In recent years, the quality and safety issues of Chinese medicinal herbs have received great attention worldwide. Thereinto, heavy metal contamination has been one of the most serious concerns. Compared to the wide research in the analysis of heavy metals in medicinal herbs, the studies on the removal of heavy metals are relatively limited. In this study, polyethylene glycol functionalized Fe 3 O 4 @MIL-101(Cr) (Fe 3 O 4 @MIL-101(Cr)@PEG) was designed and synthesized to remove heavy metals from the decoction of Ligusticum chuanxiong Hort. The in-house fabricated Fe 3 O 4 @MIL-101(Cr)@PEG was characterized by a porous structure and a large specific surface area. Then, the efficiency of the material for the removal of five heavy metals was tested under optimal adsorption conditions. Meanwhile, the content of Senkyunolide A, Senkyunolide I, and Ferulic acid, the solid content, and the HPLC fingerprints similarity were used as the quality monitoring indicators of Ligusticum chuanxiong Hort decoction before and after the heavy metal removal. Results showed that the magnetic nanomaterial had excellent removal efficiency for As 5+ (81.4 %), Cd 2+ (88.19 %), and Pb 2+ (83.79 %) and certain removal efficiency for Ni 2+ (51.59 %) and Zn 2+ (55.4 %) under the spiked concentration of 50 μg/mL. The content of Senkyunolide A, Senkyunolide I, and Ferulic acid were decreased by less than 8.00 % after the removal of heavy metals. Besides, the loss rate of solid content was only 0.18 %, and the fingerprints similarity was over 99.9 %. The results indicated that Fe 3 O 4 @MIL-101(Cr)@PEG could efficiently and selectively remove heavy metals from Ligusticum chuanxiong Hort without affecting its effective components. Due to the advantages of low-cost, simple manipulation, and good efficiency, the material can be recommended for heavy metals removal from the aqueous solutions of medicinal herbs, providing a new and promising application for the removal of exogenous contaminants in medicinal herbs.

Senkyunolide-H (SEH), a major bioactive compound extracted from Ligusticum chuanxiong, has been reported to be effective in preventing cerebral ischemic stroke (CIS). In this study, we employed network pharmacology to reveal potential mechanism of SEH against CIS on a system level and confirmed the therapeutic effects of SEH on CIS by models of cerebral ischemia-reperfusion in vivo and in vitro. Through protein-protein interaction networks construction of SEH- and CIS-related targets, a total of 62 key targets were obtained by screening topological indices and analyzed for Gene Ontology and Kyoto Encyclopedia of Genes and Genomes pathway enrichment. Gene Ontology analysis indicated that SEH might have a role in treating CIS via regulating some biological processes including regulation of transcription from RNA polymerase II promoter, epidermal growth factor receptor signaling pathway, phosphatidylinositol-mediated signaling, and some molecular function, such as transcription factor and protein phosphatase binding and nitric oxide synthase regulator activity. Meanwhile, the Kyoto Encyclopedia of Genes and Genomes analysis showed that phosphoinositide 3-kinase (PI3K)/protein kinase B (Akt) signaling pathway was significantly enriched. In addition, our result showed that SEH posttreatment significantly decreased the neurological scores, infarct volume, and neuronal death in the middle cerebral artery occlusion mice. Moreover, the PI3K/Akt/nuclear factor kappa B signaling pathway was activated by intragastric administration of 40 mg/kg SEH, as verified by Western blot. In vitro, treatment of PC12 cells with 100 μM SEH markedly reduced cell death induced by oxygen-glucose deprivation through the activation of PI3K/Akt/nuclear factor kappa B pathway, and the therapeutic effect of SEH was obviously inhibited by 10 μM LY294002. In summary, these results suggested that SEH carries a therapeutic potential in CIS involving multiple targets and pathways, and the most crucial mechanism might be through the activation of PI3K/Akt/nuclear factor kappa B (NF-κB) signaling pathway to inhibit inflammatory factor releases and increase the antiapoptosis capacity. Our study furnishes the future traditional Chinese medicine research with a network pharmacology framework.

