An axolotl limb regeneration-inspired strategy to enhance alveolar bone regeneration

Rongpu Liu,Guifang Wang,Li Ma,Guangzheng Yang,Sihan Lin,Ningjia Sun,Jiajia Wang,Huijing Ma,Xinquan Jiang,Wenjie Zhang

Guided bone regeneration (GBR) is widely applied in implant dentistry, employing barrier membranes to create an osteogenic space by preventing gingival tissue ingrowth. However, this method does not enhance the osteogenic capacity of osteoblasts, limiting sufficient bone volume in larger defects. Inspired by axolotl limb regeneration, abundant soft tissue-derived stem cells mobilized to the defect may facilitate comprehensive osteogenesis within a BMP-2-enriched environment. We developed a biomimetic channel system (BCS) to promote alveolar bone regeneration, using channel structures to activate gingival-derived stem cells under a BMP-2-enriched biological barrier. In a cell-tracing mouse model, Prrx1 + stem cells demonstrated a critical role in BMP-2-induced subcutaneous osteogenesis. Sequencing and histological analyses revealed that channel structures significantly enhance soft tissue cell proliferation and migration. Attributable to the biological barrier, BCS applications markedly improved bone formation in beagle mandibular defects. These results suggest a novel osteoinductive strategy for alveolar bone regeneration that functions without a traditional barrier membrane.

The Effect and Biocompatibility of Nanofibrous Nano-Hydroxyapatite/Polycaprolactone/Gelatin Scaffolds Multipurpose Membrane in Guiding Bone Regeneration

Cao Shuaishuai,Zhang Liang,Chen Zhihui,Li Shuyi,Jiang Huaji,Tian Yuguang,Gu Weiwang,Zhou Miao,Chen Xiaoming

To prepare a new tri-layer multifunctional membrane (TLMM), which comprises gelatin loaded with tinidazole as the outer layer, dense polycaprolactone (PCL) as the intermediate layer, and compound nHA/PCL/gelatin as the inner layer, and to investigate its applicability as a barrier membrane to guide osteanagenesis. The morphology of TLMM prepared is observed using a scanning electron microscope (SEM). The hydrophilicity and swelling ratio of are tested. The mechanical properties of TLMM are tested using a universal electronic tester. The infrared spectrogram of each component in TLMM is obtained using infrared Fourier spectrometer through analytic transmission method. MG63 cell is cultivated on the surface of the multifunctional membrane, the activity of ALP is tested, and the cellular vitality is determined through the cck-8 test. Rats with skull defect are used as the test model. The skull recovery of the rats with skull defect is examined through Micro-CT method. The expression of osteocalcin and osteopontin in the cranial tissue of rats whose skull defect is repaired through TLMM repair is tested through WB test. SEM shows that this compound membrane has a three-dimensional network structure of nanofiber, and the cross-section of the membrane has a thickness of 120 m. TLMM has good hydrophilicity and extremely low swelling ratio. Results from mechanical testing show that TLMM has excellent mechanical strength. Infrared spectrum result demonstrates that the gelatin successfully carries tinidazole, and the release mechanism of tinidazole matches the recovery process after surgery. CCK-8 test indicates that TLMM has no significant influence on the proliferation of MG63 cell, but can greatly improve the activity of cellular ALP. Micro-CT examination proves that TLMM can effectively facilitate cranial recovery among rats with skull defect. WB test shows that TLMM can significantly induce the expression of osteocalcin and osteopontin in the cranial tissue of rats with skull defect, and performs better than traditional bone powder. The results demonstrate that TLMM designed by the study has good biocompatibility, and is applicable as a barrier membrane to guide osteanagenesis.

