Effects of Salmonella bacteriophage, nisin and potassium sorbate and their combination on safety and shelf life of fresh chilled pork

Changbao Wang,Jie Yang,Xiaoyu Zhu,Yingjian Lu,Yingying Xue,Zhaoxin Lu

Salmonella is an important foodborne pathogen and a serious threat to human health worldwide. This study was to reduce Salmonella and the spoilage bacteria on fresh chilled pork using bacteriophage, nisin, and potassium sorbate (PS) along with their combinations. Microbial, chemical, and sensory qualities of the fresh chilled pork (artificially contaminated with Salmonella 3 log CFU/g) treated with bacteriophage (9 log PFU/g), nisin (5000 IU/g), PS (2 mg/g) and their combinations were evaluated. The result showed that all the samples treated with phage could significantly ( P  < 0.05) reduce Salmonella population on fresh chilled pork. The combination treatment of nisin, PS and phage (N-PS-P) could significantly lower total viable counts (TVC), TVB-N and TBARS of the chilled pork during the storage period. The TVC of sample treated by N-PS-P was reduced by 2.3 log CFU/g at 7th day. It was also found through the electronic nose detection that the N-PS-P treatment was able to significantly reduce odour and maintain good sensory of the chilled pork. Hence, the N-PS-P treatment extended the shelf life of fresh chilled pork up to 14 days. No adverse effect of the phage on the chilled pork was observed. In conclusion, this study suggests that the phage and its combination with nisin and PS have great potential to be used as a good preservative for fresh chilled pork.

Synthesis and antimicrobial applications of the inclusion complexes of β-cyclodextrin copolymers with potassium sorbate

Yu Cao,Jiayu Li,Jun Liu,Yuying Tang,Huan Liu,Yan Jiang,Hongwen Zhang

Four inclusion complexes were synthesized from β-cyclodextrin (β-CD) copolymers and potassium sorbate (PSA) using the self-assembly principle. Reactive β-CD monomers carrying unsaturated ester functional groups [β-CD-acryloyl chloride (AC)] were synthesized from β-CD and AC and copolymerized with styrene to obtain β-CD copolymers of the general formula poly(β-CD-AC- co -St). These β-CD copolymers having cavity structures were complexed with PSA to obtain the corresponding inclusion complexes of the general formula poly(β-CD-AC- co -St)/PSA. These complexes were added to the polystyrene (PS) sheets, as an antibacterial additive, to inhibit the adhesion and growth of bacteria on its surface. The chemical and physical structures of copolymers were characterized by Fourier transform infrared, nuclear magnetic resonance, and the slow release performance was investigated using ultraviolet spectroscopy. The results showed that the composition of the copolymers and their molecular weight could be controlled by changing the rate of charge while the rate and the amount of slow release could be controlled by altering the content of β-CD in the copolymer. The PS sheets containing the inclusion complexes exhibited excellent antibacterial activity that significantly reduced the adhesion of yeast and Staphylococcus aureus to its surface. With the increase of the β-CD content in the copolymers, they contained more of PSA and their bacteriostatic property became more pronounced. © 2018 Wiley Periodicals, Inc. J. Appl. Polym. Effect of nisin and potassium sorbate additions on lipids and nutritional quality of Tan sheep meat

Wei Jia,Xixuan Wu,Ruiting Li,Shuxing Liu,Lin Shi

Nisin and potassium sorbate as preservatives are used in a broad range of meat. A lipidomic evaluation was performed on Tan sheep meat treated by two types of preservatives. The addition of potassium sorbate resulted in higher lipid losses compared with nisin treatment. Furthermore, 106 significant lipids of 12 lipid classes (PC, PS, LPS, LPC, PE, PI, LPE, TG, Cer, DG, SM, Sph) with variable importance in projection scores greater than 1.0 were detected and qualified to distinguish different preservatives added meat using UHPLC-Q-Orbitrap MS/MS. LOD and LOQ were 0.12–0.32 μg kg −1 and 0.35–0.89 μg kg −1 , indicating high sensitivity and excellent analytical characteristics in the study. Nisin was confirmed to be the better preservative for prolonging the shelf life of Tan sheep meat while reducing the loss of nutrients. These results could provide a strong cornerstone for future research on preservatives in meat products.