Chuanxiong Rhizome (known as Chuanxiong in Chinese), is widely used in traditional Chinese medicines (TCMs) compounding patches. Ferulic acid (FA) and tetramethylpyrazine (TMP) are the main components of Chuanxiong. This paper aimed to prepare an FA-TMP compound patch with proven anti-inflammatory activity and elucidate the molecular mechanisms of transdermal behaviors of compounds. As confirmed by an anti-inflammatory study, the formulation was optimized by the employment of in vitro skin permeation. The addition of TMP increased the skin permeation amount of FA by approximately 1.9 times. Through FT-IR, XPS, and thermal analysis, it was demonstrated that FA mainly formed hydrogen bonds with TMP, which depressed the melting point (ΔM.P. = 14.6 °C) and hydrophilicity (Δlog D o/w = 0.44) of FA. As revealed by ATR-FTIR and 13 C NMR, compounds increased the degree of skin lipid perturbation (Δν as CH 2 = 3.28 cm −1 ) and attenuated FA binding to ceramides and keratin . To sum up, the hydrogen bonding site of FA was competed by TMP, which enhanced the ability of FA to penetrate the stratum corneum (SC) and partition into the hydrophilic dermal layer, increasing the amount of skin permeation of FA. This study is the first investigation on the combined citation of FA and TMP in patches. It will not only provide a valuable reference for research on the molecular mechanism of transdermal behaviors concerning compounds but will also help to provide a theoretical basis at the molecular level for the scientific design of TCMs formulations.

Background The dry root and rhizome of Ligusticum chuanxiong Hort., or Chuanxiong, has been used as a blood-activating and stasis-removing traditional Chinese medicine for 1000 years. Our previous studies have shown the inhibitory activity on platelet and thrombin (THR) of Chuanxiong. THR and factor Xa (FXa) play significant roles in the coagulation cascade and their inhibitors are of valuable in the treatment of thromboembolic diseases. The aim of the present study is to screen THR and FXa inhibitors from Chuanxiong. Methods Four extracts [ethyl acetate (EA), butanol (BA) and remained extract (RE) from 75% ethanol extract, and water extract (WE)] of Chuanxiong were prepared, and their THR/FXa inhibitory activities were assessed in vitro. Following silica-gel column chromatography (SC), the active EA extract and BA extract was further partitioned, respectively. Their active fractions (EA-SC1 to EA-SC5; BA-SC1 to BA-SC5) were obtained and analyzed by LC–MS. After modeling by the principal component analysis (PCA) and orthogonal partial least squares discriminate analysis (OPLS-DA), the specific marker compounds were predicted and identified. Their enzyme inhibitory was assessed in vitro and interactions with THR/FXa were investigated by molecular docking analysis. Results Chuanxiong EA extract showed strong activity against THR and BA extract was more effective in inhibiting FXa activity, and their fractions exhibited obvious difference in enzyme inhibitory activity. Furthermore, marker compounds a–h were predicted by PCA and OPLS-DA, and their chemical structures were identified. Among them, senkyunolide A, Z-ligustilide, ferulic acid and senkyunolide I (IC50 was determined as 0.77 mM) with potential THR inhibitory activity, as well as isochlorogenic acid A with FXa inhibitory activity were screened out. It was found that the four components could interact with the active site of THR, and the binding energy was lower than − 5 kcal/mol. Isochlorogenic acid A were bound to the active site of FXa, and the binding energy was − 9.39 kcal/mol. The IC50 was determined as 0.56 mM.Conclusion sTHR/FXa inhibitory components in different extracts of Chuanxiong were successfully characterized by the method of enzyme inhibition activity assays with ultra performance liquid chromatography-quadrupole time of flight mass spectrometry-based multivariate statistical analysis.

Puerarin, the most important flavonoid in Puerariae Lobatae Radix (PLR), has high biological activity but poor oral absorption. This study investigated how to enhance the absorption and pharmacokinetics of puerarin through modulating the intestinal barrier. Analyses of intestinal absorption, pharmacokinetics demonstrated enhanced puerarin absorption with daidzin, daidzein and ligustrazine (the main active ingredient in vinegar and Chuanxiong Rhizoma (CXR)). In particular, PLR combined with CXR enhanced the absorption and pharmacokinetics of puerarin through elevating puerarin solubility, regulating P-glycoprotein efflux and reducing claudin-5 expression. The combination of PLR and CXR reduced the diversity of gut microbiota and the ratio of Firmicutes to Bacteroidetes . In conclusion, combining PLR with CXR enhanced the absorption and pharmacokinetics of puerarin by modulating the intestinal barrier and influenced gut microbiota. The gut microbiota is likely to be a potential target for the combination of PLR and CXR.