Durable Hydrate-phobic Coating with In Situ Self-Replenishing Hydrocarbon Barrier Films for Low Clathrate Hydrate Adhesion

Muhammad Imran Jamil,Tao Qian,Waqar Ahmed,Xiaoli Zhan,Fengqiu Chen,Dangguo Cheng,Qinghua Zhang

Clathrate hydrate growth, deposition, and plug formation during oil and gas transportation causes blockage of pipelines. An effective strategy to solve this problem is to mitigate the hydrate formation and reduce its adhesion on pipe walls through a coating process. However, durability failure, corrosion, inability to self-heal, high cost, and strong hydrate adhesion remain unsolved issues. To address these challenges, in this work, we present an in situ self-replenishing nonfluorinated durable hydrate-phobic coating of candle soot particles. The candle soot coating reduces hydrate adhesion by promoting a thick barrier film of hydrocarbons between the hydrate and the soot coated substrate. The hydrocarbons permeating the soot coating display a high contact angle for water and inhibit the formation of water bridges between the hydrate and soot coated substrate. The spherical cyclopentane hydrate slides off easily on the candle soot coating inside the cyclopentane environment. The hydrate former, cyclopentane–water emulsion, and THF–water mixture have high contact angles as well as low hydrate adhesion on soot coating simultaneously. In addition, the coating is flow-induced long-term slippery, durable, low cost, anticorrosion, self-cleaning, and suitable for practical applications.

Multifunctional chitin-based barrier membrane with antibacterial and osteogenic activities for the treatment of periodontal disease

Tao Wu,Lin Huang,Jing Sun,Jiahui Sun,Qi Yan,Bo Duan,Lina Zhang,Bin Shi

The guided tissue regeneration technique is an effective approach to repair periodontal defect. However, collagen barrier membranes used clinically lose stability easily, leading to soft tissue invasion, surgical site infection, and failure of osteogenesis. An ideal barrier membrane should possess proper antibacterial, osteoconductive activities, and favorable biodegradation. In this study, zinc oxide nanoparticles were homogeneously incorporated into the chitin hydrogel (ChT-1%ZnO) through one-step dissolution and regeneration method from alkaline/urea solution the first time. The remaining weights of ChT-1%ZnO in 150 μg/mL lysozyme solution was 52% after 5 weeks soaking. ChT-1%ZnO showed statistical antibacterial activities for P. gingivalis and S. aureus at 6 h, 12 h, and 24 h. Moreover, ChT-1%ZnO exhibits osteogenesis promotion in vitro , and it was further evaluated with rat periodontal defect model in vivo . The cemento-enamel junction value in ChT-1%ZnO group is 1.608 mm, presenting a statistical difference compared with no-membrane (1.825 mm) and ChT group (1.685 mm) after 8 weeks postoperatively.

Moisture-Permeable, Humidity-Enhanced Gas Barrier Films Based on Organic/Inorganic Multilayers

Jiajie Wang,Xiaozhi Xu,Jian Zhang,Mengting Chen,Siyuan Dong,Jingbin Han,Min Wei

Gas barrier films with water-vapor-permeability have exhibited broad application prospects in gas separation and dehumidification. Herein, multilayer films comprised of layered double hydroxides (LDH) nanosheets and carboxymethyl cellulose sodium (CMC) were fabricated via layer-by-layer assembly. The resulting (LDH/CMC)n films show excellent gas barrier properties, which are ascribed to the significantly increased pathway for gas permeation originating from the large aspect ratio and high orientation of two-dimensional LDH nanosheets. Unlike traditional gas barrier films with nonselective blocking effect for various gases (including water vapor), the (LDH/CMC)n films exhibit an unusual moisture permselective property. The moisture-permeable property was related to the hygroscopicity of CMC and hydrophilicity of LDH, which can enrich the water molecules from the surroundings and aggrandize the osmotic pressure for water vapor, resulting in an uncommon improvement of water vapor transmission. It is interesting to find that the (LDH/CMC)n films exhibit enhanced gas (O2, CO2, CH4, and N2) barrier properties upon treatment in a humid environment, due to the formation of hydrogen bonds between the infiltrated water molecules and hydrophilic groups in CMC, thus padding the interstitial space of the CMC molecular chains and increasing the gas transmission path. The reduction of free volume and extension of the gas transmission path further enhance the gas barrier properties of (LDH/CMC)n films. Moreover, the (LDH/CMC)n films represent the water vapor permselective property in mixed gas (including O2, CO2, CH4, N2, and water vapor), while maintaining the barrier for other gases, which can be potentially applied in air dehydration and dehumidification of natural gas.