Bubble-driven solid-phase microextraction for the rapid determination of potassium sorbate in oyster sauce by HPLC-DAD

Jin-Yu Li,Yun-Jiao Ma,Feng Xiao,Xian-Bing Xu

Summary On-site sample preparation has the advantage of combining sampling and sample preparation into a single step, thereby minimising sources of error, and eliminating the risk of analyte instability. Bubble-driven solid-phase microextraction (BD-SPME) was first proposed as an on-site pretreatment method. Oyster sauce underwent on-site pretreatment to measure potassium sorbate levels via high-performance liquid chromatography with a diode array detector (HPLC-DAD), obviating the need for stirrers. The high sensitivity of the HPLC determination of potassium sorbate was obtained using BD-SPME pretreatment with a limit of detection (LOD) of 0.03 mg L −1 and a limit of quantification (LOQ) of 0.10 mg L −1 . The refined technique displayed remarkable linearity from 0.10 to 30.00 mg L −1 , boasting a coefficient of determination R 2 ≥ 0.998. In addition, the recoveries of potassium sorbate in oyster sauce using the method were in the range of 90.8–101.4%. Evidently, BD-SPME in combination with HPLC-DAD is a sensitive and promising analysis for on-site pretreatment.

A Dietary Source of High Level of Fluoroquinolone Tolerance in mcr-Carrying Gram-Negative Bacteria

Tianqi Xu,Dan Fang,Fulei Li,Zhiqiang Wang,Yuan Liu

The emergence of antibiotic tolerance, characterized by the prolonged survival of bacteria following antibiotic exposure, in natural bacterial populations, especially in pathogens carrying antibiotic resistance genes, has been an increasing threat to public health. However, the major causes contributing to the formation of antibiotic tolerance and underlying molecular mechanisms are yet poorly understood. Herein, we show that potassium sorbate (PS), a widely used food additive, triggers a high level of fluoroquinolone tolerance in bacteria carrying mobile colistin resistance gene mcr. Mechanistic studies demonstrate that PS treatment results in the accumulation of intracellular fumarate, which activates bacterial two-component system and decreases the expression level of outer membrane protein OmpF, thereby reducing the uptake of ciprofloxacin. In addition, the supplementation of PS inhibits aerobic respiration, reduces reactive oxygen species production and alleviates DNA damage caused by bactericidal antibiotics. Furthermore, we demonstrate that succinate, an intermediate product of the tricarboxylic acid cycle, overcomes PS-mediated ciprofloxacin tolerance. In multiple animal models, ciprofloxacin treatment displays failure outcomes in PS preadministrated animals, including comparable survival and bacterial loads with the vehicle group. Taken together, our works offer novel mechanistic insights into the development of antibiotic tolerance and uncover potential risks associated with PS use.

Preparation and characterization of probiotic Bacillus velezensis 906 metabolites /potassium sorbate/ polyvinyl alcohol antimicrobial blend film

Si Cheng,Chao Yin,Keying Li,Zhe Liu,Qingyan Pan,Xifeng Zuo,Ailing Guo,Huawei Ma

Films with excellent mechanical properties and antimicrobial activity have potential applications in food preservation. The antimicrobial film was prepared by blending probiotic Bacillus velezensis 906 metabolites (906), potassium sorbate (PS), and polyvinyl alcohol (PVA). The film was characterized using scanning electron microscopy (SEM), Fourier transform infrared spectroscopy (FTIR), X-ray diffraction (XRD), and thermal stability analysis. The mechanical, barrier, and antimicrobial properties of the film were tested. The results of the physicochemical analysis indicated that, compared to the pure PVA film, the film's mechanical properties decreased, while its barrier properties and hydrophobicity increased with the addition of PS and 906, respectively. The results of the antimicrobial tests revealed that the blend film exhibited significant antimicrobial activity against Vibrio parahaemolyticus, Psychrobacter maritimus , Oceanobacillus kimchii, Pseudomonas aeruginosa , and Listeria monocytogenes . SEM demonstrated good fusion between the components of the PVA/PS/906 film, resulting in a dense and crack-free structure. FTIR suggested that PS and 906 enhance molecular interactions within the film matrix. XRD revealed that the film was crystallized, with 906 increasing the crystallinity of the bonds between the film components. Thermal analysis indicated the thermal stability of the films was enhanced. Consequently, the PVA/PS/906 blend film shows significant potential for application in the preservation of aquatic products.