In order to develop novel botanical insecticides, the joint action ofLigusticum chuanxiongoil (LCO) and lemongrass oil (LO) againstAphis citricolavan der Goot was determined systematically indoors and outdoors. The chemical profiles of LCO and LO as determined by gas chromatography–mass spectrometry (GC-MS) analysis revealed that the main compounds from LCO were (Z)-ligustilide (44.58%) and senkyunolide A (26.92%), and that of LO were geranial (42.16%) and neral (32.58%), respectively. The mixture of LCO and LO showed significant synergy againstA. citricola, with a common-toxicity coefficient (CTC) value of 221.46 at the optimal ratio of LCO to LO (4:1,w:w). Based on the results of solvents and emulsifiers screening,L. chuanxiongoil·Lemongrass oil 20% emulsifiable concentrate (20% LCO·LO EC) was developed, and its stability was confirmed with tests of cold and thermal storage. Field trials indicated that the insecticidal activity of the diluted 20% LCO·LO EC (1000 fold dilution) was comparable to conventional pesticide (20% imidacloprid EC) onA. citricolaseven days after application. Thus, the mixture of LCO and LO has the potential to be further developed as a botanical pesticide. Keywords:lemongrass oil;Ligusticum chuanxiongoil;Aphis citricolavan der Goot;botanical aphicides

Thrombosis is a key pathological event in cardiovascular diseases, and is also the most important targeting process for their clinical management. New drug development in thrombosis treatment is still in great demand. According to the traditional Chinese medicine (TCM) theory, thrombosis belongs to the syndrome of blood stasis. Salvia miltiorrhiza Bunge and Ligusticum striatum DC. are two common TCM herbs with long-term documented function in promoting blood circulation and inhibiting thrombosis, especially when used together. Guanxinning Tablet, a modern Chinese drug which contains extracts of the two herbs, also showed strong therapeutic effects in coronary heart disease. However, the pharmacological mechanism is still lacking for the compatibility of the two herbs. Here, through zebrafish-based in vivo fluorescence screening, we demonstrated the synergistic effects between S. miltiorrhiza Bunge and L. striatum DC. in regulating endogenous thrombosis. Moreover, combined with high-resolution mass spectrometry, the main compounds of the botanical drugs were analyzed and screened in our model system. Interestingly, cryptotanshinone and senkyunolide I, two representative compounds, respectively derived from the two herbs, also showed synergistic antithrombotic effects. Further analysis suggested that they may regulate thrombi formation at different levels via multiple signaling pathways, including oxidative stress, platelet activation and coagulation cascade. Taken together, our findings provided solid biological supports toward the drug compatibility theory of TCM, and suggested cryptotanshinone and senkyunolide I as promising drug candidates in thrombosis management.

Background and aims: Inducing the senescence of activated hepatic stellate cells (HSCs) has emerged as a promising therapeutic strategy for liver fibrosis, with potential connections to the Yes-associated protein (YAP)-controlled cGAS-STING pathway. However, the regulatory role of cytoskeletal dynamics on HSC senescence and its potential as a target for natural products have remained poorly understood. Methods: We employed preclinical in vivo and in vitro transcriptome analyses, experimental systems, Tmem173 -/- mice and liver-specific STING knockdown mice to demonstrate the anti-fibrotic effects and mechanism of ligustilide (LIG). Results: LIG selectively bound to monomeric globular actin (G-actin), thereby preventing its polymerization into polymeric filamentous actin (F-actin), which disturbed its interaction with intermediate filament component lamin A/C and initially destroyed the nuclear membrane. Moreover, the disruption of nuclear membrane caused YAP leakage from nuclear, which in turn suppressed lamin A/C and created a deleterious feedback loop that exacerbated nuclear membrane destabilization. Consequently, nuclear double stranded DNA (dsDNA) leakage caused by the above damage cascade ultimately triggered the activation of the cGAS-STING signaling pathway, promoting senescence-associated secretory phenotypes (SASPs) release and inducing HSC senescence. Moreover, the induction of HSC senescence and anti-fibrotic effects of LIG were completely abrogated in both whole-body STING knockout and liver-specific STING knockdown mice. Conclusions: By interacting with G-actin, LIG disrupted the cytoskeleton to compromise nuclear integrity with the involvement of YAP and further stimulated the cGAS-STING pathway, leading to the release of SASPs and HSC senescence, which ultimately mitigated liver fibrosis.

