug Release

Ziyang Cao,Yinchu Ma,Chunyang Sun,Zidong Lu,Zeyu Yao,Junxia Wang,Dongdong Li,Youyong Yuan,Xianzhu Yang

Drug delivery systems with remotely controlled drug release capability are rather attractive options for cancer therapy. Herein, a reactive oxygen species (ROS)-sensitive polymeric nanocarrier TK-PPE@NPCe6/DOX was explored to realize remotely controlled drug release by light-activated size shrinkage. The TK-PPE@NPCe6/DOX encapsulating chlorin e6 (Ce6) and doxorubicin (DOX) was self-assembled from an innovative ROS-sensitive polymer TK-PPE with the assistance of an amphiphilic copolymer poly(ethylene glycol)-b-poly(ε-caprolactone) (PEG-b-PCL). Under the 660 nm red light irradiation, ROS generated by the encapsulated Ce6 were capable of cleaving the TK linker in situ, which resulted in the rapid degradation of the TK-PPE@NPCe6/DOX core. Consequently, the size of TK-PPE@NPCe6/DOX shrank from 154 ± 4 nm to 72 ± 3 nm, and such size shrinkage affected further triggered rapid DOX release. As evidenced by both in vitro and in vivo experiments, such ROS-sensitive polymeric nanocarriers with light-induced size shrinkage capability offer remarkable therapeutic effects in cancer treatment. This concept provides new avenues for the development of light-activated drug delivery systems for remotely controlled drug release in vivo.

Transformation of the shape and shrinking the size of acid-resistant metal–organic frameworks (MOFs) for use as the vehicle of oral proteins

Li Yang,Jun Guo,Liwen Wang,Sicheng Tang,Ai-fang Wang,Shengwu Zheng,Zhiyong Guo,Xingjie Zan

The oral delivery of protein-based drugs is of great significance, but faces various obstacles, including the deactivation of proteins by the low pH in the stomach and the high concentration of protease, poor transport through intestinal bio-barriers, etc. Herein, we present an acid-resistant metal–organic framework (MOF), NU-1000, in which insulin (Ins, a model protein) was loaded with high capacity (Ins@NU-1000) through the pseudo second-order kinetic model and Langmuir isotherm model. Ins@NU-1000 protects Ins from deactivation in the stomach acid environment and releases it in the intestine through the transformation of the micro-sized rod particles into spherical nanoparticles. Interestingly, the rod particles exhibit long-term retention in the intestine, and Ins is efficiently transported by the shrunk nanoparticles through intestinal bio-barriers and released into the blood, resulting in significant oral hypoglycemic effects (lasting more than 16 h after a single oral administration). Our findings demonstrate that switching the physical properties of the delivery vehicle, such as the shape and size, can contribute to the success of oral protein administration.

Targeting tumor hypoxia with stimulus-responsive nanocarriers in overcoming drug resistance and monitoring anticancer efficacy

Zhiqi Xie,Wangwei Guo,Ningning Guo,Mingyi Huangfu,Huina Liu,Mengting Lin,WenHong Xu,Jiejian Chen,TianTian Wang,Qichun Wei,Min Han,Jianqing Gao

Although existing nanomedicines have focused on the tumor microenvironment with the goal of improving the effectiveness of conventional chemotherapy, the penetration of a tumor’s core still represents a formidable barrier for existing drug delivery systems. Therefore, a novel multifunctional hypoxia-induced size-shrinkable nanoparticle has been designed to increase the penetration of drugs, nucleic acids , or probes into tumors. This cooperative strategy relies on three aspects: (i) the responsiveness of nanoparticles to hypoxia, which shrink when triggered by low oxygen concentrations; (ii) the core of a nanoparticle involves an internal cavity and strong positive charges on the surface to deliver both doxorubicin and siRNA; and (iii) a reactive oxygen species (ROS) probe is incorporated in the nanoparticle to monitor its preliminary therapeutic response in real time, which is expected to realize the enhanced efficacy together with the ability to self-monitor the anticancer activity . A more effective inhibition of tumor growth was observed in tumor-bearing zebrafish, demonstrating the feasibility of this cooperative strategy for in vivo applications. This research highlights a promising value in delivering drugs, nucleic acids, or probes to a tumor’s core for cancer imaging and treatment. Statement of Significance Hypoxia-induced chemoresistance of tumor cells still represents a formidable barrier, as it is difficult for existing drug delivery systems to penetrate the tumor hypoxia core. This study involves the hypoxia-responsive size-shrinkable nanoparticle co-delivery of DOX and siRNA to enhance the penetration of DOX deep within tumors and subsequently disturb crucial pathways of cancer development induced by hypoxia and to improve sensitization to DOX chemotherapy. Furthermore, the nanopreparation can combine the ROS probe as a self-reporting nanopreparation to realize the function of real-time feedback efficacy, which has a good application prospect in the diagnosis and treatment of cancer.