One-step synthesis of dual-emission carbon dots for ratiometric fluorescence sensing of curcumin in food samples

Zongmei Huang,Cong-Ting Wu,Jing Li,Lu-Shuang Li

Illegal addition of curcumin (Cur) in food poses a significant public health threat, creating an urgent need for sensitive, reliable detection methods. In this work, dual-emission carbon dots (DCDs) were synthesized via a one-step hydrothermal method with Rose Bengal and melamine as raw materials. Based on the distinct fluorescence responses of DCDs to Cur at two characteristic wavelengths, a novel ratiometric fluorescence sensor for Cur detection was constructed. The emission at 432 nm is primarily governed by inner filter effect, while quenching at 532 nm is associated with the fluorescence resonance energy transfer. The sensor shows a robust linear range of 0.1–20 μM with a limit of detection of 0.089 μM. The sensor was successfully employed for the detection of Cur in various food samples, including chili, ginger, and mustard powder, achieving recovery rates between 92.75% and 110.00%. This work enriches CDs synthesis strategies and provides a sensitive, cost-effective, rapid method for Cur detection, supporting food industry monitoring needs.

A colorimetric sensor for sensitive and highly selective detection of butyl hydroxyanisole in edible oil based on dual-enzyme-mimic activities of Ag2CO3 nanozymes

Zhongqin Li,Jiazhen Shi,Yinghua Song,Yue Tang,Lian Xia,Yuangen Wu

Butylated hydroxyanisole (BHA) is a synthetic phenolic antioxidant widely used in edible oils and fat-containing foods. Excessive BHA poses significant risks to human health, thus making rapid and sensitive detection of BHA crucial for health protection. Herein, the Ag 2 CO 3 nanozyme was prepared by the simple precipitation method, which exhibits both laccase-mimic and oxidase-mimic activities. It was found that the oxidase-mimic activity of Ag 2 CO 3 facilitated the conversion of TMB to its blue oxidized form (oxTMB), and subsequent addition of BHA resulted in the formation of a characteristic purple adduct. Additionally, BHA effectively suppressed the laccase-mimic activity of the Ag 2 CO 3 nanozymes. Therefore, a novel colorimetric sensor was constructed using the dual-enzyme-mimic activities as a sensing signal for rapidly detecting BHA in edible oil. The colorimetric sensor achieves limits of detection (LOD) for BHA as low as 1.1 μM in the laccase-mimic channel and 0.08 mM in the oxidase-mimic channel, both of which are substantially lower than the maximum permissible BHA level in edible oils stipulated in Chinese National Food Safety Standard GB 2760. The sensor developed in this work shows great promise for application in the field of edible oil detection.

Dual-enzyme synergistic preparation and characterization of antioxidant peptide from perilla meal and its alleviation on cellular oxidative injury

Yangfan Hu,Yulong Yao,Cheng Li,Huizhen Li,Tianyu Hou,Zhijun Zhang

To improve the comprehensive utilization of perilla meal, dual-enzyme hydrolysis was optimized to prepare antioxidant peptides (PAP). The optimal conditions were pH 8.0, 50°C, 6 h, solid-liquid ratio of 1:22 g mL −1 , 1:1 compound enzymes and 6% enzyme dosage. The dual-enzyme product presented higher hydrolysis degree and antioxidant capacity than single-enzyme groups, with excellent solubility and processing stability against food additives, metal ions and food ingredients. The ultrafiltration screened active fraction PAP-SF-4 effectively alleviated oxidative damage in H 2 O 2 -stressed 4T1 cells by regulating ROS, MDA and endogenous antioxidant indexes. A key antioxidant peptide GDF was identified, which tightly bound to Keap1 through hydrogen bonds and hydrophobic interactions. This study provides a theoretical basis for the development and application of perilla-derived bioactive peptides.