Micro- and nano-environment dual-modulated anti-tendon adhesion barrier membranes

Qiang Zhang,Kui Ma,Chun-Hei Lam,Ho-Pan Bei,Yu Liu,Xing Yang,Xin Zhao

Despite promise in preventing peritendinous adhesion, electrospun membranes face many challenges related to their complex fabrication process, untargeted/uncontrolled drug delivery and consequently low therapeutical effect. Here, a micro-and nano-environment dual-modulated barrier membrane (MNBM) with on-demand gene delivery capability is presented. Our MNBM is developed by first preparing extracellular signal-regulated kinase-2 (ERK2) siRNA-loaded gelatin methacryloyl (GelMA) nanogels via facile nano-emulsification technique, then incorporating these nanogels into poly-L-lactic acid (PLLA) fibers via simple blending electrospinning. The GelMA nanogels offer a nano-niche for ERK2-siRNA encapsulation and allow for a nano-environment controlled siRNA release by readily tuning the GelMA concentrations during nano-emulsification, while the resultant MNBM can mediate a micro-environment controlled siRNA delivery in response to the matrix metalloproteinase-2 (MMP-2) enriched micro-environment at the tendon repair site. Such MNBM can not only biologically orchestrate fibroblast behaviors by silencing the target gene expression, but also physically shield the tendon from extrinsic cell/tissue invasion. This study provides a proof-of-concept of anti-adhesion barrier membrane as an intelligent gene delivery system to offer a spatiotemporal and biophysical dual control over tendon recovery according to disease state and ensure long-term therapeutic efficacy. We envision such MNBM represents a promising therapeutic platform with great efficacy to achieve adhesion-free tendon repair.

Zinc hybrid polyester barrier membrane accelerates guided tissue regeneration

Qiao Zhang,Chaoqian Lou,Hang Li,Yanyan Li,Hongjie Zhang,Zimeng Li,Ganggang Qi,Xia Cai,Qiaojie Luo,Lijie Fan,Xiaojun Li,Weiwei Lao,Weipu Zhu,Xiaodong Li

Barrier membranes play a pivotal role in the success of guided periodontal tissue regeneration. The biodegradable barriers predominantly used in clinical practice often lack sufficient barrier strength, antibacterial properties, and bioactivity, frequently leading to suboptimal regeneration outcomes. Although with advantages in mechanical strength, biodegradability and plasticity, bioinert aliphatic polyesters as barrier materials are usually polymerized via toxic catalysts, hard to be functionalized and lack of antibacterial properties. To address these challenges, we propose a new concept that controlled release of bioactive substance on the whole degradation course can give a bioinert aliphatic polyester bioactivity. Thus, a Zn-based catalytic system for polycondensation of dicarboxylic acids and diols is created to prepare zinc covalent hybrid polyester (PBS/ZnO). The atomically-dispersed Zn 2+ ions entering main chain of polyester molecules endow PBS/ZnO barrier with antibacterial properties, barrier strength, excellent biocompatibility and histocompatibility. Further studies reveal that relying on long-term controlled release of Zn 2+ ions, the PBS/ZnO membrane greatly expedites osteogenetic effect in guided tissue regeneration (GTR) by enhancing the mitochondrial function of macrophages to induce M2 polarization. These findings show a novel preparation strategy of bioactive polyester biomaterials based on long term controlled release of bioactive substance that integrates catalysis, material structures and function customization.

