Urocanic acid, like histamine, is a degradation product of histidine that may accumulate over the storage of histidine-rich pelagic fish. Recently evidence from the dermatological and related fields has directed the attention of food scientists toward the adverse health effects of urocanic acid, mostly as a possible “missing factor” responsible for scombrotoxicosis besides histamine. However, experimental effort to study the hypothesis remains scarce, especially regarding the formation of urocanic acid in comparison to the well-documented histamine. Here we report a mixed-mode HPLC method for simultaneously analyzing urocanic acid (both the trans - and cis -isomers), histamine, and histidine in chub mackerel ( Scomber japonicus ) fillets and the use of the method to monitor their changes in concentration at typical refrigeration and abuse temperatures. The method features a simple, reliable chromatographic analysis in cation-exchange/reversed-phase/hydrophilic-interaction triple mode of retention, without the need of precolumn derivatization or addition of ion-pairing agents. At both 4 and 20 °C slow but consistent formation of trans -urocanic acid was observed over time with a relatively faster rate at 20 °C. Histamine formation, by contrast, occurred at 20 °C but ceased completely at 4 °C. In addition, we demonstrate the occurrence of trans -to- cis isomerization of UCA in chub mackerel fillets upon direct sunlight exposure: under our conditions, an isomerization yield around 13 % was achieved within hours on a sunny winter day. Overall, this study provides a chemical analysis method and relevant data for exploring the possible role of urocanic acid in the aetiology of scombrotoxicosis.

Urocanic acid, deaminated histidine and the first intermediate in the histidine utilization (Hut) pathway in bacteria, has recently been suggested to have adverse health effects by inducing scombrotoxicosis and immunosuppression after trans – cis isomerization. This work aimed to shed some light on the microbiology of its formation in scombroid fish, given that such information is very scarce. The isolation of urocanic acid producers from chub mackerel was basically based on enrichment of bacteria possessing the Hut pathway with histidine as the sole source of carbon, energy, and nitrogen. Morphological and molecular identification revealed that the isolates ( n = 12), taken from the skin, gills, and intestine, were comprised entirely of Acinetobacter from at least five species. This implies that the Hut pathway in the microbes can be particularly active and as such conferred a growth advantage. Dynamic monitoring of bacterial growth and urocanic acid production in fish juice broth indicated that urocanic acid is a primary metabolite and its production is a closely growth-associated process. This in turn implies that its production can be controlled directly by suppressing the bacterial growth. Several potential methods are proposed through a study of the growth characteristics and preservative susceptibility of representative isolates. In conclusion, this work suggests that Acinetobacter can constitute a potentially important group of urocanic acid producers in chub mackerel.

Interactions between the host and the gut microbiota influence cancer progression and treatment responses. While the combination of tyrosine kinase inhibitors (TKIs) and immune checkpoint blockade (ICB) has improved outcomes, some cancer patients still have poor responses. The underlying mediators of this heterogeneity remain unclear. Here, we demonstrate that TKIs potentiate the immunotherapy response by increasing the abundance of Muribaculum and its metabolite, urocanic acid (UCA), which reduces myeloid-derived suppressor cell (MDSC) recruitment via the CXCL1-CXCR2 axis by inhibiting p65 in tumor vascular endothelial cells. Mechanistically, UCA selectively binds to the aspartic acid 31 residue of IκBα and suppresses its phosphorylation at serine 32. Compared with non-responders, clinical ICB responders present a higher UCA concentration and a greater level of Muribaculum gordoncarteri in feces, indicating both as potential predictive biomarkers for treatment response. Collectively, our findings reveal and highlight the important role of the gut microbial metabolite UCA in response to ICB.

Di-(2-ethylhexyl) phthalate (DEHP), an environmental pollutant, has been widely detected in both environmental and clinical samples, representing a serious threat to the homeostasis of the endocrine system. The accumulation of DEHP is notably pronounced in the liver and can lead to liver damage. The lack of effective high-throughput screening system retards the discovery of such drugs that can specifically target and eliminate the detrimental impact of DEHP. Here, by developing a Cy5-modified single-strand DNA-aptamer-based approach targeting DEHP, we have identified luteolin as a potential drug, which showcasing robust efficacy in detoxifying the DEHP by facilitating the expulsion of DEHP in both mouse primary hepatocytes and livers. Mechanistically, luteolin enhances the protein degradation of hepatic urocanate hydratase 1 (Uroc1) by targeting its Ala270 and Val272 sites. More importantly, trans-urocanic acid (trans-UCA), as the substrate of Uroc1, possesses properties similar to luteolin by regulating the lysosomal exocytosis through the inhibition of the ERK1/2 signal cascade. In summary, luteolin serves as a potent therapeutic agent in efficiently detoxifying DEHP in the liver by regulating the UCA/Uroc1 axis.

