Separation of vitexin-4″-O-glucoside and vitexin-2″-O-rhamnoside from hawthorn leaves extracts using macroporous resins

Hongjuan Li,Ying Liu,Haizhu Jin,Sujing Liu,Shengtao Fang,Chunhua Wang,Chuanhai Xia

Vitexin-4″- O -glucoside and vitexin-2″- O -rhamnoside are the major flavonoids of hawthorn leaves. In this work, the adsorption and desorption characteristics of vitexin-4″- O -glucoside and vitexin-2″- O -rhamnoside on seven macroporous resins were evaluated. Among the tested resins, the HPD-400 resin showed the best adsorption and desorption capacities. Adsorption isotherms were constructed for the HPD-400 resin and well fitted to Langmuir and Freundlich models. Dynamic adsorption and desorption tests were performed on column packed with the HPD-400 resin to optimize the chromatographic parameters. After one run treatment with the HPD-400 resin, the contents of vitexin-4″- O -glucoside and vitexin-2″- O -rhamnoside in the product were increased 8.44-fold and 8.43-fold from 0.720% and 2.63% to 6.08% and 22.2% with recovery yields of 79.1% and 81.2%, respectively. These results show that the developed method is a promising basis for the large-scale purification of vitexin-4″- O -glucoside and vitexin-2″- O -rhamnoside from hawthorn leaves and other plant materials.

Roles of the Main Physical Properties of the Wet Granulation Product of Hawthorn leaf Extract Mixtures in High Shear Granulation

Zhe Li,Juan Luo,Qieying Jiang,Guowei Zhao,Zhenggen Liao,Xinli Liang,Ronggui Zeng,Dan Lv

The purpose of this study was to establish a relationship between the physical properties of powders and their behavior during high shear granulation. Hawthorn leaf extract was utilized as a model drug, and microcrystalline cellulose (MCC) and soluble starch were selected as excipients. The extract was mixed with each of the different excipients at ratios of 1:0.5, 1:1 and 1:2 by weight. The granules were prepared by high shear granulation (HSG). The wet granulated product was evaluated by monitoring responses such as the granule yield, the ratio of lumps to fine powder, the angle of repose of the granules (granule-AOR) and the Hausner ratio of the granules (granule-HR). The effect of the powder's physical properties on the granulation behavior was analyzed through stepwise regression. The critical relative humidity, particle size (d50), surface free energy, AOR, moisture content, equilibrium hygroscopic moisture content ( f ), specific surface area, pore volume, Carr index, contact angle with water and bulk density were the main factors linked to the granule product. The main physical properties of the extract powder were the important factors in HSG. The findings from this study are useful in preparing ideal granulation products by reasonably controlling the physical properties of powders. Practical Applications The present work investigated the influence of the physical properties of Hawthorn leaf extract on high shear granulation behavior. Results showed that critical relative humidity, particle size, surface free energy, angle of repose, moisture content, equilibrium hygroscopic moisture content, specific surface area, pore volume, Carr index, contact angle with water and bulk density were the main factors linked to the granule product. Taking into consideration that the food industry uses Hawthorn leaf extract not only to plant health product as nutrient supplement but to botanical drug in several countries, this work is a useful guide to prepare ideal granulation products by reasonably controlling the physical properties of powders.

Application of alkyl polyglycoside surfactant in ultrasonic-assisted extraction followed by macroporous resin enrichment for the separation of vitexin-2″-O-rhamnoside and vitexin from Crataegus pinnatifida leaves

Feng Han,Yupin Guo,Huiyan Gu,Fenglan Li,Baozhong Hu,Lei Yang

An alkyl polyglycoside (APG) surfactant was used in ultrasonic-assisted extraction to effectively extract vitexin-2″- O -rhamnoside (VOR) and vitexin (VIT) from Crataegus pinnatifida leaves. APG0810 was selected as the surfactant. The extraction process was optimized for ultrasonic power, the APG concentration, ultrasonic time, soaking time, and liquid–solid ratio. The proposed approach showed good recovery (99.80–102.50% for VOR and 98.83–103.19% for VIT) and reproducibility (relative standard deviation, n   =   5; 3.7% for VOR and 4.2% for VIT) for both components. The proposed sample preparation method is both simple and effective. The use of APG for extraction of key herbal ingredients shows great potential. Ten widely used commercial macroporous resins were evaluated in a screening study to identify a suitable resin for the separation and purification of VOR and VIT. After comparing static and dynamic adsorption and desorption processes, HPD100B was selected as the most suitable resin. After column adsorption and desorption on this resin, the target compounds VOR and VIT can be effectively separated from the APG0810 extraction solution. Recoveries of VOR and VIT were 89.27%   ±   0.42% and 85.29%   ±   0.36%, respectively. The purity of VOR increased from 35.0% to 58.3% and the purity of VIT increased from 12.5% to 19.9%.

Folium crataegi boosts skin regeneration for burn injury in rats through multiple ways

Zhihong Deng,Lifeng Zhao,Xiuxue Li,Chang-Yun Wang,Yue Zhou,Mengfei Li,Yingfei Li,Xianjun Fu

Appropriate topical dressings for burn treatments are important to accelerate skin wound recovery and prevent external infections. This study aimed to evaluate the effect and investigate the mechanism of folium crataegi ( Crataegus pinnatifida Bge. ) for the treatment of burn wounds, as well as to compare the therapeutic effects of aqueous extracts ( HLW ) and alcoholic extracts ( HLE ) from folium crataegi . The results demonstrated that both HLW and HLE groups exhibited a higher wound contraction rate than the silver sulfadiazine ( SSD ) ointment group. Moreover, HLW showed more significant wound repair effects than HLE . HLW significantly increased levels of EGF and FGF-2 in wound tissue, as well as TGF-β1 , VEGF , CAT and IL-10 in serum. Folium crataegi extract, especially aqueous extracts, exerted good anti-inflammatory, anti-oxidant and anti-bacterial effects by upregulating the expression of lag3 , txn1 and slpi , respectively. Folium crataegi extract significantly inhibits the expression of npas2 , a key gene in the circadian rhythm pathway. In conclusion, this research illustrated that the folium crataegi extract, especially aqueous extracts, had better therapeutic effects on skin burns through multiple ways, possibly including a novel mechanism related to circadian rhythm pathway. These findings suggest that folium crataegi could be a valuable source of compounds for enhancing skin regeneration through multiple ways.