Programmable prodrug micelle with size-shrinkage and charge-reversal for chemotherapy-improved IDO immunotherapy

Liangliang Dai,Xiang Li,Mengjiao Yao,Peiyun Niu,Xichen Yuan,Ke Li,Maowen Chen,Zengxiang Fu,Xianglong Duan,Haibin Liu,Kaiyong Cai,Hui Yang

IDO blockade-based immunotherapy has been impeded by the activation of antitumor immune response and low delivery efficiency of immunotherapeutic, resulting from natural biological barriers and immune resistance. Herein, a programmable drug delivery nanosystem with enhanced tumor penetration and endocytosis is constructed for chemotherapy-enhanced immunotherapy by loading immune checkpoint IDO inhibitor NLG919 in pH/redox cascade-responsive prodrug micelle . The nanosystem shrinked micelles sizes and converted charge from negative to positive for enhanced tumor penetration and endocytosis in responding to the weakly acidic tumor microenvironment . The endocytosed nanosystem dramatically disassembled and released curcumin and NLG919 in redox-rich cytoplasm. In vitro and in vivo studies demonstrate that the nanosystem not only effectively overcame biological barriers, but also significantly boosted antitumor immune response and reduced immune resistance. It was realized by the combined effects of chemotherapy-enhanced immunogenicity , and NLG919-induced IDO-blockade immunotherapy, consequently inhibiting tumor growth, metastasis and recurrence with high efficiency in vivo . The study offers a nanoplatform with deep tumor penetration, high cellular uptake and effective antitumor immune response for the advance of chemo-immunotherapy.

Dual-Stage Irradiation of Size-Switchable Albumin Nanocluster for Cascaded Tumor Enhanced Penetration and Photothermal Therapy

Peiying He,Qi Lei,Bin Yang,Tongyi Shang,Jianjun Shi,Qing Ouyang,Wei Wang,Liecong Xue,Fanhui Kong,Zeyu Li,Junda Huang,Lihan Liu,Jimin Guo,C. Jeffrey Brinker,Kaisheng Liu,Wei Zhu

The triple-negative breast cancer (TNBC) microenvironment makes a feature of aberrant vasculature, high interstitial pressure, and compact extracellular matrix, which combine to reduce the delivery and penetration of therapeutic agents, bringing about incomplete elimination of cancer cells. Herein, employing the tumor penetration strategy of size-shrinkage combined with ligand modification, we constructed a photothermal nanocluster for cascaded deep penetration in tumor parenchyma and efficient eradication of TNBC cells. In our approach, the photothermal agent indocyanine green (ICG) is laded in human serum albumin (HSA), which is cross-linked by a thermally labile azo linker (VA057) and then further modified with a tumor homing/penetrating tLyP-1 peptide (HP), resulting in a TNBC-targeting photothermal-responsive size-switchable albumin nanocluster (ICG@HSA-Azo-HP). Aided by the enhanced permeability and retention effect and guidance of HP, the ca. 149 nm nanoclusters selectively accumulate in the tumor site and then, upon mild irradiation with the 808 nm laser, disintegrate into 11 nm albumin fractions that possess enhanced intratumoral diffusion ability. Meanwhile, HP initiates the CendR pathway among the nutrient-deficient tumor cells and facilitates the transcellular delivery of the nanocluster and its disintegrated fractions for subsequent therapy. By employing this size-shrinkage and peptide-initiated transcytosis strategy, ICG@HSA-Azo-HP possesses excellent penetration capabilities and shows extensive penetration depth in three-dimensional multicellular tumor spheroids after irradiation. Moreover, with a superior photothermal conversion effect, the tumor-penetrating nanocluster can implement effective photothermal therapy throughout the tumor tissue under a second robust irradiation. Both in vivo orthotopic and ectopic TNBC therapy confirmed the efficient tumor inhibition of ICG@HSA-Azo-HP after dual-stage irradiation. The synergistic penetration strategy of on-demanded size-shrinkage and ligand guidance accompanied by clinically feasible NIR irradiation provides a promising approach for deep-penetrating TNBC therapy.

Dual-Responsive Size-Shrinking Nanocluster with Hierarchical Disassembly Capability for Improved Tumor Penetration and Therapeutic Efficacy

Xiaoyan Sun,Jiulong Zhang,Chunrong Yang,Ziyuan Huang,Menghao Shi,Shuang Pan,Haiyang Hu,Mingxi Qiao,Dawei Chen,Xiuli Zhao

It is generally known that, for nanoparticles in cancer therapy, sufficient tumor penetration needs a minor particle size, while long in vivo circulation time needs a larger particle size. It is hard to balance them because they are standing on either side of a seesaw. To address these two different requirements, a dual-responsive size-shrinking nanocluster can self-adaptively respond to a complicated tumor microenvironment and transform its particulate property to overcome sequential in vivo barriers and reach a preferable antitumor activity. The nanocluster (RPSPT@SNCs) could preferentially accumulate into tumor tissue and dissociate under extracellular matrix metalloproteinase-2 (MMP-2) to release small-sized micelle formulations (RPSPTs). RPSPT possesses favorable tumor penetration and tumor targeting capability to deliver the antitumor agent paclitaxel (PTX) into deep regions of solid tumor. The intracellular redox microenvironment can also accelerate drug accumulation. The prepared RPSPT@SNCs possesses enhanced cell cytotoxicity and tumor penetration capability on MCF-7 cells and a favorable antitumor activity on the xenograft tumor mouse model.