Restoration of periodontitis-induced bone defects with multifunctional barrier membranes by alleviating oxidative stress and remodelling macrophage metabolism

Lei Han,Jimin Huang,Zhiyong Zhang,Guijuan Feng,Yi Zheng,Yufang Zhu,Huang Li

Periodontal bone regeneration is hindered by the inflammatory microenvironment with high reactive oxygen species levels. Current barrier membranes lack sufficient antioxidative and bioactive properties, leading to poor bone regeneration. To overcome this, a novel guided bone regeneration membrane was developed by combining manganese-incorporated bioactive glass (MnBG) with biodegradable poly(lactic- co -glycolic acid (PLGA) and polycaprolactone (PCL). This PLGA/PCL/MnBG (PPMnBG) membrane could protect bone marrow mesenchymal stem cells (BMMSCs) and RAW264.7 cells from oxidative damage, promote cell proliferation, adhesion, and osteogenic differentiation, and inhibited M1 macrophage polarization. It enhanced macrophage oxidative stress resistance and suppressed glycolysis in M1 macrophages via the JAK-STAT1/NLRP3 pathway, reducing inflammation. In vivo studies using SD rat alveolar bone defect confirmed the membrane's efficacy in alleviating oxidative stress, reprogramming macrophage metabolism, and promoting bone regeneration. These findings highlighted the PPMnBG membrane's potential in treating inflammatory alveolar bone defects by mitigating oxidative stress, enhancing osteogenesis, and modulating macrophage metabolism.

Controlling the Pore Structure of Collagen Sponge by Adjusting the Cross-Linking Degree for Construction of Heterogeneous Double-Layer Bone Barrier Membranes

Xi Zhao,Xinying Li,Xiaofen Xie,Jinfeng Lei,Liming Ge,Lun Yuan,Defu Li,Changdao Mu

Guided bone regeneration (GBR) has been regarded as a valuable way to effectively induce bone remodeling. The key factor of GBR is to place a barrier membrane between the soft tissue and bone defect, preventing the untimely intrusion of fibroblasts and permitting the prior settlement of internal osteoblasts. Notably, heterogeneous double-layer GBR membranes with a compact upper layer and a loose lower layer exhibit enhanced effectiveness in blocking fibroblasts and promoting the growth of osteoblasts. Herein, we present porous and interconnected collagen-based sponges with controllable pore size for the fabrication of absorbable GBR membranes with a heterogeneous double-layer structure. Dialdehyde carboxymethyl cellulose was used to fix collagen-based sponges. The pore size of the sponges can be well controlled by adjusting the cross-linking degree, which is decreased with an increase of cross-linking degree. The sponges show enhanced mechanical properties, inhibited swelling ability and biodegradation, good blood compatibility, and good cytocompatibility. The sponges are feasible to form a heterogeneous double-layer structure with a loose lower layer and a compact upper layer. Interestingly, the lower layer with the pore size of 200–300 μm can promote the adhesion, proliferation, and differentiation of osteoblasts MC3T3-E1 cells, while the upper layer with the pore size of 20–50 μm makes a great contribution to the growth of myoblast C2C12 cells. Overall, the collagen-based sponges have the potential to be used to fabricate heterogeneous double-layer bone barrier membranes for bone remodeling.

Bredigite bioceramic-based barrier membrane promotes guided bone regeneration by orchestrating an immuno-modulatory and osteogenic microenvironment