An antibody-free sensing strategy based on FeNPs@Pt and thiol-Michael addition: Visual detection of acrylamide in food

Liu Yang,Baoshan He,Ming Lu,Zhengyong Liang,Yao Liu,Wenjie Ren,Yiwei Xu

The core–shell FeNPs@Pt shows excellent catalytic activity and stability. • Selective detection via thiol–Michael addition without enzymes or aptamers. • The color development is rapid and convenient, taking only 150 s to complete. • The suppression–recovery mechanism amplifies the signal and enhances sensitivity. In this study, we propose an innovative Acrylamide (AA) detection platform based on platinum-coated magnetic iron nanoparticles (FeNPs@Pt). The introduction of a platinum shell not only preserves the intrinsic catalytic properties of FeNPs but also significantly enhances their structural stability and catalytic activity. Compared with traditional antibody–aptamer-based detection methods, this platform employs a thiol–Michael addition reaction combined with a nanocatalyst-mediated cascade colorimetric process. This approach overcomes key limitations of antibody–aptamer technologies, such as high cost and operational complexity, enabling rapid and low-cost detection of AA. The mechanism involves a specific Michael addition reaction between AA and 2,2′-(ethylenedioxy)diethanethiol (EDT), which mitigates the inhibitory effect of EDT on the catalytic activity of FeNPs@Pt. Subsequently, the addition of 3,3′-dimethylbenzidine (TMB) triggers a colorimetric reaction within 150 s, rendering the result visible to the naked eye. This sensing platform exhibits a linear response over the range of 1–200 μM, with a detection limit as low as 0.63 μM. Notably, the FeNPs@Pt–EDT–TMB-based colorimetric detection platform not only provides an efficient and low-cost method for acrylamide detection but also offers an innovative solution for on-site food safety monitoring, demonstrating significant application potential. Investigation of Emulsifying Properties and Stability of Fish Gelatin and Tea Saponin Complex Emulsion System

Kaixuan Wang,Shengnan Zhan,Jianyuan Yang,Qiaoming Lou,Tao Huang

In this study, environmental stability, rheological properties, and structural characterization of co-stabilized emulsions using fish gelatin (FG) and tea saponin (TS) were investigated. The results demonstrated that the addition of TS significantly enhanced the emulsifying properties of FG, and the FG-TS 0.1% emulsion had the smallest particle size. TS and FG co-stabilized emulsions provided resistance to salt and high temperatures. Optical microscopy and CLSM showed that the addition of TS made FG more effectively adsorb at the oil–water interface, leading to the formation of more uniform oil droplet sizes. Additionally, the addition of TS increased the viscosity of FG emulsions, which reduced emulsion flocculation. Results of intrinsic fluorescence, FTIR, and surface hydrophobicity revealed that the addition of TS altered the secondary structure of FG, enhancing surface hydrophobicity and improving emulsification. In conclusion, the moderate addition of TS significantly enhanced the emulsification and rheological properties of FG, suggesting new potential applications for FG in various industries.

Photodegradation mechanism of organic contaminants mediated by chlorinated algal organic matter

Dong Wan,Chengjie Yu,Yafei Zhao,Gaofei Song,Wujuan Mi,Yuxuan Zhu,Changzi Liu,Yonghong Bi

Algal organic matter (AOM) significantly influences the photochemical behavior of dissolved organic matter in aquatic environments. This study investigated the effects of chlorination on the photophysical and photochemical properties of AOM derived from Microcystis aeruginosa , compared these alterations with those observed for natural organic matter (NOM), and examined their impact on the photodegradation of organic contaminants, with a particular focus on N,N -diethyl-m-toluamide (DEET) as a model substrate. The results demonstrated that chlorination substantially altered the photochemical reactivity of AOM. AOM and NOM exhibit distinct reactivities, reflecting their varied molecular compositions and functional groups. Specifically, chlorination reduced the aromaticity (SUVA 254 decreased by ∼42%) and molecular weight (decreased by ∼30%) of AOM, resulting in a shift of fluorescence peaks to lower wavelengths. It also enhanced the formation of singlet oxygen ( 1 O 2 ) and hydroxyl radical ( • OH). Chlorinated extracellular organic matter (EOM) exhibited a remarkable increase in • OH quantum yield, with a 200-fold enhancement at a high free available chlorine (FAC) dose (FAC/TOC ratio of 2.0). The photodegradation of DEET, involved H-abstraction and hydroxylation by • OH, was significantly accelerated in chlorinated EOM, highlighting the critical role of chlorinated AOM in driving photosensitized degradation processes. The findings emphasized the role of chlorination in altering AOM's photochemical properties, with significant implications for the enhanced transformation of contaminants in natural and engineered aquatic systems.