Urocanic acid (UA), existing in trans - or cis -isoform, is of fairly recent interest to food researchers because of its potential public health hazards of scombrotoxicity and immunotoxicity, as well as associating with fish spoilage. This work is among the first efforts to study the analytical chemistry of UA in fish. With 0.6 M perchloric acid UA was extracted, and co-extracted fish matrix components were efficiently removed through a reactive extraction of UA. The optimum conditions for the reactive extraction, which allowed an 80% recovery of UA, were sample pH adjustment to 9, twice extractions with 32% (w/w) di (2-ethylhexyl) phosphate in hexanol, and a back-extraction with 0.1 M hydrochloric acid at 1:1 phase ratio. A chaotropic hexafluorophosphate salt was added to acidic water-acetonitrile mobile phases to improve the reversed-phase chromatography of UA, which otherwise was poorly retained. Optimum separation conditions were obtained for fish samples and enabled a fast (10 min), convenient-to-use chromatography that clearly outperforms cumbersome legacy ion-pair chromatography. Intended for routine use in our laboratory, the proposed method passed an in-house validation test for linearity, matrix effect (on reactive extraction), accuracy, precision, and detectability.

Histamine food poisoning (HFP) or scombrotoxic fish poisoning is a major public health concern associated with seafood and seafood products containing copious amounts of free histidine , which may be decarboxylated into scombrotoxic histamine, the main culprit of HFP. Moreover, the deamination product of the histidine, trans -urocanic acid, is assumed to be scombrotoxic by degranulating mast cells to release endogenous histamine after isomerizing to the cis -form, as well as known to have immunosuppressive effects. In recognition of the potential health risks associated with the histidine degradatives, this work aimed to provide a method for the simultaneous determination of histidine, histamine, and urocanic acid isomers in food, based on precolumn dansylation and subsequent reversed-phase HPLC analysis. The dansylation conditions were optimized for yield with the design of experiments approach consisted of a center point-enhanced 2 4 factorial design followed by sequential steepest ascent searches. Satisfactory chromatographic separation of fish sauce was achieved within 9 min by retention modeling of both the analytes and potentially interfering peaks. The method was fully validated with limits of detection between 0.004 and 0.017 mg 100 g −1 fish sauce, and successfully applied to several fish sauce samples, in which the histamine and urocanic acid levels were found low and unlikely of concern, with correspondingly low levels of the precursor histidine.

The aim of this study is to construct a pH- and reduction-responsive nanodrug delivery system to effectively deliver a ginsenoside (Rh2) and enhance its cytotoxicity against human hepatocarcinoma cells (HepG2). Here, pullulan polysaccharide was grafted by urocanic acid and α-lipoic acid (α-LA) to obtain a copolymer, α-LA-conjugated N-urocanyl pullulan (LA-URPA), which was expected to have pH and redox dual response. Then, the copolymer LA-URPA was used to encapsulate ginsenoside Rh2 to form Rh2 nanoparticles (Rh2 NPs). The results showed that Rh2 NPs exhibited an average size of 119.87 nm with a uniform spherical morphology. Of note, Rh2 NPs showed a high encapsulation efficiency of 86.00%. Moreover, Rh2 NPs possessed excellent pH/reduction dual-responsive drug release under acidic conditions (pH 5.5) and glutathione (GSH) stimulation with a low drug leakage of 14.8% within 96 h. Furthermore, Rh2 NPs with pH/reduction dual response had higher cytotoxicity than Rh2 after incubation with HepG2 cells for 72 h, indicating that Rh2 NPs had a longer circulation time. After the treatment with Rh2 NPs, the excessive increase of reactive oxygen species and the decrease of superoxide dismutase, glutathione (GSH), and mitochondrial membrane potential suggested that the mitochondrial pathway mediated by oxidative stress played a role in this Rh2 NP-induced apoptosis. In conclusion, this study provides a new strategy for improving the application of ginsenoside Rh2 in the food and pharmaceutical fields.

Qiwei Tongbi oral liquid (QWTB), a classical traditional Chinese medicine (TCM) formula, has a good therapeutic effect on rheumatoid arthritis (RA) and is widely used in China. To comprehensively elucidate the therapeutic mechanism of QWTB in the treatment of RA, the effects of QWTB on biomarkers and metabolic pathways in a rat model of kidney deficiency arthritis were investigated in this study. The effects of QWTB on pharmacodynamic indicators, including paw swelling, arthritis score; interleukin-1β, interleukin-6, interleukin-17 F, tumor necrosis factor-α, tartrate-resistant acid phosphatase 5b, bone alkaline phosphatase, bone-specific alkaline phosphatase, bone glaprotein, urea, and creatinine levels; and histopathology, suggested that QWTB significantly improved renal function, inhibited the inflammatory response, and reduced bone loss. In total, 39 differential metabolites were screened by comparing the endogenous components between blank and model rat plasma, among which 16 metabolites were altered by QWTB. The metabolism pathway analysis revealed that α-linolenic acid metabolism, phenylalanine metabolism, sphingolipid metabolism, histidine metabolism and glycerophospholipid metabolism were greatly disturbed. Thus, the biomarkers investigated included (1) α-linolenic acid, (2) hippuric acid, (3) phosphatidylethanolamine (15:0/22:2(13Z,16Z)), (4) phenylpyruvic acid, (5) sphinganine, and (6) urocanic acid. QWTB affected three abnormal biomarkers: (3), (4), and (6). Phenylphruvic acid, sphinganine and urocanic acid were significantly associated with pharmacodynamic indicators, as shown by Pearson correlation analysis. These results indicated that RA-related biomarkers had certain reliability and biological significance. In summary, QWTB regulated the metabolic disorders in rats with RA. Its therapeutic mechanism may involve the regulation of phenylalanine metabolism, histidine metabolism, and glycerophospholipid metabolism. The results of this study are useful for understanding the therapeutic mechanisms of TCM.