Longwei Hu,Yun Zhu,Yibo Guo,Chenping Zhang,Yang Wang,Zhen Zhang

Barrier membranes play an important role in guided bone regeneration (GBR) and have been primarily considered as physical barriers. Due to their inherent “foreign body” properties, barrier membranes introduced during GBR procedures unavoidably modify the local immune microenvironment, with consequent impacts on osteogenesis. In bone tissue engineering applications, inorganic bioceramics have been shown to possess the abilities to modulate the local immune response and promote the differentiation of stem cells along osteogenic lineages. Moreover, these bioceramics can be easily incorporated into scaffold membranes, thus demonstrating significant potential as a barrier membrane material for GBR. Herein, an inorganic bredigite (BRT, Ca 7 MgSi 4 O 16 ) bioceramic-containing scaffold was fabricated and characterized, and its effects on the immune response and osteogenesis during GBR were investigated. The results indicated that the BRT-containing scaffolds promoted the migration and osteogenic differentiation of bone marrow-derived stem cells (BMSCs). Single-cell RNA sequencing analysis indicated a unique microenvironment and subpopulation of BMSCs cultured on the BRT scaffolds, which exhibited increased osteo/chondrogenic and angiogenic differentiation potential. Furthermore, the BRT-containing scaffolds induced macrophage polarization into the pro-regenerative M2 phenotype, which subsequently promoted the migration and osteogenic differentiation of BMSCs. Additionally, in an in vivo rat model, the BRT-containing membrane was confirmed to enhance the immune microenvironment and support bone regeneration in a cranial critical-size defect. Collectively, these findings indicate that the BRT-containing scaffold membrane possesses dual immunomodulatory and osteogenic properties, offering a potentially beneficial GBR membrane for clinical application.

Janus fibre/sponge composite combined with IOPNs promotes haemostasis and efficient reconstruction in oral guided bone regeneration

Hanbang Chen,Jia Yan,Shuying Hu,Shangwen Sun,Fang Zhou,Jun Liu,Shijia Tang,Qiao Zhou,Haonan Ding,Feimin Zhang,Ning Gu

Placing a barrier membrane between an alveolar bone wound and gingival soft tissue is a common and effective method to prevent fibroblast invasion and promote new bone formation. However, the combined use of a barrier membrane with bone-forming substitutes remains a challenge for dentists during surgery. Thus, a Janus fibre/sponge composite combining iron oxide nanoparticles (IONPs, γ-Fe 2 O 3 ) was fabricated to prevent epithelial invasion and promote alveolar reconstruction. In the scaffold, a poly (lactic-co-glycolic acid)/polycaprolactone (PP) electrospun layer was covalently bound with a chitosan sponge containing γ-Fe 2 O 3 by polydopamine. The resilient chitosan sponge can easily adhere to the wound surface and encompass haemostatic capability by swelling and forming blood clots to block bleeding. The introduction of IONPs promoted the osteogenesis of rBMSCs, as shown by ALP analysis and cytokine assay. The PP layer maintained its structure integrity and prevented epithelial cells from invading into blood clots or fibroblasts from penetrating through the critical pore size. Assessment with a rat model having one-sided calvarial bone injury further confirmed the effectiveness of Janus scaffolds in facilitating the bone regeneration in the postoperative areas.

Asymmetric barrier membranes based on polysaccharide micro-nanocomposite hydrogel: Synthesis, characterization, and their antibacterial and osteogenic activities

Zhiqi He,Xiaolin Zhou,Yi Wang,Jian Lin,Siyan Huang,Rongdang Hu,Yunlong Zhou,Qiuping Qian,Hui Deng

Guided tissue regeneration (GTR) strategies enable periodontal tissue regeneration, generally by providing barrier membranes. However, currently available membranes have limited osteoconductive and antibacterial potential. To address these challenges, we fabricated a new asymmetric barrier membrane. Agarose hydrogel functions as the main body of the barrier membrane. Hollow carbonated hydroxyapatite (CHA) prepared by hydrothermal method , was sedimented in agarose to exhibit an asymmetrical structure. And ε-poly-lysine (ε-PLL) was chosen as an antimicrobial agent to equip the membrane with long-lasting antibacterial activity . With the increased dose of CHA addition, the barrier membrane shows better biocompatibility, and higher mechanical properties. We demonstrated the osteoconductivity and antibacterial properties of the membrane in vitro and in vivo. In summary, our findings suggest that the barrier membrane has good osteoconductive and antibacterial properties, indicating its potential for periodontal tissue engineering.