In this study, the sorption properties of Cr(VI), As(III), and Pb(II) on chitosan-modified magnetic biochar (CMBC) derived from residues of Ligusticum chuanxiong Hort. were investigated. CMBC was found to be a valuable material for removing three heavy metals from water simultaneously. Kinetic analysis suggested Cr(VI), As(III), and Pb(II) were chemisorbed onto CMBC, while isotherm data conformed well to Langmuir model, the maximum adsorption capacity of CMBC was found to be 65.74 mg/g for Cr(VI), 49.32 mg/g for As(III), and 69.45 mg/g for Pb(II). Experiments, characterization, and density functional theory (DFT) calculations were employed to explore the mechanisms. Furthermore, CMBC demonstrated excellent removal rates of over 95% for Cr(VI), 99% for As(III) and Pb(II) from contaminated water bodies. This work shows that CMBC holds significant potential for wastewater treatment of heavy metals and provides an effective solution for the utilization of Chinese herb residues in environmental remediation.

Xanthine oxidase (XO) has been well-recognized as a key enzyme in the pathogenesis of hyperuricemia. In this study, Chuanxiong Rhizome, a tonic food widely consumed in China, was found to inhibit XO activity. The potential XO inhibitory ingredients were screened by UPLC-ZenoTOF system and bioactivity assays. Coniferyl ferulate (CF), a main constitute of Chuanxiong Rhizome, was identified as a potent XO inhibitor with an IC 50 value (1.97 ± 0.11 μM) lower than that of allopurinol (2.49 ± 0.07 μM). Enzyme-kinetic assays showed that CF reversibly inhibited XO in an uncompetitive manner. And hydrogen bonds played crucial roles on the binding interaction according to the fluorescence titration and molecular docking. Moreover, the combination of CF with allopurinol exhibited a synergistic inhibitory effect on XO. Our results revealed for the first time the XO inhibitory effect of CF from Chuanxiong Rhizome, which shed light on its potential application for developing anti-hyperuricemia agents.

Human monoamine oxidase B (hMAO-B) has emerged as a pivotal therapeutic target for Parkinson's disease. Due to adverse effects and shortage of commercial drugs, there is a need for novel, highly selective, and reversible hMAO-B inhibitors with good blood-brain barrier permeability. In this study, a high-throughput at-line nanofractionation screening platform was established with extracts from Chuanxiong Rhizoma, which resulted in the discovery of 75 active compounds, including phenolic acids, volatile oils, and phthalides, two of which were highly selective novel natural phthalide hMAO-B inhibitors that were potent, selective, reversible and had good blood‒brain permeability. Molecular docking and molecular dynamics simulations elucidated the inhibition mechanism. Sedanolide (IC 50 = 103 nmol/L; SI = 645) and neocnidilide (IC 50 = 131 nmol/L; SI = 207) demonstrated their excellent potential as hMAO-B inhibitors. They offset the limitations of deactivating enzymes associated with irreversible hMAO-B inhibitors such as rasagiline. In SH-SY5Y cell assays, sedanolide (EC 50 = 0.962 μmol/L) and neocnidilide (EC 50 = 1.161 μmol/L) exhibited significant neuroprotective effects, comparable to the positive drugs rasagiline (EC 50 = 0.896 μmol/L) and safinamide (EC 50 = 1.079 μmol/L). These findings underscore the potential of sedanolide as a novel natural hMAO-B inhibitor that warrants further development as a promising drug candidate.

Ultrasonic-assisted extraction technology was employed to prepare Ligusticum chuanxiong Hort polysaccharide. Single factor test and orthogonal experimental design were used to optimize the extraction conditions. The results showed that the optimal extraction conditions consisted of ultrasonic temperature of 80 °C, ultrasonic time of 40 min and water to raw material ratio of 30 mL/g. Three novel polysaccharides fractions, LCX0, LCX1 and LCX2, were isolated and purified from the crude polysaccharides using DEAE-52 cellulose and Sephadex G-100 column chromatography. The molecular weight and monosaccharide composition of three LCX polysaccharides fractions were analyzed with gel permeation chromatography (GPC) and HPLC analysis, respectively. Furthermore, the antioxidant and in vitro anticancer activities of the polysaccharides were investigated. Compared with LCX0, LCX2 and LCX1 showed relative higher antioxidant activity and inhibitory activity to the growth of HepG2, SMMC7721, A549 and HCT-116 cells. It is suggested that the novel polysaccharides from rhizome of L. chuanxiong could be promising bioactive macromolecules for biomedical use.
